Determination of Protein Content in Pea by Near Infrared Spectroscopy

  • Near infrared reflectance spectroscopy (NIRS) was used as a rapid and non-destructive method to determine the protein content in intact and ground seeds of pea (Pisum sativum L.) germplasms grown in Korea. A total of 115 samples were scanned in the reflectance mode of a scanning monochromator at intact seed and flour condition, and the reference values for the protein content was measured by auto-Kjeldahl system. In the developed ground and intact NIRS equations for analysis of protein, the most accurate equation were obtained at 2, 8, 6, 1 math treatment conditions with standard normal variate and detrend scatter correction method and entire spectrum (400-2,500 nm) by using modified partial least squares regression (n=78). External validation (n=34) of these NIRS equations showed significant correlation between reference values and NIRS estimated values based on the standard error of prediction (SEP), $R^2$, and the ratio of standard deviation of reference data to SEP. Therefore, these ground and intact NIRS equations can be applicable and reliable for determination of protein content in pea seeds, and non-destructive NIRS method could be used as a mass analysis technique for selection of high protein pea in breeding program and for quality control in food industry.

Skin depth profiling by using fiber optic probes in the near infrared

  • Recently we showed the prototype portable device for the determination of human skin moisture by using near infrared spectroscopy. In order to optimize the acquiring condition of NIR spectrum of skin and control the target information of water depending the site such as epidermis and dermis, skin depth profiling was investigated changing the distance between illuminations and receiving of radiation in the terminal of fiber probe. The colleted light information could be controlled by changing the distance of the fiber optic probes. It was confirmed that the longer distance we used, the deeper site from the skin surface we could get information from in this study. Four kinds of probes with distances such as 0.03 mm, 0.1 mm, 0.5 mm, and 1.0 mm were used. In addition, the gap size from 0.3 mm to 3.0 mm was studied to control the intensity of water absorbance effectively and to avoid saturation of water absorption. We also investigated the reference materials depending the reflectance ratio for water absorption not to be saturated because of the strong absorptivity of water. Furthermore, spectroscopic information regarding free water and bound water around 1850 nm was investigated by using the different distance of fiber optic probes. This study would be great help to control the spectroscopic information of water to be measured depending the site where water exists.

BRACKETT LINE-BASED MBH ESTIMATORS AND HOT DUST TEMPERATURES OF TYPE 1 AGNs FROM AKARI SPECTROSCOPIC DATA

  • We provide results of near-infrared (NIR) spectroscopic observations of 83 nearby (0.002< z <0.48) and bright (K <14 mag) type 1 active galactic nuclei (AGNs). For the observations, we used the Infrared Camera (IRC) on AKARI allowing us to obtain the spectrum in the rarely studied spectral range of $2.5-5.0{\mu}m$. The $2.5-5.0{\mu}m$ spectral region suffers less dust extinction than ultra violet (UV) or optical wavelength ranges, and contains several important emission lines such as $Br{\beta}$ ($2.63{\mu}m$), $Br{\alpha}$ ($4.05{\mu}m$), and polycyclic aromatic hydrocarbon (PAH; $3.3{\mu}m$). The sample is selected from the bright quasar surveys of Palomar Green and SNUQSO, and AGNs with black hole (BH) masses estimated from reverberation mapping method. We measure the Brackett line properties for 11 AGNs, which enable us to derive BH mass estimators and investigate circum-nuclear environments. Moreover, we perform spectral modeling to fit the hot and warm dust components by adding photometric data from SDSS, 2MASS, WISE, and ISO to the AKARI spectra, and estimate hot and warm dust temperatures of ~1100K and ~220 K, respectively.

High-resolution Near-infrared Spectroscopy of IRAS 16316-1540: Evidence of Accretion Burst

  • The high-resolution near-infrared (NIR) spectroscopy can reveal the evidence of the accretion burst (e.g., the broadened absorption features produced by the Keplerian disk motion) although the moment of the outburst was not caught. The embedded protostar IRAS 16316-1540 observed with the Immersion Grating Infrared Spectrograph (IGRINS, $R={\Delta}{\lambda}/{\lambda}{\sim}45000$) shows the broad absorption features in atomic and CO transitions, as seen in FU Orionis objects (FUors), indicative of an outburst event. We examine whether the spectra of IRAS 16316-1540 arise from the rotating inner hot gaseous disk. Using the IGRINS spectral library, we show that the line profiles of IRAS 16316-1540 are more consistent with an M1.5 V template spectrum convolved with a disk rotation profile than the protostellar photosphere absorption features with a high stellar rotation velocity. We also note that the absorption features deviated from the expected line profile of the accretion disk model can be explained by a turbulence motion generated in the disk atmosphere. From previous observations that show the complex environment and the misaligned outflow axes in IRAS 16316-1540, we suggest that an impact of infalling clumpy envelope material against the disk induces the disk precession, causing the accretion burst from the inner disk to the protostar.

  • The international coffee trade is conducted almost exclusively with green coffee. The main coffee producing countries include Brazil, Columbia, Indonesia, Mexico and the Ivory Coast. About 99 % of the coffee grown throughout the world belong to two coffee plant varieties that are commonly known as Arabica and Robusta. The quality of green coffee can be assessed according to several ISO standards (1,2,3,4,5). However, no official international standards for the authenticity of green coffee have been issued. It is important to know the country of origin of the coffee for the purposes of fair international trade. The geographic origin of the coffee is often stated on the label of coffee products such as speciality roasted and soluble coffees. Near Infrared Spectroscopy (NIR) is an accepted technique for quantitative analysis of various parameters in routine QC analysis of food products. It would appear to be a promising candidate as a tool for identification of green coffee origin and numerous feasibility studies have appeared in the literature on its use for soluble, roasted and green coffee variety identification as well as identification of arabica or robusta coffees. NIR spectrophotometers when configured in the reflectance mode are able to perform a complete profile of the NIR spectrum on whole beans. The data can then be interpreted by discriminant chemometrics data analysis. This is the approach used in the present study.

  • 답토양부식물(畓土壤腐植物)에 대(對)한 일련(一連)의 토양화학적연구(土壞化學的硏究)에서 본실험(本實驗)은 수개(數個) 고위(高位)및 저위답토양(低位畓土壞)에서 부식물(腐植物)을 추출(抽出)하고 hymatomelan 산(酸)과 humin 산(酸)으로 분획(分劃)하여 가시(可視), 자외(紫外) 및 적외부(赤外部)에시 흡광(吸光) spectrum을 측정(測定)하여 비교검토(比較檢討)하였다. 토양비옥도(土壞肥沃度)의 차이(差異)에 따라 부식분획물(腐植分劃物)의 흡광(吸光)은 어느 광파역(光波域)에서도 별(別)다른 상이성(相異性)이 존재(存在)하지 않으며 자외광흡수(紫外光吸收) spectrum이 퇴비(堆肥)와 다른 유형(類型)의 토양(土壞)에서 분리(分離)한 부식산(腐植酸)의 spectrum과는 다르게 hymatomelan 산(酸)이 나 humin 산(酸) 모두 $235\;m{\mu}$에서 최고(最高), $395\;m{\mu}$에서 최저(最低)의 만곡접(灣曲點)을 나타내었다. 적외광흡수(赤外光吸收) spectrum은 양(兩) fraction간(間)에 상이점(相異點)을 보여 파수(波數) $1720\;cm^{-1}$에 특징적(特徵的)인 Peak가 존재하(存在)하며 이보다 장파역(長波域)($7.5{\sim}9.5{\mu}$)에서도 상이(相異)한 양상(樣狀)이었다. 또한 이 spectrum 들을 다른 연구자(硏究者)들에 의(依)한것과 대조(對照)하여 상이성(相異成)을 고찰(考察)하였다.

Fourier transform infrared spectroscopy를 이용한 SiNx박막의 수소농도 연구 (Study of the hydrogen concentration of SiNx film by Fourier transform infrared spectroscopy)

  • 실리콘 웨이퍼 위에 Plasma Enhanced Chemical Vapor Deposition (PECVD) 방법으로 증착 된 SiNx 박막의 수소 함량을 측정하였다. 제작된 SiNx 박막은 Fourier Transform Infrared Spectroscopy (FT-IR) 사용하여 박막의 수소함량과 결합상태를 확인하였으며, Atomic Force Microscopy (AFM) 측정을 통하여 박막의 표면 거칠기를 비교, 시료의 조성비 평가를 위하여 Rutherford Backscattering Spectrometry (RBS)을 사용하였다. 또한 SiNx박막의 조성확인을 위하여 Photoluminescence(PL)를 이용하여 FT-IR spectrum의 결과와 비교 해석하였다. FT-IR에서 NH의 수소함량(at%)이 0.92 %에서 0.64 %로 낮아질 수록 AFM을 이용한 표면 거칠기는 12.8 $\AA$에서 10.8 $\AA$로 낮아지고, Si양이 상대적으로 많아지는 것을 PL에서 확인하였으며, RBS에서도 시뮬레이션을 통해 비슷한 결과를 얻을 수 있었는데, 이는 FT-IR을 사용함으로써 SiNx 박막의 수소 함량의 측정이 가능하다는 것을 보여주므로 단위공정에서 신속하게 SiNx 박막 분석이 가능함을 알 수 있었다.

근적외선 반사분광분석법에 의한 정육 및 그 구성성분의 반사 spectra에 관한 연구 (Studies on the Near Infrared Diffuse Reflection Spectroscopy for Meat Components)

  • 이 연구의 목적은 식품성분의 분석을 위한 신속간편법으로 근적외선 반사 분광분석을 이용함에 있어 흡광도에 영향을 미치는 시료의 물리적, 화학적 요인을 규명하고자 한다. 온도를 달리한 정육시료를 근적외선 파장 범위인 $1100{\sim}2500\;nm$에서 흡광도를 측정한 결과 온도가 상승함에 따라 흡광도가 대체로 증가하였다. 시료의 세절시간이 길어질수록 흡광도는 감소하였다. 정육, 육단백질, 지방 및 수분의 근적외선 스펙트럼에서 나타난 흡수띠는 분자내의 C-H, N-H, O-H 및 C=O 결합의 overtone과 combination 진동에 의한 것으로 확인되었다. 정육 구성성분 중 육단백질의 흡광도가 가장 낮았으며 지방은 1700 nm와 2300 nm 부근에서 특징적인 ${\cdot}CH_2{\cdot}$ 흡수띠를 보였고 1450 nm와 1930 nm에서 물분자에 의한 강한 흡수띠가 나타났다. 결과적으로 정육의 근적외선 스펙트럼의 흡광도는 함유된 수분과 지방에 의한 영향을 많이 받는 것으로 사료된다.

근적외선 분광법 기반 비침습식 혈당 검출 센서 시스템 (Non-invasive Blood Glucose Detection Sensor System Based on Near-Infrared Spectroscopy)

  • 비침습식 혈당 검출 기술 중 광학 기법은 생물학적 매체를 통과할 때 빛의 반사와 흡수 및 산란 특성을 이용하는 방법으로 통증이나 측정의 불편함을 감소시키고 감염 위험이 없어 혈당 검출 연구의 주요 흐름이 되고 있다. 이 중 근적외선 분광법은 혈당 분자와 유사한 흡수 기능을 공유하는 단백질과 산의 간섭들로 감지된 신호 분석 시 복잡성이 증가하는 단점이 있다. 본 연구에서는 근적외선의 피부 흡수로 발생할 수 있는 혈당검출 기능저하를 완화시키기 위해 다중 근적외선 대역의 비침습식 센서시스템을 설계하고 제작하였다. 제작한 시스템의 검증을 위해 혈액 조사를 실시하였으며, 혈액 내의 혈당 반응 정도를 스펙트럼 데이터로 수집하고, 데이터와 혈당과의 상관관계 관점에서 정량적으로 본 연구의 성과를 검증하였다.

Classification of Red Wines by Near Infrared Transflectance Spectroscopy

  • During the recent years, wine analysis has played an increasing role due the health benefits of phenolic ingredients in red wine [1]. On the other hand there is the need to be able to distinguish between different wine varieties. Consumers want to know if a wine is an adulterated one or if it is based on the pure grape. Producers need to certificate their wines in order to ensure compliance with legal regulations. Up to now, the attempts to investigate the origin of wines were based on high-performance liquid chromatography (HPLC), gas chromatography (GC) and pyrolysis mass spectrometry (PMS) [l,2,3]. These methods need sample pretreatment, long analysis times and therefore lack of high sample throughput. In contradiction to these techniques using near infrared spectroscopy (NIRS), no sample pretreatment is necessary and the analysis time for one sample is only about 10 seconds. Hence, a near infrared spectroscopic method is presented that allows a fast classification of wine varieties in bottled red wines. For this, the spectra of 50 bottles of Cabernet Sauvignon, Lagrein and Sangiovese (Chianti) were recorded without any sample pretreatment over a wavelength range from 1000 to 2500 nm with a resolution of 12 cm$\^$-1/. 10 scans were used for an average spectrum. In order to yield best reproducibility, wines were thermostated at 23$^{\circ}C$ and a optical layer thickness of 3 mm was used. All recorded spectra were partitioned into a calibration and validation set (70% and 30%). Finally, a 3d scatter plot of the different investigated varieties allowed to distinguish between Cabernet Sauvignon, Lagrein and Sangiovese (Chianti). Considering the short analysis times this NRS-method will be an interesting tool for the quality control of wine verification and also for experienced sommeliers.
