In Vivo 레벨에서 1-아닐리노-8-나프탈렌 설포네이트(ANS)의 간내 이행 및 담즙배설 과정의 속도론적 해석 (Kinetic Analysis of the Hepatic Uptake and Biliary Excretion of 1-Anilino-8-Naphthalene Sulfonate (ANS) in Vivo)

  • The purpose of the present study was to investigate the hepatic uptake and biliary excretion of l-anilino-8-naphthalene sulfonate (ANS) in vivo. The plasma concentration and liver concentration of ANS were determined after its i.v. bolus administration at a dose of $30\;{\mu}mol/kg$ in rats. The hepatic uptake clearance $(CL_{uptake})$ of ANS was 0.1 ml/min/g liver. On the basis of the unbound concentration of ANS, the permeability-surface area product $(PS_{influx})$ was calculated to be l0.4 ml/min/g liver, being comparable of in vitro data. On the other hand, we determined the plasma concentration, liver concentration and biliary excretion rate of ANS at steady-state after its i. v. infusion $(0.2-1.6\;{\mu}mol/min/kg)$ in rats. The excretion clearance $(CL_{excretion})$ of ANS showed Michaelis-Menten kinetics with increasing the infusion rate. The permeability-surface area product $(PS_{excretion})$ based on the unbound concentration in the liver was calculated to be 0.0165 ml/min/g liver, which is negligible compared with the intrinsic clearance $(CL_{int}=3.3\;ml/min/g\;liver)$ by rat liver microsomes. The sequestration process of ANS, therefore, was considered to be mainly due to the metabolic process in the liver $(PS_{seq}{\risingdotseq}CL_{int})$. Furthermore, $PS_{efflux}$ value calculated from $PS_{influx}$ and $PS_{seq}$ was 4.4 ml/min/g liver, which was comparable of in vitro data. In conclusion, in vivo parameters such as $PS_{influx}$, $PS_{efflux}$ and $PS_{seq}$ in the present study showed good in vivo-in vitro relationship. Thus, the kinetic analysis method proposed in the present study would be useful to analyze the hepatic transport of drugs in vivo.

  • 표고버섯의 미토콘드리아는 설탕밀도 선형기울기 원심분리법으로 정제하여, 광감응성 mitochondrial ATPase의 유기물 효과, 광증감제 효과 및 파장 변화에 따른 $K^+$ 이온의 유입 효과를 실험하였다. 1. 이 효소는 10m mol dithiothreitol 및 0.1m mol quinacrine에 의하여 각각 139% 및 128%의 활성도를 증가시켰다. 2. $100\;{\mu}g$의 oligomycin과 1m mol phlorizin은 이 효소의 활성을 각각 48% 및 45% 억제시켰다. 3. 광증감제인 0.1m mol phenazine methosulfate는 이 효소의 활성도를 36% 촉진시켰다. 4. $K^+$ 이온 유입 효과의 최적 파장은 690nm였고, 이때의 최적 pH 및 최적 온도는 각각 7.2 및 $55^{\circ}C$였다.

  • 1. 최적광조사 조건, 470 nm에서 15초 동안 빛을 조사한 미토콘드리아성 ATP synthase는 1mmole DCPI에 의하여 그 활성이 85% 증가되었다. 2. 1mmole DNP, $10\;{\mu}mole$ HHQNO 및 $100\;{\mu}g/ml$의 oligomycin은 이 효소의 활성을 각각 61, 41% 및 12% 억제시켰다. 3. 최적 조건의 빛에 의한 $Fe^{3+}$$Fe^{2+}$ 이온의 유입효과에서, 5mmole $Fe^{3+}$ 및 0.5mmole $Fe^{2+}$ 이온에 의하여 이 효소 활성이 각각 14% 및 12% 증가되었다. 4. 광증감제인 phenazine methosulfate의 존재하에서 470nm의 빛을 조사한 후의 효소의 활성도가 크게 증가되는 것으로 보아, 미토콘드리아성 ATP synthase내에 미지의 흡광물질이 존재함을 추정할 수 있었다.

Korean Red Ginseng extract and ginsenoside Rg3 have anti-pruritic effects on chloroquine-induced itch by inhibition of MrgprA3/TRPA1-mediated pathway

  • Background: It was previously found that Korean Red Ginseng water extract (KRGE) inhibits the histamine-induced itch signaling pathway in peripheral sensory neurons. Thus, in the present study, we investigated whether KRGE inhibited another distinctive itch pathway induced by chloroquine (CQ); a representative histamine-independent pathway mediated by MrgprA3 and TRPA1. Methods: Intracellular calcium changes were measured by the calcium imaging technique in the HEK293T cells transfected with both MrgprA3 and TRPA1 ("MrgprA3/TRPA1"), and in primary culture of mouse dorsal root ganglia (DRGs). Mouse scratching behavior tests were performed to verify proposed antipruritic effects of KRGE and ginsenoside Rg3. Results: CQ-induced $Ca^{2+}$ influx was strongly inhibited by KRGE ($10{\mu}g/mL$) in MrgprA3/TRPA1, and notably ginsenoside Rg3 dose-dependently suppressed CQ-induced $Ca^{2+}$ influx in MrgprA3/TRPA1. Moreover, both KRGE ($10{\mu}g/mL$) and Rg3 ($100{\mu}M$) suppressed CQ-induced $Ca^{2+}$ influx in primary culture of mouse DRGs, indicating that the inhibitory effect of KRGE was functional in peripheral sensory neurons. In vivo tests revealed that not only KRGE (100 mg) suppressed CQ-induced scratching in mice [bouts of scratching: $274.0{\pm}51.47$ (control) vs. $104.7{\pm}17.39$ (KRGE)], but also Rg3 (1.5 mg) oral administration significantly reduced CQ-induced scratching as well [bouts of scratching: $216.8{\pm}33.73$ (control) vs.$115.7{\pm}20.94$ (Rg3)]. Conclusion: The present study verified that KRGE and Rg3 have a strong antipruritic effect against CQ-induced itch. Thus, KRGE is as a promising antipruritic agent that blocks both histamine-dependent and -independent itch at peripheral sensory neuronal levels.

멕시코 중부의 토양 침식과 고환경 변화 (Soil Erosion and Environmental Change in Central Mexico)

  • 멕시코 중부의 환경사를 파악하고자 할 때, 호수퇴적물을 이용하는 고환경 분석은 필수적이다. 이 지역의 고대문명과 스페인인의 식민사 등에 대한 기록이 거의 남아있지 않기 때문이다. 본 연구에서는 호수로의 토사 유입량을 분석하여 연구 지역에서 발생했던 토양 침식과 환경변화를 시기별로 밝혀 냈다. 토사유입량은 연대분석 결과와 작열감량 분석 결과를 기초로 산출하였다. 다양한 연대측정(Lead210, 바브층, 화산재층, 지진연도)을 수행하여 정확한 토사유입량를 계산할 수 있었으며 지역 환경에 중요한 영향을 미쳤던 사건들의 상세한 연대를 파악할 수 있었다. 연구 결과에 따르면, 유역의 환경을 훼손시킨 가장 큰 요인은 점령 전후의 옥수수 농경이었다 한편, 스페인인의 목축활동이 미친 영향은 최소한 호수 유역 내에서는 그리 크지 않았다. 본 연구에서는 여러 종류의 연대분석을 다각적으로 시도하여 보다 정확한 고환경 정보를 획득할 수 있었다.

Lysophosphatidylcholine Increases $Ca^{2+}$ Current via Activation of Protein Kinase C in Rabbit Portal Vein Smooth Muscle Cells

  • Lysophosphatidylcholine (LPC), a metabolite of membrane phospholipids by phospholipase $A_2$, has been considered responsible for the development of abnormal vascular reactivity during atherosclerosis. $Ca^{2+}$ influx was shown to be augmented in atherosclerotic artery which might be responsible for abnormal vascular reactivity. However, the mechanism underlying $Ca^{2+}$ influx change in atherosclerotic artery remains undetermined. The purpose of the present study was to examine the effects of LPC on L-type $Ca^{2+}$ current $(I_{Ca(L)})$ activity and to elucidate the mechanism of LPC-induced change of $I_{Ca(L)}$ in rabbit portal vein smooth muscle cells using whole cell patch clamp. Extracellular application of LPC increased $I_{Ca(L)}$ through whole test potentials, and this effect was readily reversed by washout. Steady state voltage dependency of activation or inactivation properties of $I_{Ca(L)}$ was not significantly changed by LPC. Staurosporine (100 nM) or chelerythrine $(3{\mu}M)$, which is a potent inhibitor of PKC, significantly decreased basal $I_{Ca(L)}$, and LPC-induced increase of $I_{Ca(L)}$ was significantly suppressed in the presence of PKC inhibitors. On the other hand, application of PMA, an activator of PKC, increased basal $I_{Ca(L)}$ significantly, and LPC-induced enhancement of $I_{Ca(L)}$ was abolished by pretreatment of the cells with PMA. These findings suggest that LPC increased $I_{Ca(L)}$ in vascular smooth muscle cells by a pathway that involves PKC, and that LPC-induced increase of $I_{Ca(L)}$ might be, at least in part, responsible for increased $Ca^{2+}$ influx in atherosclerotic artery.

원료 생약 추출물의 최적화를 위한 한국 5개 지역에서 채집된 대계 추출물들의 항우울 효과 비교 (Comparison of the Antidepressant Activity of Cirsii herbal Extracts Collected from 5 Different Regions of Korea Determining the Optimal Herbal Preparation)

  • Previously, we identified that Cirsium japonicum (CJ) extract posses antidepressant properties. In order to look for the optimal hebal preparation, CJ extracts collected from Koheung, Yesan, Cheju, Pyungchang and Pocheon were compared. The psychopharmacological activities of each extract were investigated using the open field test, the rota-rod test and the forced swimming test. Effect of each extract on $Cl^-$ influx in neuroblastoma cells were also identified. All of the extracts were orally administered at dosages of 50, 100, 200, 400 mg/kg of body weight. Supplementation of CJ did not significantly alter psychomotor behaviors such as movement, rearing frequency and total turn angle on the open field. Supplementation of CJ's extract from Pyungchang (100 mg/kg, p.o.), Cheju (200 and 400 mg/kg p.o.) and Yesan (100 and 200 mg/kg, p.o.) significantly decreased immobile duration and increased mobile duration in the forced swimming test. All of extracts have no influence on NE uptake in HEK-293 cells, but increased $Cl^-$ influx in neuroblastoma cell. The extract from Yesan demonstrated the most potent effect on $Cl^-$ influx. These results indicate that CJ's extracts from Pyungchang, Cheju and Yesan may alleviate depression, and the extract from Yesan may be the most effective preparation.

Ginsenoside Rd protects cerebral endothelial cells from oxygen-glucose deprivation/reoxygenation induced pyroptosis via inhibiting SLC5A1 mediated sodium influx

  • Background: Ginsenoside Rd is a natural compound with promising neuroprotective effects. However, the underlying mechanisms are still not well-understood. In this study, we explored whether ginsenoside Rd exerts protective effects on cerebral endothelial cells after oxygen-glucose deprivation/reoxygenation (OGD/R) treatment and its potential docking proteins related to the underlying regulations. Method: Commercially available primary human brain microvessel endothelial cells (HBMECs) were used for in vitro OGD/R studies. Cell viability, pyroptosis-associated protein expression and tight junction protein degradation were evaluated. Molecular docking proteins were predicted. Subsequent surface plasmon resonance (SPR) technology was utilized for validation. Flow cytometry was performed to quantify caspase-1 positive and PI positive (caspase-1+/PI+) pyroptotic cells. Results: Ginsenoside Rd treatment attenuated OGD/R-induced damage of blood-brain barrier (BBB) integrity in vitro. It suppressed NLRP3 inflammasome activation (increased expression of NLRP3, cleaved caspase-1, IL-1β and GSDMD-N terminal (NT)) and subsequent cellular pyroptosis (caspase-1+/PI + cells). Ginsenoside Rd interacted with SLC5A1 with a high affinity and reduced OGD/R-induced sodium influx and potassium efflux in HBMECs. Inhibiting SLC5A1 using phlorizin suppressed OGD/R-activated NLRP3 inflammasome and pyroptosis in HBMECs. Conclusion: Ginsenoside Rd protects HBMECs from OGD/R-induced injury partially via binding to SLC5A1, reducing OGD/R-induced sodium influx and potassium efflux, thereby alleviating NLRP3 inflammasome activation and pyroptosis.

Enhancing effect of Panax ginseng on Zip4-mediated zinc influx into the cytosol

  • Background: Zinc homeostasis is essential for human health and is regulated by several zinc transporters including ZIP and ZnT. ZIP4 is expressed in the small intestine and is important for zinc absorption from the diet. We investigated in the present study the effects of Panax ginseng (P. ginseng) extract on modulating Zip4 expression and cellular zinc levels in mouse Hepa cells. Methods: Hepa cells were transfected with a luciferase reporter plasmid that contains metal-responsive elements, incubated with P. ginseng extract, and luciferase activity was measured. Using 65ZnCl2, zinc uptake in P. ginseng-treated cells was measured. The expression of Zip4 mRNA and protein in Hepa cells was also investigated. Finally, using a luciferase reporter assay system, the effects of several ginsenosides were monitored. Results: The luciferase activity in cells incubated with P. ginseng extract was significantly higher than that of control cells cultured in normal medium. Hepa cells treated with P. ginseng extract exhibited higher zinc uptake. P. ginseng extract induced Zip4 mRNA expression, which resulted in an enhancement of Zip4 protein expression. Furthermore, some ginsenosides, such as ginsenoside Rc and Re, enhanced luciferase activity driven by intracellular zinc levels. Conclusion: P. ginseng extract induced Zip4 expression at the mRNA and protein level and resulted in higher zinc uptake in Hepa cells. Some ginsenosides facilitated zinc influx. On the basis of these results, we suggest a novel effect of P. ginseng on Zip4-mediated zinc influx, which may provide a new strategy for preventing zinc deficiency.

지역 간 인구이동, 경지면적, 외국인 근로자의 관계 분석 (Interrelationship Between Regional Population Migration, Crop Area, and Foreign Workers)

  • 지역 인구와 경지면적 간의 상호작용을 이해하는 것은 지역 경제 활성화와 농업의 지속가능성 향상을 위해 중요하다. 기존의 연구들은 지역의 인구이동이나 경지면적 변화 각각의 주제에 중점을 두었으며, 이들의 상호작용에 관한 연구는 미흡했다. 또한, 새로운 노동 공급원인 외국인 근로자의 증가를 대상으로 한 지역 단위의 정량적 연구도 부족했다. 이에 본 연구에서는 지역의 인구 변화, 경지면적, 그리고 외국인 근로자 수 변화의 상호작용을 패널 자기 상관 모형(Panel Vector Auto Regression Model)을 활용하여 분석하였다. 분석 결과, 지역의 인구유입률과 경지면적은 서로에게 부정적인 영향을 발생시키지만, 농업부문의 외국인 근로자 증가는 경지면적을 증가시키는 것으로 나타났다. 또한, 밭 면적의 증가는 외국인 근로자를 증가시키는 것으로 나타났다. 이러한 결과는 지역의 인구감소와 경지면적 감소 현상이 상호영향을 미치고 있으며 외국인 근로자의 유입이 농촌지역의 구조적 문제해결에 긍정적인 영향을 미칠 가능성이 있음을 시사한다.
