Studies on Coliform Mastitis: II. Pathological Findings on The Effects of Dexamethasone, Iron and Transferrin in Mastitis of Lactating Rabbits (Coliform 유방염(乳房炎)에 관한 연구(硏究) : II. Dexamethasone, Iron 및 Transferrin이 유방염(乳房炎)에 미치는 영향(影響)에 관한 병리학적소견(病理學的所見))

  • In this study lactating female rabbits and strains of coliforms previously isolated from the cases of acute and chronic mastitis in dairy cattle were employed. The pathological changes were observed on the mastitis experimentally induced with the coliform strains and the mamary glands after infusions of E. coli suspension together with dexamethasone, dextran iron or transferrin were grossly and microscopically observed. From the results reported, the following points are concluded. In the bacterial suspension-infused groups by E. coli, K. pneumoniae and Ent. aerogenes, respectively, the affected quarters of udder showed grossly swelling, hyperemia, hemorrhage, focal necrosis and firmness. The microscopic findings of early stage of the mastitis were appearance of large numbers of heterophils in the glandular lumina and ducts accompanied by degeneration, necrosis and desquamation of epithelial cells and also infiltration of heterophils, hemorrhage and edema in the interstitial tissue and destruction of alveoli. Later, proliferation of fibroblasts, plasma cells, lymphocytes, eosinophils and macrophages appeared in the glandular tissue and with these cells necrotic foci of glandular tissue were surrounded by highly proliferated connective tissue. In addition, granulomatous inflammatory changes could be observed in the glandular tissue from the 7th day after infusion. The difference of the inflammatory response among the groups did not recognized. In the groups infused with dexamethasone and E. coli suspension the inflammatory response was slighter at the inflammatory change with alveolar destruction and hemorrhage was more rapid and severer than E. coli alone. Also in the groups infused with dextran iron and E. coli suspension the inflammatory change was more rapid and severer than E. coli alone and the histological changes were not recognized in the groups infused with dextran iron alone. Reaction of the iron staining was diffusely strong positive within the glandular alveolar lumina in the groups of dextran iron alone, hut was slightly positive toward epithelial cells in the groups of dextran iron and E. coli infusion. In the group infused with transferrin and E. coli suspension, the inflammatory response was tighter, but the peroxidase activity of the heterophils in the glandular lumina was more or less stronger than E. coli alone.

Coronary Arterial Lesions of Kawasaki Disease Observed in a Mouse Model of Sepsis: A Pilot Study and a Review of the Literature (패혈증 마우스 모델에서 가와사키병의 관상동맥 병변 관찰: 예비연구와 문헌고찰)

  • Purpose: Coronary arterial lesions (CALs) were reported to have developed in children with systemic inflammatory diseases, as well as those with Kawasaki disease (KD). The purpose of this study was to confirm that the CAL development in children with KD occurs in a mouse model of sepsis presenting typical systemic inflammatory response syndrome (SIRS). Methods: To induce the sepsis mouse model with SIRS, 6-week-old C57BL/6 mice were intraperitoneally injected with endotoxin. We compared histological findings of the major organs between the control and the sepsis groups and examined CAL in the heart of the septic mice. Results: Infiltrating inflammatory cells were relatively increased in the heart, liver, and kidneys of the sepsis group, compared with those of the control group. We confirmed lymphocytic infiltration in the myocardium (myocarditis) and the pericardial soft tissue of the heart. Furthermore, coronary artery of the septic mouse was identified, but CAL was not observed. Conclusions: In this study, we failed to confirm the existence of CAL in a mouse model of sepsis. However, it is well-known that CALs are seen in many kinds of diseases that cause SIRS. Our findings suggest further investigation into the clinical significance of CAL in various systemic inflammatory diseases, including KD.

The Anti-inflammatory Effect of Nypa fruticans Wurmb. Fruit on Lipopolysaccharide-induced Inflammatory response on RAW 264.7 cells (LPS로 유도한 염증반응에서 해죽순의 항염증 효과)

  • Objective : Nypa fruticans Wurmb. Fruit (NF) has been used as a conventional medicine to treat inflammatory peridontal diseases in Myanmar and Eastern Asia. However, the anti-inflammatory effect of NF aqueous extract on lipopolysaccharide (LPS)-induced inflammatory responses was not well-investigated. Therefore, this study was aimed to investigate the anti-inflammatory effect of NF on LPS-induced inflammatory responses on RAW 264.7 cells.Methods : To induce inflammation on the macrophage cell line, RAW 264.7 cells were treated with 500 ng/mL of LPS. Water extracts of NF was treated 1 h prior to treatment of LPS. Cell viability was measured by MTT assay. Production of nitrite was measured with Griess assay and pro-inflammatory cytokines such as interleukine (IL)-1β and IL-6, and tumor necrosis factor (TNF)-α was measured by enzyme-linked immunosorbent assay (ELISA) and real-time polymerase chain reaction (PCR). In addition, we examined the inhibitory mechanisms of NF by western blot and immunocytochemistry.Result : Water Extract from NF itself did not have any cytotoxic effect at the concentration of 200 ㎍/ml in RAW 264.7 cells. Treatment of NF inhibited the production of nitrite, and pro-inflammatory cytokines inlcuding IL-1β, IL-6 and TNF-α in a dose dependant. In addition, NF treatment inhibited the LPS-induced activation and translocation of nuclear factor (NF)-κB.Conclusion : In summary, our result suggest that treatment of NF could reduce the LPS-induced inflammatory responses via deactivation of NF-κB. This study could suggest that NF could be a beneficial drug or agent to prevent inflammation.

Changes of Electrolytes, Hematological Indices, and Cytokines following Dietary Magnesium Deficiency in Rats

  • Magnesium (Mg) plays an essential role in physiological and metabolic reactions. Recently, there has been an increased interest in the role of Mg deficiency, particularly the relationship between serum Mg value and inflammatory response. This study was designed to determine the relationship between serum Mg deficiency with inflammatory response, electrolytes and hematological alteration over long-term periods. Sixteen male Sprague-Dawley rats were divided into two groups: control (n=8), and Mg deficiency group (MgD group, n=8). Chow and normal water (tap water) were regularly provided to the control group and Mg-depleted chow and third distilled water were regularly provided for 60 days to the MgD group. Body weights, Serum Mg, $K^+$, inorganic phosphorus (IP) and total iron binding capacity (TIBC) levels in the MgD group were lower than those of the control group (P<0.05). Granulocyte fraction and MCV, RDW and PDW levels were higher, whereas lymphocyte fraction, erythrocyte, hemoglobin and MCHC levels were lower in the MgD group than in the control group (P<0.05). MCP-1 and TNF-${\alpha}$ levels in the MgD group were greater than those of the control group (P<0.05). In conclusion, the results of the present study suggest that Mg deficiency over a long-term period had not altered total leukocyte concentration in the blood, but had detrimental effects, including disturbances of electrolytes balance, disturbance of iron indices, potential anemia and elevation of pro-inflammatory cytokine. However, further studies should be performed to determine the relationship between serum Mg deficiency and major organ damage or alteration.

Apelin-APJ axis inhibits TNF-alpha-mediated expression of genes involved in the inflammatory response in periodontal ligament cells

  • Periodontitis is an inflammatory disease of the supportive tissues surrounding the teeth, and is characterized by irreversible destruction of the gingiva, periodontal ligament (PDL), and alveolar bone, which results in the loss of teeth. In the present study, we elucidated the correlation between periodontitis and apelin (APLN), an adipokine and a regulatory peptide, respectively, which are involved in inflammation and bone remodeling. The expression of APLN is negatively correlated with periodontitis progression in gingival tissue. In addition, treatment with TNF-α downregulated the expression of APLN in PDL cells and gingival fibroblasts, indicating the protective role played by APLN against periodontitis progression. The overexpression of APLN or treatment with exogenous APLN suppressed the TNF-α-mediated catabolic gene expression of MMP1, IL6, and PTGS2 in PDL cells. Moreover, the inhibition of the APLNA-PJ axis by ML221, an APJ inhibitor, induced catabolic gene expression in PDL cells. Thus, the results of this study provided evidence to support APLN as a regulatory factor of the inflammatory response during periodontitis.

Streptozotocin-Induced Pro-Inflammatory Response in Rat Blood and its Attenuation by External Albumin Administration

  • Previous report has suggested that the albumin levels were reduced in the peripheral blood mononuclear cells (PBMCs) and consequently oxidative stress was elevated in streptozotocin (STZ)-induced diabetic rats as albumin is the predominant antioxidant in plasma. In this study, we suggest that the levels of pro-inflammatory cytokine such as interleukin-6 (IL-6) and tumor necrosis factor-${\alpha}$ ($TNF-{\alpha}$) were increased by approximately 3.4- and 2.9-fold, respectively, in the serum of STZ-diabetic rats, compared to those of normal rat. In addition to the cytokines, the levels of C-reactive protein (CRP) were also about 3.6-fold higher, indicating that STZ induced a pro-inflammatory response in rat blood. However, when purified rat albumin was externally co-administrated with STZ through the tail vein, the serum levels of IL-6, $TNF-{\alpha}$, and CRP were markedly reduced, although the values were still higher than those of normal (non-diabetic) rats. Albumin administration also decreased STZ-induced oxidative stress in serum and PBMCs. Moreover, the decrease in cytokine and CRP levels was dependent on the dose of injected albumin. These results suggest that STZ-induced pro-inflammation and oxidative stress in rat blood might be attenuated by treatment with exogenous albumin.

NRF2 activation by 2-methoxycinnamaldehyde attenuates inflammatory responses in macrophages via enhancing autophagy flux

  • A well-controlled inflammatory response is crucial for the recovery from injury and maintenance of tissue homeostasis. The anti-inflammatory response of 2-methoxycinnamaldehyde (2-MCA), a natural compound derived from cinnamon, has been studied; however, the underlying mechanism on macrophage has not been fully elucidated. In this study, LPS-stimulated production of TNF-α and NO was reduced by 2-MCA in macrophages. 2-MCA significantly activated the NRF2 pathway, and expression levels of autophagy-associated proteins in macrophages, including LC3 and P62, were enhanced via NRF2 activation regardless of LPS treatment, suggesting the occurrence of 2-MCA-mediated autophagy. Moreover, evaluation of autophagy flux using luciferase-conjugated LC3 revealed that incremental LC3 and P62 levels are coupled to enhanced autophagy flux. Finally, reduced expression levels of TNF-α and NOS2 by 2-MCA were reversed by autophagy inhibitors, such as bafilomycin A1 and NH4Cl, in LPS-stimulated macrophages. In conclusion, 2-MCA enhances autophagy flux in macrophages via NRF2 activation and consequently reduces LPS-induced inflammation.

The role of 27-hydroxycholesterol in meta-inflammation

  • 27-Hydroxycholesterol (27OHChol), a prominent cholesterol metabolite present in the bloodstream and peripheral tissues, is a kind of immune oxysterol that elicits immune response. Recent research indicates the involvement of 27OHChol in metabolic inflammation (meta-inflammation) characterized by chronic responses associated with metabolic irregularities. 27OHChol activates monocytic cells such that they secrete pro-inflammatory cytokines and chemokines, and increase the expression of cell surface molecules such as pattern-recognition receptors that play key roles in immune cell-cell communication and sensing metabolism-associated danger signals. Levels of 27OHChol increase when cholesterol metabolism is disrupted, and the resulting inflammatory responses can contribute to the development and complications of metabolic syndrome, including obesity, insulin resistance, and cardiovascular diseases. Since 27OHChol can induce chronic immune response by activating monocyte-macrophage lineage cells that play a crucial role in meta-inflammation, it is essential to understand the 27OHChol-induced inflammatory responses to unravel the roles and mechanisms of action of this cholesterol metabolite in chronic metabolic disorders.

A study on the pulp response of posterior composite resins in relation to the thickness of remaining dentin (잔존상아질 후경에 따른 구치용 복합레진의 치수반응에 관한 연구)

  • The pulp response of posterior composite resins in relation to the thickness of remaining dentin was studied with 120 teeth from 6 dogs, Class V. cavities were prepared on the cervical area of facial surfaces. The thickness of remaining dentin was controlled with Caries Meter$^{(R)}$. The cavities of group A were prepared to show the electrical impedance of 22-26$K{\Omega}$(thickness of remaining dentin:0.4-0.5mm). The cavities of group B, 50-55$K{\Omega}$(thickness of remaining dentin: 0.8-0.9mm). Zinc - Oxide Eugenol cement, Estilux$^{(R)}$ posterior, Heliomolar$^{(R)}$ radiopaque, P-30$^{(R)}$ and Scotchbond$^{(R)}$+P-30$^{(R)}$ were filled in each cavity. After 3days, 1 week, 2 weeks, 4 weeks, 9 weeks and 13 weeks, the teeth and pulp tissue were processed routinely and stained with Hematoxylin and Eosin. Pathological tissue changes were observed with light microscope. The following results were obtained. I. The pulp response of group A cavties was severer than that of group B cavities. 2. In the pulp of group A cavities which were filled with Zinc-Oxide Eugenol Cement, only vascular changes were observed after 3 days and 1 week, severe acute inflammation after 4 weeks, moderate acute inflammation after 9 weeks, and chronic inflammation and formation of granulation tissue after 13 weeks. 3. In the pulp of group A cavities which were filled with Estilux$^{(R)}$ posterior, only vascular changes were observed after 3 days and 1 week. But the inflammatory response has became much severer with the elapsed experimental period. 4. In the pulp of group A cavities which were filled with Heliomolar$^{(R)}$ radiopaque, the inflammatory response with the elapsed experimental period was not severer than that of the pulp of group A cavities which were filled with other materials. 5. In the group B cavities, the difference of pulp response by filling materials was not recognizable. In the group A cavities, the pulp response of Estilux$^{(R)}$ posterior was severest and in order P-30$^{(R)}$, Heliomolar$^{(R)}$ radiopaque was slighter.

  • Chronic inflammatory processes are associated with pathology of Alzheimer's disease(AD). The expression of both cyclooxygenase-2(COX-2) and phospholipase A2(PLA2) appears to be strongly activated during AD, indicating the importance of inflammatory gene pathways as a response to brain injury. Stimulation of heterotrimeric G protein-coupled receptors including muscarinic receptors activates cytosolic PLA2 and receptor-mediated activation of PLA2 generates free fatty acids (i.e., arachidonic acid).
