Up-regulation of cell adhesion molecules and proinflammatory cytokines contributes to enhanced monocyte adhesiveness and infiltration into the skin, during the pathogenesis of various inflammatory skin diseases, including atopic dermatitis. In this study, we examined the anti-inflammatory effects of butein, a tetrahydroxychalcone, and its action mechanisms using TNF-α-stimulated keratinocytes. Butein significantly inhibited TNF-α-induced ICAM-I expression and monocyte adhesion in human keratinocyte cell line HaCaT. Butein also decreased TNF-α-induced pro-inflammatory mediators, such as IL-6, IP-10 and MCP-1, in HaCaT cells. Butein decreased TNF-α-induced ROS generation in a dose-dependent manner in HaCaT cells. In addition, treatment of HaCaT cells with butein suppressed TNF-α-induced MAPK activation. Furthermore, butein suppressed TNF-α-induced NF-kappaB activation. Overall, our results indicate that butein has immunomodulatory activities by inhibiting expression of proinflammatory mediators in keratinocytes. Therefore, butein may be used as a therapeutic agent for the treatment of inflammatory skin diseases. [BMB Reports 2015; 48(9): 495-500]

This study investigated the anti-inflammatory effects of Xanthoceras sorbifolia of seeds oil on RAW264.7 macrophages and TPA (12-O-tetra decanoylphorbol-acetate)-induced ear edema mice. MTT assay method to measure cytotoxicity was formed in RAW264.7 cell. The anti-inflammatory effect was measured by ability to inhibit production nitric oxide (NO) in RAW264.7 cell. Hexane and eight-percent methanol fractions from Xanthoceras sorbifolia seeds oil did not show cytotoxicity. Hexane and eight-percent methanol fractions were showed significantly inhibitory effect on NO production. TPA-induced acute edema was developed in the mouse ears, and Xanthoceras sorbifolia seeds oil dissolved in acetone was applied to inflamed ears. It was found out that Xanthoceras sorbifolia seeds oil could significantly reduce th ear swelling, compared to the control. Overall results indicate that the Xanthoceras sorbifolia seeds oil has anti-inflammatory activity and could be used as a resource of anti-inflammatory materials.
Objectives This study was designed to investigate whether the Cheongajihwang-Tang (CT) has an inhibitory effect association with oxidation or inflammation in RAW264.7 cells. Methods Cytotoxic activity of CT extract on RAW264.7 cells was evaluated by using 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) solution. Nitric oxide production was measured using Griess reagent system. The total phenolic contents and 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was measured to evaluate the anti-oxidative effects of CT. Dichlorofluorescin diacetate (DCFH-DA) has been used as a substrate for measuring intracellular oxidant production. Results Cheongajihwang-Tang does not impair the cell viability in tested concentration. CT showed anti-oxidative effects in vitro by decreasing electron donating ability, and also showed anti-inflammatory effects suppressing NO and ROS expression in LPS induced RAW264.7 activation. CT inhibited the generation of intracellular ROS production as dose dependant manner. Conclusions CT has anti-oxidative effects and anti-inflammatory activities. These results indicate that CT extract has an anti-inflammatory activities via anti-oxidative effects.
Equisetum hyemale Linne. (EH) (Equisetaceae) has been used for the treatment of eye and skin disease, chronic eczema, pneumoconiosis and asthma in Korea and China. Human leukemic mast cells are widely distributed in the connective tissues of mammals and other vertebrates. Phorbol 12-myristrate 13-acetate (PMA) and calcium ionophore A23187 stimulated Human leukaemic mast cell line-1 (HMC-1) can produce a variety of inflammatory mediators and several pro-inflammatory and chemotactic cytokines such as TNF-$\alpha$, IL-6 and IL-8. Since TNF-$\alpha$, IL-6 and IL-8 are major factors during the inflammatory process, we studied the effects of EH on TNF-$\alpha$, IL-6 and IL-8 release in HMC-1 stimulated with PMA and A23187. The result of this study indicate that EH inhibits TNF-$\alpha$, IL-6 and IL-8 in activated HMC-1 cells via $I{\kappa}B$/Nuclear factor-${\kappa}B$ pathway. Therefore, EH might contribute significantly to the prevention or treatment of mast-cell mediated inflammatory diseases and EH has potential use in the therapy of chronic allergic inflammation.

Xanthone is a kind of polyphenolic compounds that contain a distinctive chemical structure with a tricyclic aromatic ring found in a few higher plant families e.g. gentian root. This compound had a variety of biological activity, for instance antioxidant, antibacterial, anti-inflammatory, and anticancer effects. However, the effect of xanthone on mast cell-mediated allergic inflammation and its associated mechanism have not been elucidated. Therefore, the aim of this study was to elucidate the anti-allergic inflammatory effects and the underlying molecular mechanism of xanthone in PMACI-stimulated human mast cells-1 (HMC-1). In this result, xanthone treatment decreased the production of histamine, pro-inflammatory cytokines such as tumor necrosis factor-a (TNF-${\alpha}$), IL-6, and IL-8 and expressions of TSLP in PMACI-stimulated HMC-cells. In addition, xanthone significantly suppressed the phosphorylation of MAPKs and the activation of NF-${\kappa}B$ signal pathway in activated mast cells. Furthermore, xanthone inhibited the activation of caspase-1, an IL-$1{\beta}$ converting enzyme, in PMACI-stimulated HMC-1 cells. These findings provide evidence that xanthone could be a potential therapeutic agent for allergy-related inflammatory disorders.
Marine sources as potential treatment options for various diseases have been a subject of growing interest. However, information on the anti-inflammatory mechanism employed by Undaria pinnatifida (UP) remains limited. The present study was conducted to investigate the mechanisms of UP on the mast cell-mediated inflammatory response. To determine the pharmacological mechanism of UP in inflammatory reaction, we evaluated the effects of UP on interleukin (IL)-6, IL-8 and tumor necrosis factor (TNF)-α production and nuclear factor-κB (NF-κB) and caspase-1 activation in calcium ionophore A23187 plus phorbol 12-myristate 13-acetate-stimulated human mast cells-1 (HMC-1). The results showed that UP suppressed IL-6, IL-8 and TNF-α production in a dose-dependent manner. Moreover, UP significantly attenuated NF-kB/caspase-1 activation in stimulated HMC-1. Collectively, these findings provide experimental evidence that UP may be a useful candidate for the inflammation-related diseases treatment.

Actinidia polygama has long been used in traditional Korean medicine to treat rheumatoid arthritis and gout. Although numerous chemical compounds in the fruit extracts of A. polygama have been characterized and their role in inhibiting nitric oxide (NO) production has been reported, the anti-inflammatory properties of A. polygama extracts remain to be explored. In this study, we investigated the in-vivo effect of A. polygama extract on lipopolysaccharide (LPS)-induced inflammation in BV-2 microglial cell lines. We discovered that 100% ethyl alcohol extract of A. polygama effectively attenuates the release of NO and is superior to both water extract and 50% ethanol extract. Using MTT assay, western blot, and ELISA on LPS-induced BV-2 microglial cells lines, we established the ability of A. polygama extract to markedly suppress the expressions of inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), and pro-inflammatory cytokines, such as tumor necrosis factor alpha and interleukin-6. These results reveal that the anti-inflammatory property of A. polygama in BV-2 microglial cells is due to the downregulation of iNOS, COX-2, MAPK protein, and pro-inflammatory cytokines.

Recent studies suggest that inflammatory events are implicated in a variety of human diseases including cancer and neurodegenerative diseases. It has been reported that expression of inducible cyclooxygenase (COX) and nitric oxide (NO) synthase and subsequent production of prostaglandins (PG)s and NO, respectively are elevated in many inflammatory disorders.(omitted)
In facioscapulohumeral muscular dystrophy (FSHD), prominent inflammatory cellular infiltrates mimicking inflammatory myopathies are often observed in muscle biopsies. We report extensive inflammatory changes in a 16-year-old girl who was genetically confirmed as to have FSHD. Immunohistochemical staining revealed that this could be clearly distinguished from inflammatory myopathies, both in terms of cell subsets and the expression of antigenic targets. Our observations strongly suggest that the inflammatory cellular infiltrates in FSHD differ from those observed in inflammatory myopathies.
Inflammation plays an important role in immune system's response to tissue injury and biological stimuli. However, excessive inflammation can cause tissue damage. Therefore, the development of naturally derived anti-inflammatory agents have received broad attention. In this study, we investigated the anti-inflammatory mechanism of Rubi Fructus (RF) extract on the mast cell-mediated inflammatory response. To determine the regulatory mechanism of RF in inflammatory reaction, we evaluated the effects of RF on secretion of interleukin (IL)-8, IL-6 and tumor necrosis factor (TNF)-α and activation of nuclear factor-κB (NF-κB) and caspase-1 in activated human mast cells-1 (HMC-1). The results showed that RF attenuated IL-8, IL-6 and TNF-α secretion in a concentration-dependent manner. Moreover, RF significantly attenuated caspase-1and NF-κB activation in activated HMC-1. Conclusively, the present results provide evidence that RF may be a promising agent for anti-inflammatory therapy.
