Impact of mesenchymal stem cell senescence on inflammaging

  • Life expectancy has dramatically increased around the world over the last few decades, and staying healthier longer, without chronic disease, has become an important issue. Although understanding aging is a grand challenge, our understanding of the mechanisms underlying the degeneration of cell and tissue functions with age and its contribution to chronic disease has greatly advanced during the past decade. As our immune system alters with aging, abnormal activation of immune cells leads to imbalance of innate and adaptive immunity and develops a persistent and mild systemic inflammation, inflammaging. With their unique therapeutic properties, such as immunomodulation and tissue regeneration, mesenchymal stem cells (MSCs) have been considered to be a promising source for treating autoimmune disease or as anti-aging therapy. Although direct evidence of the role of MSCs in inflammaging has not been thoroughly studied, features reported in senescent MSCs or the aging process of MSCs are associated with inflammaging; MSC niche-driven skewing of hematopoiesis toward the myeloid lineage or oncogenesis, production of pro-inflammatory cytokines, and weakening their modulative property on macrophage polarization, which plays a central role on inflammaging development. This review explores the role of senescent MSCs as an important regulator for onset and progression of inflammaging and as an effective target for anti-aging strategies.

Cordycepin from Medicinal Fungi Cordyceps militaris Mitigates Inflammaging-Associated Testicular Damage via Regulating NF-κB/MAPKs Signaling in Naturally Aged Rats

  • Inflammaging in male reproductive organs covers a wide variety of problems, including sexual dysfunction and infertility. In this study, the beneficial effects of cordycepin (COR), isolated from potential medicinal fungi Cordyceps militaris, in aging-associated testicular inflammation and serum biochemical changes in naturally aged rats were investigated. Male Sprague Dawley rats were divided into young control (YC), aged control (AC), and COR (5, 10, and 20 mg/kg) treated aged rat groups. Aging-associated serum biochemical changes and inflammatory parameters were analyzed by biochemical assay kits, Western blotting, and real-time RT-PCR. Results showed a significant (p < 0.05) alteration in the total blood cell count, lipid metabolism, and liver functional parameters in AC group when compared with YC group. However, COR-treated aged rats ameliorated the altered biochemical parameters significantly (p < 0.05 and p < 0.01 at 5, 10, and 20 mg/kg, respectively). Furthermore, the increase in the expression of inflammatory mediators (COX-2, interleukin (IL)-6, IL-1β, and tissue necrosis factor-alpha) in aged rat testis was significant (p < 0.05) when compared with YC group. Treatment with COR at 20 mg/kg to aged rats attenuated the increased expression of inflammatory mediators significantly (p < 0.05). Mechanistic studies revealed that the potential attenuating effects exhibited by COR in aged rats was mediated by regulation of NF-κB activation and MAPKs (c-Jun N-terminal kinase, extracellular signal-regulated kinase 1/2, and p38) signaling. In conclusion, COR restored the altered serum biochemical parameters in aged rats and ameliorated the aging-associated testicular inflammation proving the therapeutic benefits of COR targeting inflammaging-associated male sexual dysfunctions.

Flavonoids: Broad Spectrum Agents on Chronic Inflammation

  • Flavonoids are major plant constituents with numerous biological/pharmacological actions both in vitro and in vivo. Of these actions, their anti-inflammatory action is prominent. They can regulate transcription of many proinflammatory genes such as cyclooxygenase-2/inducible nitric oxide synthase and many cytokines/chemokines. Recent studies have demonstrated that certain flavonoid derivatives can affect pathways of inflammasome activation and autophagy. Certain flavonoids can also accelerate the resolution phase of inflammation, leading to avoiding chronic inflammatory stimuli. All these pharmacological actions with newly emerging activities render flavonoids to be potential therapeutics for chronic inflammatory disorders including arthritic inflammation, meta-inflammation, and inflammaging. Recent findings of flavonoids are summarized and future perspectives are presented in this review.

미세먼지 유발 피부노화에 대한 쌍별귀뚜라미의 예방 효과 (Anti-skinaging effects of Gryllus bimaculatus on ERM-CZ100-exposed human diploid fibroblasts)

  • 국내 미세먼지의 증가는 만성 호흡기 질환과 피부 염증 및 노화를 유발하여 국민 전체의 건강을 위협하는 심각한 문제로 대두되었다. 본 연구는 미세먼지 유발 피부 염증과 노화에 대해 식용곤충인 쌍별귀뚜라미 70% 에탄올 추출물 (AE-GBE)의 미세먼지에 대한 활성산소 소거능, 세포내 β-galactosidase 효소 활성, MMP-1 발현, 콜라겐 생성, 그리고 염증성 반응에 대해 알아보았다. AE-GBE는 HDF 세포에서 ERM-CZ100에 의해 유도될 수 있는 활성산소종, DNA 단편화 수준 및 β-galactosidase 활성을 유의하게 감소시켰다. 또한 IL-1β, IL-6, TNF-α와 같은 전염증성 사이토카인의 생성과 이들 사이토카인에 의해 발현되는 것으로 알려진 COX2 단백질의 발현을 현저히 감소시켰으며, MMP-1을 억제하여 콜라겐 분해를 막았다. 따라서 본 연구결과는 쌍별귀뚜라미 추출물이 미세먼지 유발 피부 염증에 대한 잠재적인 치료 표적이 될 수 있으며 더 나아가 피부 노화를 늦추는 데 긍정적인 효과를 가질 수 있음을 시사한다.
