The Experiment and Design Formula of Rectangular CFT Columns Reinforced by Carbon Fiber Sheets (탄소섬유쉬트로 보강된 각형 CFT기둥의 실험 및 설계식)

  • Axial load tests and cyclic load tests for FRP reinforced rectangular CFT columns were carried out The main parameters were width-thickness ratio of a steel tubeand FRP layer numbers for the axial load tests and were concrete strength and FRP layer numbers for cyclic load tests. The maximum strength and ductility capacity were compared between the current CFT columns and the FRP reinforced CFT columns. Finally, the axial design formulas were presented for the FRP reinforced CFT columns.

Mutagenic effects of industrial wastewaters by using umu-test (umu-test에 의한 일부 배출시설별 폐수의 변이원성 조사연구)

  • Genotoxicity/mutagenicity of organic chemicals in industrial wastewater was investigated using umu-test with a Salmonella typhimurium TA1535 strain. The tester strain was derived by introducing plasmid pSK 1002, which carried a umu C - lac Z fusion gene into S typhimurium TA1535, and tester strain in the presence microsomal activation proved to be the more sensitive maker of genotoxicity. Genotoxic responses were observed in concentrated with a blue-rayon column, from 14 plants tested. The results were as follow; 1. Genotoxic responses were observed in concentrated from nine plants(64.3%) tested. 2. The results show that genotoxic activity was particulary high in the untreated wastewaters and decreased in the treated wastewaters(35.7%) 3. No significant correlation was found between genotoxicity and water ollution indicators, such as COD and BOD.

Analysis of γ-Aminobutyric Acid Content in Fermented Plant Products by HPLC/UV

  • ${\gamma}$-Aminobutyric acid (GABA) content in fermented plant products and their main plant materials (aerial part of Acanthopanax sessiliflorus, fruit of Crataegus pinnatifida, and whole plant of Morus alba) was determined by high-performance liquid chromatography. GABA was quantified using a reverse-phase column with a gradient elution program (water:acetonitrile =90:10 to 0:100 for 40 min). UV detection was conducted at 280 nm. GABA content was measured in fermented plant products (15.07 mg/g), aerial part of A. sessiliflorus (4.49 mg/g), fruit of C. pinnatifida (10.59 mg/g), and whole plant of M. alba (2.31 mg/g). The presence of GABA in fermented plant products, including A. sessiliflorus, C. pinnatifida, and M. alba is important in industrial application for health supplements.

  • The propose of this study was to investigate the feasibility of using waste lime core to remove phosphorus from wastewater in continuous flow reaction. The phosphorus was found to be removed from municipal wastewater by hydroxyapatite crystallization and precipation. Waste lime core size 1, 2 showed phosphorus removal rate of about 90% during early 11 hrs of run time. In addition, breakpoint time was decreased by increased inflow rate regardless of waste lime core size.

  • A room and pillar mining method has been adopting at the Jeungsun limestone mine. To check stability of pillar with multiple and irregular openings, the size, shape and spacing of rib pillar were first designed using some empirical suggestions. The Finite Difference Method(FDM)was used to analyze the pillar stability. Twelve different cases with the variation of K(horizontal/vertical stress)values, different height and different spacing of pillar were used in this study. Finally Mohr-Coulomb criterion was adopted to calculate the safety factors. Horizontal and vertical displacement, maximum and minimum principal stresses, range of plastic zone and safety factors were calculated at each case. As a result of analysis, the size of one block is 160m long, 70m wide, 40m high with 20m wide rib pillar and 20m square column pillar. The overall recovery at this case can be estimated about 40%.

Soluble Expression of Human Angiostatin and Endostatin by Maltose Binding Protein (MBP) Fusion in E. coli (Maltose Binding Protein 융합단백질에 의한 인간유래의 앤지오스타틴과 앤도스타틴의 대장균에서 수용성 단백질발현)

  • Rapid production of therapeutic proteins such as angiostatin and endostatin angiogenic inhibititors has been highly demanded for cancer treatment. In this regard, recombinant human angiostatin and endostatin were successfully expressed as soluble forms by maltose binding protein (MBP)-mediated fusion expression in Escherichia coli. PCR amplified, angiostatin and endostatin genes from human placenta cDNA library were inserted into an expression vector pMAL-c2e to construct prokaryotic expression vectors, pMAL-c2e/AS and pMAL-c2e/ES, respectively. Recombinant angiostatin and endostatin were efficiently expressed in E. coli origami (DE3) after IPTG induction and protein expression were confirmed by SDS-PAGE analyses. The expressed recombinant proteins were purified near homogenity using an amylose affinty column chromatography. In contrast that previous E. coli expressions were all insoluble, our results first time demonstrated that MBP fused human angiostatin and endostatin were soluble in E. coli.

Cloning and expression of cellulase genes from Erwinia carotovora in E. coli (Erwinia carotovora 유래의 cellulase 유전자의 클로닝 및 대장균에서의 발현)

  • New cellulase genes, named as CelV2 and CelN1, respectively, were isolated from Erwinia carotovora ATCC15713 and expressed in E. coli. The CelV2 and CelN1 gene were PCR amplified with degenerated primers and PCR products were sequenced and expressed in E. coli. Two new cellulase genes showed 97% homologies with previously reported Erwinia cellulase genes. The recombinant cellulase were purified with Ni-NTA column chromatography and its enzymatic properties were characterized. The optimum temperature of two enzymes were about $50^{\circ}C$ degree and optimum pH were around pH7.0. The newly isolated celluase genes could be used for enhancing substrate range of alcohol-producing bacteria such as Zymomonas mobilis.

Prediction of Consolidational Settlement of Dredged and Reclaimed Ground (준설매립토지반의 압밀침하량 예측)

  • For soils with high void ratios, the inverse method of utilizing results obtained from centrifuge model test was used to find the constitutive relation of effective stress - void ratio - permeability whereas conventional oedometer test and constant rate of strain consolidation test were also used to fine its relation at ranges of relatively low void ratio. Results of column test about settlement of interface and pore pressure and distribution with time were compared with numerically estimated values to confirm such a constitutive relation as obtained from the inverse method. Consolidational settlement in dredged and reclaimed ground, where the consolidation was in progress, was predicted by using the numerical technique implemented with the finite strain consolidation theory.

Cloning and expression of new laccase gene (soncotA) from Bacillus sonorensis KCTC13918 in E. coli (Bacillus sonorensis KCTC13918로부터 새로운 laccase유전자 (soncotA)의 클로닝과 대장균에서의 발현)

  • A new putative laccase gene (soncotA) which show 78% homology with that from Bacillus licheniformis (liccotA) was isolated from draft genome sequence of Bacillus sonorensis KCTC 13918. A 1,545 bp of PCR product corresponding 514 amino acids was cloned into NdeI-NotI site of pET21c and expressed as soluble form in E. coli. About 59 kDa size of recombinant laccase was purified into homogenity by Ni-NTA column and laccase activity was confirmed by zymography. The enzymatic properties of recombinant laccase were characterized. The specific activity of B. sonorensis laccase was 0.033 fold lower than that of Bacillus licheniformis laccase. The finding of new laccase gene broadened the enzymatic diversity of Bacillus species laccases.

  • This study showed a way to develop web site of shopping mall through systematic identification of the interface elements affecting user's performance and subjective sensibility for Internet shopping mall. To quantify the effects of factors, simulators were designed and used in experiments. It was shown that location of 'Table of Contents'(left > right = up = down) and 'Menu' type(non_Drop down and page Movement = Drop down and page Movement > Drop down and non_age Movement) were the significant factors. However, whether 'Frame' was used or not, there was no significant difference. Also, in the evaluation of subjective sensibility, 'Background color' was a significant factor. And for 'Header & Scanning Column color', yellow color had a tendency to enhance satisfaction for 'simplicity', while green or blue color strengthened 'balance' feeling. But 'text style' was not significant.
