고성능 액체 크로마토그래피에 의한 지방족 화합물의 간접 분광광도 측정에 관한 연구 (A Studies on Indirect Photometric Detection of Aliphatic Compounds by High-Performance Liquid Chromatography)

  • 고성능 액체 크로마토그래피에 있어서 지방족 화합물인 알코올 케톤 및 에스테르의 간접 분광광도 측정에 관하여 조사하였다. 비발색단 또는 약한 발색단을 가지고 있는 이들 시료들은 검출시약으로 UV 흡수물질인 p-클로로페놀 또는 p-니트로페놀을 이동상에 첨가하므로 UV 검출기로서 간접적으로 검출할 수 있었고 검출한계로 Submicrogram 수준까지 얻을 수 있었다. 또한 최적조건하에서 몇가지 혼합시료들을 좋은 분리도와 비교적 높은 감도로서 분리 및 정량할 수 있었다.

  • An ion chromatographic method based on indirect photometric detection of UV transparent anions was developed. Separation of anion was accomplished on strong anion exchange column (Waters SAX) using UV detector at 254 nm. Among examined UV-active additives (Dns-H, Dns-glu, Dnsser, Dns-val), Dns-glu showed the highest sensitivity. Studies on the effects of the pH and ionic strength of eluent revealed that the increase of pH and ionic strength of the eluent decreased capacity factor. The best eluent for the separation of acetate, fluoride, chloride, nitrate and bromide was $1\;{\times}\;10^{-4}M$ Dns-glu in 5 mM phosphate buffer (pH 6.30). The detection limit of chloride ion was 2.1 ng in this condition.

  • The separation and determination of inorganic cations by a capillary zone electrophoresis was studied. Cations were separated by the capillary column and detected by indirect UV method. The running buffer of 0.03 M creatinine and 0.02 M-hydroxy isobutylic acid solution (pH 4.8) was used to improve the mobility of ions in the column. A potential of 18 kV was applied at anode. A complete separation of cations ranged within 15 min and detection limits were between 0.1 and 1.0 ppm. The applicability of the method for real samples was demonstrated.

  • Bromide, chloride, fluoride, nitrite, nitrate, sulfite, sulfate, perchlorate, chlorate 및 chlorite가 포함된 시료를 capillary zone electrophoresis(CZE)와 indirect UV detection 방법으로 분리하였다. 음이온류의 분리는 running buffer로 5 mM ammonium dichromate, 10 mM ammonium acetate, 20 mM diethylenetriamine, 10% methanol(pH 9.3)을 사용하였으며 전압은 cathode에 15 kV를 공급하였다. 또한 최초로 CZE에서 음이온의 정량이 가능하도록 각 이온들에 대한 regression과 detection limit를 결정하였고, 이 분석방법은 톨루엔을 흡입한 사람의 뇨, 부검 혈액, 폐수 및 폭약 등 시료에 적용하였으며, 음이온의 함량을 ion chromatograph(IC)와 비교 검토하였다.

Analysis of Inorganic Anions in Various Drinking Waters by Capillary Electrophoresis

  • The quantitation of inorganic anions in various drinking waters were investigated using capillary electrophoresis(CE) and the results were compared with ion chromatography(IC). With CE, in contrast to IC, was demonstrated rapid analysis, good efficiency, a low detection limit and the low comsumption of a solvent and samples. CE analysis was used 5 mM sodium chromate(pH 8.0) containing 20 mM tetraalkylammoniumbromide at -25kV applied voltage with indirect UV detection at 254 nm. This results in exceedingly short analysis time within 3 min. with efficiencies approaching 200,000 theoretical plates. The coefficients of variants of migration time are less than 0.8% and those of peak area are less than 2.3%. Detection limits for quantitative determination were 300 ppb-50 ppm level. These optimum conditions are applicable to various samples without pretreatment.

  • 간접자외선흡광검출 이온크로마토그래피를 7가지 음이온($H_2PO_4^-$, $Cl^-$, $NO_2^-$, $NO_3^-$, $I^-$, $SO_4^{2-}$, $SCN^-$)분리에 적용하였다. 컬럼은 저용량의 강염기성 음이온 교환체를 가진 상용화된 TSK-guardgel QAE-SW (ca. 0.3 meq./g; 4.6 mm i.d${\times}$150 mm)를 사용하였다. 선택적이고 신속한 이온분리방법을 연구하기 위하여 naphthalene trisulfonic acid(NTS) 및 naphthalene disulfonic acid(NDS) 용리액에 대한 분리거동을 조사하였다. 더 나아가, 0.20 mM NTS 용리액에 5% 아세토리트릴을 첨가하여 음용수에 적용한 결과, 양이온과 음이온을 동시에 분리 할 수 있었다.

  • Determination of chloride ion in concentrated NaOH solution by capillary electrophoresis has been studied. The analysis was performed by indirect UV absorption detection using chromate buffer at 254nm. The matrix effect of the sample has been observed so that the sensitivity in strong NaOH solutaion has decreased up to 10% of that in distilled water. The pH effect of the sample on the sensitivity of CE peaks has been investigated. The method for increasing the sensitivity have been investigated and the optimum pH and concentration of the buffer were 7.5 and 10mM, respectively. A cationic surfactant cetyltrimethylammonium bromide(CTAB), was added to a buffer solution in order to reverse the electroosmotic flow(EOF) in the capillary. This results in a short analysis time and better peak shapes. Using this optimum condition, the determination of chloride ion in real environmental sample has been performed, which is captured in strong NaOH absorbent prepared for absorbing gas from chimney. The standard addition method has been applied for the quantitative analysis, and it was obtained the good reproducibility.
