Objective: This study was conducted to compare different methods on effective energy evaluation of corn silage for beef cattle. Methods: Twenty Wandong bulls (Chinese indigenous yellow cattle) with initial body weight of $281{\pm}15.6kg$, were assigned to 1 of 5 dietary treatments with 4 animals per treatment in a randomized complete block design. Five dietary treatments included group 1 with corn silage only diet, group 2 with corn silage-concentrate basal diet (BD) and 3 groups with 3 test diets, which were the BD partly substituted by corn silage at 10%, 30%, and 60%. The total collection digestion trial was conducted for 5 d for each block after a 10-d adaptation period, and then an open-circuit respiratory cage was used to measure the gas exchange of each animal in a consecutive 4-d period. Results: The direct method-derived metabolizable energy and net energy of corn silage were 8.86 and 5.15 MJ/kg dry matter (DM), expressed as net energy requirement for maintenance and gain were 5.28 and 2.90 MJ/kg DM, respectively; the corresponding regression method-derived estimates were 8.96, 5.34, 5.37, and 2.98 MJ/kg DM, respectively. The direct method-derived estimates were not different (p>0.05) from those obtained using the regression method. Using substitution method, the nutrient apparent digestibility and effective energy values of corn silage varied with the increased corn silage substitution ratio (p<0.05). In addition, the corn silage estimates at the substitution ratio of 30% were similar to those estimated by direct and regression methods. Conclusion: In determining the energy value of corn silage using substitution method, there was a discrepancy between different substitution ratios, and the substitution ratio of 30% was more appropriate than 10% or 60% in the current study. The regression method based on multiple point substitution was more appropriate than single point substitution on energy evaluation of feedstuffs for beef cattle.
This study was carried out to examine a molecular marker system for parentage test in Jeju Black cattle (JBC). Based on the preliminarily studies, we finally selected for construction of a novel genetic marker system for molecular traceability, identity test, breed certification, and parentage test in JBC and its related industrial populations. The genetic marker system had eight MS markers, five indel markers, and two single nucleotide polymorphisms (SNPs; g.G299T and g.del310G) within MC1R gene which is critical to verify the breed specific genotypes for coat color of JBC differing from those of exotic black cattle breeds such as Holstein and Angus. The results showed lower level of a combined non-exclusion probability for second parent (NE-P2) of $4.1202{\times}10^{-4}$ than those previously recommended by International Society of Animal Genetics (ISAG) of $5.000{\times}10^{-4}$ for parentage, and a combined non-exclusion probability for sib identity (NE-SI) of $2.679{\times}10^{-5}$. Parentage analysis has been successfully identified the JBC offspring in the indigenous population and cattle farms used the certified AI semens for production using the JBC-derived offspring for commercial beef. This combined molecular marker system will be helpful to supply genetic information for parentage test and traceability and to develop the molecular breeding system for improvement of animal productivity in JBC population.
Anaplasmosis is a tick-borne disease mainly of cattle, sheep, and goats. Anaplasmosis in goat had been reported at last year by authors. The disease brought the economic losses in the goat farm in Chonbuk province. In order to treat the disease, a long-acting formulation terramycin injectable solution was used experimental animals which indigenous goats were sick with moderate anemia. We were devided into two groups was treated with single dosage (20mg /kg of body weight). one group was treated with single injection, the other group was treated with twice of the same dosage intramusculary injection. The results indicate that the use of long-acting terramycin would minimize clinical signs of anaplasmosis infection in goat. After treament the differrences of hematological appearences and parasitaemia were observed in the effect of terramycin treatment. obviously, increased value of RBC. HB and HCT, and parasitaemia by means of Giemsa staining and acrydine orange staining was observed decreased due to treatment.
Two 1-year-old Korean native steers in the same herd presented severe hemorrhagic diarrhea. Case 1 had severe dehydration and died after 3 days, whereas case 2 had anorexia, depression, and severe diarrhea with mucus and blood. Only case 2 was necropsied, and bovine viral diarrhea virus-2a (BVDV2a) was detected in the tissues of its alimentary tract. Gross lesions, including erosion, ulceration, and extensive hemorrhage, were observed in the digestive tract mucosa. Immunohistochemistry revealed marked positive staining for BVDV2a antigen in the large intestine. These findings are indicative of hemorrhagic disease caused by BVDV2a in a native Korean steer.
This study was evaluated to know the effects of vitamin E(VE) on the lipid peroxidation in blood and sirloin of castrated korean indigenous beef cattle. Experimental groups were divided into VE 500IU(A), 1,500IU additative feeding group(B) and non-VE-treated control group(C). After oral administration to the cattle for 120 and 150 days, body weight gains, VE contents in plasma and sirloin, and thiobarbituric acid(TBA) value were examined according to the exhibition period(1-7 days) in refrigerated showcase between aging and non-aging group. The results obtained from this study were summarized as follows ; 1. Body weight gain per day of control compared with VE additative feeding A and B groups were showed no significantly differences. 2. The concentrations of VE in plasma after oral administration with VE for 120 days were significantly increased(p<0.05) in A and B groups. There were higher(p<0.n) 4.22$\mu\textrm{g}$/$\m\ell$ in A and 6.22$\mu\textrm{g}$/$\m\ell$ in B group than the control(3.0$\mu\textrm{g}$/$\m\ell$). And the concentrations of VE in plasma for 150 days were significantly increased(p<0.05) in VE additative feeding groups. There were higher 4.89$\mu$g/$m\ell$ in A and 7.05$\mu\textrm{g}$/$\m\ell$ in B group than the control(3.15$\mu\textrm{g}$/$\m\ell$). 3. The concentrations of VE in sirloin for 120 days were significantly increased(p<0.05) in A and B groups. There were higher 1.84$\mu\textrm{g}$/g in A group and 2.40$\mu\textrm{g}$/g in B group than the control(0.78$\mu\textrm{g}$/g). And the concentrations of VE in sirloin for 150 days were significantly increased(P<0.05) in A and B groups. There were higher 1.94$\mu\textrm{g}$/g in A group and 2.63$\mu\textrm{g}$/g in B group than the control(1.00$\mu\textrm{g}$/g). 4. TBA values, the indicator of lipid peroxidation, in non-aging sirloin according to the exhibition period(1-7 days) in refrigerated showcase after oral administration with VE additative feed for 120 days were lower 0.73 in A and B groups than 0.82 in control at the third day after exhibition. In the same group, TBA values were significantly(p<().05) tower 0.77 and 0.75 in A and B groups than 1.22 in control at the seventh day after exhibition. Equally, in the aging group, there were significantly(p<0.05) showed lower TBA values 1.05 and 0.99 in A and B groups than 1.87 in control at the seventh day after exhibition. 5. After oral administration with VE additative feed to the cattle for 150 days, TBA values in non-aging sirloin according to the exhibition period(1-7 days) in refrigerated showcase were significantly(p<0.05) decreased to 0.84 and 0.88 in A and B groups than 1.26 in control at the seventh day after exhibition. In the aging group, there were significantly(p<0.05) showed lower TBA values 0.95 and 0.99 in A and B groups than 1.79 in control at the seventh day after exhibition.
The aim of the current study was to analyze the diversity of the major surface protein (Msp) gene in Theileria buffeli, which is known as the major antigenic protein recognized by the immune system of the host. In addition, we characterized the diversification of the Msp gene and its relationship to with the pathogenicity of Theileria. Complete blood counts (CBC) and Theileria 18S rRNA PCR sequence analysis were performed for 177 Korean indigenous cattle (KIC) in Jeju Island. A total of 28 KIC (16 anemic and 12 non-anemic KIC) were then randomly selected based on 18s rRNA PCR positive samples for sequence analysis of the Theileria Msp gene, which was performed twice for each specimen. The resulting 56 Msp gene sequences were classified into five antigenicity types (type I to V), according to the variable region (517-571 bp), which exhibited high similarity (${\geq}$ 98.9%) to several available GenBank sequences (Theileria spp. from China-EU584237; T. sergenti from China-DQ078264; Theileria spp. from Thailand-AB081329; Theileria spp. from Japan-AB218442; T. sergenti from Japan-AB016280). The 56 Msp sequences consisted of 22, 15, 9, 8, and 2 cases of type I to type V Msp genes, respectively. The most prevalent type in both anemic and non-anemic KIC was type I (37.5% in anemic and 41.7% in non-anemic). Among the remaining types, type II was the most prevalent (37.5%) in anemic KIC, while type IV was the most prevalent (25%) in non-anemic KIC. The results of our study help confirm the diversity of Msp gene types and demonstrate that the gene type distribution of Msp genes varies among Theileria-infected KIC in Jeju Island.
The aim of the present study was to investigate whether Bovine Spongiform Encephalopathy (BSE) is present in this country and to analyze the Global BSE Risk (GBR) status in Bangladesh. A total of 2,000 brain samples were collected from cattle older than 30 months of age, slaughtered for human consumption in the district slaughter houses from 2005 to 2006. The brainstem (obex), Pyriform lobe, cerebrum and cerebellum were subjected to histopathological study. Samples that showed some nonspecific lesions were subjected to immunohistochemistry and only brain stem to ELISA for the detection of abnormal prion protein $PrP^{sc}$. In passive surveillance, annual overall diseases of cattle, buffalo, sheep and goats in Bangladesh were collected from Department of Livestock Services (DLS), Dhaka to investigate the occurrences of neurological diseases. Import related data were collected from "National Export Promotion Bureau" Kawran Bazar, Bangladesh Bank and DLS to analyze the importing products of animal origin (cattle, buffalo, sheep and goats) from different countries to find whether or not the imported products posed any risk for the BSE. In an actire surveillance conducted in slaughter house, histopathologically BSE specific lesions were not detected in any of the brain samples, but other nonspecific lesions were observed. No $PrP^{sc}$ was detected from the samples by immunohistochemistry and ELISA. DLS report also supported the absence of BSE in cattle and buffalo and scrapie in sheep and goats in Bangladesh. It was also clearly recorded that Bangladesh imported livestock products from countries in GBR level I and II but not from countries in GBR level III and IV. From this study it apparently seems that BSE is not currently present in the indigenous animals in Bangladesh and poses no or negligible risk to human and animal health.

Objective: Recently, indigenous Korean grass Sasa quelpaertensis Nakai (SQ) has garnered much interest as a roughage source for livestock to mitigate its adverse effects on habitat diversity. Thus, the objective of the present study was to evaluate the ruminal fermentation, palatability, and nutrient digestibility of SQ for Korean native beef cattle (Hanwoo) using in vitro rumen fermentation, in situ rumen degradability, and in vivo feeding trials. Methods: Using in vitro tests with rumen fluid as the inoculum for 48 h, ruminal fermentation of SQ was evaluated and compared with that of other roughage sources commonly used in Korea (i.e., rice straw, Timothy hay, and Italian ryegrass [IRG]). Additionally, an in situ trial 96 h was performed using three cannulated Hanwoo steers. Further, an in vivo trial was performed using eight Hanwoo steers to compare the palatability of SQ with rice straw in total mixed ration (TMR) and forage-concentrate separate feeding conditions. Finally, an in vivo digestibility trial of SQ fed as TMR of two particle sizes was performed with four Hanwoo steers. Results: In vitro and in situ trials revealed that SQ was comparable or superior to rice straw in terms of the ruminal fermentation characteristics of pH, gas production, total volatile fatty acid content, and effective ruminal dry matter digestibility (DMD), although its fermentability was lower than that of Timothy hay and IRG. In the palatability test, steers showed a greater preference for SQ when given as TMR. The total tract DMD of SQ fed as TMR was 75.9%±1.37%, and it did not differ by particle size. Conclusion: The feed value of SQ as a roughage source for Hanwoo steers is comparable or superior to that of rice straw, particularly when provided as TMR.
Thanks to recent advances in next-generation sequencing (NGS) technology, diverse livestock species have been dissected at the genome-wide sequence level. As for cattle, there are currently four Korean indigenous breeds registered with the Domestic Animal Diversity Information System of the Food and Agricultural Organization of the United Nations: Hanwoo, Chikso, Heugu, and Jeju Heugu. These native genetic resources were recently whole-genome resequenced using various NGS technologies, providing enormous single nucleotide polymorphism information across the genomes. The NGS application further provided biological such that Korean native cattle are genetically distant from some cattle breeds of European origins. In addition, the NGS technology was successfully applied to detect structural variations, particularly copy number variations that were usually difficult to identify at the genome-wide level with reasonable accuracy. Despite the success, those recent studies also showed an inherent limitation in sequencing only a representative individual of each breed. To elucidate the biological implications of the sequenced data, further confirmatory studies should be followed by sequencing or validating the population of each breed. Because NGS sequencing prices have consistently dropped, various population genomic theories can now be applied to the sequencing data obtained from the population of each breed of interest. There are still few such population studies available for the Korean native cattle breeds, but this situation will soon be improved with the recent initiative for NGS sequencing of diverse native livestock resources, including the Korean native cattle breeds.
The aim of this study was to examine the epidemiological features of Eimeria in calves with acute diarrhea. Samples were collected from between 15 days and 90 days old calves (n=83) in Gimje area from March 2006 to March 2007. Feces of bloody diarrhea were examined for the presence of Eimeria oocysts using a sucrose flotation method. Out of 83 calves, 62 (74.6%) had Eimeria oocysts. In the results of monthly analysis, the highest prevalence (12.0%) of Eimeria oocysts was found on June. In the seasonal infection rate, spring was the highest prevalence (30.1%), followed by summer (24.0%). Furthermore, the highest prevalence (44.5%) was found in calves from between 31 - 60 days old in the analysis by ages. However, there was no significant differences between female and male sex even though the prevalence was slightly bigger in female than in male. The prevalence of the present study to detect Eimria oocysts for infection may have been affected by weather-conditions in the spring. Young calves should be separated to minimize the infection from cattle as much as possible. Additional studies are necessary to find other factors for infection and combining molecular methods with a highly sensitive system for Eimeria detection could be a reliable and economic way of Eimeria eradication.
