Protective Effect of Mulberry and Lithospermum erythrorhizon Extracts on Anti-aging against Photodamage (오디 및 자초추출물의 피부 광노화 보호효과)

  • We investigated the protective effect of UVB inducing photodamage from mulberry extract (ME) and Lithospermum erythrorhizon extract (LE). The contents of total anthocyanin and shikonin as a color compound of ME and LE were 4.92 mg/g and 9.58 mg/g, respectively. The electron donating ability and superoxide radical scavenging activity of ME were 84.32% and 76.34%, respectively. The oxygen radical absorbance capacity of the ME ($545.37{\mu}moles$ TE/g) was higher than LE ($427.18{\mu}moles$ TE/g). MMP-1 production in the HS68 cells were exposed to UVB suppressed by treatment with $200{\mu}g/mL$ of ME (68.6%) and LE (32.7%). ME and LE were applied to a skin aging mouse model, which was induced by the irradiation of UVB to the backs of hairless mice. The value of skin erythema index, wrinkle depth and thickness, epidermis thickness, and collagenous fiber damage in the experiment groups (MEL: ME 3%, MEM: ME 5%, MEH: ME 7%, LEL: LE 3%, LEM: LE 5%, LEH: LE 7%) were remarkably reduced than in the control group (only UVB exposure group), while water capacity increased. The level of total wrinkles depth in the skin was decreased to be 30% of the control group by MEH and LEM. These results suggest that ME and LE are useful cosmetic materials for skin protection against UVB-inducing.

The Comparison of Growth and Quality Characteristics during the Storage of Pleurotus ostreatus Cultivated in the Remnants of Medicinal Herb Extracts (한약박에서 재배한 느타리버섯의 성장 및 저장 중 품질 특성의 비교)

  • This study was conducted to determine whether Pleurotus ostreatus (oyster mushroom), cultivated in various ratios with herbal extract remnants instead of cotton supplemented with nutrients (the control), improved mycelial growth, mushroom yields and longevity during storage. In addition, we investigated the transfer of medicinal herb components into the mushrooms since they contained non-specific medicinal herbs and their composition could not be controlled. Mushrooms cultivated with 70% and 100% medicinal herb remnants had faster growth rates, higher yields and less failure in the development of the fruit body than the control group. There were no differences in HPLC chromatogram among the methanol extracts of Pleurotus ostreatus in all groups. In addition, glycyrrhizin, an indicative compound of licorice which was a major herb among the herbal remnants, was not detected in any of the extracts. Pleurotus ostreatus that was cultivated with 70% and 100% herbal extract remnants had improved storage longevity in comparison with the control. They exhibited the least weight loss during storage among the groups and they maintained firmness in the stipe and pileus. However, the sources of media did not alter the color difference of the stipe and pileus or the quality index of the outward appearance during storage. In conclusion, cultivating media that contained over 70% of medicinal herb extract remnants increased the growth rates and yields of Pleurotus ostreatus. In addition, these mushrooms had enhanced storage longevity due to their firmness. Therefore, medicinal herb extract remnants should be utilized in the cultivating media of various mushrooms.

  • Pharmacologic coronary vasodilation in conjunction with myocardial scintigraphy has become an accepted alternative to dynamic exercise testing for the diagnosis of coronary artery disease. Although dipyridamole has traditionally been used for this purpose, it causes frequent side effect, which at times can be life-threatening. Moreover, dipyridamole dose not elicit maximal coronary vasodilation in a substantial number of patients receiving the usual i.v. dose. Adenosine is an endogenously produced compound that has significant effects as a coronary vasodilator and rapid onset action and extremely short half-life (< 10 seconds). The diagnostic accuracy and safety profile of adenosine $^{99m}Tc-MIBI$ myocardial scintigraphy were evaluated and comparison with exercise $^{99m}Tc-MIBI$ was performed. Twenty-eight subjects underwent $^{99m}Tc-MIBI$ imaging after adenosine infusion and exercise $^{99m}Tc-MIBI$ imaging. Adenosine was infused intravenously at a dose of 0.14mg/kg/body weight per minute for 6 min and MIBI was injected at 3 minute. Adenosine caused an incerease in heart rate ($64{\pm}12$ at baseline versus $74{\pm}16$ beats/min at peak effect, p<0.001), a mild decrease in systolic and diastolic blood pressure and a slightly increase in PR interval(p; NS). Side effects were reported in 92% of patients and were mostly mild in nature and promptly resolved within 1 or 2 minutes of termination of adenosine infusion. Facial flushing (53%), chest pain (36%), mild dyspnea (39%), headache (21%), throat discomfort (21%) were frequent symptoms. ST segment depression (> 1 mm) and second degree AV block in electrocardiography occured in 11% of the patients, respectively. The overall sensitivity and specificity for individual coronary stenoses in 16 patients underwent coronary angiography were 88% and 95%, respectively. The agreement ratio of segmental perfusion between adenosine and exercise images was 92% (Kappa index=0.82). In conclusion, $^{99m}Tc-MIBI$ myocardial perfusion scintigraphy with intravenous adenosine is a feasible, safe and highly accurate noninvasive technique for the detection of coronary artery disease and results are at least comparable with those of exercise $^{99m}Tc-MIBI$ scintigraphy.

Assessment of health risk associated with arsenic exposure from soil, groundwater, polished rice for setting target cleanup level nearby abandoned mines

  • This study focused on health risk assessment via multi-routes of As exposure to establish a target cleanup level (TCL) in abandoned mines. Soil, ground water, and rice samples were collected near ten abandoned mines in November 2009. The As contaminations measured in all samples were used for determining the probabilistic health risk by Monte-Carlo simulation techniques. The human exposure to As compound was attributed to ground water ingestion. Cancer risk probability (R) via ground water and rice intake exceeded the acceptable risk range of $10^{-6}{\sim}10^{-4}$ in all selected mines. In particular, the MB mine showed the higher R value than other mines. The non-carcinogenic effects, estimated by comparing the average As exposure with corresponding reference dose were determined by hazard quotient (HQ) values, which were less than 1.0 via ground water and rice intake in SD, NS, and MB mines. This implied that the non-carcinogenic toxic effects, due to this exposure pathway had a greater possibility to occur than those in other mines. Besides, hazard index (HI) values, representing overall toxic effects by summed the HQ values were also greater than 1.0 in SD, NS, JA, and IA mines. This revealed that non-carcinogenic toxic effects were generally occurred. The As contaminants in all selected mines exceeded the TCL values for target cancer risk ($10^{-6}$) through ground water ingestion and rice intake. However, the As level in soil was greater than TCL value for target cancer risk via inadvertent soil ingestion pathway, except for KK mine. In TCL values for target hazard quotient (THQ), the As contaminants in soil did not exceed such TCL value. On the contrary, the As levels in ground water and polished rice in SD, NS, IA, and MB mines were also beyond the TCL values via ground water and rice intake. This study concluded that the health risks through ground water and rice intake were greater than those though soil inadvertent ingestion and dermal contact. In addition, it suggests that the abandoned mines to exceed the risk-based TCL values are carefully necessary to monitor for soil remediation.

Effects of Processing Temperature and Browning Inhibitor on Quality Properties of Fresh-cut Burdock Roots (가공 온도 및 갈변 억제제가 신선절단 우엉의 품질 특성에 미치는 영향)

  • Fresh-cut burdock roots were processed at 4 and $25^{\circ}C$, respectively. The fresh-cut burdock roots were treated with different browning inhibitors (3% citric acid, 3% sodium chloride, 0.3% cystein, and 3% sodium acetate solutions), and the changes in their quality and browning characteristics were investigated. The respiration rate and browning index of the cut roots prepared at $4^{\circ}C$ were lower than those of the cut roots prepared at $25^{\circ}C$. The soluble solid content was higher in the cut roots prepared at $4^{\circ}C$ than in those prepared at $25^{\circ}C$. The weight loss and flesh firmness were not affected by the processing temperatures. Among the browning inhibitors, 0.3% cystein showed the best browning-retarding effect. There was no difference in phenolic compound content between the browning-inhibitor-treated roots and the nontreated roots, but the PPO activity was higher in the latter than in the former. Therefore, the processing temperatures of fresh-cut burdock roots affected their quality and browning development, and the combination of a low processing temperature and the use of the proper browning inhibitor should be applied for the higher quality of the produce.

Influence of Abnormally Low Temperatures on Growth, Yield, and Biologically Active Compounds of Strawberry (이상 저온조건이 딸기의 생육, 수량 및 생리활성 성분에 미치는 영향)

  • The present study aimed to investigate the effects of low temperature on the growth, yield, quality, and biologically active compounds of strawberry and obtain basic information for developing a technology for stable growth of strawberry in greenhouses. Growth of strawberry, including leaf number, area, and length, plant height, and dry weight was better at the optimum growth temperature of $20^{\circ}C$ than at a lower temperature of $15^{\circ}C$. At the low temperature of $15^{\circ}C$, the cultivar 'Maehyang' was more tolerant and displayed better growth rate than 'Seolhyang'. At $15^{\circ}C$, the fruit production per week and fruit weight was lower than that at $20^{\circ}C$. In contrast, fruit length and diameter were not significantly different between the two growth temperatures. Growth temperature also did not affect the fruit color index, Hunter L, a, b value, or fruit firmness. However, the sugar content of strawberries grown at $15^{\circ}C$ was higher by 0.8 and 1.5 Brix for 'Seolhyang' and 'Maehyang', respectively, than of those grown at $20^{\circ}C$. There was no difference in the content of fisetin, a biologically active compound, for 'Seolhyang' at both growth temperatures, however, the fisetin content of 'Maehyang' was higher at $20^{\circ}C$ than at $15^{\circ}C$. Cinchonine and ellagic acid content of 'Seolhyang' was higher at $20^{\circ}C$ than at $15^{\circ}C$, whereas that of 'Maehyang' was higher at $15^{\circ}C$ than at $20^{\circ}C$. Quercetin content showed no significant differences with respect to growth temperature, however, it tended to increase at $20^{\circ}C$. The cinnamic acid content of 'Seolhyang' was higher at $15^{\circ}C$ than at $20^{\circ}C$, whereas that of 'Maehyang' increased at $20^{\circ}C$. Collectively, the biologically active compounds of strawberry were affected by growth temperature. Moreover, the content of these compounds tended to increase at $20^{\circ}C$, the optimum growth temperature, rather than at the sub-optimal growth temperature of $15^{\circ}C$.

Effects of Roasting Temperature on Phycochemical Properties of Job's tears (Coix lachryma jobi L. var ma-yeun) Powder and Extracts (볶음온도에 따른 율무 분말과 침출액의 이화학적 특성)

  • This study was carried out to evaluate the effect of the masting temperature on the phycochemical properties of the roasted Job's tears. Raw seeds were roasted for 20 min at 150, 170, 190, 210 or $230^{\circ}C$, were milled and extracted with hot water. The L and a values of the powder were sharply decreased or increased at the masting temperature of above $190^{\circ}C$, respectively. The b value was maximum at $190^{\circ}C$. Water absorption capacity of the powder and browning index of the extract were proportionally increasing with increasing the masting temperature. The pH of the extracts was decreased at the masting temperature of above $190^{\circ}C$. Total sugar content of the extract tended to be decreased until $170^{\circ}C$ and then be increased from $190^{\circ}C$. Content of phenolic compound of the extract was increased at the masting temperature of above $210^{\circ}C$. At the sensory evaluation of the extract, aroma and taste of samples masted at $170^{\circ}C$ and above $190^{\circ}C$, respectively were higher than those masted at the others. Overall acceptability of the extract was highest at $190^{\circ}C$.

  • The ultimate research goal of the current study was a development of hydrolyzed whey protein powder (7%-GNANA) manufactured with normal content of sialic acid, a marker compound, that is naturally occurring at 7% concentration in GMP obtained from the milk protein. GMP is a safe food, used worldwide in infant and baby foods, etc. The test substance was prepared using (7% sialic acid containing) GMP as a raw material, and then using alcalase, an enzyme approved as a food additive, after separation of sialic acid with 100% efficiency and 7%-GNANA (containing 7% sialic acid and protein; product name: HELICOBACTROL-7) provided by MEDINUTROL Inc. (Korea). Bacterial reverse mutation (Ames) test was conducted in accordance with GLP Guideline using the test substance specified above. To identify its mutagenic potential against microorganisms, histidine auxotrophic strains of Salmonella Typhimurium, TA98, TA100, TA1535, and TA1537, and tryptophan auxotrophic strain of Escherichia coli, WP2uvrA, were used. The bacterial reverse mutation (Ames) test was performed by dividing the test substances into five different concentration groups (0, 61.7, 185, 556, 1,670, $5,000{\mu}g/plate$). Results of this experiment did not reveal repetitive increase of colony generating values or positive criteria for reverse mutagenicity for any concentration of test substances in any of the five strains, regardless of the presence of a metabolic activation system, and no dose-dependency was identified. In conclusion, the safety of 7%-GNANA test substance was verified by bacterial reverse mutation test conducted before registration of 7%-GNANA as a food additive.

Effect of Heat Treatment on the Antioxidant Properties of Yacon (Smallanthus sonchifolius) (열처리에 따른 야콘의 항산화 활성 변화)

  • This study was investigated the effects of heat treatment on polyphenolic compounds and antioxidant activities of yacon. Raw yacon was heated at $100^{\circ}C$ and $121^{\circ}C$ for 15, 30, or 60 min by using an autoclave. The browning intensity, levels of free and bound phenolic and flavonoid compounds, and 1,1-diphenyl-2-picrylhydrazyl(DPPH) and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS) radical-scavenging activity of yacon extracts following heat treatment were measured. The browning index, free polyphenolic content, and antioxidant activities in the extracts were significantly increased (p<0.05) with both increased heating temperature and time, while bound polyphenolic levels were decreased. The levels of free phenolic and flavonoid compounds in the yacon extract heated to $121^{\circ}C$ for 60 min were increased by 1.2 and 1.1 folds compared to raw yacon respectively. Moreover, DPPH and ABTS radical-scavenging activities of the yacon heated to $121^{\circ}C$ for 60 min were 1.7 fold and 2.0 fold higher, respectively, than those of raw yacon. A significant (p<0.01) correlation was observed among browning intensity, free and bound phenolic and flavonoid levels, and DPPH and ABTS radical-scavenging activities in heated yacon extract. Thus, heat treatment can be used as a method to enhance the antioxidant compound content in and the antioxidant activity of yacon.

  • It has recently been considered urgent to establish an accurate definition and scope of functional foods in the livestock industry in Korea. The tertiary function of food is gradually emphasized with various changes in conditions such as international competitive power enhancement of the whole world, including Korea. Therefore, the purpose of this study was carried out to gather basic information toward the establishment of standardization, management plans, and a system adapted to the Korean state through researching management systems of functional meat and meat products in the European Union (EU) and the Oceania Area (OA). A brief summary of this study is as follows. In the EU and OA, special management systems and laws about functional livestock and products were once nonexistent; only 'nutrition claims' and 'health claims' operated in the nutritional consolidation side of foods. Also, it was once thought that functional index materials and permissions established in the application of functional products sufficed, because management standards for enriched food were not established. Therefore, standardization of functional live stock products needs to be considered based on the case of developed countries in the EU and OA. It also seems that the above functional indicative substances can be applied, based on normal standards of indicative substances of functional products and the normal standards of indicators of functional substances added to livestock products presented in this study.
