각막상피의 창상치유에 중요한 역할을 하는 것으로 알려진 Nerve Growth Factor(NGF)의 내인성 변화와 국소적 투여효과를 알아보았다. 6마리의 Beagle 을 사용하여 비교군인 오른쪽 눈에만 6mm의 기계적인 각막 창상을 만들었고 왼쪽 눈은 대조군으로 사용하였다. 눈물은 수술후 1주일동안 매일, 각막 상피는 수술중 그리고 수술후 7일에 각각 채취하여 NGF 농도를 enzyme-linked immunosorbent assay (ELISA)를 사용하여 측정하였다. 2차 실험으로, 16마리의 Bealgle을 사용하여 오른쪽 눈에 위와 동일한 창상을 만든 후, 창상 부위가 회복될 때까지 recombinant human (rh) NGF (n=4), murine NGF (n=6) 그리고 anti-NGF blocking antibody (n=6)를 6시간마다 점안하였으며, 왼쪽 눈은 bovine serum albumin (BSA)를 점안하여 대조군으로 사용하였다. 창상 면적은 NIH image software를 이용하여 분석하였다. 대조군에 비하여, 비교군의 눈물내 NGF는 치유초기에 현저하게 증가하였으며, 각막 상피의 NGF도 유의적인 증가를 보였다. 그러나 rh NGF, murine NGF, anti-NGF blocking antibody처치군과 BSA처치군 간에는 각막창상 치유에 유의적인 차이가 인정되지 않았다. 각막창상후, rh NGF 혹은 murine NGF의 추가적인 국소점안은 정상개의 각막상피 치유에 효과가 없었으나, 창상후 초기 치유기동안 눈물과 손상받은 각막상피에서 내인성 NGF의 급격한 보상성 증가로 미루어 NGF는 각막 상피의 창상치유에 중요한 역할을 하는 것으로 사료된다.
Anaplasmosis 은 흡혈성 절지동물인 진드기, 이파리, 모기 등에 의해 매개되는 리케차성 인수공통 전염병이다. 철새는 anaplasmosis의 매개체인 진드기의 숙주이다. 제주도의 다양한 철새 분류군 중에서 지빠귀과 조류의 진드기 감염률이 높다. 특히 마라도는 봄철 남방구에서 북방구로 이동하는 철새의 중간기착점으로 지리적으로 중요한 위치에 있다. 따라서 본 연구에서는 제주도의 대표적 이동철새인 지빠귀과 새들의 Anaplasma spp. 감염여부를 조사하였다. 우리는 마라도에서 34마리의 혈액과 제주야생동물구조센터에서 구조 채취된 6개의 혈액 시료를 대상으로 하였다. 그 결과, 40개체의 지빠귀 중 7개체가 감염이 확인되었으며, 감염률은 17.5%로 나타났다. 7개체 모두 Anaplasma phagocytophilum으로 동정되었다. 이러한 결과는 제주도를 통과하는 대표적인 철새인 지빠귀과 새들이 A. phagocytophilum을 육지로 전파할 수 있음을 시사하며, 다른 이동성 철새들간의 질병전파의 보균자로 작용할 수 있기에 철새, 텃새 및 가축으로의 전파여부를 지속적으로 모니터링할 필요가 있다.
Purpose: Thoracic aortic injury is a life-threatening injury that has been traditionally treated by using surgical management. Recently, thoracic endovascular aortic repair (TEVAR) has been conducted pervasively as a better alternative treatment method. Therefore, this study will focus on analyzing the outcome of TEVAR in patients suffering from a blunt thoracic aortic injury. Methods: Of the blunt thoracic aortic injury patients admitted to Eulji University Hospital, this research focused on the 11 patients who had received TEVAR during the period from January 2008 to April 2014. Results: Seven of the 11 patients were male. At the time of admission, the mean systolic pressure was $105.64{\pm}24.60mm\;Hg$, and the mean heart rate was $103.64{\pm}20.02per$ minute. The median interval from arrival to repair was 7 (4, 47) hours. The mean stay in the ICU was $21.82{\pm}16.37hours$. In three patients, a chimney graft technique was also performed to save the left subclavian artery. In one patient, a debranching of the aortic arch vessels was performed. In two patients, the left subclavian artery was totally covered. In one patient whose proximal aortic neck length was insufficient, the landing zone was extended by using a prophylactic left subclavian artery to left common carotid artery bypass before TEVAR. There were no operative mortalities, but a patient who was covered of left subclavian artery died from ischemic brain injury. Complications such as migration, endovascular leakage, collapse, infection and thrombus did not occur. Conclusion: Our short-term outcomes of TEVAR for blunt thoracic aorta injury was feasible. Left subclavian artery may be sacrificed if the proximal landing zone is short, but several methods to continue the perfusion should be considered.
This study was conducted to investigate the optimal conditions of collagen extraction from scales of yellowfin tuna (Thunnus albacares) using surface response methodology. Four independent variables of NaOH concentration and pretreatment fime in alkali pretreatment and enzyme concentration and treatment time in enzyme hydrolysis were used to predict a model equation for the collagen yield. The determinant coefficient ($R^2$) for the equation was 0.906. The values of the independent variables for the maximum yield were 0.32 N NaOH, 16.38 h alkali pretreatment time, 0.18% enzyme concentration, and 31.02 h enzyme treatment time. In the physicochemical properties of tuna scale collagen, sodium dodecyl sulfate-polyacrylamide gel electrophoresis of tuna scale collagen showed the same migration distances as that of calf skin collagen. The amide A, I, II, and III regions of tuna scale collagen in Fourier transform infrared measurements were shown in the peaks of 3,414 $cm^{-1}$, 1,645 $cm^{-1}$, 1,553 $cm^{-1}$, and 1,247 $cm^{-1}$, respectively. The amount of imino acids in tuna scale collagen was 18.97% and the collagen denaturation temperature was $33^{\circ}C$. The collagen solubility as a function of NaCl concentration decreased to 4% NaCl (w/v) and the collagen solubility as a function of pH was high at pH 2-4 and sharply decreased from pH 4 to pH 7. Viscosity of the collagen solution decreased continuously until $30^{\circ}C$ and this decreasing rate slowed in the temperature range of $35-50^{\circ}C$.
Manganese superoxide dismutase (MnSOD) is a vital enzyme that protects cells from free radicals through eliminating superoxide radicals ($O^{2-}$). Hirudin, a kind of small active peptide molecule, is one of the strongest anticoagulants that can effectively cure thrombus diseases. In this study, we fused Hirudin to the C terminus of human MnSOD with the GGGGS linker to generate a novel dual-feature fusion protein, denoted as hMnSOD-Hirudin. The hMnSOD-Hirudin gene fragment was cloned into the pET15b (SmaI, CIAP) vector, forming a recombinant pET15b-hMnSOD-Hirudin plasmid, and then was transferred into Escherichia coli strain Rosetta-gami for expression. SDS-PAGE was used to detect the fusion protein, which was expected to be about 30 kDa upon IPTG induction. Furthermore, the hMnSOD-Hirudin protein was heavily detected as a soluble form in the supernatant. The purification rate observed after Ni NTA affinity chromatography was above 95%. The hMnSOD-Hirudin protein yield reached 67.25 mg per liter of bacterial culture. The identity of the purified protein was confirmed by western blotting. The hMnSOD-Hirudin protein activity assay evinced that the antioxidation activity of the hMnSOD-Hirudin protein obtained was $2,444.0{\pm}96.0U/mg$, and the anticoagulant activity of the hMnSOD-Hirudin protein was $599.0{\pm}35.0ATU/mg$. In addition, in vitro bioactivity assay showed that the hMnSOD-Hirudin protein had no or little cytotoxicity in H9c2, HK-2, and H9 (human $CD_4{^+}$, T cell) cell lines. Transwell migration assay and invasion assay showed that the hMnSOD-Hirudin protein could suppress human lung cancer 95-D cell metastasis and invasion in vitro.
연구배경 : 양성 기도협착질환 환자에서 새로 개발된 Natural 기도스텐트의 임상결과를 Dumon 스텐트와 비교하였다. 방 법 : 양성 기도협착으로 스텐트를 삽입한 94명의 환자 (39 Dumon, 55 Natural)를 대상으로 임상증상, 시술 전 검사결과, 시술방법, 그리고 추적관찰시의 임상경과를 후향적으로 조사하였다. 결 과: 원인질환으로 기관지결핵 후 기도협착 (74%)이 가장 많았고, 기관삽관 후 기관협착 (20명), 기관절개술 후 기관협착 (2명)등이 있었다. 스텐트를 삽입한 후, 전체 94명의 환자 중 82명 (87%)에서 주관적으로 호흡곤란이개선되었고, Dumon 스텐트군(35/39, 90%)과 Natural 스텐트군 (47/55, 86%) 사이의 차이는 없었다. 모두 48명 (51%)에서 중앙값 14개월(범위6-37개월)후에 스텐트를 성공적으로 제거할 수 있었고, Dumon 스텐트군 (54%, 16개월 후) 과 Natural 스텐트군 (49%, 12개월 후)간의 차이는 없었다. 스텐트 삽입 후 부작용은 스텐트의 위치 변동이 45명 (48%)에서, 육아종 과형성이 41명 (44%)에서, 점액저류에 의한 스텐트 내경 50% 이상의 감소가 17명 (18%)에서, 그리고 스텐트 삽입 전후부위의 협착이 40명 (43%)에서 발생하였다. Dumon 과 Natural 스텐트를 삽입한 환자들 사이에서 부작용의 분포의 차이는 없었다. 결 론 : 새로 개발한 Natural 기도스텐트는 양성기도협착 환자에서 기존의 Dumon 스텐트와 대등한 임상 결과를 보여주고 있어, 기도 협착이 있는 환자의 치료에 유용하게 사용할 수 있을 것으로 사료된다.
향후 도래할 수소경제에서 가장 유망한 기술 중에 하나인 고체수소저장 시스템들의 전체성능은 고체수소화물 내부의 열 및 물질전달 속도에 크게 영향을 받으며, 최적화된 시스템 설계를 위해서 이들에 대한 연구들이 선행되어야 한다. 본 연구에서는 Pt-CNTs 수소저장물질을 이용한 수소저장시스템에 대한 모델링 및 2차원 비정상상태 전산해석을 수행하였다. 기존 상용화된 CFD 소프트웨어를 이용하여 충전동안 발생하는 열 및 물질전달에 대한 현상들을 연구하였으며, 최적화된 수소저장시스템 설계는 고압에서 대류에 의한 냉각효과를 최대화하여 시스템 내부의 온도 상승과 충전시간 지연을 개선할 수 있음을 밝혀냈다. 아직까지 CNT 기반의 수소저장시스템에 대한 연구들이 보고되고 있지 않은 상황에서, 본 연구는 향후 CNT 기반의 고체수소저장시스템 최적 설계에 대한 방안들을 제시한다.

Objective: To investigate the long-term outcomes of percutaneous treatment of benign biliary strictures using temporary placement of a retrievable expanded polytetrafluoroethylene (PTFE) covered stent. Materials and Methods: We retrospectively analyzed the outcomes of 148 patients (84 male and 64 female; age range, 11-92 years) who underwent percutaneous transhepatic placement and removal of a retrievable PTFE-covered stent for the treatment of benign biliary strictures between March 2007 and August 2019 through long-term follow-up. Ninety-two patients had treatment-naïve strictures and 56 had recurrent/refractory strictures. Results: Stent placement was technically successful in all 148 patients. The mean indwelling period of the stent was 2.4 months (median period, 2.3 months; range, 0.2-7.7 months). Stent migration, either early or late, occurred in 28 (18.9%) patients. Clinical success, defined as resolution of stricture after completing stent placement and removal, was achieved in 94.2% (131 of 139 patients). The overall complication rate was 15.5% (23 of 148 patients). During the mean follow-up of 60.2 months (median period, 52.7 months; range, 1.6-146.1 months), 37 patients had a recurrence of clinically significant strictures at 0.5-124.5 months after removal of biliary stent and catheter (median, 16.1 months). The primary patency rates at 1, 3, 5, 7, and 10 years after removal of biliary stent and catheter were 88.2%, 70.0%, 66.2%, 60.5%, and 54.5%, respectively. In the multivariable Cox proportional hazard regression analysis, sex, age, underlying disease, relation to surgery, stricture type, biliary stones, history of previous treatment, and stricture site were not significantly associated with the primary patency. Conclusion: Long-term outcomes suggest that percutaneous treatment of benign biliary strictures using temporary placement of retrievable PTFE-covered stents may be a clinically effective method.
본 논문은 이동 컴퓨터 통신 환경에서 IP(Internet Protocol) 데이터그램을 전송하기 위한 가상 셀 시스템(virtual cell system0의 성능 분석을 다룬다. 하나의 가상 셀은 이웃한 다수의 물리적 셀(physical cell)들의 집합으로서, 원격브리지(remote bridge)로 구현된 기지국(base station)들을 멀티캐스트 기능을 갖는 고속 데이터그램 패킷 데이터망으로 연결하여 구성된다. 가상 셀 시스템에서의 호스트 이동성은 기지국들 사이에 분산되어 있는 계층적 위치정보를 기반으로 동작하는 데이터링크 계층의 가상 셀 프로토콜(Virtual Cell Protocol )을 통하여 지원된다〔1〕. 이러한 가상 셀 시스템은 물리적 셀들 사이에 임의의 호스트 이동성 패턴과 데이터전송 패턴이 주어진 경우에 전체 시스템의 통신 비용을 최소화할 수 있도록 논리적으로 유연한 가상 셀 시스템의 구축을 가능하게 한다〔2〕. 본 논문에서는 가사 dtpf 시스템의 성능 모델로서 BCMP 개방 복합 클래스 대기 행렬 네트워크(BCMP open multiple class queueing network)를 채택하고, 물리적 셀들 사이의 호스트 이동성 패턴과 데이터전송 패턴에 대하여 임의의 토폴지와 최적화된 토폴로지로 구축된 가상 셀 시스템의 성능을 비교 분석한다. 특히 이동 호스트 수, 이동 속도, 그리고 데이터전송 양과 같은 다양한 시스템 파라메타를 변화시키면서 이에 따라 생성되는 데이터 메시지, 핸드오프(handoff) 메시지, 그리고 주소 용해(address resolution) 메시지 각각에 대하여 망 구성요소의 이용도(utilization)와 시스템 처리 시간(system response time)을 비교 분석한다.
