Pepper anthracnose caused by Colletotrichum species is one of the most important limiting factors for pepper production in Korea, its management being strongly dependent on chemicals. The aim of this work was to evaluate the possibilities of using silver nanoparticles instead of commercial fungicides. In this study, we evaluated the effect of silver nanoparticles against pepper anthracnose under different culture conditions. Silver nanoparticles (WA-PR-WB13R) were applied at various concentrations to determine antifungal activities in vitro and in the field. The application of 100 ppm concentration of silver nanoparticles produced maximum inhibition of the growth of fungal hyphae as well as conidial germination in comparison to the control in vitro. In field trials, the inhibition of fungi was significantly high when silver nanoparticles were applied before disease outbreak on the plants. Scanning electron microscopy results indicated that the silver nanoparticles caused a detrimental effect on mycelial growth of Colletotrichum species.
We have investigated the in vitro cytotoxic effect of liposome-encapsulated N-(phospho-nacetyl)-L-aspartic acid (PALA) against C-26 murine colon cancer cells, and have compared it in this regard to free PALA. Three different PALA-containing liposomal formulations using distearoylphosphatidylcholine (DSPC), distearoylphosphatidylglycerol (DSPG), and polyethyle-neglycol-derivatized distearoylphosphatidylethanolamine (PEG-DSPE) were made and their cytotoxicity was measured. In 72 hr continuous exposure experiment with C-26 cells, the 50% growth inhibitory concentration ($IC_50$) of DSPG-PALA liposome formulation was $0.09\mu\$, which showed about 65-fold more potent than unencapsulated free PALA ($5.1\mu\$). Similar degree of increase in cytotoxicity was also observed in 1 hr exposure experiment. However the $IC_50$ of PEG-DSPE-PALA liposome and DSPC-PALA liposome were $10.7\mu\$and $11.8\mu\$respectively, which showed slightly less potent than unencapsulated free PALA. Physical characteristics of PALA-liposomes, such as the size and drug:lipid ratio were also determined. In conclusion, negatively-charged DSPG-PALA liposome showed the highest cytotoxic effect among tested on the C-26 cells in vitro.
Objectives: The purpose of this study was to investigate the antithrombotic effect of Carthamus tinctorius. Methods: SD rats were used to investigate the inhibitive effect of platelet aggregation (in vitro & in vivo), prothrombin time, platelet count. Mice were used to investigate the survival rate after injection of collagen & epinepbrine. Results: 1. The inhibitive effect of platelet aggregation (in vitro & in vivo) were increased significantly in all groups. 2. Prothrombin time was decreased in both groups. 3. Platelet count was decreased significantly in sample A. 4. Survival rate after injection of collagen & epinepbrine was increased in sample B. Conclusion: According to the above results, it is suggested that Carthamus tinctorius has antithrombotic effects.
The effect of ginseng extract and sodium ascorbate (vitamin C) administered separately or in combination on the some cancer cells cultured in vitro have been examined. Mouse leukemic cells (L1210 and P388), human rectal cancer cells (HRT-18) and human colon cancer cells (HCT-48) were used for the experiment. When given separately, the growth rate for each kind of cancer cell was inhibited In proportion to the concentration of ginseng extract or vitamin C. The inhibitory effect on the growth rate of the cancer cells was stronger in ginseng extract than in vitamin C except for the HCT-48 cells. Based on the cytotoxic activity, combined administration of ginseng extract and vitamin C demonstrated a synergistic inhibition of cancer cell growth. The cytotoxic activities of ginseng extract and vitamin C on the mouse leukemic cells were more sensitive than on human colon cancer cells. And the sensitivity of cytotoxic activity was somewhat different in different cancer cell lines.
The development of isolated blastomeres from mammalian preimplantation embryos has been basically studied for the multiplication of embryos from superior animals. Therefore, this study was investigated the effect of co-culture with mouse fetal fibroblast cells(MFFC) on in vitro development of blastomeres from mouse preimplantation embryos. Mature female ICR mice were treated with hormone to induce superovulation and embryos were collected at each 2, 4, and 8-cell stage. Then, after removing zona pellucida with protease, blastomeres were isolated by micropipetting, or reconstituted with different stage blastomere, and incubated for 72 hrs either in T6 or TCM199 or on the monolayer of MFFC, which was prepared with fibroblast cells from 14∼14 day mouse fetus. After incubation, we examined their development rates every day and the nuclei numbers of each blastocyst by Hoechst-33342 staining. In the development rates of blastomeres, there were no significant differences between media but the higher rateswere found in the monolayer of MFFC, regardless of reconsititution. In addition, blastomeres cultured with MFFC had slightly greater number of nuclei than those cultured in single media. Generally, the higher development rates of blastomeres were found from earlier stage embryos than the later ones, regardless of culture conditions. Reconsitituted blastomeres had more nuclei but did not show the higher development rates, compared to the single blastomeres. Taken together, our results suggest that co-culture with MFFC have a beneficial effect on the in vitro development of blastomeres from mouse embryos.
Objective : Paeoniae radix alba(PRA) can enrich the blood and regulate menstruation, astringe yin and arrest sweating, calm the liver and arrest pain. This study was designed to investigate effects of PRA on skin whitening and elasticity using melanoma cells. Methods : In this experiment, effect of PRA on cell viability, inhibition of melanin synthesis and inhibitory effect on tyrosinase and elastase. Results : 1. More than $1,000\;{\mu}g/ml$ of PRA treated group showed lowered proliferation rates significantly compared to non-treated control group. 2. All of treated groups were lower levels of melanin synthesis respectively. 3. PRA did not show inhibitory effect on tyrosinase activities in vitro. But, PRA suppressed tyrosinase activities in B16F10 cells significantly. 4. PRA suppressed elastse type 1 activities in dose-dependent manner in vitro. But, PRA slightly suppressed elastase type 4 activities in vitro, and PRA also slightly suppressed elastase activities in vivo. Conclusion : These results suggest that PRA can inhibit melanin synthesis through ihhibitory action on tyrosinase activity and inhibt elastase activity, and also suggest that these results can be used for the study on maintaining skin whitening or elasticity.
GRF (Korean Red Ginseng mixed formula) consists of six herbs such as Ginseng Radix rubra Koreana, Lycii Fructus, Artemisiae Capillaris Herba, Poria, and Glycyrrhizae Radix and Hoveniae Fructus. For the evaluation of hepatoprotective effect of GRF, the study was performed on protective effect against hepatic damage induced by galactosamine in vitro and ccl4 in vivo and also elucidate antioxidant activity. In vitro assay with 1.1 mM galactosamine, protection (%) was 44% (GR), and 58% (GRF-A) at 50 ug/ml. GRF effectively protected fatty degenertion and necrosis in murine hepatic damage induced by ccl4. For the -antioxidant study, GRF inhibited hemolysis of erythrocyte and decolored DPPH (2,2-diphenyl-l-picrylhydrazyl) free radical in a dose dependent manner more effetively than GR alone in vitro. GRF and GR significantly suppressed the time course $(1\;hr{\sim}6\;hr)-level$ of MDA (malondialdehyde) following AAPH (2,2'-azo-bis-(2-amidino -propane) dihydrochloride) treatment in vivo as compared with control data. From the results it can be concluded GR and GRF exerted the hepatoprotective effect by dint of antioxidant activity.
Effect of grape seed extract (GSE) against Candida albicans was examined under in-vitro and in-vivo conditions. The GSE was extracted in ethanol. In-vitro results from an agar diffusion susceptibility assay showed the GSE inhibited C. albicans growth. This anticandidal effect was at dose-dependency. In experiments with animals, mice that received the GSE (0.5 mg per mice), intravenously (i.v.), before i.v.-infection wish viable C. albicans yeast cells survived longer than diluent (buffer)-received control mice. In contrast, when GSE was given to mice after the mice were infected with the yeast cells, these mice showed a similar survival rate as compared to control mice that received no treatment with the GSE. Taken together, these data indicate that GSE has prophylactic effect but not therapeutic effect against disseminated candidiasis.
This study was conducted to investigate the effect of energy source on development of in vitro development of in vitro matured and fertilized porcine 2-cell embryos. The relative preferences of glucose, lactate and pyruvate for in vitro development of porcine 2-cell embryos were determined. The results obtained are as follows. 1. 33.3, 20.8 and 29.2% of porcine embryos reached morula stage in addition to lactate, glucose, and both glucose and lactate in the culture medium as energy source, respectively. 2. 38.5, 15.4 and 26.9% of porcine embryos reached morula stage in addition to pyruvate, glucose, and both glucose and pyruvate in culture medium as energy source, respectively. 3. 42.9, 21.4 and 28.6% of porcine embryos reached morula stage in addition to pyruvate and lactate, glucse alone, and glucose, lactate and pyruvate in culture medium as energy source, respectively.
This study evaluated SPF (Sun Protection Factor) of various quntity and kind of sunscreen ingredients which is used in O/W and W/O emulsion by systematic and quantitative approach. Octylmethoxycinnamate(OMC), Buthylmethoxydibenzoylmethan (BMDM), Octocrylene (OC), Octylsalicylate(OS), Octyl Triazone (OT), Titanium dioxide (TD) are used for the experiment. As a result, when different chemical sunscreen ingredients are added to OMC, the synergy effect of SPF was high in order of BMDM, OC, OS, OT. There was no significant difference in O/W and W/O emulsions. It can be a guide to use sunscreen ingredients effectively when the relation between the results of in vitro SPF and in vivo SPF is comprehended.
