The edible medicinal piano with antitumor activity used in Korea

  • The present study has been undertaken to detect edible medicinal plants with antineoplastic property on the basic of a number of traditional Korean medical literatures, besides studies on development of anti-cancer medical wild plants growing in Korea and to prove experimentally their efficacy by in vitro and in vivo tests.235 species from 45 family 79 genus were screened primarily as edible sources of antitumor effect. Among those the crude. extracts of 40 spp. showed considerable cytotoxicity in vitro and especially Pegangkuen(Patrinia scabiosaefolia), Deod-eog(Codonopsis lanceolata), Okssusu(Zea may), and Geureong(Eragrositis ferru-ginea) exhibited significant antitumor activity against sarcoma 180 asites mice. However, additional researches should be mode for the confirmation of their availability as antitumor plants.

  • The activity-guided fractionation of some medicinal plants led to yield five kinds of natural stilbene compounds namely 3, 5-dihydroxy-4'-methoxystillbene(I), rhapontigenin(II), reveratrol(III), rhaponticin(IV) and piceid(V) and two common flavonoids, apigenin(VI) and luteolin(VII) as active principles of the antitumor property, in vitro, against five kinds of human tumor cell lines, A-549, SK-OV-3, SK-MEL-2, XF-498 and HCT15.

In Vitro Antitumor Properties of an Isolate from Leaves of Cassia alata L

  • Leaf extracts of Cassia alata L (akapulko), traditionally used for treatment of a variety of diseases, were evaluated for their potential antitumor properties in vitro. MTT assays were used to examine the cytotoxic effects of crude extracts on five human cancer cell lines, namely MCF-7, derived from a breast carcinoma, SK-BR-3, another breast carcinoma, T24 a bladder carcinoma, Col 2, a colorectal carcinoma, and A549, a nonsmall cell lung adenocarcinoma. Hexane extracts showed remarkable cytotoxicity against MCF-7, T24, and Col 2 in a dose-dependent manner. This observation was confirmed by morphological investigation using light microscopy. Further bioassay-directed fractionation of the cytotoxic extract led to the isolation of a TLC-pure isolate labeled as f6l. Isolate f6l was further evaluated using MTT assay and morphological and biochemical investigations, which likewise showed selectivity to MCF-7, T24, and Col 2 cells with $IC_{50}$ values of 16, 17, and 17 ${\mu}g/ml$, respectively. Isolate f6l, however, showed no cytotoxicity towards the non-cancer Chinese hamster ovarian cell line (CHO-AA8). Cytochemical investigation using DAPI staining and biochemical investigation using terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL)-a method used to detect DNA fragmentation-together with caspase assay, demonstrated apoptotic cell death. Spectral characterization of isolate f6l revealed that it contained polyunsaturated fatty acid esters. Considering the cytotoxicity profile and its mode of action, f6l might represent a new promising compound with potential for development as an anticancer drug with low or no toxicity to non-cancer cells used in this study.

  • A new series of methotrexate analogues was prepared in which the ${\beta}-position$ of the glutamic acid moiety is substituted by the aryl groups. The glutamic acid moiety was modified in order to enhance the lipophilic property. Reaction of N-acetylglicine ester[1] with ethyl 3-arylacrylate derivatives[2] produced trans-3-aryl-2-carboxy-5-pyrrolidone derivatives[3], which were hydrolyzed to give ${\beta}-aryl-glutamic$ acid derivatives[4]. The compounds[4] were treated with the p-aminobenzoic acid moiety and then with the pteridine ring moiety to give ${\beta}-arylmethotrexate$ derivatives[6,7]. These compounds were tested for antibacterial activity against Streptococcus faecium and for antitumor activity against murine leukemias and against human tumor cell lines in vitro. Several compounds showed significant antibacterial activity.

  • Thirteen kinds of naturally occurring or derivatised thiterpenes, reported to have an antitumoral property, were reinvestigated on the basis of their direct cytotoxicity or the inhibitory activity on cell growth against five kinds of cultured human tumor cells, i.e., A-549, SK-OV-3, SK-MEL-2, XF-498 and HCT15, in vitro. Ursonic acid III, betulinic acid VIII, betulonic acid X and glycrrhetinic acid XI were exhibitied a marked inhibition on cell growth.

  • As part of a research program to develope 3rd generation anti tumor platinum complexes, a series of platinum complexes which have 4, 5-bis-(aminomethyl)- 1, 3-dioxolane derivatives as bidenate amine ligands, represented by the general structual formula was prepared. The R$_1$ and/or R$_2$ substituents in this series of platinum complexes can be hydrogen. alkyl, of jointly formed cyclohexane. Two Xs can be a bidenate leaving ligand such as 1, 1-cyclobutanedicarboxylate, malonate, dimethylmalonate, ethylmalonate, glycolate, L-lactate, or N-methyliminodiacetate. From based on the pharmacological and toxicological studies, we have chosen SKI 2053R, cis-malonato[(4R, 5R)-4, 5-bis(aminomethyl)-2-isopropyl-1, 3-dioxolane] platinum(II) complex (NSC D644591) as a candidate for clinical evaluation. The antitumor activity of a new anti tumor platinum complex, cis-malonato [(4R, 5R)-4, 5-bis(aminomethyl)-2-isopropyl-1, 3-dioxolane] platinum(II) (SKI 2053R, NSC D644591), was compared with those of cisplatin and carboplatin using murine tumors. We evaluated three platinum complexes against L1210/CPR, a subline of L1210 leukemia resistant to cisplatin for their abilities to overcome tumor resistance to cisplatin. The in vitro cytotoxicity of SKI 2053R to L1210 cell line was 2.5-fold less potent thann that of cisplatin, and was 10-fold more cytotoxic than that of carboplatin. SKI 2053R retained similar cytotoxic effect and anti tumor activity to L1210/CPR cell line, like the cytotoxicity of SKI 2053R to L1210 cell line, while either cisplatin or carboplatin had not property to overcome the acquired cisplatin-resistance. SKI 2053R exhibited greater or comparable antitumor activity than cisplatin or carboplatin in murine tumor models.

  • All fifteen flavonoids (1-15) have been isolated from the roots of Sophora flavescens (Leguminosae) as active principles of the cytotoxic property toward human tumor cell lines such as A549, SK-OV-3, SK-Mel-2, XF498 and HCT15, in vitro. By means of spectral analyses, particularlyby the aid of various two dimensional NMR experiments, all $^1H-NMR$ ad $^{13}C$ -NMR signals of 1-15 were completely assigned, and thus the structures of 1-15 were established unambiguously.

  • The activity-oriented fractionation of Psoralea corylifolia led to an isolation of a (+) bakuchio 1 as an activ eprinciple of its antitumoral property in vitro 1 was observed to exhibit a mild cytotoxicity against five kinds of cultured human cancer cell lines, i. e. the A549, SK-OV-3, SK-MEL-2, XF-498 and HCT15. The synthesized 2, 3-epoxide of (+)-bakuchiol 3 showed the similar activity as the (+) bakuchiol 1, whereas the other oxidation derivatives 4 and 5 including the acetyl (+) bakuchiol 2 showed a decreased activity.

한국산 겨우살이 전초의 Methanol 추출물로부터 암세포증식 저해성분의 분리 (Active Principles of the Methanol Extract of Korean Mistletoe Responsible for the Inhibitory Effect on the Proliferation of Human Tumor Cell Lines)

  • A bioassay-guided fractionation of the whole extract of Viscum album var. coloratum Ohwi (Loranthaceae) led to the isolation of two triterpenoidal components; oleanolic acid (1) and ${\beta}-amyrin$ acetate (2), and a flavonoid, homoflavoyadorinin B (3) as well as large quantity of free fatty acid mixtures as active ingredients of the extract responsible for the antitumoral property. The EtOAc soluble fraction and BuOH soluble fraction of the extract demonstrated a significant inhibition on the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system), and HCT-15 (colon) in vitro, whereas the remaining water soluble fraction exhibited a poor inhibition. The intensive phytochemical investigation of the EtOAc soluble fraction and BuOH soluble fraction of the extract indicated that the oleanolic acid (1) and large amounts of free fatty acid mixtures might be attributed to the in vitro antitumoral property of the whole extract of Viscum album var. coloratum.

Anti-tumor Substance from Panax Kinsenk Roots

  • Antitumor polyacetylenlc alcohol, panaxytriol (Ci7 H2603), was isolated and purified from a Powder of the root of Panax ginseng C.A. Meyer. Panaxytriol Possesses unusual Property of being soluble in both water and organic solvents. Panaxytriol inhibited the growth of various kinds of human cultured cell lines in dose-dependent fashion in vitro. The in vivo effects of panaxytriol were tested against C57BU6 mice transplanted with Bl6 melanomas. Panaxytriol (8 and 40 mg/kg) administered intramuscularly (im) produced significant tumor growth delays in mice. Although a detailed mechanism of growth inhibition by panaxytriol is unknown, preliminary results appear to implicate a surface membrane site of action. And its action seems to be more dose-dependent than time-dependent. Finally, panaxytriol pharmacokinetics was evaluated in mice given single 8 mgrkg doses intraperitoneally(ip) or im. Serum panaxytriol content was measured using both tumor growth inhibitory assay and a gas chromatographic method. The maximum serum panaxytriol content after ip and im administration was 35.0 and 1.61 ug/ml respectively. These results indicate that the compound may act as cytotoxic substance even in-patients.
