  • The study was designed to investigate the effects of progesterone and estrogen on the uterus of rats by immunohistochemical methods using Proliferating Cell Nuclear Antigen (PCNA) antibody. Eighteen female rats(Wistar), weighing initially about 300g, were ovariectomized. These rats were divided into four groups, progesterone-treated group, estrogen-treated group, estrogen+progesterone-treated group, and control group, progesterone-treated group was injected with 1mg of progesterone per rat per day for 2 days and estrogen-treated group with $20{\mu}g$ of $17{\beta}-estradiol$ for 3 days and estrogen+progesterone-treated group with $17{\beta}-estrdiol$ for 3 days and then with progesterone for 2 days as above. In gross findings, the uteri were markedly hypertrophied by estrogen treatment but were not affect in size by progesterone treatment. Immunohistochemical investigation was performed on the cell types with higher appearance of PCNA positive reaction cells in four groups. The groups with higher appearance of the stromal cells were ordered as estrogen-treated group, progesterone-treated group, estrogen+progesterone-treated group, and control group. The muscle cells were ordered as progesterone-treated group, estrogen-treated group, estrogen+progesterone-treated group, and control group. Positive reaction cells of the stromal cells were total 4.6 times higher than those of muscle cells. Therefore, the affect of the hypertrophy on the uterus by estrogen was larger than those of progesterone and affect on the uterus by stromal cells were larger than those of muscle cells. The group with more PCNA positive reaction cells of luminal epithelial cells were ordered as control group, progesterone-treated group, estrogen+progesterone-treated group, and estrogen-treated group, and glandular epithelial cells were ordered as estrogen+progesterone-treated group, progesterone-treated group, control group, and estrogen-treated group. It was suggested that estrogen and progesterone did not affect on the proliferating cells of luminal epithelial cells and affection of progesterone on the development of glandular epithelial cell was larger than that of estrogen.

  • Diagnosing Hirschprung's disease is one of the clinical challenges of this disorder. In the stomach and the intestines, Cathepsin D was readily detected in cytoplasm of the rat gastric and in intestinal ganglion cells of the autonomic nervous system. The objectives of the present study were to examine cathepsin D expression in ganglion cells of the submucosal and myenteric plexuses of the intestine of children and to determine the utility of immunohistochemical staining of cathepsin D for detection of immature ganglion cells. Paraffin blocks of 35 intestinal segments were reviewed for immunohistochemical staining with polyclonal antibody to cathepsin D and hematoxylineosin stainings from the compatible specimens. There were 9 aganglionic segments and 9 ganglionic segments of neonates with Hirschsprung's disease, 8 intestinal segments with non-Hirschsprung's disease in neonates and 9 intestinal segments with non-Hirschsprung's disease infants over the age of 10 months. All ganglion cells showed intense granular cytoplasmic reactivity for cathepsin D regardless of maturity and all aganglionic segments had no expression for cathepsin D in the submucosal and myenteric plexuses of the intestine. However, histiocytes within the laminar propria and submucosa stained positively for cathepsin D. In conclusion, intestinal ganglion cells in children have reactivity for cathepsin D, threrfore immunohistochemical staining for cathepsin D can be used for identification of ganglion cells in neonates.

  • In order to study the immunohistochemical effects of herbal drug-acupuncture of Radix paeoniae lactiflorae on gastric ulcer induced by HCl-aspirin in rats. this experiment was done by herbal drug-acupuncture to Wisu($B_{21}$). Chung-Wan($CV_{12}$), Chok-Samni($S_{36}$) loci to measure histological features of ulcer lesion, the change of numbers of parietal cell, chief celI, gastrin and somatostatin-immunoreactive cell. The obtained results were as follows; 1. The ulcerative lesions of gastric mucosa were decreased to WiSU($B_{21}$) loci followed by Chung-Wan($CV_{12}$) and Chok-Samni($S_{36}$) loci compared to the control groups. 2. In the numbers of parietal cell, the most remarkable decrease was observed to Wisu($B_{21}$) loci followed by Chung-Wan($CV_{12}$) and Chok-Samni($S_{36}$) loci compared to the control groups. 3. In the numbers of chief cell, the most remarkable increase was observed to Wisu($B_{21}$) loci followed by Chung-Wan($CV_{12}$) and Chok-Samni($S_{36}$) loci compared to the control groups. 4. In the numbers of gastrin-immunoreactive cell, the most remarkable decrease was observed to Wiu($B_{21}$) loci followed by Chung-Wan($CV_{12}$) and Chok-Samni($S_{36}$) loci compared to the control groups. 5. In the numbers of somatostatin-immunoreactive cell, the most remarkable decrease was observed to Wisu($B_{21}$) loci followed by Chung-Wan($CV_{12}$) and Chok-Samni($S_{36}$) loci compared to the control groups.

No Association between BRCA1 Immunohistochemical Expression and Tumor Grade, Stage or Overall Survival in Platinum-Treated Epithelial Ovarian Cancer Patients

  • Background: The aim of this work is to assess the frequency of BRCA1 protein immunohistochemical (IHC) expression in epithelial ovarian cancer (EOC) and to evaluate the association of BRCA1 expression with clinical and pathological characteristics and the overall survival (OS) of patients treated with postoperative platinum-based chemotherapeutic agents. Materials and Methods: This retrospective study was conducted on 35 cases of epithelial ovarian cancer selected from the files of the Pathology Department, Faculty of Medicine, Mansoura University, Egypt. Immunohistochemistry (IHC) was performed for BRCA1 gene protein. BRCA1 expression was compared to patient's age, tumor histology, grade, stage and OS time. Statistical analysis was carried out with the SPSS version 16.0 to assess significant associations. Results: BRCA1 nuclear expression was detected in 40% of EOC, in which a mild increase in the percentage of positive cases was observed with serous histology, stage IV, and grade 3 carcinomas. There was a significant statistical difference in BRCA1 expression with regard to histological subtypes of EOC (p=0.048), but not grade or stage. Mean OS and survival rate were slightly better for BRCA1 expressing group, but there was no statistically significant difference (p=0.528). Conclusions: No association between BRCA1 immunohistochemical expression and tumor grade, stage or overall survival was noted in platinum-treated epithelial ovarian cancer patients.

Uterine Cervix Metastasized from Signet Ring Cell Adenocarcinoma - 1 Case Report - (자궁경부에 전이된 반지세포암종 - 1례 보고-)

  • This study is a report about a specific patient whose primary stomach adenocarcinoma metastasized to uterine cervix adenocarcinoma. A thirty-nine year old female patient was initially diagnosed as having metastatic adenocarcinoma in the supraclavicular lymph node. Upon further examination, she was diagnosed with stomach adenocarcinoma. 8 months later, a cervix punch biopsy was performed. The stains used for examination were H&E stain, PAS stain, Alcian blue stain, Mucicarmine stain, Papanicolaou's (Pap.) stain, and as immunohistochemical stains, cytokeratin 7 and 20 were done. In the H&E stain, the tumor cells showed prominent and eccentric nuclei, thin nuclear membrane in abundant mucous cytoplasm, and cylinder shape. In the PAS stain, intracytoplasmic mucin vacuoles were stained with pink, and in Alcian blue and Mucicarmine stains, intracytoplasmic mucin vacuoles were stained with blue and red. As in the above results, she was diagnosed with undifferentiated adenocarcinoma. As found on the cytologic smear preparation of the uterine cervix stained by Papanicolaou's stains, the background was relatively clear, the number of malignant cells was relatively low, and large and eccentric nuclei in abundant cytoplasm were observed. Upon observing the tissue preparation of the uterine cervix biopsy by H&E stain, a clear background, large and eccentric nuclei, and a signet ring cell types were observed, and the number of malignant cells were fewer than in the primary uterine cervix adenocarcinoma. The vacuoles in cytoplasm were observed. The nuclear membrane and chromatin were thick and very rough, and upon observation by cytokeratin 7 and 20 of immunohistochemical stain, the tumor cells indicated a positive rate of 70% and 20%, respectively. According to these results, also she was diagnosed with metastasized uterine cervix adenocarcinoma. In summary of the results of pathologic findings on stomach biopsy and cytologic, histopathologic, and immunohistochemical finding on uterine cervix biopsy, the adenocarcinoma of her uterine cervix could assert the adenocarcinoma of signet ring cell type that was metastasized from the primary undifferentiated adenocarcinoma in stomach.

Oral Amelanotic Malignant Melanoma in a Dog: Melan A Immunohistochemical Findings (개에서 발생한 구강 멜라닌결핍 악성흑색종 예 : Melan A 면역화학조직 염색 고찰)

  • A 10-year-old intact male mixed breed dog was presented with a three-month history of massive oral mass. Physical examination revealed extending mass from the right upper gingiva. No metastasis was found at the time of presentation. Histopathologic examination of biopsied tissue from the oral mass was consistent with a neuroendocrine tumor with generalized epithelioid cells and few spindle cells. There were highly mitoses and no visible melanin granules with H&E staining. Immunohistochemical staining for Melan A was performed on section of tumor and was strongly positive. Diagnosis was made as amelanotic malignant melanoma based on histopathology with Melan A immunohistochemistry. This case study indicates that the Melan A immunohistochemical staining may be valuable to diagnose amelanotic malignant melanoma in dogs.

  • Purpose : Deoyeongjeon(DYJ : 大營煎) is used in female infertility, caused by ovulation disorder. so this study is to examine what are the effects of the Deoyeongjeon(DYJ) on the vulation and Ovary in Rats Methods : 4weeks Female Sprague-Dawley 12 rats of weighting 160-l80g, were divided into three groups including the DYJ oral administration(4ml/kg) groups(4heads) and DYJ oral administration(8m/lkg) groups(4heads). then we observed changes in the serum concentrations of FSH, LH, and estradiol(E2) and the histological changes of ovary and the immunohistochemical staining for progesterone receptor in ovary of rats. Results : 1. Blood FSH level significantly increased in experimental group as compared with control group. 2. In blood LH level, experimental group showed no efficacy as compared with control group. 3. In blood estradiol(E2) level, experimental group showed no efficacy as compared with control group. 4. In histological observations of ovary, ovulation increased in experimental group as compared with control group, which showed no efficacy. 5. In observations of immunohistochemical staining for progesterone receptor in ovary, immunohistochemical staining score (ISS) of atretic follicles significantly showed a tendency to decrease in experimental group as compared with control group. Conclusion : DYJ influences the pituitary gland and ovary to increase the ovulation of rats.

Methods : 4weeks Female Sprague-Dawley 12 rats of weighting 160-l80g, were divided into three groups including the GSH oral administration(4ml/kg) groups(4heads) and GSH oral administration(8ml/kg) groups(4heads). Then we observed changes in the serum concentrations of FSH, LH, and estradiol($E_2$) and the histological changes of ovary and the immunohistochemical staining for progesterone receptor in ovary of rats. Results : 1. GSH didn't make a difference as compared with control group in serum FSH level. 2. GSH didn't make a difference as compared with control group in serum LH level. 3. GSH significantly increased serum $E_2$ level. 4. GSH significantly increased ovulation in histological observations of ovary. 5. GSH tended to decrease immunohistochemical staining score (ISS) of atretic follicles in immunohistochemical staining for progesterone receptor in ovary. Conclusion : GSH influences ovary to increase the ovulation of rats.
