Poor Prognostic Factors in Surgically Resected Stage I Non-small Cell Lung Cancer: Histopathologic and Immunohistochemical Analysis

  • Background: A better understanding of the histopathology and molecular biology of lung cancer might improve our capability to predict the outcome for any individual patient. The purpose of this study was to evaluate several histopathologic and molecular markers in order to assess their prognostic value in stage I non-small cell lung cancer. Materials and Methods: One hundred ten patients at the Kyungpook National University Hospital were enrolled in the study. Histopathologic factors and molecular markers were selected. Results: Univariate analysis showed that the T stage, differentiation, visceral pleural invasion, and survivin expression were significantly associated with recurrence. Multivariate analysis demonstrated that differentiation and survivin overexpression emerged as independent prognostic factors of recurrence. Conclusion: In resected stage I non-small cell lung cancer, poor differentiation and survivin overexpression have been identified as independent predictors of poor disease-free survival.

Relaxation Effects of Eucomiae Cortex in Isolated Rabbit Corpus Cavernosum Smooth Muscle (杜冲의 토끼 음경해면체 평활근 이완효과)

  • This study was aimed to investigate the relaxation effects of Eucomiae Cortex (EC) extract in isolated rabbit corpus cavernosum smooth muscle and its mechanism. To evaluate the relaxation of EC extract in rabbit corpus cavernosum, EC extract was treated in corporal strips which were precontracted with phenylephrine(PE). To study its mechanism, Nω-nitro-L-arginine (L-NNA) was pretreated after infuse of EC extract and compared with non-treated. In calcium chloride (Ca2+) -free krebs solution, EC extract and Ca2+ 1 mM were infused by turns after Ca2+ 1 mM was treated into corporal strips contracted by PE. Cell ability, nitric oxide (NO) and epithelial nitric oxide synthase (eNOS) on human umbilical vein endothelial cell (HUVEC) were measured by MTT assay, Griess reagent system and histochemical, immunohistochemical methods. EC extract showed a significant relaxation effects on the corporal strips, this effects were inhibited by pretreatment of L-NNA. EC extract inhibited the increase of contraction by Ca2+ influx in Ca2+-free krebs solution, and eNOS positive reaction in corpus cavernosum, NO production in HUVEC increased by treatment of EC extract. These result suggest that the relaxation effects of EC extract in isolated corpus cavernosum smooth muscle are involved in increase of eNOS and NO production, blocking of extracellular Ca2+ influx.

Regional distribution and relative frequency of the gastrin, secretin and pancreatic polypeptide-immunoreactive cells in the gastrointestinal tract of the fetus of Korean native goat (한국재래산양 태자의 위장관에 있어서 gastrin, secretin 및 pancreatic polypeptide 면역반응세포의 분포 및 출현빈도에 관한 연구)

  • The regional distributions and relative frequencies of the gastrin, secretin and pancreatic polypeptide(PP)-immunoreactive cells in the gastrointestinal tract of the fetus(180 days of gestation) of Korean native goat were studied with immunohistochemical(ABC) methods. Gastrin-immunoreactive cells were detected in fundus, pylorus and duodenum and these cells were most predominant in pylorus. Secretin-immunoreactive cells were observed in pylorus, duodenum and ileum. PP-immunoreactive cells were restricted to fundus. These immunoreactive cells were situated in surface epithelium and mucosal gland regions. The regional distribution and relative frequency of PP-immunoreactive cells was somewhat different to the adult Korean native goat. Immunoreactive cells in the surface epithelial regions were open typed cells which were spindle shaped cells but closed typed cells which were round or/to spherical shaped cells were observed in the mucosal gland regions.

Innervation of Neuroepithelial Bodies in Bronchiolar Epithelium of Human Fetal Lung (인태아 폐의 신경상피소체와 신경종말에 관한 미세구조적 연구)

  • Ultrastructure of nerves and their associated cells in the bronchiolar epithelium of the human fetal lung were studied with ultrastructural and immunohistochemical methods. The neuroendocrine cells were scattered along the basal part of non-ciliated respiratory epithelium and appeared as single cell (solitary neuroendocrine cell) or groups (neuroepithelial bodies). The solitary neuroendocrine cells were devoid of any detectable innervation, while the neuroepithelial bodies were associated with nerve ending containing morphologically afferent (sensory) and efferent (motor) intraepithelial terminals. The afferent nerve endings contained abundant mitochondria with long cristae. The efferent nerve endings were characterized by the presence of synaptic vesicles. Both types of nerve endings formed synaptic junction between nerve endings and neuroepithelial bodies cells. Serial sections of the intraepithelial nerves revealed that both morphologically afferent and efferent types of nerve endings may be formed by the same nerve fiber. By immunohistochemistry, bombesin and serotonin were localized in solitary neuroendocrine cells and neuroepithelial bodies of human fetal lung from various prenatal age groups. These results suggest that the neuroepithelial bodies cells of the human fetal lung have neuroreceptor function.

  • This study was performed to elucidate the histopathologic distribution of immunoglobulins, particularly IgA, IgG and IgM in the periapical lesions, including 22 periapical granulomas and 18 periapical cysts. The immunoperoxidase staining method using reagents manufactured and supplied by Danish DAKO company was employed in this study. In comparison with the immunohistochemical methods, this method was proved to be reliable and convinient one to detect immunoglobulins in the tissue. The following results were obtained: 1. In the 22 periapical granulomas, IgG was found in 20 cases (90.9%), IgA in 16 cases (72.7%) and IgM in 19 cases (86.3%). 2. In the 18 periapical cysts, IgG was found in 16 cases (88.8%), IgA in 13 cases (72.2%) and IgM in 15 cases (83.3%). 3. The distribution of immunoglobulins both in periapical granulomas and periapical cysts was in great diversity according to the lesion and area. 4. More immunoglobulins were found in the exudative area with moderate or severe infiltrations of plasma cells and lymphocytes and less concentration of immunoglobulins were seen in the area with leukocytes infiltration and tissue destruction. 5. The area with collagenolysis and reparative activity contained more abundant IgG and IgM than IgA in periapical granulomas. 6. IgG was dominant in the granulomatous connective tissue and immunoglobulins were not easily found in the fibrous capsule in periapical cysts.

An Immuno-Electron Microscopic Study of Expression of $\alpha$-tubulin on Ciliogenesis in Newborn Rat Trachea (신생 횐쥐의 기관섬모에서 $\alpha$-tubulin의 발현과 섬모형성 양상에 대한 면역전자현미경적 연구)

  • Background: This study was undertaken to detect the patterns of ciliogenesis in newborn rats trachea. Materials and Methods: The experimental animals(Sprague-Dawley strain) were divided five groups, one day, two day, three day, five day and seven day newborns as experimental groups. To obtain differential distribution of $\alpha$-tubulins in the ciliated cells and patterns of ciliogenesis, we used immunohistochemical stain method with mouse anti $\alpha$-tubulin monoclonal antibody as the primary antibody and gold particles conjugated goat anti mouse IgG as the secondary antibody. And we observed the specimens by electron microscope (Hitach-600 Model). Results : 1) The distribution of the $\alpha$ -tubulin reactions in apical zone was slightly decreased from three day after birth. 2) From 5th day after birth, the decreasing number of gold particles in intermediate zone was remarkable. 3) On the comparison with the other zones, the number of gold particles in the Golgi zone for seven days showed no statistical difference. Conclusion : The ciliogenesis of the tracheal ciliated cells in early newborn rat were made via centriolar and acentriolar pathways, in late groups, from five day after birth, the major ciliogenesis pattern might be centriolar pathway.

Ontogeny, distributions and relative frequencies of the somatostatin-immunoreactive cells in the proventriculus of the chicken embryos with incubation periods (닭 태자 샘위의 somatostatin 면역반응세포의 출현시기, 분포 및 상대적 빈도에 관한 연구)

  • Ontogeny, distributions and relative frequencies of somatostatin-immunoreactive cells were investigated in the proventriculus of the chicken embryos with incubation periods. Samples were taken from 10 groups(10 days of incubation to hatching) and studied by immunohistochemical methods. The findings were as follows. Somatostatin-immunoreactive cells were observed from 12 days of incubation in the proventricular glands and after that increased with incubation periods. The first observation time of these cells in the epithelium were at 15 days of incubation in the basal portion but in 16 and 17 days of incubation, no immunoreactive cells were observed in the epithelium but after that a few immunoreactive cells were observed in the basal portion and gastric gland regions. The shapes of these cells were spherical to spindle in the proventricular glands and spherical to round in the epithelium and gastric gland.

Effects of Jiyutang on DSS-induced Colitis of the Mouse (Dextran Sulfate Sodium으로 유발된 생쥐의 대장염에 미치는 지유탕(地楡湯)의 효과)

  • Objectives : We examined the effect of Jiyutaug(JYT) on the experimental ulcerative colitis induced by dextran sulfate sodium(DSS). Methods : Experimental colitis was induced in mice by daily treatment with 3% DSS in the drinking water for 5days. Afterward, the mice were divided into two groups: the control group was administered water and the sample group was administered JYT for 7 days. Results : The sample group provided JYT for 7 days demonstrated faster recovery of body weight compared with the control group. Histologic change showed faster regeneration of crypt and surface epithelial cells, decreased edema of the submucosa, and decreased Iymphatic follicles of mucosa compared with the control group. immunohistochemical stain using COX-2 gene was decreased. Regeneration of surface epithelial cells and goblet cells in mucosa was observed by transmission electron microscope. Conclusion : These results indicate therapeutic effect of JYT on DSS-induced colitis in mice.

  • Objectives : The aim of this study was to investigate the effect of various electroacupuncture stimulation on neuronal nitric oxide synthase(nNOS) in cerebral cortex, brain stem, cerebellum of spontaneously hypertensive rats. Methods : We evaluated the changes of nNOS-positive neurons using a immunohistochemical method. The staining intensity of nNOS positive neurons was assessed in a quantitative fashion using a microdensitometrical method based on optical density by means of an image analyzer. Results : The average optical density of nNOS-positive neurons of 100 Hz (bipolar square wave 0.2 ms duration and 100 Hz frequency) electroacupuncture treatment group significantly decreased in most cortical areas comparison between the manual acupuncture and 2 Hz (bipolar square wave 0.2 ms duration and 2 Hz frequency) electroacupuncture groups. In the brain stem, the optical density of nNOS-positive neuron at superficial gray layer of the superior colliculus area, dorsolateral periaqueductal gray area and paralemniscal nucleus were same as cerebral cortex. Conclusion : We conclude that the morphological evidence for nNOS-positive neurons may be have regional change in cerebral cortex brain stem and cerebellum according to various electroacupuncture stimulations.

Changes of Somatostatin-Immunoreactive Cells on the Stomach of Ovariectomized Rats

  • The changes on the regional distributions and frequencies of somatostatin-immunoreactive (IR) cells in the fundus and pylorus of the stomach of osteoporotic Sprague-Dawley rats induced by ovariectomy were studied by immunohistochemical methods. The experimental animals were divided into two groups, one for non-ovariectomized group (Sham) and the other for ovariectomized group (OVX). Samples were collected from the fundus and pylorus regions at the 10 th week after ovariectomy or sham-operation. Somatostatin-IR cells were observed in both regions of the stomach regardless of ovariectomy. Most of these IR cells in the mucosa of the fundus or pylorus were generally spherical or spindle in shape (open type cell) while cells found in the gastric gland regions were round in shape (close type cell). Significantly lower number (P<0.01) of somatostatin-IR cells were detected in OVX as compared with Sham in the fundus and pylorus. In the present study, the density of somatostatin in the stomach was markedly decreased. Therefore, these changes in density of somatostatin-IR cells detected in this study may support the speculation that the development of gastrointestinal symptoms in osteoporosis such as impairments of calcium and some lipids, frequently encountered in patients with postmenopausal osteoporosis because the changes in gastrointestinal endocrine density would reflect the change in the capacity of producing these hormones and regulating gut motility and digestion.
