Background: Breast cancer is the most common neoplasm in women and the most frequent cause of death in those between 35 and 55 years of age. All multicellular organisms have an innate immune system, whereas the adaptive or 'acquired' immune system is restricted to vertebrates. This study focused on the effect of conditioned medium isolated from cultured breast cancer cells on NB4 neutrophil-like cells. Materials and Methods: In the current study neutrophil-like NB4 cells were incubated with MCF-7 cell-conditioned medium. After 6 h incubation the intracellular receptor TLR2, was analyzed. Results: The results revealed that MCF-7 cell-conditioned medium elicited expression of TLR2 in NB4 cells. Conclusions: This treatment would result in the production of particular stimulants (i.e. soluble cytokines), eliciting the expression of immune system receptors. Furthermore, the flow cytometry results demonstrated that MCF-7 cell-conditioned medium elicited an effect on TLR2 intracellular receptors.
NK (Natural killer) cells are innate immune cells and play important roles as the first immune cells to act when cancer occurs. In many cancer patients, NK cells can be seen to be inactivated, suggesting that NK cells are important in cancer treatment. In order to overcome the disadvantages of NK cells in cancer treatment, it is critical to develop strategies that enhance the proliferation and cytolytic function of NK cells. We applied niclosamide to measure the degree of NK cell activation, and obtained unexpected results of increased NK cell numbers and anti-tumor activity. Although further investigation is required to uncover the detailed mechanisms, our results suggest that Niclosamide is a promising candidate to increase the efficacy of cancer immunotherapy using NK cells.
Objective : To investigate immune enhancing effects of Boummyunyuck-dan (BMD) Methods : In this study I investigated the effect of BMD on cell proliferation and viability. In addition, I investigated production of cytokines (IL-2, IL-4 and $IFN-{\gamma}$), NO, and $TNF-{\alpha}$ in human T-cell leukemia, MOLT-4 cells. The cells were cultured for 24h in the presence or absence of BMD. Result : BMD increased the cell viability by 15% (P<0.05) and enhanced IL-2, IL-4 and $IFN-{\gamma}$ production compared with media control in a dose-dependent manner (P<0.01) at 24h. BMD also increased mRNA and protein expression levels of $IFN-{\gamma}$ in MOLT-4 cells. In addition, I also assessed the effects of BMD on production of NO and $TNF-{\alpha}$ from the peritoneal macrophages because NO and $TNF-{\alpha}$ as a potent macrophage-derived immune reaction regulatory molecule has received increasing attention. However, BMD had no effect on NO and $TNF-{\alpha}$ production in the cells. Conclusion : These data indicate that BMD has some immune-enhancing effect, and that its action may be due to the proliferation and cytokine production of T cells.
By using several solvents, barley extracts containing the anticomplementary activities in classical pathway were prepared (250 $\mu\textrm{g}$/ml): methanol (83.1%), ethanol (71.9%), water extract (25.4%), M-1 (250 $\mu\textrm{g}$/ml), and the soluble part of methanol extract which showed the highest activity (83.4%) and the yield. Anticomplementary activity of methanol extract as well as protease digestion in classical pathway showed 82.4% and 78.4% in the concentration of 250 $\mu\textrm{g}$/ml, respectively. It was found that protein was not involved in anticomplementary activity in the classical pathway and the methanol extract made an impact on classical pathway, but not on alternative pathway. For the immune-stimulating effect, the T cell proliferation effect of the protease digestion displayed little effect irrespective of the dose. In addition, the T cell proliferation effect of methanol extract showed 13-fold higher proliferation effect compared with positive control. It was revealed that the substance containing protein serves as an important factor for the immune proliferation. Therefore, the anticomplementary activity ${\beta}$-glucan in classical pathway and alternative pathway displayed the lowest activity, showing 2.2%, 22.3% respectively. However, the immune-stimulating effect of ${\beta}$-glucan showed the T cell stimulating effect 13 times higher than positive control.
We investigated the immune enhancement effects of JaSaengHwan (JSH). The forced swimming test (FST) has been used as a screening model for new immune enhancement agents. We found that JSH (0.1 mg/ml) significantly reduced the immobility time in the FST compared to the control. Also, we investigated the effect of JSH on the proliferation of T cell and production of cytokines in human T-cell line, MOLT-4 cells and mouse peritoneal macrophages. JSH (1 mg/ml) significantly increased the cell proliferation by $46.78{\pm}6.41%$ (p<0.05) and also significantly increased the interleukin (IL)-2, IL-4 and interferon $(IFN)-{\gamma}$ production compared with media control (about 2-fold for IL-2, 3-fold for IL-4 and 1.5-fold for $IFN-{\gamma}$, p<0.05) at 24 h. In addition, JSH increased the production of IL-12 on the mouse peritoneal macrophages (by 3.6-fold for IL-12, p<0.05). In conclusion, these data indicate that JSH may have an immune-enhancement effect.
The purpose of this experiment was to investigate both the immunomodulatory effect of evening primrose(EP) oil and the effects of EP oil on immunoregulation by cyclophosphamide in mice. EP oil at doses of 0.1, 0.2 and 0.4 ml/kg were orally administered to ICR male mice once daily for 28 consecutive days. Cyclophosphamide was injected intraperitoneally to ICR mice with a single dose of 5 mg/kg at 2 days before secondary immunization. Mice were sensitized and challenged with sheep red blood cells(S-RBC). Immnune responses were evaluated by humoral and cellular immune responses and non-specific immune response. The results of this study were summarized as follows; (1) The humoral immune responses such as hemagglutination titer(HA), hemolysin titer(HY), Arthus reaction and plaque forming cell(PFC) were significantly enhanced in the low dose EP oil administered groups(0.1 and 0.2 ml/kg). However, in the high dose EP oil administered group(0.4 ml/kg) the responses were significantly lowered. (2) In the case of cellular immune responses, delayed type hypersensitivity reaction(DTH) was significantly decreased in EP oil whereas rosette forming cell(RFC) was remarkably enhanced. (3) Activities of natural killer cells and phagocyte were generally enhanced in EP oil. In addition, serum albumin and globulin were also increased.
The present study was performed to evaluate how cellular and humoral immune responses were perturbed by immunization of mixed periodontal bacterial biofilms. Each group of mice was immunizared with 1) Poqhyromonas gingivalis (P. gingivaliis) grown as a planktonic culture, 2) Fusobacterium nucleatum (F. nucleatum), 3) P. gingivalis grown as a biofilm, or 4) mixed P. gingivalis plus F. nucleatum grown as a biofilm culture, respectively. Immune mouse sera were collected from each mouse. Spleens were harvested to isolate T cells and consequently stimulated with antigen presenting cells and P. gingivalis whole cell antigen to establish P. gingivalis-specific T cell lines. There were no significant differences in the mean anti- gingivalis IgG antibody titers among mouse groups. Immunization of mice with pure P. gingivalis biofilm or mixed P gingivalis plus F. nucleatum biofilm resulted in significant reduction o f antibody avidity and opsonophagocytois function. INF-$\gamma$production by P. gingivalis-specific T cell lines was also substantially recluced in mouse groups immunized with the biofilm. It was concluded that P. gingivalis biofilm perturbs the cellular and humoral immune responses in periodontal disease.
Lung cancer is one of the most commonly diagnosed cancers and the leading cause of cancer-related deaths worldwide. Although progress in the treatment of advanced non-small cell lung cancer (NSCLC) has been made over the past decade, the 5-year survival rate in patients with lung cancer remains only 10%-20%. Obviously, new therapeutic options are required for patients with advanced NSCLC and unmet medical needs. Cancer immunotherapy is an evolving treatment modality that uses a patient's own immune systems to fight cancer. Theoretically, cancer immunotherapy can result in long-term cancer remission and may not cause the same side effects as chemotherapy and radiation. Immunooncology has become an important focus of basic research as well as clinical trials for the treatment of NSCLC. Immune checkpoint inhibitors are the most promising approach for cancer immunotherapy and they have become the standard of care for patients with advanced NSCLC. This review summarizes basic tumor immunology and the relevant clinical data on immunotherapeutic approaches, especially immune checkpoint inhibitors in NSCLC.
CD4+ T helper (Th) cells play a crucial role in the modulation of innate and adaptive immune responses through the differentiation of Th precursor cells into several subsets, including Th1, Th2, Th17, and regulatory T (Treg) cells. Effector Th and Treg cells are distinguished by the production of signature cytokines and are important for eliminating intracellular and extracellular pathogens and maintaining immune homeostasis. Stimulation of naive Th cells by T cell receptor and specific cytokines activates master transcription factors and induces lineage specification during the differentiation of Th cells. The master transcription factors directly activate the transcription of signature cytokine genes and also undergo post-translational modifications to fine-tune cytokine production and maintain immune balance through cross-regulation with each other. This review highlights the post-translational modifications of master transcription factors that control the differentiation of effector Th and Treg cells and provides additional insights on the immune regulation mediated by protein argininemodifying enzymes in effector Th cells.
Head and neck squamous cell carcinoma (HNSCC) is the sixth most common cancer globally with high morbidity and mortality. Immune surveillance is well recognized as an important mechanism to prevent development or progression of HNSCC. HNSCC can escape the immune system through multiple mechanisms including development of tolerance in T cells and inhibition of T-cell-related pathways, generally referred to as checkpoint inhibitors. Recent clinical trials have demonstrated a clear advantage in advanced HNSCC patients treated with immune checkpoint blockade. Right at the front of the new era of immunotherapy, we will review current knowledge of immune escape mechanisms and clinical implication for HNSCC.
