The Epstein-Barr virus-transformed lymphoblastoid cell line (LCL) is one of the major genomic resources for human genetics and immunological studies. Use of LCLs is currently extended to pharmacogenetic studies to investigate variations in human gene expression as well as drug responses between individuals. We evaluated four common internal controls for gene expression analysis of selected hematopoietic transcriptional regulatory genes between B cells and LCLs. In this study, the expression pattern analyses showed that TBP (TATA box-binding protein) is a suitable internal control for normalization, whereas GAPDH (glyceraldehyde-3-phosphate dehydrogenase) is not a good internal control for gene expression analyses of hematopoiesis-related genes between B cells and LCLs at different subculture passages. Using the TBP normalizer, we found significant gene expression changes in selected hematopoietic transcriptional regulatory genes (downregulation of RUNX1, RUNX3, CBFB, TLE1, and NOTCH2 ; upregulation of MSC and PLAGL2) between B cells and LCLs at different passage numbers. These results suggest that these hematopoietic transcriptional regulatory genes are potential cellular targets of EBV infection, contributing to EBV-mediated B-cell transformation and LCL immortalization.
We have established in culture a spontaneously immortalized bovine embryonic fibroblast (BEF) cell line that has lost p53 and $p16^{INK4a}$ functions. MyoD is a muscle-specific regulator capable of inducing myogenesis in a number of cell types. When the BEF cells were transduced with MyoD they differentiated efficiently to desmin-positive myofibers in the presence of 2% horse serum and 1.7 nM insulin. The myogenic differentiation of this cell line was more rapid and obvious than that of C2C12 cells, as judged by morphological changes and expression of various muscle regulatory factors. To confirm that lack of the p53 and $p16^{INK4a}$ pathway does not prevent MyoD-mediated myogenesis, we established a cell line transformed with SV40LT (BEFV) and introduced MyoD into it. In the presence of 2% horse serum and 1.7 nM insulin, the MyoD-transduced BEFV cells differentiated like the MyoD-transduced BEFS cells, and displayed a similar pattern of expression of muscle regulatory proteins. Taken together, our results indicate that MyoD overexpression overcomes the defect in muscle differentiation associated with immortalization and cell transformation caused by the loss of p53 and Rb functions.
We have established three immortal bovine muscular epithelial (BME) cell lines, one spontaneously immortalized (BMES), the second SV40LT-mediated (BMEV) and the third hTERT-mediated (BMET). The morphology of the three immortal cell lines was similar to that of early passage primary BME cells. Each of the immortal cell lines made cytokeratin, a typical epithelial marker. BMET grew faster than the other immortal lines and the BME cells, in 10% FBS-DMEM medium, whereas neither the primary cells nor the three immortal cell lines grew in 0.5% FBS-DMEM. The primary BME cells and the immortal cell lines, with the exception of BMES, made increasing amounts of p53 protein when treated with doxorubicin, a DNA damaging agent. On the other hand, almost half of the cells in populations of the three immortal cell lines may lack $p16^{INK4a}$ regulatory function, compared to primary BME cells that were growth arrested by enforced expression of $p16^{INK4a}$. In soft-agar assays, the primary cells and immortal cell lines proved to be less transformed in phenotype than HeLa cells. The three immortal epithelial-type cell lines reported here are the first cell lines established from muscle tissue of bovine or other species.
지방줄기세포는 다양한 세포로 분화할 수 있어서 세포치료제로 주목받고 있으나 세포 증식 속도가 느리고 세포의 수명이 짧아 임상 적용에 어려움이 있다. 본 연구팀은 선행연구에서 지방줄기세포의 증식속도를 향상시키고 세포 수명을 연장시키기 위해 SV40의 T항원 유전자를 사람의 ADSC에 도입하여 불사화시킨 ADSC 세포주를 제작하였으며 ADSC-T라고 명명하였다. 본 연구에서는 ADSC-T 세포의 분화능력을 검증하고, ADSC-T 세포를 배양하여 획득한 무혈청 배양액의 항염증 효능을 평가하였다. 그 결과, ADSC-T 세포주는 지방세포와 골세포로 분화할 수 있는 능력을 보유하고 있어 줄기세포의 가장 중요한 특징인 분화능력을 유지하고 있음을 알 수 있었다. 또한 ADSC-T의 배양액은 염증을 유발하는 NF-κB의 활성을 억제하였고, NF-κB의 표적 유전자인 COX-2와 iNOS의 발현도 억제하였다. 또한 ADSC-T 배양액은 ERK, JNK, p38의 인산화를 저해하여 세포 내 염증 유발 신호 전달 회로인 MAPKs 회로를 억제하는 것으로 나타났으며, 염증을 유발하는 cytokine인 TGF-β, TNF-α, IL-6, IL-13의 발현도 억제하는 효과를 보여 강한 항염증 효능이 있을 것으로 추정되었다. 실제로, 아토피 모델 쥐인 Nc/Nga 마우스를 DNCB로 처리하여 아토피 피부염을 유발한 뒤 ADSC-T 배양액을 도포한 결과 강한 치료 효과를 나타내었다. 이상의 결과로부터, 불사화된 ADSC-T 세포주는 지방줄기세포의 장점과 효능을 유지하고 있으며 향후 지방줄기세포의 세포 수급 문제를 해결할 수 있을 뿐 아니라 임상 적용이 가능한 배양액의 생산에도 널리 이용될 수 있을 것으로 사료된다.
목 적 : Hyper IgM syndrome(HIGM)은 크게 세 부류로 나누는데, CD40L 분자의 돌연변이로 초래되며 X-linked로 유전되는 형태를 HIGM1이라고 하고, 상염색체성 열성 형태로 유전되면서 CD40L는 정상적으로 표현되는 형태로 activation-induced cytidine deaminase(AID) 유전자에 이상 때문에 오는 경우를 HIGM2로 분류하고 있다. 다른 한 부류는 X-linked HIGM 증후군의 매우 드문 한 형태로서 발한 저하성 외배엽 이형성증이 동반된 경우로, 이 질환은 전사인자인 nuclear factor ${\kappa}B$의 활성화에 관여하는 nuclear factor ${\kappa}B$ essential modulator를 coding하는 유전자의 돌연변이 때문에 오는 것으로 알려져 있다. 연구자들은 HIGM2와 유사하지만 AID 유전자에 변이는 없는 새로운 형태의 HIGM 환자의 말초 B세포를 이용하여 병인을 조사하고자 본 연구를 시도하였다. 방 법 : 환자의 말초혈액 단핵구를 분리하고, EBV로 immortalization을 시켜 Cycle TEST PLUS DNA Reagent kit를 이용하여 cell cycle을 분석하였다. 환자의 말초혈액 T 세포에서 CD40L의 표현을 immunostaining으로 알아보고, RNA를 추출하여 RT-PCR을 하고 direct sequencing을 통하여 CD40L 유전자의 돌연변이 부위를 찾아보았다. 아울러 AID 유전자의 돌연변이를 찾기 위하여 같은 방법으로 sequencing하고 조사하였다. 환자의 림프절을 병리학적인 검사를 시행하고 CD3, CD23, CD40, Fas-L, bcl-2, BAX의 표현을 알아보기 위하여 immunostaining을 실시하였다. 결 과: 림프절의 광학적 소견은 반응성 여포증식의 소견을 보였으며, 여포와 단구양 증식은 B-세포 표시인자인 L26에 양성을 보였고, 대부분의 형질세포는 IgM에 양성을 보였다. 여포는 CD40, Fas, BAX에 양성반응을 보이고, bcl-2와 Fas-L에 음성반응을 보였다. 말초혈액 B 림프구를 이용하여 cell cycle을 분석한 결과 정상(17.9%)에 비하여 G2/mitosis phase(M3 in figure)가 현저하게 감소(8.5%)되어 있는 양상을 보여주고 있으며, IL-4로 자극한 경우에는 정상인에서 보여주는 양상으로 회복되는 양상을 볼 수 있었다. 단핵수에서 CD40L의 표현은 정상이었고 CD40L 유전자에도 돌연변이는 발견할 수 없었으며, HuAID 유전자에도 돌연변이를 발견할 수 없었다. 결 론 : 환자의 말초혈액 B림프구를 통한 연구 결과, 기존에 보고 되어진 HIGM2 형태와 임상적으로는 비슷하지만, 정상적인 AID유전자를 보이고, G2/mitosis phase가 정상에 비하여 감소된, 새로운 형태의 HIGM이라고 여겨진다.
