Immobilization of Glucose Oxidase on Multi-Wall Carbon Nanotubes for Biofuel Cell Applications

  • Glucose oxidase was immobilized on the carboxylated multi-wall carbon nanotubes (MWNT-COOHs) in the presence of a coulping reagent, 1-ethy1-3-(3-dimethylaminopropy1) carbodiimide. Significant amounts of glucose oxidase were also immobilized on MWNT-COOHs without the coupling reagent. Various conditions for the immobilization of glucose oxidase were optimized. Optimal pH for the maximal activity of the immobilized glucose oxidase shifted to 7 from the optimal pH of 6 for the maximal activity of free enzyme due to the carboxy1 groups on the surface of MWNT-COOHs. An electrode of graphite rod with a diameter of 6 mm was fabricated using the immobilized glucose oxidase. The cyclic voltammetry study of the enzyme electrode revealed that the oxidation of glucose and subsequent transfer of electrons from the oxidation of glucose to the electrode were possible by the immobilized glucose oxidase without a mediator, implying that the enzyme electrode can be utilized for the development of biofuel cells.

  • 제주도 근해에서 다량 채취되고 있는 해양갈조류인 톳의 살아있지 않은 건조한 생물질을 Ca-alginate법(Ca-ALG), Ba-alginate법(Ba-ALG), polyethylene glycol법(PEG), $\kappa$-carrageenan법(CARR) 등으로 고정화시켜 고정화방법에 따른 Cu의 흡착기능을 검토하였다. 생물질의 첨가량이 증가함에 따라서 Cu의 제거율은 증가하였으나 흡착량은 감소하였다. 그러나 초기농도가 증가함에 따라서는 cu의 흡착량이 증가하였다. 고정화법에 따른 Cu의 흡착량은 Ca-ALG>Ba-ALG>PEG>CARR순으로 감소하엿다. 고정화된 생물질에 으한 Cu의 흡착은 Freundlich 흡착 등온식 보다는 Langmuir 흡착 등온식을 잘 따름을 알 수 있었다.

Immobilization and Characterization of Tannase from a Metagenomic Library and Its Use for Removal of Tannins from Green Tea Infusion

  • Tannase (Tan410) from a soil metagenomic library was immobilized on different supports, including mesoporous silica SBA-15, chitosan, calcium alginate, and amberlite IRC 50. Entrapment in calcium alginate beads was comparatively found to be the best method and was further characterized. The optimum pH of the immobilized Tan410 was shifted toward neutrality compared with the free enzyme (from pH 6.4 to pH 7.0). The optimum temperature was determined to be $45^{\circ}C$ for the immobilized enzyme and $30^{\circ}C$ for the free enzyme, respectively. The immobilized enzyme had no loss of activity after 10 cycles, and retained more than 90% of its original activity after storage for 30 days. After immobilization, the enzyme activity was only slightly affected by $Hg^{2+}$, which completely inhibited the activity of the free enzyme. The immobilized tannase was used to remove 80% of tannins from a green tea infusion on the first treatment. The beads were used for six successive runs resulting in overall hydrolysis of 56% of the tannins.

UV/TiO2와 유동층 반응기를 이용한 안료의 광촉매 탈색 (Photocatalytic Decolorization of Dye usingUV/TiO2 and Fluidized Bed Reactor)

  • The photocatalytic decolorization of Rhodamine B (RhB) was studied using immobilized $TiO_2$ and fluidized bed reactor. Immobilized $TiO_2$(length: 1$\~$2 mm, width: 1$\~$3 mm, thickness: 0.5$\~$2 mm) onto silicone sealant was employed as the photocatalyst and a 30 W germicidal lamp was used as the light source and the reactor volume was 4.8 L. The effects of parameters such as the amounts of photocatalyst, initial concentration, initial pH, superficial velocity, $H_2O_2$ and anion additives. ($NO_3^{-},\;SO_4^{2-},\;Cl^{-},\;CO_3^{2-}$) The results showed that the optimum dosage of the immobilized $TiO_2$ were 87.0 g/L. Initial removal rate of RhB of the immobilized $TiO_2$ was 1.5 times higher than that of the powder $TiO_2$ because of the adsorption onto the surface of immobilized $TiO_2$ In the conditions of acidic pH, initial reaction rate was increased slowly and reaction time was shorted. The effect of anion type on the reaction rate was not much.

Identification of Dinitrotoluene Selective Peptides by Phage Display Cloning

  • Biomolecules specific to explosives can be exploited as chemical sensors. Peptides specific to immobilized dinitrotoluene (DNT) were identified using a phage display library. A derivative of DNT that contained an extended amine group, 4-(2,4-dinitrophenyl)butan-1-amine, was synthesized and immobilized using a self-assembled monolayer surface on gold. Filamentous M13 phages displaying random sequences of 12-mer peptides specific to the immobilized DNT-derivate were isolated from the M13 phage library by biopanning. A common peptide sequence was identified from the isolated phages and the synthesized peptides showed selective binding to DNT. When the peptide was immobilized on a quartz crystal microbalance (QCM) chip, it showed a binding signal to DNT, while toluene barely showed significant binding to the QCM chip. These results demonstrate that peptides screened by biopanning against immobilized DNT can be useful for quick and accurate detection of DNT.

Removal of Strontium Ions by Immobilized Saccharomyces Cerevisiae in Magnetic Chitosan Microspheres

  • A novel biosorbent, immobilized Saccharomyces cerevisiae in magnetic chitosan microspheres was prepared, characterized, and used for the removal of $Sr^{2+}$ from aqueous solution. The structure and morphology of immobilized S. cerevisiae before and after $Sr^{2+}$adsorption were observed using scanning electron microscopy with energy dispersive X-ray spectroscopy. The experimental results showed that the Langmuir and Freundlich isotherm models could be used to describe the $Sr^{2+}$ adsorption onto immobilized S. cerevisiae microspheres. The maximal adsorption capacity ($q_m$) was calculated to be 81.96 mg/g by the Langmuir model. Immobilized S. cerevisiae was an effective adsorbent for the $Sr^{2+}$ removal from aqueous solution.

  • The performance of immobilized fungal cells on celite beads for the production of gibberrelic acid was investigated in flasks and 7-L stirred-tank reactor. Repeated incubations of immobilized fungal cells increased cell concentrations and volumetric productivity. The maximum volumetric productivity obtained in the immobilized-cell culture was 3-fold greater than that in suspended-cell culture. The concentration of cotton seed flour (CSF), among the various nutrients supplied, most significantly influenced productivity and operational stability. Notably, insoluble components in CSF were found to be essential for production. CSF at 6 g/L with 60 g/L glucose was found to be optimal for gibberellic acid production and stable operation by preventing excessive cell growth.

  • In this report, we describe the use of gold nanoparticles (AuNPs) immobilized on pH. responsive, cross-linked poly(4-vinylpyridine) (P4VP) thin films, as a potential application for pH nanosensors. The methodology is based on the variation in surface plasmon resonance of immobilized AuNPs with changing the interparticle distances, caused by the swelling/deswelling of the pH responsive P4VP polymer films. The change in optical properties of the immobilized AuNPs in response to the pH of surrounding media was investigated by a simple yet powerful tool; UV-vis absorption spectroscopy. The swelling of the P4VP chains at pH 2 causes an increase in the interparticle distances of immobilized AUNPS ($\sim20nm$) and hence leads to a blue shift of 48 nm in their surface plasmon resonance band peak. On the other hand, when the surrounding media was altered from pH 2 to 10, a red shift of absorption maxima was observed. The changes were rapid, and the effect was reversible. This system could prove to be useful in fabricating nanosensors for detecting the pH or pH changes of surrounding aqueous medium.

Equilibrium and Kinetic Studies of the Biosorption of Dissolved Metals on Bacillus drentensis Immobilized in Biocarrier Beads

  • Biocarrier beads with dead biomass, Bacillus drentensis, immobilized in polymer polysulfone were synthesized to remove heavy metals from wastewater. To identify the sorption mechanisms and theoretical nature of underlying processes, a series of batch experiments were carried out to quantify the biosorption of Pb(II) and Cu(II) by the biocarrier beads. The parameters obtained from the thermodynamic analysis revealed that the biosorption of Pb(II) and Cu(II) by biomass immobilized in biocarrier beads was a spontaneous, irreversible, and physically-occurring adsorption phenomenon. Comparing batch experimental data to various adsorption isotherms confirmed that Koble-Corrigan and Langmuir isotherms well represented the biosorption equilibrium and the system likely occurred through monolayer sorption onto a homogeneous surface. The maximum adsorption capacities of the biocarrier beads for Pb(II) and Cu(II) were calculated as 0.3332 and 0.5598 mg/g, respectively. For the entire biosorption process, pseudo-second-order and Ritchie second-order kinetic models were observed to provide better descriptions for the biosorption kinetic data. Application of the intra-particle diffusion model showed that the intraparticle diffusion was not the rate-limiting step for the biosorption phenomena. Overall, the dead biomass immobilized in polysulfone biocarrier beads effectively removed metal ions and could be applied as a biosorbent in wastewater treatment.

효모 Candida tropicalis 고정화 담체를 이용한 Airlift 미생물반응기의 톨루엔 제거 및 미생물 성장 (Toluene Removal and Microbial Growth of Candida tropicalis Immobilized with Polymer Media in Airlift Bioreactors)

  • This study was conducted to improve biological degradation efficiency of toluene as a model volatile organic compound (VOC) using yeast Candida tropicalis and to suggest an effective method for bioreactor operation. The yeast strain was immobilized with polyethylene glycol (PEG), alginate, and powdered activated carbon (PAC). The yeast-immobilized polymer media were used as fluidized materials in an airlift bioreactor. Polymer media without PAC were also made and operated in another airlift bioreactor. The two bioreactors showed toluene removal efficiencies ranging 80-96% at loading rates of $10-35 g/m^3-hr$, and the bioreactor containing the polymer media with PAC achieved higher removal efficiency. Protein contents in the liquid phase showed that the bioreactor using the yeast-immobilized polymer media with PAC had a higher rate of microbial growth initially than that without PAC. In addition, the microbial growth rate inside of the polymer media with PAC was five times higher than that without PAC. Consequently, the polymer media containing the yeast strain and PAC could enhance removal efficiencies for VOCs, and the immobilization method improve microbial activity and stability for a long-term operation of biological systems.
