The purpose of this study was to investigate the potential hypolipidemic effect of adlay extract in streptozotocin-induced diabetic rats. Fifty six rats were fed either a control diet or adlay extract diets of : Methanol 1%(Ml%) ; Methanol 2%(M2%) ; Methanol 4%(M4%) ; Chloroform-Methanol 1%(CMl%) ; Chloroform-Methanol 2%(CM2%) ; Chloroform-Methanol 4%(CM4%) for 3 weeks. The amount of extracts added was 1%, 2% or 4% by diet weight respectively. The levels of glucose, total cholesterol(TC), high-density-lipoprotein cholesterol (HDL-C), free-fatty acid(FFA) and triglyceride(TG) in plasma, liver and skeletal muscle were compared. Among diabetic rats, there were no significant differences in the plasma level of glucose and TC regardless of a different extraction procedure or different amount of extracts added. While the plasma 7G level tended to increase with times passed in diabetic control group, was not increased with times passed in CM groups and was significantly lower in CM groups at 3rd week. Compared to the diabetic control group, the levels of FFA tended to be lower in all M groups and were not different in all CM groups. The levels of HDL-cholesterol were not different in all M groups and were significantly lower in all CM groups than diabetic control group. Compared to diabetic control group, liver triglyceride level was lower in M4% group and no significant difference was seen in Ml%, M2% and all CM groups. Muscle triglyceride level tended to be lower in Ml%, M2% and CM2% group and significantly was lower in M4% and CM4% group. Thus, it can be suggested that a CM extract of adllay could have a potential hypotriglyceridemic effect on diabetic subjects. (Korean J Nutrition 31(5) : 921∼926, 1998)
Twenty college women were led experimental diet which composed ot basal diet plus different kinds of dietary rats at 27% Cal. Equal amount of 13.5g of corn oil, perilla oil or fish oil was supplied for 2 weeks as a source of n6 linoleic acid(LA). n3 $\alpha$-linolenic acid (LL). or n3 EPA + DHA. respectively. Plasma total Chol level was reduced by perilla and fish oils, significantly only by fish oil. Plasma Chol level was rather increased by corn oil(P<0.05), but was decreased by double amount of corn oil supplement. Therefore, hypocholesterolemic effect of fatty acids was in the order of n3 EPA+DHA>n3 LL>n6 LA and influenced by the degree of fat unsaturation. Plasma TG level was also significantly decreased by n3 EPA+ DHA and increased by n6 LA. Hypotriglyceridemic effect of fatty acids was also in the order of n3 EPA + DHA> n3 LL>n6 LA and influenced by the unsaturation. However, the reduction of plasma TG was more influenced by the fatty acid structure rather than the fat unsaturation. There were no significant effects on lipoprotein pattern 3nd chemical compositions of lipoprotein by different dietary PUFAs. but fish oil diet significantly increased the relative proportion of HDL-Chol. In conclusion. cholesterol- lowering effect of dietary PUFAS seemed to be a function of total fat unsaturation but hypotriglyceridemic effect seemed to be more linked to the ratty acid structure rather than the degree of unsaturation. The hypolipidemic effect of n3 PUFAs was significant so that fish oil or perilla oil may have important nutritional applications in the prevention and treatment of atherosclerotic disease.
To observe the effect of dietary n6 linoleic acid, n6 gamma-linolenic acid and n3 alphalinolenic acid aon plasma lipid composition and platelet aggregation, twenty college women were divided into 4 groups and treated for 2 weeks with experimental diets supplying fat at 23% cal which were different only in fatty acid composition. Dietary fat was corn oil(CO) as a source of n6 linoleic acid(LA), perilla oil(PO) for n3 alpha-linolenic acid(ALA) and evenign primrose oil(EPO) for n6 gamma-linolenic acid(GLA). Plasma cholesterol level was slightly decreased by PL(13.5g) but significantly increased by equal amount of CO. However, there was similar hypocholeaterolemic effect when double amount of CO(27.0g), was supplemented. Therefore, total fat unsaturation may be more important factor for plasma cholesterol-lowering effect than the structure of fatty acid itself. Plasma cholesterol level was not lowered by supplement of GLA in CO diet. There was similar trend in hypotriglyceridemic effect by PO and CO as in plasma cholesterol. Plasma TG level was rather increased but not significantly by GLA supplement to CO diet. Overall, plasma lipid-lowering effect was greater by ALA than LA and GLA effect was not greater than by LA. GLA supplement did not significantly improve lipid compositions to prevent against CHD. There was no significant change both in fatty acid composition in platelet and ADP-induced platelet aggregation by GLA supplement to corn oil diet and by ALA in PO diet in young women.
The present study was designed to observe the effects of soyprotein and casein with or without cholesterol on serum and liver lipids in male rats. The 6 experimental groups were as fellows ; SF ; soyprotein, cholesterol-free diet. SC ; soyprotein, 0.5% cholesterol added diet. CF ; casein, cholesterol-free diet CC ; casein,0.5% cholesterol added diet. SCF ; protein mixture of soyprotein and casein(1 : 1), cholesterol-free diet SCC ; mixed protein, 0.5% cholesterol added diet. The hypocholesterolemic and hypotriglyceridemic effects of soyprotein were observed at 3 weeks, but these effects disappeared at 6 weeks. The hypocholesterolemic effect of soyprotein was more obvious when the 0.5% cholesterol was supplemented in the diets. The serum free cholesterol level was not affected by the dietary protein source or the dietary cholesterol, therefore, the difference in serum total cholesterol among groups seems due to the difference in cholesterol esters. There was a tendency of a higher percentage of HDL in soyprotein groups compared to casein groups at 1 week, however, this tendency disappeared with time. The liver cholesterol and triglyceride contents were not differ among cholesterol-free diet groups, however, with addition of cholesterol, those of soyprotein groups were significantly lower than casein groups. The higher serum arginine/lysine ratio of soyprotein groups may offer the part of explanation of its hypocholesterolemic effect.
Effects of hesperetin and naringenin on the concentration of triacylglycerol in the serum and liver were studied in male golden hamster fed with the semipurified diet containing at 1% level of them for 3 weeks. The concentration of triacylglycerol in serum of the naringenin group decreased by 31%, whereas that in liver increased by 37% compared to the control group. The concentration of triacylglycerol in the serum and liver of the hesperetin group was slightly lower than the control group. The activity of microsomal phosphatidate phosphohydrolase in the liver, which is a key enzyme for biosynthesis of triacylglycerol, was significantly inhibited in the hesperetin group, whereas it was not affected in the naringenin group. The effect of hesperetin on phosphatidate phosphohydrolase was also measured in vitro. Hesperetin decreased the activity of phosphatidate phosphohydrolase with a dose-dependent manner. Both naringenin and hesperetin did not statistically affect the daily food consumption, body weight, liver weight, and total cholesterol in the serum. The observation accounts for the hypotriglyceridemic effect of hesperetin in the hyperlipidemic hamster.
The purpose of this study was to investigate the effect of chloroform(CHC1$_3$) fraction of Alisma canaliculatum All. Braun et Bouche(Ac) with selenium(Se) on the plasma glucose and lipid levels in streptozotocin(STZ)-induced diabetic rats. Male Sprague-Dawley rats were divided into five groups. The normal and diabetic rats were separated into four groups: the STZ-control group, the Ac group, the Ac-Se group and the Se group. Diabetes was induced in the male rats by an injection of STZ into the tail vein at a dose of 45 mg/kg. The CHCl$_3$fraction of Ac(250 mg/kg) was administered orally for 14 days. The supplementation was achieved with the AIN-93 recommended diet by adding 2 mg/kg diet of selenium as Na$_2$SeO$_3$. which was prepared freshly everyday. The body weight, hematocrit(Hct), glucose, insulin, cholesterol, HDL-cholesterol, triglyceride(TG) and free fatty acids(FFA) concentrations in plasma were measured. The aminotransferase activities were also analyzed. The changes of body weight in the experimental groups were not significantly different from that of the STZ-control group, but diabetes hyperphagia accompanied changes with body weight loss in Ac-Se group. The levels of Plasma cholesterol were not significantly different among the experimental groups. The concentrations of FFA in the Ac-Se group increased significantly compared with the STZ-control group. The effect of Se alone significantly increased aspartate aminotransferase activity and alanine aminotransferase activity. The results showed that the treatment of CHCl$_3$fraction of Ac in combination with Se has no synergistic effect. There was a tendency for the plasma glucose levels to decrease when Se was administered into diabetic rats. Supplementation of Se in diabetic rats did not elicit a significant increase in plasma insulin levels and exhibited hypotriglyceridemic effect.
The number of diabetics in Korea is about 3 to 5 percent of the population, and the incidence is increasing yearly due to changes of life style and food intake. Diet is a key element in the management of diabetes, yet the appropriate diet for diabetes remains controversial. We have recently shown that a diet rich in protein of animal origin(casein) seems beneficial to controling plasma glucose and lipids in streptozotocin-induced diabetic rats. It therefore seemed desirable to find out whether the beneficial effect of high casein diet in experimental diabetes could also be reproduced with a vegetable source of protein(soy). The purpose of this study is to compare these results with the results of our previous study. In the present study, non-diabetic and streptozotocin-induced diabetic rats were studied in order to examine the effects of altering the level(20% vs 60%) of dietary soy protein on blood glucose, lipids, and hormones. Results of the present study showed that a high soy protein diet decreased triglyceride concentration in diabetic rats. However, diabetic rats fed a high soy protein diet were not hypocholesterolemic compared to rats fed a control diet. Moreover, diabetic rats fed a high soy protein diet had significantly increased plasma glucose concentration compared to rats fed a control diet. This study was not able to discern a specific effect of dietary protein level on insulin, glucagon, or insulin/glucagon ratio. Except for the hypotriglyceridemic effect, the results were not similar to the findings of our previous study which showed a beneficial effect on streptozotocin-induced diabetic rats fed a high casein diet.
To compare the hypolipidemic effects of n6 linoleic acid. n3 $\alpha$-linolenic acid and n3 eicosapentaenoic acid in rats fed high carbohydrate(70% Cal) diet. male Sprague Dawley rats were fed different experimental diets for 6 weeks. which were different only in fatty acid composition. The dietary fats were beer tallow(BT) as a source of saturated fatty acid (SFA), corn oil(CO) for n6 linoleic acid(LA), perilla oil(PO) for n3 $\alpha$-linolenic acid(LL) and fish oil(FO) for n3 eicosapentaenoic acid(EPA) and docosahexaenoic acid (DHA) Plasma total cholesterol(Chol) level was increased by n6 LA but decreased by n3 LL and n3 EPA and most effectively reduced by n3 EPA. HDL-Chol level was raised by n6 LA, but there was no significant change in HDL-Chol levels by n3 LL and was lowered by n3 EPA. Plasma TG level was reduced by n6 LA, but lipogenesis in liver was not affected by n6 LA. However, plasma TG level was lowered by n3 LL and EPA. Both lipogenic enzyme activity and liver TC level were also decreased by n3 PUFA. The relative proportions of TG in VLDL was significantly lowered by n3 EPA. but the proportions of Apo B in VLDL was not changed by n3 EPA. Overall. the hypolipidemic effect was in the order of EPA+ DHA(n3) >LL(n3) >LA(n6) and fish oil and perilla oil rich in n3 PUFA may have important nutritional applications in the prevention and treatment of hypertriglyceridemia.
This study was to compare the effects of dietary n-6 and n-3 fatty acids and fat unsaturation on plasma lipids and chemical composition of VLDL and LDL fraction and lipogenic enzymes activity in rat liver under the conditions providing 1) a similar amount of n-6, n-3 fatty acids(LA, ALA, EPA+DHA) in diets and 2) the various degree of fat unsaturation. Male Sprague-Dawley rats weighing 420g were treated for 6-n with six experimental diets providing 25% of energy as fat and which were different only in fatty acid composition. The fats used for a source of each fatty acid were beet tallow for saturated fatty acid corn oil for n-6 linoleic acid(LA) perilla oil for n-3 $\alpha$-linolenic acid(ALA) and fish oil n-3 eicosapentaenoic acid (EPA) and n-3 docosahexaenoic acid(DHA). Plasma cholesterol level was increased by corn oil to compare with beef tallow but was decreased by perilla oil or fish oil. Plasma TG level was significantly decreased by perilla oil or fish oil. Fish oil significantly reduced the level of HDL-Chol and the proportion of Chol in LDL fraction and that of TG in vVLDL fraction. Overall there was a singificant negative correlation between the level of each plasma lipid(Chol TG, VLDL-TG, LDL-C) and the degree of fat unsaturation. However this rerlationship is not always true when compared the hypolipidemic effect of each fatty acid at a similar level of fat unsaturation. There was a trend such taht glucose 6-P dehydrogenase 6-phosphogluconate dehydrogenase and malic enzyme activites were reduced by n-3 fatty acids. Perilla oil significantly increased the incorporation of c20:5 and c22:5 into liver tissue and fish oil suignificantly increased the incorporation of c20:5, c22:6 into liver tissue and the effect of long chain n-3 fatty acid incorporation was greater by fish oil. therefore the hypotriglyceridemic effect of n-3 fatty acid could be resulted from the interference of hepatic lipogenesis by long-chain n-3 fatty acids and the reduced proportion of TG in VLDL fraction and its effect was greater by n-3 EPA+DHA than n-3 ALA even though plasma Chol and TG levels were also influenced by the degree of dietary fat unsaturation.
To compare the hypolipidemic effects of n6 and n3 PUFA at different fat levels, male Sprague Dawley rats were fed either low fat (LF, 10% Cal) or high fat (HF, 40% Cal) diet which was different only in fatty acid composition for 6 weeks. Dietary fats were beef tallow, corn oil, perilla oil, and fish oil concentrate as a source of saturated fatty acid, n6 linoleic acid(LA). n3 ${\alpha}-linolenic$ acid(LL) and n3 eicosapentaenoic acid(EPA)+docosahexaenoic acid(DHA), respectively. VLDL fraction was separated by ultracentrifugation and chemical composition was determined by thin layer chromatography. Plasma cholesterol level was increased by n6 LA but decreased by n3 LL and n3 EPA in LF and HF diets, and the hypocholesterolemic effect of n3 EPA was most significant in HF diet. HDL-Chol level was raised by n6 LA in LF and HF diets, but significantly reduced by n3 EPA in HF. Plasma TG level was reduced by n6 LA n3 LL and EPA in LF and HF with the reduction of lipogenic enzyme activity only by n3 PUFAs. The proportion of TG in VLDL fraction was significantly lowered by n3 EPA in LF and HF. The proportion of apo-B in VLDL fraction was not changed in LF, but was significantly decreased in HF by n3 EPA. Therefore, the hypotriglyceridemic effect of n3 PUFA could be from the reduced lipogenesis in liver and resulted in the depressed secretion of TG as VLDL in LF and HF with significant lower production of apoB in HF diet.
