Hypolipidemic and hypoglycemic effects of Orostachys japonicus A. Berger extracts in streptozotocin-induced diabetic rats

  • The hypolipidemic and hypoglycemic effects of two dietary dosages (0.1% and 0.5%) of water and 80% ethanol extracts from hot-air dried Orostachys japonicus A. Berger were evaluated in the serum and organ tissues of streptozotocin-induced diabetic rats. The STZ-induced diabetic groups supplemented with the O. japonicus extracts showed significantly higher body weight compared to a diabetic control group at the end of experiment. The extracts exhibited substantial hypoglycemic effects by significant reductions of fasting blood glucose levels at all time points tested compared to the initial stage before treatment of the extracts. Declines of serum and hepatic triglyceride levels were greater than declines of total cholesterol in the groups treated with the 0.5% O. japonicus extract (DBW2 and DBE2) when compared to the DBC group. Hepatic glycogen content was higher in the groups treated with O. japonicus extract, while lipid peroxide content was decreased in these treated groups compared to the DBC group. Hepatic antioxidant activity was significantly increased in the groups supplemented with the O. japonicus ethanol extract The hypolipidemic and hypoglycemic effects of the O. japonicus ethanol extract were significantly greater than the effects of the water extract. Based on this study, it seems that O. japonicus ethanol extract, due to its higher phenolic and flavonoid components than the water extract, may control blood glucose and alleviate hyperlipidemia in diabetes.

혐기처리한 뽕잎의 혈당강하 효과 (Hypoglycemic Effect of Mulberry Leaves with Anaerobic Treatment in Alloxan-induced Diabetic Mice)

  • The hypoglycemic effect of mulberry leaves using alloxan-induced diabetic mice was investigated. The hypoglycemic effect of mulberry leaves was significantly affected with anaerobic incubation up to for 6 hrs. The body weight of mice treated with 20% anaerobic conditioned mulberry leaves was higher than that in the control group. Mulberry leaves which were treated with anaerobic condition significantly decreased blood glucose level in alloxan-induced type I diabetes mice. Furthermore, glucose tolerance was significantly recovered by feeding mulberry leaves anaerobically treated. Pathological analysis revealed that the disappearance of insulin-secreting beta-cells in the islets of Langerhans from the alloxan induced diabetic mice was strongly inhibited in the mulberry leaves feeding group. The content of 1-deoxynojirimycin increased up to 5% in detached mulberry leaves incubated under nitrogen gas for 6 hrs. Nevertheless, it is not clear whether the synthesis of 1-deoxynojirimycin is increased or the degradation of it is decreased. Overall, results obtained from this study indicate that the hypoglycemic effect was increased by the mulberry leaves with anaerobic treatment.

Hypoglycemic Effect of Yacon Tuber Extract and Its Constituent, Chlorogenic Acid, in Streptozotocin-Induced Diabetic Rats

  • Smallanthus sonchifolius (Yacon, Asteraceae) was originally cultivated in South America and used in food and traditional medicine by Andean inhabitants. Yacon is potentially beneficial for the management of diabetes and is composed of fructooligosaccharides, proteins, minerals and phenolic compounds. The aim of this study was to investigate the hypoglycemic effect of Yacon tuber extract (YTE) and its constituent, chlorogenic acid (CGA), in streptozotocin (STZ)-induced diabetic rats. In this study, a HPLC method was developed for simultaneous determination of major active phenolic components, CGA and caffeic acid in YTE. We investigated the hypoglycemic effect of YTE and CGA in STZ-induced diabetic rats and studied glucose tolerance test (GTT). The effect of orally administered multiple doses of YTE and CGA on plasma biochemical parameters was examined using diabetic rats. We also measured free radical scavenging activity by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. Oral administration of YTE (200 mg/kg) and CGA (10 mg/kg) for 6 weeks produced a significant hypoglycemic effect in STZ-induced diabetic rats. YTE and CGA-treated groups exhibited significantly decreased plasma glucose surge during the GTT. Total cholesterol (TC) and triglyceride (TG) concentrations were significantly decreased by 33% and 49%, respectively, in YTE-treated rats. TC and TG concentrations were also significantly decreased by 26 % and 41%, respectively, in CGA-treated rats. In the DPPH assay, free radical scavenging activity of CGA was similar to that of vitamin E, a positive control. This study suggests that YTE and its constituent, CGA, may be a useful option for management of hyperglycemia and diabetic nephropathy.

표고버섯 균사체의 생산과 당뇨 유발 흰쥐에 대한 혈당강하 효과 (Production of Lentinus edodes Mycelia in Submerged Culture and It's Hypoglycemic Effect in Diabetic Rats)

  • 표고버섯 균사체가 항당뇨 효과에 미치는 영향을 검토하기 위해 당뇨가 유발된 흰쥐에 표고버섯 균사체을 7일간 급여시킨 후 혈장 glucose, total cholesterol, triglyceride 및 ALT, AST 활성을 관찰하였고, 표고버섯 균사체 생장의 최적 배양 조건을 조사하였다. 균사체 생장을 위한 최적의 pH와 온도는 5와 $30^{\circ}C$로 나타났으며, 발효조배양시에는 최대 균사체 생장(8.13g/l)이 진탕배양일 때보다 6일 단축되었다. Hypoglycemic 효과에서 혈장 glucose는 당뇨 유발군에서 표고버섯 균사체 급여군이 대조군보다 23.0%가 유의하게 감소하였으며, 혈장 total cholesterol과 triglyceride는 20.3%와 27.1%로 낮게 나타났다. 또한 혈장 ALT와 AST 활성은 당뇨 대조군에 비해 표고버섯 균사체 급여군이 29.6%와 34.3% 낮은 활성을 보였다.

Hypoglycemic Effect of Fermented Soymilk Extract in STZ-induced Diabetic Mice

  • This study investigated the hypoglycemic effect of fermented soymilk extract (FSE) in STZ-induced diabetic mice. FSE was prepared via fermentation of soymilk with Bacillus subtilis followed by methanol extraction. The hypoglycemic effect was determined by inhibitory activities against ${\alpha}$-glucosidase and ${\alpha}$-amylase as well as the alleviation of postprandial glucose level. The non-fermented soymilk extract (SE) was used as control in this experiment. FSE showed higher (p<0.05) inhibitory activities than SE against ${\alpha}$-glucosidase and ${\alpha}$-amylase. The $IC_{50}$ values of FSE for ${\alpha}$-glucosidase and ${\alpha}$-amylase were 0.77 ancd 0.94 mg/mL, respectively, which were comparable or even superior to those of acarbose (0.79 and 0.68 mg/mL, respectively). In addition, a further suppression on the postprandial blood glucose levels were observed in the FSE than SE group for both STZ-induced diabetic mice and normal mice. Furthermore, FSE significantly lowered the incremental area under the curve (AUC) in the diabetic mice and the AUC in normal mice corroborated the hypoglycemic effect of FSE (p<0.05). Results from this study suggest that FSE may help decrease the postprandial blood glucose level via inhibiting ${\alpha}$-glucosidase and ${\alpha}$-amylase and the usefulness of FSE was proven to be better than SE.

큰번데기동충하초(Cordyceps militaris)의 혈당강하효과 (Hypoglycemic Effect of Cordyceps militaris)

  • Cordyceps militaris (CM) has been used as a tonics in the traditional medicine. To investigate the anti-diabetic principle of CM, activity guided fractionation was conducted. Hot water extract of CM was fractionated into 3 parts: above 100,000(A), $100,000{\sim}20,000(B)$, below 20,000(C) in molecular weight using in membrane filter system. All fractions showed mild hypoglycemic activity in streptozotocin (STZ)-induced diabetic rats by oral administration (300 mg/kg). The fraction C which was most active among them was fractionated again into two parts, C-1 and C-2 by Sephadex LH 20 column chromatography. The fraction C-1 showed hypoglycemic activity but C-2 did not show activity compared with control in STZ mice. In glucose-fed hyperglycemic mice, fraction C, C-1 and C-2 also showed significant glucose lowering activity. Their decreasing rates of plasma glucose level after 1 hours administrations of fraction C, C-1 and C-2 were 24.5%, 29.3% and 22.0%, respectively (Tolbutamide: 48.4%). These results suggested that CM has both insulin like and insulin release promoting activity and could be developed as an antidiabetic agent.

Picrotoxin의 경련(痙攣) 및 血糖增加作用(혈당증가작용)에 미치는 Lithium과 Ethanol의 영향(影響) (Influncences of Litium and Ethanol on the Convulsion and Hyperglycemia Induced with Pircotoxin)

  • It is well known that picrotoxin, an amaroid substance of Anamirta cocculus, is a classic stimulant on the central nervous system accompanying convulsive activity, and it liberates catecholameine from the adrenal mdulla through its central action to increase blood sugar level. Schou reported that lithium and alcohol have the similar inhibitory property on the $Na^+,\;K^+$-ATPase activity, and recently, the therapeutic efficacies of lithium on the alcohol withdrawal syndrome and the chronic alcoholics have been studied. Many studies about the hypoglycemic effect of lithium and alcohol were reported but the interaction between those hypoglycemic action is little known. Therefore, in this paper, the hypoglycemic effect of lithium and ethanol on the hyperglycemia induced with picrotoxin, and the interaction of them in those hypoglycemic action were investigated with reference to the anticonvulsive action of them. The results were obtained as follows: 1. The convulsive dose (: $CD__{50}$) of picrotoxin in mice was slightly increased by the pretreatment of lithium or ethanol. 2. The blood sugar level was markedly increased by picrotoxin but the level was sugar level was significantly decreased by lithium, ethanol or both. 3. The hyperglycemic effect of picrotoxin was significantly potentiated by the lithium pretreatment, but the potentiation effect of lithium was markedly suppressed by the additional injection of ethanol after lithium injection and more markedly suppressed by the premedication of ethanol before lithium injection 4. The hyperglycemic effect of picrotoxin was markedly inhibited by the ethanol pretreatment, and the inhibitory effect of ethanol was significantly strenthened by the additional injection of lithium after ethanol injection, but on the contrary, the inhibitory effect was completely disappeared by the premedication of lithium before ethanol injection.

The Hypoglycemic Effect of Adly Diet is not Significant when the Amount of Total Fiber Consumption is Controlled

  • The purpose of this study was to investigate the potential hypoglycemic effect of adlay diets when total fiber consumption was controlled in streptozotocin-induced diabetic rats. Forty eight rats were fed for 3 weeks with either controlled in streptozotocin -induced diabetic rats. Forty eight rats were fed for 3 weeks with either control diets or experimental diets : raw mille adlay (RMA) raw whole adlay(RWA) , steamed milled adlay(SMA) or roasted mille adlay(OMA). The composition of the AIN-76 diet was modified to ensure the same composition of protein, carbohydrate , rat, and fiber between the control diet and experimental diets. The concentrations of glucose , insulin, glycogen, and protein in plasma, liver, or skeletal muscle were compared . Compared to diabetic control rats, plasma postprandial glucose levels tended to be decreased in RMA, RWA, SMA and OMA rats until the 2nd week, but no difference was shown at the 3 rd week. There was no significant difference in insulin levels among those groups. After glucose loading, the plasma glucose level of SMA was lower than that of diabetic control rats throughout 2 hrs. Liver glycogen was lower than control values in RMA and RWA rats and not different in SMA and OMA rats. The muscle protein level of RMA, RWA, SMA, and OMA rats tended to be lower than in diabetic control rats. There was no significant difference in muscle glycogen among groups. These results suggest that the hypoglycemic effect of an adlay diet is not significant when the amount of total fiber consumption is controlled.

돼지감자 분말 첨가 쿠키의 품질특성 및 혈당에 미치는 영향 (The Quality Characteristics and Hypoglycemic Effect of Cookies Containing Helianthus tuberosus Powder)

  • This study was conducted to evaluate the quality characteristics and the hypoglycemic effect of cookies containing Helianthus tuberosus powder (HTP). To examine the effect of adding HTP, cookies with various HTP contents (0%, 10%, 20%, 30%, 40%) were made and compared for their quality characteristics. The pH of cookie doughs decreased significantly (p<0.05) as the content of HTP increased. However, the density did not show significant difference among samples. The spread ratio and loss rate in cookies showed significant increase (p<0.05) as the content of HTP increased. However the leavening rate and the hardness of the cookies decreased significantly (p<0.05) when HTP content increased. Color measurement also showed significant difference among samples, although the cookie sample with 10% HTP powder showed the highest scores from consumer tests and liking score, the cookie samples with 30% HTP were selected to measure the hypoglycemic effect of Helianthus tuberosus compared to control cookies and bread. Ten healthy subjects in their twenties participated in measuring blood glucose levels after intake of bread, control cookie, or 30% HTP cookie. Blood glucose level was measured after fasting 12 hours, before eating, and 30, 60, 90, and 120 minutes after taking the samples. As a result, the blood glucose levels at 30min after meals showed significant (p<0.05) difference among treatments: the lowest when taken the cookies added 30% HTP. However there was no difference in the blood glucose levels at 60min, 90min, and 120min after the sampling.

Streptozotocin 유발 당뇨 흰쥐에서 Tabebuia avellandae의 항당뇨 효과 (Hypoglycemic Effect of Tabebuia avellandae on Streptozotocin-Induced Diabetic Rats)

  • Hypoglycemic effect of Tabebuia avellandae was investigated in the streptozotocin(STZ)-induced diabetic rats. Diabetes was induced in male Sprague-Dawley rats by injections of STZ (45 mg/kg, i.v.). Rats weighing 200-250 g were divided into 6 groups: normal, STZ-control, hexane fr., CHCl$_3$fr., BuOH fr. and $H_2O$ fr. group. Normal and STZ-control rats received 3% Tween 80 only. Four groups of diabetic rats were administered orally at doses of 100, 400, 300 and 400 mg/kg/day of hexane, CHC1$_3$, BuOH and $H_2O$ fr. respectively. Fractions were administered orally to the rats for 7 days after STZ injection. All rats were anesthetized with ether, blood samples were taken by cardiac puncture for clinical chemistry and the rats were killed by exsanguination. Liver, kidney, heart and spleen were removed, weighed and analyzed. We measured glucose, protein, cholesterol and triglyceride levels in the plasma and glycogen, cholesterol and triglyceride levels in liver. The extent of blood glucose decrement in rats administered $H_2O$ fraction was greater than that in the STZ-control rats. The serum cholesterol and triglyceride levels were significantly lowered by administration of $H_2O$ fraction compared with those of STZ-control group. Treatment of rats with Tabebuia avellandae fractions caused decreases in STZ-induced elevation of cholesterol and triglyceride. Liver triglyceride level was significantly lowered hexane and BuOH fraction group compared with STZ-control group. These results suggest that $H_2O$ fraction of Tabebuia avellandae has the hypoglycemic action against STZ-induced diabetic rats.
