IAA Synthesis and Polyamine Metabolism in Higher Plants: Effects of IAA on the Activity of Diamine Oxidase in Seedlings of Soybean (고등식물에서 IAA합성과 Polyamine 대사에 관한 연구: 대두의 유식물에서 IAA가 Diamine Oxidase의 활성에 미치는 영향)

  • Vie investigated the effect of indole acetic acid (IAA) on diamine oxidase (DAO; EC 1.4.3.6) in the seedling of soybean (Glycine max L. forma Paldalkong). DAO activity was not detected in the resting soybean seeds. During germination it appeared in the elongating zone on day 3, increased up to day 4 and decreased thereafter. Endogenous IAA content has been shown the same pattern as DAO activity. However, cadaverine content was reduced on day 4. To investigate the effect of IAA on DAO activity, the segments of hypocotyl on day 3 were soaked in various concentration of IAA. The activity increased at low concentrations ($10^{-7}-10^{-6}M$) of 1M as compared to that of control but not at high concentrations ($10^{-5}-10^{-4}M$) of IAA. Differing from DAO activity, cadaverine content increased as concentrations of IAA increased. On the other hand, ethylene was induced by IAA at high concentrations. To study the effect of ethylene on DAO activity, we cotreated IAA and 2,S-Norbonadiene (NDE). In this case, DAO activity was not affected.fected.

In vitro grown thickened taproots, a new type of soil transplanting source in Panax ginseng

  • Background: The low survival rate of in vitro regenerated Panax ginseng plantlets after transfer to soil is the main obstacle for their successful micropropagation and molecular breeding. In most cases, young plantlets converted from somatic embryos are transferred to soil. Methods: In vitro thickened taproots, which were produced after prolonged culture of ginseng plantlets, were transferred to soil. Results: Taproot thickening of plantlets occurred near hypocotyl and primary roots. Elevated concentration of sucrose in the medium stimulated the root thickening of plantlets. Senescence of shoots occurred following the prolonged culture of plantlets. Once the leaves of plantlets senesced, the buds on taproots developed a dormant tendency. Gibberellic acid treatment was required for dormancy breaking of the buds. Analysis of endogenous abscisic acid revealed that the content of abscisic acid in taproots with senescent shoots was comparatively higher than that of taproots with green shoots. Thickened taproots were transferred to soil, followed by exposure to gibberellic acid or a cold temperature of $2^{\circ}C$ for 4 mo. Cold treatment of roots at $2^{\circ}C$ for 4 mo resulted in bud sprouting in 84% of roots. Spraying of 100 mg/L gibberellic acid also induced the bud sprouting in 81% roots. Conclusion: Soil transfer of dormant taproots of P. ginseng has advantages since they do not require an acclimatization procedure, humidity control of plants, and photoautotrophic growth, and a high soil survival rate was attained.

Identification of Proteins Phosphorylated by Protein Kinase C in Soybean (대두 유식물에서 Protein Kinase C에 의해서 인산화되는 단백질의 동정)

  • The previous report (Chung and Lee, 1992) in our laboratory demonstrated that the protein kinase C (PKC) activator, TPA, promotes the elongation of corn coleoptiles significantly. To understand the role of TPA on the growth, substrates of PKC were investigated using PKC partially purified from soybean by DEAE-52 cellulose column. The enzyme activity increased about 5-fold in the presence of $Ca^{2+}$, phosphatidylserine and diolein compared with that in the absence of these reagents. Phosphorylation of both cytosol and membrane proteins by the purified PKC increased in the presence of $Ca^{2+}$ compared with that of EGTA treatment. However, the phosphorylation did not increase markedly by treatment with TPA or phosphatidylserine and diolein in the presence of $Ca^{2+}$ compared with $Ca^{2+}$ alone. The decrease, in phosphorylation of 100, 61 and 43 Kd proteins of the cytosol, and 140, no, 66, 47 and 32 Kd membrane proteins in hypocotyls, and 140, no, 66, 47, 33, 31 and 16 Kd membrane proteins in the root was observed in the presence of PKC inhibitor staurosporine (5T A). These results suggest that subatrates of PKC in soybean may be 110, 63 and 41 Kd proteins of the cytosol, and 140, 110, 66, 47 and 32 Kd membrane proteins in the subapical region of the hypocotyl, and 140, 110, 66, 47, 33, 31 and 16 Kd membrane proteins of the root.e root.

Effect of Salinity on the Seed Germination and Seedling Growth of Pinus densiflora for. erecta Uyeki (염도가 금강소나무의 종자발아와 유묘성장에 미치는 영향)

  • The effect of salinity on the seed germination and seedling growth of Pinus densiflora for. erecta Uyeki were studied under the controlled conditions in the growth chamber. The seeds were sorted into three classes in weight such as large ($15.49\pm$1.63mg), medium($10.61\pm$1.38mg), and small ($6.57\pm$1.33mg) to determine the role of seed weight in germination and seedling growth of the pine. Polymorphic seeds of the pine were germinated an salinity range of 0 to 1.5% NaCl under various temperature ($10^{\circ}$ $-25^{\circ}$ with $5^{\circ}$C interval of constant temperature, and $10^{\circ}$ $-20^{\circ}$C, $15^{\circ}$ $-25^{\circ}$C of alternating temperature) in order to determine their germinability and seeding growth. In control plot, there was little difference of germination percentage among the seed weight classes, but in saline plot, the larger seeds generally had a higher percentage and rate of germination. There occurred synergistic interaction between salinity and temperature in the germination and the increase of temperature enhanced germination of seeds at the same salinity level. Alternating temperature regimes of $15^{\circ}$-$25^{\circ}$C yielded maximum germination and no germination was occurred at $10^{\circ}C$. The germination at alternating temperature showed higher germination percentage than at constant temperature. The percentage and of germination decreased drastically with increased salinity level to 1.00%; no germination was occurred at 1.50% salinity level. The growth fo sddelings from larger seeds was better than that of smaller seeds at the same salinity and temperature. The hypocotyl and radicle were more sensitive than cotyledon to the increased salinity stress.

Analysis of Dietary Fiber of 66 Korean Varieties of Sprout Beans and Bean Sprouts

  • The present study was conducted to determine a high-fiber variety of sprout bean and bean sprout. Sixty-six varieties of Korean sprout beans and thirty varieties of Korean bean sprouts were analyzed for their total dietary fiber (TDF) using a combination of enzymatic and gravimetric methods adopted by AOAC. The average and range of the TDF contents of the 66 varieties of sprout beans were 21.73$\pm$2.43% and 16.60~29.2% (dry basis), respectively. The top five types of sprout beans with high TDF anions the 66 samples were KLG10658 (29.2%), Dawonkong (28.46%), Sohokong (25.66%), Moohankong (25.465%) and Samnamkong (24.94%). The mean TDF content of sprout beans was relatively high in the variety with yellow seed coat color and a smaller seed size (<15 g/100 seeds) than the ones with other colors and of medium or large seed size. The average and range of the TDF content of the 30 varieties of bean sprouts were 24.48$\pm$3.9% and 18.3~33.38%, respectively. Dawonkong sprouts showed the highest TDF content (33.38%). The average TDF content of bean sprouts was 1.16 times higher than that of the corresponding seed beans. The TDF content of bean sprouts did not significantly change during the days of culture. The TDF content in the different parts of the bean sprout were high in the order of roots, hypocotyl and cotyledon.

  • Treatment of ethylacetate extract of Asiasarum sieboldi inhibited the germination and the growth of radish seeds. Two phenylpropanoids were isolated from ethylacetate extract. Their structures were identified as safrole and o-methyleugenol by spectroscopic evidence. From the test to inhibitory effect, o-methyleugenol had inhibited the germination and the growth of radish seeds, while safrole did not. The germination rate and radicle length of radish seeds were decreased to 63.0%, 31.5 % of control at 5mg/ml of o-methyleugenol, respectively. At the same concentration, o-methyleugenol inhibited the hypocotyl growth up to 100%.

Comparison of Physicochemical and Sensory Characteristics of Soybean Sprouts from Different Cultivars (나물콩 품종별 콩나물의 물리화학적 및 관능적 특성 비교)

  • Physicochemical and sensory characteristics of soybean sprouts from domestic and foreign soybean cultivars were investigated. 100-seed weights of domestic cultivars were much larger than that of Canadian cultivar and recommended cultivars were larger than traditional cultivars. Water absorption of soybeans increased sharply to initial 6 hrs during steeping, and did not change much thereafter. In all soybean cultivars 16 amino acids were detected and the contents of Asp, Glu, Lys and Arg were more than 50% of total amino acid contents. All soybean cultivars showed some difference in composition of fatty acids and the content of linoleic acid was highest. The contents of K and P were high and those of Fe and Zn were low. The compression force of hypocotyl after cultivation was the highest in Eunha and the lowest in Junjeori and the compression force of cotyledon was the highest in Danyeop and the lowest in Canadian cultivar. Qualitative discription analysis for cooked soybean sprouts and soups showed significant difference in 6 characteristics of yellowness of cotyledon, size of cotyledon and so on.

  • The changes in peroxidase and catalase activities, and isoperoxidase patterns in different parts of mung bean seedling caused by the treatment with plant growth substances, $GA_3$ and ABA, were examined. As germination proceeded, the activity of peroxidase in all part except hypocotyl was increased, while that of catalase decreased. The separate application of $GA_3$ and ABA increased the activity of peroxidase which was more influenced by $GA_3$ than by ABA only in cotyledon, while that of catalase was more affected by ABA than by $GA_3$. Electrophoretic study revealed that the number of isoperoxidase was increased continuously in all parts during development. A greater influence was exerted on the intensity of isozyme than the number of isozyme by the hormonal treatment.

  • Plants were regenerated from hypocotyl protoplast culture of cabbage (F$_1$hybrid Green Challenger) and were transplanted on fields. The ploidy level of regenerated and seed-grown plants was measured by flow cytometry. In total 125 regenerated plant, diploid (2n), tetraploid(4n), and mixoploid (2n+4n) were 72.8%, 25.6%, and 1.6%, respectively. Most of the regenerated plants had normal phenotype, whereas several plane showed abnormal phenotype such as heavy leaf incision, savoy, and wave. The regenerated plants with abnormal phenotype and different ploidy level were analysed by isozyme and RAPD, but no significant difference was found.

  • Transgenic lettuce plants were successfully obtained from hypocotyl explants inoculated with Agrobacterium tumefaciens, which harbored a binary vector plasmid with Bcl-2 gene, related to apoptosis. After culture and selection on MS medium a number of kanamycin-resistant plantlets were regenerated. Polymerase chain reaction, Southern blot analysis and Northern blot analysis were used to identify and characterize the transgenic plants with the integrated Bcl-2 gene. Over 100 transgenic plants have been established in soil and flowered in the greenhouse. T1 progeny of 100 transgenic lettuce inbred lines were inoculated with Sclerotinia sclerotiorum. Expression of the Bcl-2 peptide in transgenic lettuce plants provides high levels of field resistance against Sclerotinia sclerotiorum, causal agent of the agronomically important fungal disease of lettuce.
