Suppression of IgE and GATA-3-dependent Th2 Cell Regulation by PM-E and PM-70M Isolated Polygonum Multiflorum (하수오에서 분리한 PM-E와 PM-70M이 GATA-3의존성 Th2세포 조절과 IgE 억제 효과)

  • This study examined the effects of GATA-3-dependent Th2 cells and IgE inhibition by using PM-E and PM-70M isolated from Sasao. It was analyzed that the 70 % methanol layer contained adsorbed chromatograms of the fraction of sodium hypochlorite that inhibited GATA-3 transcription factor activity. As a result, PM-70 %MFL fraction seems to have an antiallergic effect by inhibiting GATA-3 which regulates Th2 cytokine. PM-30A, PM-70A, and PM-30A layers by inhibiting IgE secretion in B cells by co-culturing the fractions with anti-CD40/rmIL-6 and TNF-${\alpha}$ mRNA and IgE were not different from control. However, expression of IL-6 and TNF-${\alpha}$ mRNA was $0.69{\pm}0.058$ (p<0.001) and $0.72{\pm}0.58$ (p<0.05), respectively. The amount of IgE secretion was $94.6{\pm}16.0$, which was significantly decreased by 45.6% (p<0.01) compared with the control group. This study suggests that the PM-70% MFL layer in the fraction of Sasao contains the components that inhibit the differentiation and activity of B cells.

Recovery of Metallic Pd with High Purity from Pd/Al2O3 Catalyst by Hydrometallurgy in HCl (염산 침출용액을 이용한 Pd/Al2O3 촉매에서 고순도 팔라듐 회수)

  • Palladium (Pd) has been widely used in various industrial applications such as jewelry, catalyst, and dental materials despite its limited resources. It has been gaining attention to recover Pd with high purity from the spent materials. This study investigated the optimum conditions for the leaching and recovery of metallic Pd. The leaching parameters are HCl concentration, temperature, time, concentration of oxidants, and pulp density. 97.2% of Pd leaching efficiency was obtained in 3 M HCl with 3 vol% oxidants at 80℃ for 60 min. The ratio of hydrogen peroxide to sodium hypochlorite played a critical role in the leaching efficiency due to the supply of Cl- ions in the leachate. Moreover, the complete recovery of Pd in the leachate was achieved at 80℃ with 0.3 formic acid/leachate after adjusting the pH value of 7. This situation was ascribed to the decomposition of formic acid into hydrogen gas and carbon dioxide at 80℃. ICP-AES and XRD characterized the recovered Pd powder, and the purity of the recovered powder was found to be 99.6%. Consequently, the recovered Pd powder with high purity could be used in circuits, catalyst precursors, and surgical instruments.

A Study on the Frictional Characteristics of Fiber Reinforced Composites under Corrosive Environment (부식 환경 하에서의 섬유강화복합재료의 마찰 및 마모 특성 연구)

  • The treated water inside the ballast electrolytic cell creates a highly alkaline atmosphere due to hydroxide generated at the DSA(Dimension Stable Anode) electrode during electrolysis. In this study, a composite material that can replace the weakness of the PE-coated steel pipe used in the existing ballast pipe was prepared. The test samples are BRE(Basalt fiber reinforced epoxy), BRP(Basalt fiber reinforced unsaturated polyester), GRE(Glass fiber reinforced epoxy), and GRP(Glass fiber reinforced unsaturated polyester). And then it was immersed in NaOH for 720 hours. The friction test of each specimen was conducted. The Friction coefficient analysis according to material friction depth and interfacial adhesion behavior between resin and fiber were analyzed. As a result, the mechanism of interfacial separation between resin and fiber could be analyzed. In the case of the unsaturated polyester resin with low interfacial bonding strength the longer the immersion time in the alkaline solution, the faster the internal deterioration caused by the deterioration that started from the surface, resulting in a decrease in the friction coefficient. It is hoped that this study will help to understand the degradation behavior of composite materials immersed in various chemical solutions such as NaOH, acid, and sodium hypochlorite in the future.

Cleaning Methods to Effectively Remove Peanut Allergens from Food Facilities or Utensil Surfaces (식품 시설 또는 조리도구 표면에서 땅콩 알레르겐을 효과적으로 제거하는 세척 방법)

  • Peanut is a well-known food allergen that causes adverse reactions ranging from mild urticaria to life-threatening anaphylaxis. Consumers suffering from peanut allergies should thus avoid consuming undeclared peanuts in processed foods. Therefore, effective cleaning methods are needed to remove food allergens from manufacturing facilities. To address this, wet cleaning methods with washing water at different temperatures, abstergents (peracetic acid, sodium bicarbonate, dilute sodium hypochlorite, detergent), and cleaning tools (brush, sponge, paper towel, and cotton) were investigated to remove peanuts from materials used in food manufacture, including plastics, wood, glass, and stainless steel. Peanut butter was coated on the surface of the glass, wood, stainless steel, and plastic for 30 min and cleaned using wet cleaning. The peanut residue on the cleaned surfaces was swabbed and determined using an optimized enzyme-linked immunosorbent assay (ELISA). Cleaning using a brush and hot water above 50℃ showed an effective reduction of peanut residue from the surface. However, removing peanuts from wooden surfaces was complicated. These results provide information for selecting appropriate materials in food manufacturing facilities and cleaning methods to remove food allergens. Additionally, the cleaning methods developed in this study can be applied to further research on removing other food allergens.

Efficacy of Commercial Sanitizers for the Inactivation of Listeria monocytogenes on the Processing Equipment for Enoki Mushrooms (팽이버섯 재배 현장에서 Listeria monocytogenes의 성장을 억제하기 위한 살균 처리 기술 개발)

  • The consumption of enoki mushrooms has been associated with cases of listeriosis produced by Listeria monocytogenes, highlighting the significance of sanitizing food-contact surface, such as the velcro used in welding processing of enoki mushrooms, to ensure microbial safety. We investigated the inhibitory activity of nine chemical disinfectants at regular concentrations against L. monocytogenes isolated from a mushroom farm environment. The bacterial suspension was prepared in phosphate buffered saline and mushroom extract broth and inoculated onto the velcro surface. After inoculation, most disinfectants reduced the initial 8 log CFU/coupon concentration by less than 2 log CFU/coupon during a 5-min treatment. Slightly acidic hypochlorous water showed a reduction of approximately 4 log CFU/coupon when tested for more than 30 min at the maximum allowable concentration of 200 mg/L. Sodium hypochlorite solution showed a reduction of approximately 5 log CFU/coupon when used at 100 mg/L for 60 min. Peracetic acid, at the maximum allowable concentration of 300 mg/L, showed the most effective reduction of 5 log CFU/coupon or more when the surface was treated with 37.5 mg/L for 30 min. These results indicate that peracetic acid can be used as the disinfectant strategy to control cross-contamination of L. monocytogenes on the velcro surface of plastic wrappers used in the welding processing of enoki mushrooms.

Regeneration of adventitious root from Calystegia soldanella L. in Jeju island and mass proliferation method using bioreactor system (제주지역 갯메꽃(Calystegia soldanella L.) 유래 부정근 재분화 및 생물반응기 시스템 이용 대량증식법)

  • Calystegia soldanella L. is a perennial herbaceous halophyte belonging to the convolvulaceae family, which mainly grows in coastal sand dunes in Korea. Shoots and rhizomes are edible, and roots called 'Hyoseon Chogeun' are known to have medicinal effects such as antipyretic, sterilization, and diuretic. In addition, physiological activities of antioxidant, anti-inflammatory, antiviral, antifungal and PTP-1B (protein tyrosine phosphate-1B) inhibition have been reported. In this study, in vitro induction cell lines of C. soldanella L. collected from the coastal sand dunes in Jeju island was redifferentiated into adventitious roots that can be used as medicinal resources. Also the biomass of mass-proliferated adventitious roots using a bioreactor were evaluated. Plants of C. soldanella L. were collected from the crevice of the seashore in the coastal area of Taeheung 2-ri, Namwon-eup, Seogwipo-si. Then, it was separated into leaves, stems, rhizomes, and roots, and surface sterilized with 70% ethyl alcohol and 2% NaOCl (sodium hypochlorite). After washing with sterilized water, each organ section was cultured in Hormone-free MS medium (Murashige & Skoog Medium). As a result, the induction response rates were evaluated at 85% and 55%, respectively, in terms of callus formation and shoot generation in the rhizome segment. In the case of the adventitious roots morphological characteristics induced by single-use treatment of auxin-based plant growth regulators IBA and NAA from redifferentiated shoots were compared. Most efficient adventitious root culture method as a rooting rate, number, length, and biomass proliferation in the bioreactor system was confirmed when treated by culturing in MS salts, Sucrose 30 g·L-1, and IBA 1mg·L-1 for 4 weeks. In this study, the medium composition and culture period were confirmed using a bioreactor system to mass-proliferate adventitious roots derived from C. soldanella L. in Jeju island. Also this adventitious root line developed a new medicinal material could increase value of the bio-industry ingredient through quantitative and qualitative screening of phyto-bioactive compounds.

Antibacterial Mechanism and Salad Washing Effect of Bitter Orange Extract Against Salmonella Typhimurium (광귤 추출물의 Salmonella Typhimurium에 대한 항균 메커니즘 및 샐러드 세척 효과)

  • In this study, the antibacterial activity and mechanisms of bitter orange extract, a natural antibacterial agent, were investigated, with a focus on its potential application in washing water for controlling Salmonella Typhimurium contamination of salad, a ready-to-eat food. The minimum inhibitory concentration (MIC) of bitter orange extract against S. Typhimurium was determined using the broth dilution method. Subsequently, S. Typhimurium was exposed to various concentrations of bitter orange extract (1/16 MIC-2 MIC) and growth curves were measured. Following treatment with bitter orange extract, we investigated its antibacterial mechanism by measuring intracellular reactive oxygen species (ROS) levels, alterations in membrane potential and integrity, and nucleic acid leakage in S. Typhimurium. Additionally, salads artificially contaminated with S. Typhimurium were treated with different concentrations of bitter orange extract using the dipping method for various durations to assess the reduction effect. The MIC of bitter orange extract against S. Typhimurium was 195.313 mg/L, and bacterial growth was completely inhibited at a concentration of 1 MIC. Furthermore, an increase in bitter orange extract concentration correlated with elevated intracellular ROS levels, membrane potential disruption, membrane damage, and nucleic acid release. Importantly, salads treated with bitter orange extract exhibited a significant reduction in S. Typhimurium counts compared to the control, and prolonged treatment times resulted in further reductions in bacterial counts. Bitter orange extract was more effective than sodium hypochlorite and can be used as a safer salad wash. These findings indicate the potential treatment of salads to prevent foodborne illnesses.

SCANNING ELECTRON MICROSCOPIC STUDY ON THE EFFICACY OF ROOT CANAL WALL DEBRIDEMENT OF ROTARY NI-TI INSTRUMENTS WITH DIFFERENT CUTTING ANGLE (엔진 구동형 니켈-타이타늄 합금파일의 절삭각에 따른 근관성형 효과에 관한 전자현미경적 연구)

  • The purpose of this in vitro study was to compare the effects of root canal cleanness following two Ni-Ti rotary instruments with different rake angle. Thirty-six sound, extracted human premolars with single root were randomly divided into three groups. The used rotary instruments were HEROShaper (Group 1, Micro-Mega, Besancon, France, n=12) and ProFile (Group 2, Maillefer, Ballaigues, Switzerland, n=12). Control group (n=12) was only extirpated with barbed broach (Mani, Matsutani Seisakusho Co., Japan) Group 1 & 2 teeth were prepared to a #40/.04 taper at the apex followed by 1 mm using crown-down technique. After canal preparation and frequent irrigation with 5.25% sodium hypochlorite, the roots split longitudinally into a bucco-lingual direction. Root halves were cross-sectioned in apical third portion again. All root specimens were processed for SEM investigation and photographed. Separate evaluations by one endodontist were undertaken for smear layer on prepared walls with a five score-index for each using reference photograph in root halves. The penetration depth of smear layer into dentinal tubules was also estimated in the other halves. Following results were obtained: 1. Smear layer was observed on all the prepared walls with two experimental groups except control group. 2. Smear layer characteristics in two experimental groups; 1) HEROShaper group showed snowy, dusty appearance and were shown open dentinal tubuli on the prepared walls of almost specimens, and the thickness of smear layer covering onto dentinal surfaces was within 1-2 ${\mu}m$ in a few specimens. 2) ProFile group showed shiny, burnished appearance and complete root canal wall covered by a homogenous smear layer with no open dentinal tubuli in all specimens. The penetration of smear layer into dentinal tubules was found in all specimens and the thickness was at 2-4 ${\mu}m$ in all specimens. These results demonstrated that a completely clean root canal could not be achieved regardless of positive or negative rake angle, which is in accordance with the majority of previous studies on root canal cleanliness In conclusion, through irrigation with antibacterial solutions or chelating agents is recommended to remove the smear layer on prepared canal wall in spite of Ni-Ti instrumentation.

Bactericidal Activity of Strongly Acidic Electrolyzed Water on Various Vegetables and Kitchen Apparatus (채소 및 주방기구에 대한 강산성전해수의 살균효과)

  • The properties and bactericidal activities of strongly acidic electrolyzed water (SEW) against food-borne pathogenic bacteria, vegetables and kitchen apparatuses were investigated. The available chlorine concentration, pH and oxidation reduction potential (ORP) of SEW were $35{\pm}1.2\;ppm$, $2.3{\pm}0.2$, and $1,140{\pm}20.4\;mV$, respectively. Five strains of food-borne bacteria with initial cell number of 7.00 log CFU/mL were not detected except Bacillus cereus after treatment with SEW for 60 sec. The numbers of Bacillus cereus were reduced to 2.08{\pm}1.00 log CFU/mL at the same condition. In vegetables, SEW treatment after washing strongly in alkalic electrolyzed water (AEW) showed better bactericidal effects than SEW only. The viable cell on stainless steel bowl ($3.86{\pm}2.49\;\log\;CFU/100\;cm^2$) and cup for water ($2.40{\pm}1.80\;\log\;CFU/100\;cm^2$) were not detected by SEW treatment (35 ppm of available chlorine concentration) for 30 sec, but survival of more than 1.00 log CFU/$100\;cm^2$ of viable cell was shown by washing of sodium hypochlorite solution at the same condition. On the other hand, the coliform group bacteria ($5.08{\pm}4.00\;CFU/100\;cm^2$) were detected on rubber globe only, and more than 2.00 log CFU/$100\;cm^2$ of viable cell and coliform group bacteria on it survived, though it was washed with flowing SEW for 30 sec.

THE ANTIMICROBIAL EFFECT OF HORSERADISH (ARMORACIA RUSTICANA) ROOT EXTRACTS AS AN ENDODONTIC IRRIGANT (근관세척제로써 서양산 고추냉이(Horseradish, Armoracia rusticana) 뿌리 추출물의 항균효과)

  • Microorganisms are the main causative factors of pulpal and periapical diseases, therefore successful endodontic treatment is depend on the effective elimination of intracanal bacterial populations. Many studies have been reported antimicrobial effect of Allyl isothiocyanate (AIT) which the principle ingredient of Horseradish (Armoracia rusticana) root extracts. The purposes of this study are to evaluate the antimicrobial effectiveness of Horseradish root extracts against Enterococcus faecalis in root canals of extracted human teeth and compare to sodium hypochlorite (NaOCl). Extracted human mandibular premolar root canals were infected with E. faecalis for 21 days, and then irrigated with Horseradish root extracts, NaOCl solution and saline. After canal irrigation, first samples (S1) were taken. After first sampling, the canals were additionally incubated 7 days, and then second samples (S2) were taken. The samples were inoculated on EHI agar plate to determine the colony forming units (CFU). 1. Mean values of CFU in S1 were $5.815{\times}10^3$ CFU/ml at Horseradish groups, and $3.465{\times}10^3$ CFU/ml at NaOCI groups. There was no statistically significant differences (p=0.086). 2. Mean values of CFU in S2 were $3.100{\times}10^3$ CFU/ml at Horseradish groups, and $5.252{\times}10^5$ CFU/ml at NaOCI groups. There was statistically significant difference (p<.05). 3. There was no statistically significant differences (p=0.076) between S1 and S2 at Horseradish groups in the mean values of CFU. However, there was statistically significant differences (p<.05) between S1 and S2 at NaOCI groups in the mean values of CFU.
