Detection of the Factors Related to spermatization in Sclerotinia trifoliorum -I. Course of Fertilization (Sclerotinia trifoliorum의 Spermatization에 관여하는 요인(要因)의 검색(檢索) -I. 균(菌)의 수정과정(受精過程))

  • The process of fertilization and changes in anatomical structure of sclerotia during the apothecial formation in Sclerotinia trifoliorum, the causal fungus of sclerotial rot of forage legumes, were investigated. The time of fertilization could be estimated with fair accuracy by the sequencial spermatization of the sclerotia which kept at 15C in saturated moisture. In the case of one strain used in this experiment, fertilization between the sclerotia and spermatia were estimated to take place at around 18days after the sclerotia were placed under the conditions for apothecial induction (15C, saturated moisture). The fertilizable state was maintained for about 45 days and the spermatization thereafter did not induce the apothecial formation. When the sclerotia reached fertilizable state, a number of interwoven hyphal nests were developed within the medulla of sclerotia, regardless of the sexuality of the cultures. Comparing the process of multiplication and growth of the hyphal nests in homothallic and heterothallic culture, they were identified as ascogonium. These ascogonia were persisted for about 45 days. This observation was well coincided with the duration of fertilizable state elucidated by the sequencial spermatization experiment.

Isolation and In vitro and In vivo Antifungal Activity of Phenylacetic acid Produced by Micromonospora aurantiaca Strain JK-1

  • The actinomycete strain JK-1 that showed strong inhibitory activity against some plant pathogenic fungi and oomycetes was isolated from Jung-bal Mountain in Ko-yang, Korea. The strain JK-1 produced spores singly borne on sporophores and the spores were spherical and 0.9-1.2 11m in diameter. The cell wall of the strain JK-1 contained meso-diaminopimelic acid. The actinomycete strain JK-1 was identified as the genus Micromonospora based on the morphological, physiological, biochemical and chemotaxonomic characteristics. From the 168 rDNA analysis, the strain JK-1 was assigned to M aurantiaca. The antibiotic MA-1 was purified from the culture broth of M aurantiaca JK-1 using various purification procedures, such as Diaion HP20 chromatography, C18 flash column chromatography, silica gel flash column chromatography and Sephadex LH-20 column chromatography. $^{1}H-$, $^{13}C-NMR$ and EI mass spectral analysis of the antibiotic MA-1 revealed that the antibiotic MA-1 is identical to phenylacetic acid. Phenylacetic acid showed in vitro inhibitory effects against fungal and oomycete pathogens Alternaria mali, Botrytis cinerea, Magnaporthe grisea, Phytophthora capsici and yeast Saccharomyces cerevisiae at < 100 $\mug$ $ml^{-1}$. In addition, phenylacetic, acid completely inhibited the growth of Sclerotinia sclerotiorum, Bacillus subtilis, Candida albicans, Xanthomonas campestris pv. vesicatoria at < $\mug$ $ml^{-1}$. Phenylacetic acid strongly inhibited conidial germination and hyphal growth of M grisea and C. orbiculare. Phenylacetic acid showed significantly high levels of inhibitory' effect against rice blast and cucumber anthracnose diseases at 250 $\mug$ $ml^{-1}$. The control efficacies of phenylacetic acid against the two diseases were similar to those of commercial compounds tricyclazole, iprobenfos and chlorothalonil .n the greenhouse.

Application and Analysis of Rhizopus oryzae Mycelia Extending Characteristic in Solid-state Fermentation for Producing Glucoamylase

  • Enhanced application of solid-state fermentation (SSF) in industrial production and the influence of SSF of Rhizopus K1 on glucoamylase productivity were analyzed using the flat band method. A growth model was implemented through SSF of Rhizopus K1 in this experiment, and spectrophotometric method was used to determine glucoamylase activity. Results showed that in bran and potato culture medium with 70% moisture in a loose state, ${\mu}$ of mycelium reached to $0.15h^{-1}$ after 45 h of culture in a thermostatic water bath incubator at $30^{\circ}C$. Under a low-magnification microscope, mycelial cells appeared uniform, bulky with numerous branches, and were not easily ruptured. The generated glucoamylase activity reached to 55 U/g (dry basis). This study has good utilization value for glucoamylase production by Rhizopus in SSF.

Screening of Antifungal Microorganisms with Strong Biological Activity against Oak Wilt Fungus, Raffaelea quercus-mongolicae

  • Since the mass mortality of Quercus mongolica has been first reported in Gyeonggi province at 2004, the disease spread rapidly over Korean peninsula annually. Ambrosia beetle (Platypus koryoensis) was known as the insect vector of oak wilt fungus, Raffaelea quercus-mongolicae, and control methods of the disease had mainly been focused on eradication of insect vector. However, for the efficient management of the disease, combined control methods for both of the pathogenic fungus and insect vector are strongly required. As one of the efforts to suppress the pathogenic fungus, antifungal activities of Streptomyces isolated from oak forest soil were assayed in this study. Optimum culture condition for the selected isolates was also studied, As a result, Streptomyces blastmyceticus cultured in PDB (Potato Dextrose Broth) at $25^{\circ}C$ for 1 week showed the strongest antifungal activity against oak wilt fungus. Mycelial growth inhibition rates (MGIRs) of Streptomyces isolates were compared on culture media supplemented with heated and unheated culture filtrates of S. blastmyceticus. MGIRs on culture media with unheated culture filtrates were generally higher than those on culture media with heated culture filtrates. Antagonistic mechanism to get involved in the inhibition of hyphal growth and spore formation of the pathogen is due to the antifungal metabolites produced by Streptomyces. This study will provide the fundamental information in developing biocontrol agents for the environment-friendly management of oak wilt disease.

Genomic Organization of Penicillium chrysogenum chs4, a Class Ⅲ Chitin Synthase Gene

  • Class Ⅲ chitin synthases in filamentous fungi are important for hyphal growth and differentiation of several filamentous fungi. A genomic clone containing the full gene encoding Chs4, a class Ⅲ chitin synthase in Penicillium chrysogenum, was cloned by PCR screening and colony hybridization from the genomic library. Nucleotide sequence analysis and transcript mapping of chs4 revealed an open reading frame (ORF) that consisted of 5 exons and 4 introns and encoded a putative protein of 915 amino acids. Nucleotide sequence analysis of the 5′flanking region of the ORF revealed a potential TATA box and several binding sites for transcription activators. The putative transcription initiation site at -716 position was identified by primer extension and the expression of the chs4 during the vegetative growth was confirmed by Northern blot analysis. Amino acid sequence analysis of the Chs4 revealed at least 5 transmembrane helices and several sites for past-transnational modifications. Comparison of the amino acid sequence of Chs4 with those of other fungi showed a close relationship between P chrysogenum and genus Aspergillus.

Effect of Antimicrobial Peptide from Coptidis Rhizoma on Candida albicans Infection (황련 유래 Antimicrobial Peptide의 Candida albicans 감염 억제효과)

  • We previously reported the protein isolated from Coptidis Rhizoma (CRP), which has antifungal activity against a fungal pathogen, Candida albicans. In the current study, we investigated what portion in the CRP is responsible for the antifungal activity. For the investigation, the CRP was fractionated on a Shepadex G-50 column. Data resulting from the fractionation, seven fractions were obtained. Fractions (Fr.) I, II, and III eluted initially from the column showed no inhibitory effect on the growth of C. albicans, whereas Fr. IV, V, and VI eluted later revealed inhibition of the growth, and Fr. IV and VI showed potent antifungal activity by broth susceptibility analysis. However, Fr. VI was contained in the CRP more than Fr. IV, which led us to select the VI for the following experiments. In a murine model of a subcutaneous candidiasis caused by C. albicans, the Fr. VI displayed a therapeutic effect on nude mice pretreated with anti-neutrophil monoclonal antibody (RB68C5) and then infected subcutaneously with live C. albicans. At day 16, these mice were healed almost up to 78% of the infected area when compared to infected area of control nude mice that received diluent (Dulbecco's Phosphate-Buffered Saline; DPBS), instead of the Fr. VI (P<0.01). The Fr. VI blocked hyphal formation from blastoconidial form of C. albicans (P<0.01), which might prevent penetration of hyphae to the deeper site of skin and thus helps the healing. In the ionic strength test, the effect of Fr. was influenced by $Ca^{2+}$ ion just like other known antimicrobial peptides, but the influence was affected at an extremely high concentration such as 500 mM. Thus, such ion-concentration is considered to be meaningless in the clinical situation. Considering all data together, Coptidis Rhizoma is appeared to produce an antimicrobial peptide that has therapeutic effect on subcutaneous infection caused by C. albicans.

Antimicrobial Activities of Wood Vinegar and Application as Natural Fungicides and Food Preservatives (목초액의 항균활성 및 농약, 식품보존제로의 이용 가능성)

  • Antifungal and antibacterial activities of wood vinegars were investigated in this study in order to apply for the natural fungicides and food preservatives. The antifungal activities of wood vinegars were measured as a hyphal growth inhibition rate using four plant pathogenic, four wood rot and one mushroom pathogenic fungi. Inhibition effect on the fungi growth was explicitly observed at the concentrations higher than 0.5% of unrefined wood vinegar and higher than 1.0% of refined wood vinegar, respectively, suggesting that unrefined wood vinegars showed the higher antifungal activities than that of refined wood vinegars. The highest inhibition effect on the fungi growth was shown in Libertella betulina which is a mushroom pathogenic fungus. In addition, the wood vinegars from Pinus densiflora showed the higher antifungal activities against the plant pathogenic and wood rot fungi than that from Quercus spp.. On the other hand, the antibacterial activities of wood vinegars were determined by a paper disc method using the three gram positive and five gram negative bacteria. The unrefined wood vinegars showed a prominent effect on the suppression of bacteria growth at the concentration of 10%, while the suppression of bacteria growth was not observed in the refined wood vinegars. These results also suggest that the unrefined wood vinegars showed the higher antibacterial activities than the refined wood vinegars. The antibacterial activities of wood vinegars against gram positive bacteria were higher than those of wood vinegars against gram negative bacteria. From these results, it can be inferred that the wood vinegars has the strong antimicrobial activities and can be applied as the natural fungicides and food preservatives.

Characterization of a new variety 'Chung' by intra-specific hyphal anastomosis in Pleurotus ostreatus (계통간 교잡에 의한 느타리 품종 '청'의 육성 및 그 특성)

  • The Oyster mushroom is saprophytic fungus. The pileus is stemmed at the side and later depressed. It grows to 5-15 cm, and is of grey, grey-lilac, blackish-grey, steel grey, grey-brown, and blue-blackish. Various kinds of Oyster mushrooms such as golden, pink, brown, grey, white, and blue make marketing an interesting challenge depending upon the market niche. A new commercial strain "Chung" of oyster mushroom was developed by hyphal anastomosis. It was improved with hybridization between monokaryotic strain derived from Pleurotus ostreatus ASI 2194 and ASI 2487. The pileus of parental strain ASI 2194 and ASI 2487 was grey and light blue-grey, respectively. Most of intra-specific hybrids between strain ASI 2194 and ASI 2487 were showed similar pileus color of parents. By the way, the pileus color of variety 'Chung' was blue to bluish grey. The optimum temperatures of mycelial growth and fruiting body development was $25{\sim}30^{\circ}C$ and $12{\sim}16^{\circ}C$, respectively.

Isolation of Siderophore-producing Pseudomonas fluorescens GL7 and Its Biocontrol Activity against Root-rot Disease (Siderophore 생산성 생물방제균 Pseudomonas fluorescens GL7의 선발 및 식물근부병의 방제)

  • For the development of a multifunctional biocontrol agent, the siderophore-producing strain GL7 was isolated from a rhizosphere on chrome azurol S agar. The GL7 was identified as a strain of Pseudomonas fluorescents on the basis of their reactions to standard physicochemcial tests from Bergey's manual, API diagnostic test, and fatty acid analysis. P. fluorescents GL7 considerably inhibited spore germination and hyphal growth of phytopathogenic fungus Funsarium solani in a dual culture. In pot trials of bean with P. fluorescens GL7, the disease incidence was significantly reduced down to 5% from 70% of incidence in the untreated control. P. fluorescens GL7 also enhanced plant growth to nearly 1.5 times than that of the untreated control, promoting elongation and development of the roots. These results suggest that the plant growth-promoting P. fluorescens GL7 can play an important role in the biological control of soil-borne plant disease in a rhizosphere.

Protoplast Regeneration and Reversion in Pleurotus cornucopiae (노랑느타리버섯의 원형질체(原形質體) 재생(再生) 및 환원(還元)에 관한 연구(硏究))

  • Protoplasts of P. cornucopiae were reverted to normal hyphal growth and reversion frequency was $0.04{\sim}19%$. The complete medium stabilized with 0.6 M sucrose was most effective for regeneration of protoplasts. When hypertonic mushroom complete medium not containing agar was overlaid, regeneration frequency of protoplasts was the highest rate among the others of topagar. The protoplast reversion frequency and mycelial growth of P. cornucopiae were increased when various amino acids, nucleic acid components and vitamin compound were added to the hypertonic minimal medium. The relation between sources increasing reversion frequency and sources accelerating mycelial growth was similar in amino acids and nucleic acid components but it was different in vitamins. The protoplast reversion frequency showed the highest rate when all sources were added to the regeneration minimal medium. Microscopically, regeneration patterns of protoplasts showed formation of a bud-like structure, direct germination, yeast-like cell chain of the protoplast, and the production of both direct germ tube and yeast-like cell chain from a protoplast.
