이소플라본 보충이 난소절제 흰쥐의 골대사에 미치는 영향 (The Effect of Isoflavone Supplementation on Bone Metabolism in Ovariectomized SD Rats)

  • 이소플라본 보충이 골다공증의 예방 및 치료에 미치는 영향을 관찰하고자 Sprague-Dawley 암컷을 수술없이 일반식이를 제공한 군(CON), sham-operation 후 일반식이를 제공한 군(SH), 난소절제한 뒤 일반식이를 제공한 군(OVX), 난소절제 후 isoflavone을 공급한 군(OL, OM, OH)으로 나눈 뒤, 난소절제 4일 및 8주 후부터 8주간 이소플라본을 공급하여 혈액의 ALP, sy 중의 hydroxyproline의 변화, 골밀도, 골강도 및 골무게 등을 연구하였다. 골다공증의 예방에 대하여 isoflavone의 효과를 관찰한 결과, ALP는 난소를 절제한 OVX군에서는 CONrns과 SH군에 비해 유의적으로 높은 값을 보였다. 그러나, 난소절제 후 4일부터 8주간 isoflavone을 보충시킨 결과 ALP 활성과 urinary hydroxyproline 함량은 SH군과 비슷하게 낮아졌다. 특히 OM(0.8mg/kg diet)군의 경우 ALP활성과 urnary hydroxyproline 함량이 가장 낮아서 OVX군과 유의적인 차이를 보였다.(p<0.05). 난소적출 후 8주 후에 골밀도를 측정한 결과 난소절제군의 골밀도가 CONrns과 SH군에 비해 유의적으로 낮은 것으로 나타났다. Isoflavone을 보충한 결과 골밀도가 증가하였으나 유의적인 차이는 없어다. 골다공증의 치료효과에 대한 연구결과 alkaline phosphatase activity의 감소, 골밀도, 골강도 및 골무게가 증가하였으나 유의적인 차이는 나타나지 않았다. 이상의 결과는 난소절제 후에 일어나는 골격 대사의 이상은 isoflavone을 보충시킴으로써 어느 정도 정상화시킬 수 있음을 시사한다. 따라서 적절한 양의 isoflavone 보충은 폐경 후 여성의 골대사에 유익한 효과를 줄 것으로 풀이된다.

난소 절제 흰쥐의 골대사에 미치는 별갑의 영향 (Effects of Amydae Carapax on Bone Metabolism in Ovariectomized Rats)

  • Objectives : This study was undertaken to investigate the effects of Amydae Carapax (AC) on parameter related to bone metabolism in ovariectomized rats. Methods : We measured alkaline phosphatase activity and contents of estrogen, calcium, hydroxyproline, osteocalcin, calcitonin and parathyroid hormone after the ovariectomized rats were treated with AC for 30 days. Results : Serum estrogen, calcium and calcitonin contents in ovariectomized rats significantly decreased, but increased after AC treatment. [Significant increase of serum alkaline phosphatase activity, parathyroid hormone activity and osteocalcin content in ovariectomized rats was remarkably decreased by AC treatment. Increase of urinary calcium and hydroxyproline content in ovariectomized rats was decreased by AC treatment.] Conclusions : These results shows that AC has the ability to counteract abnormal calcium metabolic processes due to sex hormone inequality, promoting bone absorption and inhibiting bone formation.

Effects of Steroid Hormones on Collagen Biosynthesis in Rat Aorta and Uterus

  • Effects of steroid hormones on the collagen biosynthesis in aorta and uterus were studied with ovariectomized Sprague-Dawley rats. Effects of administration of hormones, such as estrogen, testosterone and prednisolone, to the ovariectomized animals were studied, comparing with the control. Each group was injected with ³H-proline and sacrificed, followed by removals of aorta and uterus. Separations and quantitative analyses of proline and hydroxyproline were performed by means of thin layer chromatography; and radioactivities of the separated amino acids were assayed by liquid scintillation counter. Normally the incorporation of ³H-proline into hydroxyproline was greater in uterus than in aorta, and collagen turnover rate of uterus was observed rapid as well than that of aorta. In the two tissues from ovariectomized rats, the incorporation rate of ³H-proline into hydroxypoline was markedly decreased than that of the former. Changes in the turnover rate of collagen in these tissues were not observed. Decrease in ³H-proline incorporation into collagen in ovariectomized rats was markedly antagonized by estrogen, but not influenced by prednisolone in the tissues tested.

홍화씨의 경골골절치유에 미치는 영향 (Effects of Safflower-seed on the Fracture Healing in Rat Tibia)

  • We investigated the effect of safflower-seed on fracture healing of fracture model in rat. Fracture healing was evaluated by examining the degree of wound healing macroscopically, radiography, bone histomorphometry and biochemical examination. After 1, 3, 5, 7 days, the would healing was accelerate in safflower-seed diet group. Radiography does not reveal the difference in fracture healing between two group. After 2 weeks, safflower-seed had a significant, stimulatory effect on external callus formation (p<0.05). But after 4, 6, 8 weeks, no difference was observed between normal and safflower-seed dietgroup in callus size. Urinary hydroxyproline, osteocalcin and total alkaline phosphatase decreased significantly (p<0.05) in safflower-seed treated group at 2 week after tracture.

  • This study is designed to investigate the effects of Saengmaegsan extract on cultured myocardial cell sheets affected by oxygen tension change. The results were as follows : 1. Saengmaegsan meaningfully restored the cellular activity decrease in the cultured myocardial cells affected by high oxygen tension. 2. Saengmaegsan meaningfully lessened the degree of total protein decrease in the myocardial cells caused by high oxygen tension. 3. Saengmaegsan meaningfulJy refrained from syntetic decrease of 4-hydroxyproline in both low oxygen tension group and high oxygen tension group.

  • Objectives : Cirsii Japonici Herba (CJH) is one of medicinal plants that has been frequently used for styptic purposes in Asian countries. In order to evaluate a hepatoprotective effects of CJH in the liver fibrotic diseases, the present study investigated how CJH improves a hepatic function in the dimethylnitrosamine(DMN) treated rat. Methods : CJH were orally administered to rats that has been treated with DMN. Subsequently, the amount of blood L-asparate aminotransferase (AST), L-alanine aminotransferase (ALT), and hydroxyproline were quantitated. Several histopathological markers for examining the degree of hepatic fibrosis were investigated by H-E and Masson-Trichrome staining. Results: DMN treatment caused a increase of relative liver weight to the body at 14 days after DMN induction, Administration of CJH with 100mg/kg and 1,000mg/kg dose decreased significantly the AST level elevated by DMN injection(p<0.01). But ALT level was not improved. The hydroxyproline level was reduced by a simultaneous treatment of CJH with DMN for 7 days, but not recovered completely to its normal value, CJH administration improved conspicuously the DMN-induced histopathological changes of liver such as granuloma, but cell necrosis and fibrosis were not improved with CJH 1,000mg/kg dose. Conclusion: These results indicate that CJH has protective effect on liver injury and can inhibit liver fibrosis Induced by DMN in rats.

장기간의 고.저단백식이섭취가 난소절제쥐의 Ca 및 골격대사에 미치는 영향 (A Study on the Long-Term Effects of Dietary Protein Level on Ca and Skeletal metabolism in Ovariectomized Rats)

  • This study was performed to investigate the effect of dietary protein level on the metabolic changes of Ca and skeletons in postmenopausal women, using ovariecotomized rats as an animal model. The female rats of 200∼250g were fed either 8%(L) or 50%(H) casein diet for 15 weeks(1st experiment). At 15th week, the rats of each diet group were undergone ovariectomy or sham-operation and they were continued to feed the same experimantal diet for 9 more months(2nd experiment). Ca metabolism, kidney function and bone composition were determined at the end of 1st experiment, 3rd and 9th month of 2nd experiment. After 1st experiment, high protein group showed higher urinary Ca and protein excretion, however, there was no difference in GFR and urinary hydroxyproline excretion. The weights, ash and Ca content of femur, scapular and vertebra tended to be higher in high protein groups which tells that high protein promoted skeletal growth. In 2nd experiment, high protein group showed higher urinary Ca and protein excretion and lower Ca absorption and balance. GFR was not affected by dietary protein and ovariectomy but increased with time, as well as kidney weight which shows the continuous development of kidney at this age of 15 month in rats. There were no difference in urinary hydroxyproline, serum ALP, and PTH among experimental groups. The weights of femur, scapular, 4th vertebra increased with time, showing the skeleton continues to grow at this age in rats. However, Ca contents, Ca/wt, Ca/ash were decreased with time and tended to be lower in high protein group especially in femur. In conclusion, prolonged feeding of high protein diet deteriorated Ca metabolism and induced bone loss as time after menopause is extended.

산처리 공정에 따라 추출한 돈피 젤라틴의 이화학적 특성에 관한 연구 (Effect of Acid Treatment Process on the Physicochemical Properties of Gelatin Extracted from Pork Skin)

  • 본 연구는 gelatin의 생산공정에서 각종 산 처리가 최종 gelatin의 품질에 어떤 영향을 주는지를 평가함으로써 양질의 gelatin을 생산하는데 기초를 제공하고자 하였다. Gelatin gel의 물성은 lactic acid로 처리한 gelatin이 높은 hardness, cohesiveness, brittleness를 나타내었고 다음으로는 citric acid, acetic acid 순으로 나타났다. Gelatin의 색도는 citric acid, 처리한 gelatin이 acetic acid나 lactic acid로 산침한 젤라틴 보다 높은 명도를 나타냈다. a-value는 acetic acid로 산침한 gelatin이 가장 높게 나타났으며 황색도를 나타내는 b-value는 lactic > acetic acid > citric acid 순으로 높게 나타났다. Gelatin의 점도를 측정한 결과 전단속도(shear rate)에 대한 전단응력(shear stress)의 관계는 모두 전단속도가 증가함에 따라 전단응력이 증가하는Newton성 유체로 판단되었다. 그 기울기는 acetic acid로 산침한 것이 가장 높게 나타났으며 lactic acid, citric acid 순으로 높게 나타났다. Gelatin내의 hydroxyproline 함량을 살펴보면 acetic acid(0.186 g/100 g sample)로 가장 높은 수치를 나타나며 다음으로 lactic acid, citric acid 순으로 나타났다. 그러나, sample중 crude protein 함량을 비교하면 acetic acid(68.74%), lactic acid(80.08%), citric acid(85.47%)로 나타났다. 이것은 gelatin의 열수 추출시 가장 작게 용출된 acetic acid가 가장 높은 collagenous connective tissue 함량(1.35 g/100g)을 나타내었다.

  • Methylisothiocyanate에 의한 단백질의 아미노산 배열 결정시 순차적으로 분리되어 나오는 methylthiohydantion 아미노산을 기체 크로마토그라피로 효과적으로 정성 및 정량하기위하여 새로운 silylating reagent인 N-methyl-N-(tert.-butyldimethylsily)trifluoroacetamide를 사용하여 N-tert.-butyldimethylsily MTH 유도체로 silylation 한 후 HP-1 capillary column으로 분석하였다. Cystine을 제외한 21개의 단백질 구성 아미노산을 동정할 수 있었고 지금까지 packed column에서 TMS 유도체로 동정할 수 없었던 arginine도 분리 동정되었다. 2개 이상의 peak를 나타낸 것으로는 hydroxyproline, proline, isoleucine, glycine 및 tyrosine이었고 이중 hydroxyproline은 많은 수의 peak들로 분리되었다. Lysine, histidine 및 arginine은 주입량 $5.0\;nmole{\sim}15.0\;nmole$의 범위에서 나머지는 $2.5\;nmole{\sim}7.5\;nmole$의 범위에서 상관관계를 측정한 결과 고도의 직선 상관관계를 나타내었다(p<0.001). TMS 유도체에 의한 분석은 많은 불순 peak들 때문에 정량분석에 이용할 수 없었다.

Poly-gamma-glutamic acid의 방사선 조사에 의한 인대 조직 손상 보호 효과 (Prevention Effect of Poly-gamma-glutamic Acid on Porcine Ligament Tissue Damage Induced by Gamma Irradiation)

  • This study was conducted to determine the prevention effect of poly-gamma-glutamic acid (${\gamma}-PGA$) on tissue damage induced by gamma irradiation for development of xenograft. Porcine tendons were treated at various doses of ${\gamma}-PGA$ (0.1, 0.5, 1 and 5%) and then gamma-irradiated (30 kGy). Prevention effects on tissue damage were measured as the result of tensile strength, hydroxyproline contents and viscosity of ${\gamma}-PGA$. Tensile strength was remarkably decrease in gamma-irradiated porcine ligament, but increased by ${\gamma}-PGA$ treated one. Among the ${\gamma}-PGA$ treatment doses, 1% treated group showed the highest values of tensile strength compared to non-treated group. Hydroxyproline contents was significantly increased by gamma irradiation, but decreased by the ${\gamma}-PGA$ treatment. Particularly, 1 and 5% ${\gamma}-PGA$ treated group were exhibited lower values of hydroxyproline contents than other group. In the result of viscosity, gamma-irradiated ${\gamma}-PGA$ (1%) was remarkably increased. Base on the results, it demonstrated that gamma irradiation induces severe alteration of mechanical property and collagen contents on porcine ligament, but ${\gamma}-PGA$ can effectively prevent these tissue damage.
