Free Radical Scavenging Activity of Enzymatic Hydrolyzates of Hot Water Extract from the Shell of Reeve's Turtle (Chinemys reevesii)

  • The shell of Reeve's turtle has been used as a traditional folk medicine in Korea. We produced a hot water extract from Reeve's turtle shell according to the traditional medical practice. To release bioactive peptides, the hot water extract was enzymatically hydrolyzed with various proteases, and the free radical scavenging activity of the hydrolysate was investigated against 1,1-diphenyl-2-picryl-hydrazyl (DPPH), hydroxyl and peroxyl radicals. The free radical scavenging activity of the enzymatic hydrolysates varied from 1 to 79% depending on the enzymes, free radical species, and concentration. The $EC_{50}$ values demonstrated that the enzymatic hydrolysates of hot water extract from the shell of Reeve's turtle are potential antioxidants.

  • Studies were carried out to clarify the effect of ginsenoside-Rbl -Rbr and acid hydrolyzatps of ginsenoside-Rbl, -Rb2 (HRbl, HRbf) on lipolysis and lipogenesis induced by epinephrine, glucagon, ACTH (adrenocorticotrophic hormone), TSH (thyroid-stimulating hormone) and insulin in rat adipose tissilr. HRbl , HRb2 slightly inhibited lipolysis induced by epinephine. glucagon and TSH. ACTH-induced lipolysis in fat tissue slices was significantly inhibited by ginsenoside -Rbl, -Rb2, HRbl and HRb2, particulary HRb2. None of ginsenoside-Rbl, -Rb2, HRbl and HRb2 accelerated insulin-stimulated lipogenesis in fat calls. Among ginseng products, extract powder (freeze dried), extract powder (spray dried), red ginseng powder inhibited ACTH-induced lipolysis in adipose tissue slices, but red ginseng extract not affect them.

  • The effect of natural condiments on peptic hydrolysis of casein in vitro has been studied, peptic activity was determined by the colorimetric method using folin reagent. The obstructive compounds on colorimetry from hydrolyzates of casein were separated by gel filteration method using sephadex G-10 column and 0.05M NaCl. Zinger and red pepper $(0.1{\sim}1.0%)$ were found to have slightly supressing effect for protein digestive action of pepsin. But garlic, green onion and onion acted as synergist for protein digertive action of pepsin.

대두 펩타이드와 소화율이 흰쥐의 혈청 콜레스테롤 농도에 미치는 영향 (Effect of Digestibility of Soybean Peptides on the Concentration of Serum Cholesterol in Rats)

  • In order to investigate the hypocholesteremic effect of soybean perptides, soybean protein(ISP), casein(CNP), their peptic hydrolyzates fractionated by acid precipitation at different pH's(SHT, SH8, SH6, SH4, CHT, CH5, CH4) and amino acid mixtures of the same composition as the proteins(SAA, CAA) were fed to rats and the concentration of serum cholesterol was measured. Then in vitro digestibility and molecular weight distribution of the peptides by pepticpancreatic hydrolysis was measured by FPLC. The lower the in vitro digestibility of peptides is, the lower the concentration of serum cholesterol becomes(r=0.986) and the higher the ratio of macropeptides is, the lower the concentration of serum cholesterol becomes(r=-0.932) in rats. These results suggest that the in vitro digestibility of peptides has close relationship to the concentration of serum cholesterol in rats and non-digestible meacropeptides or polypeptides especially more than 1 kDa, formed through digestion in gut, may lower the serum cholesterol in rats.

유채박의 알칼리 가수분해물을 이용한 합판용 접착제의 개발 (Development of Adhesive Resins Formulated with Rapeseed Flour Akali Hydrolyzates for Plywood Panels)

  • 현재 석유화학계 접착제는 합판과 같은 목질계 판상재료 생산을 위하여 주로 사용되고 있다. 그러나 석유화학계 접착제의 기본 원료인 원유의 가격 상승과 포름알데히드 방산과 같은 문제로 대체 접착제에 대한 개발이 절실한 상황이다. 본 연구는 석유화학계 접착제를 대체하기 위하여 바이오 디젤 부산물인 유채박을 이용하여 접착제를 조제하였으며, 이 접착제를 합판 제조에 적용한 후 접착능 및 포름알데히드 방산량을 조사함으로써 유채박의 합판용 접착제 제조를 위한 원료화 가능성을 확인하고자 하였다. 유채박 접착제 조제를 위하여 먼저 유채박을 수산화나트륨으로 화학적으로 개량한 후, phenol formaldehyde (PF) prepolymer와 혼합하여 접착제를 제조하고 그 접착제를 합판 제조에 적용하였다. 유채박 접착제는 가수분해 조건과 PF prepolymer의 몰비에 따라 26.08~36.12%의 고형분 함량을 보유하였으나, 점도가 매우 높은 것으로 조사되었다. 한편 유채박 접착제로 제조된 합판의 인장 전단강도와 목파율은 유채박의 가수분해 조건과 PF prepolymer의 종류와 상관없이 KS 규격의 보통 합판 품질기준을 상회하였다. 포름알데히드 방산량은 1.8몰의 포름알데히드와 1몰의 phenol로 조제한 PF prepolymer를 사용한 접착제에서 유채박의 가수분해 조건과 상관없이 KS 규격의 E0 기준보다 낮은 것으로 나타났다. 결과적으로 유채박이 합판용 접착제의 원료로서 사용될 수 있을 것으로 생각되나, 열압시간을 단축시키기 위한 유채박 접착제의 고형분 함량을 증가시키는 방안과 접착제의 목재 침투 정도와 인장 전단강도 사이의 관계를 확인하는 현미경적 미세구조에 대한 연구가 유채박 접착제의 상용화를 위해 필요할 것으로 판단된다.

두부비지 가수분해물과 페놀수지로 조제한 마루판 화장용 접착제의 접착성능 (Bonding Quality of Adhesives Formulated with Okara Hydrolyzates and Phenol-formaldehyde Resins for Bonding Fancy Veneer onto High-density Fiberboard)

  • 본 연구에서는 두부비지 가수분해물과 페놀수지로 조제된 마루판 화장용 접착제의 접착성능을 확인하기 위하여 수행하였다. 먼저 두부비지를 황산과 수산화나트륨으로 화학적으로 개량한 후 가교제로 phenol formaldehyde (PF) prepolymer와 혼합하여 접착제를 조제하였는데, 이 접착제는 고형분 함량을 기준으로 35%의 황산 가수분해물(AC), 35%의 수산화나트륨 가수분해물(AK) 그리고 30%의 PF로 조제하였다. 이렇게 조제된 접착제를 고밀도섬유판에 $300g/m^2$의 비율로 도포한 후 참나무 무늬목을 접착시켜 $7kg/m^2$의 압력으로 $120^{\circ}C$에서 열압하여 마루판을 제조하였다. 제조된 마루판에 대하여 formaldehyde/phenol 몰비(1.8, 2.1, 2.4), 퇴적시간(0분, 10분, 20분), 열압시간(90초, 120초)이 평면인장강도, 준내수 박리율, 포름알데히드 방산량에 미치는 영향에 대해 조사하였다. 두부비지 접착제를 이용하여 제조된 목질마루판의 평면인장강도는 KS 규격의 최소값을 상회하였으며, 대부분의 포름알데히드 방산량 값은 KS 규격의 E0 기준에 근접하는 것으로 나타났다. 따라서 두부비지 접착제가 마루판 화장용 접착제로 사용될 수 있다는 가능성을 보여 주었으나, 상용화를 위하여 내수성의 향상과 포름알데히드 방산량의 저감을 위한 연구가 필요할 것으로 생각한다.

  • 최근 환경오염 및 인체 유해성으로 사용이 제한 받고 있는 목재방부제인 chromated copper arsenate (CCA) 그리고 CCA의 대체방부제로 사용되고 있는 고가의 Copper Azole (CuAz)과 alkaline Copper quaternary (ACQ)를 대체하기 위하여 유기폐기물인 두부비지, $CuSO_4$ 그리고/또는 Borax를 이용하여 조제하였다. 이렇게 조제된 두부비지 목재방부제의 사용 가능성을 확인하기 위하여 갈색부후균에 대한 방부효능을 조사하고, 그 방부제가 처리된 목재시편을 FE-SEM과 EDS를 이용하여 현미경적 분석을 수행하였다. 두부비지 방부제로 처리한 목재시편은 Postia placenta와 Gloeophyllum trabeum에 대해 우수한 방부효능을 보였으며, 비지의 가수분해를 위해 사용된 산농도 및 온도의 증가, 그리고 Borax의 첨가는 방부효능을 향상시켰다. 두부비지 방부제로 처리된 시편의 현미경적 관찰을 통하여 비지 가수분해물과 구리의 반응을 통해 용탈과정 중에 용출에 대한 내성을 향상시킨다는 것을 확인할 수 있었다. 상기 결과를 토대로 두부비지 방부제가 경제성과 환경적 측면에서 목재방부제로 사용될 수 있다고 생각한다.

Purification and Characterization of Extracellular Chitinase Produced by Marine Bacterium, Bacillus sp. LJ-25

  • Abstract Extracellular chitinase was purified from the culture liquid of the marine bacterium, Bacillus sp. LJ-25 , and its enzymatic properties were examined. The purified chitinase exhibited a single band on SDS-PAGE and the molecular weight was estimated to be approximately 50 kDa. The optimum pH and temperature for the enzymatic activity were 7.0 and $35^{\circ}C$, respectively. The activity of the chitinase was strongly inhibited by $Zn^{2+}$ and slightly inhibited by $Ba^{2+},{\;}Co^{2+},{\;}Mn^{2+},{\;}and{\;}Cu^{2+}$. The purified chitinase did not hydrolyze $p-nitrophenolN-acetyl-{\bata}-D-glucosaminide{\;}(GlcNAc)_2$ and Micrococcus lysodeikticus cells, which are known to be the substrates for exo-type chitinase. Among the hydrolyzates of colloidal chitin, $(GlcNAc)_2$ was in the highest concentration with small amounts of GlcNAc and $(GlcNAc)_3$..

Isolation of Soil Bacteria Secreting Raw-Starch-Digesting Enzyme and the Enzyme Production

  • Two strains (No. 26 and 143) of bacteria which secrete both pectinase and raw-starch-digesting amylase simultaneously, were isolated from various domestic soil samples. The two bacteria were identified as Pasteurella ureae judging by their morphological and physiological characteristics. The optimal culture conditions for the production of raw-starch-digesting enzyme by the Pasteurella ureae 26 were using $NH_4NO_3$ as the nitrogen source at $37^{\circ}C$ with the pH of 7.5, and 15 of C/N ratio. Since the enzyme was produced only when raw or soluble starch was used as a carbon source, but not when glucose or other sugars was used, the enzyme was considered to be an inducible enzyme by starch. Thin layer chromatography of the hydrolyzed product of starch by the raw-starch-digesting enzyme of the strain No. 26 showed that glucose, maltose and other oligosaccharides were present in the hydrolyzates, and therefore the enzyme seemed to be ${\alpha}-amylase$. The enzyme had adsorbability onto raw com starch in the pH range of 3 to 9.

유제품의 기능성 펩타이드 (Health Functional Peptides from Milk Products)

  • Various peptides derived from food are among the most potent physiologically active agents known, and include anticancer peptides, angiotensin converting enzyme(ACE) inhibitor exhibiting antihypertension action, opioid peptides, antithrombotic peptides, hypocholesterolemic peptides, immunomodulators, calcium absorption enhancers, and other peptides. Hydrophobic peptides extracted from a Cheddar-type cheese slurry were fractionated by gel chromatography and repeated HPLC. A peptide fraction from HPLC showed high cytotoxicity on the tumor cell lines such as a human colon carcinoma, and comprised of Tyr, Ser, Leu, Gly, and others. Hypocholesterolemic peptides were isolated from peptic hydrolyzates of casein and soy proteins. Macropeptides of 1,000${\sim}$5,000 dalton were effective on reducing the cholesterol level of mouse serum. Peptides showing high Krigbaum hydrophobicity and ANS surface hydrophobicity resulted in high hypocholesterolemic effect and fecal steroid concentrations. Caseinomacropeptides(CMP) were isolated from whey powder and treated with soluble and immobilized trypsin to obtain antithrombotic peptides. One fraction from the CMP hydrolyzed with immobilized trypsin for 24h exhibited high antithrombotic activity with 52.5% inhibition of platelet aggregation. These result suggested that peptides from various milk products could be utilized as a good bioactive agents for developing health functional foods.
