수종의 Aspergillus 속 균 사이의 핵전이에 의한 종간잡종 형성 (Construction of Interspecific Hybrids detween Aspergillus spp. by Nuclear transfer)

  • Interspecific hybrids between the ASpergillus spp., A. awamori, A. usamii and A. oryzae, were obtained by nuclear transfer technique. Nuclei isolated from an auxotrophic mutant strain were transferred into the protoplasts of a recipient strain of different species. The frequency of interspecific hybrid formation by nuclear transfer was $2*10^{-5}$ $-7*10^{-4}$ In contrast, no interspecific hybrid was isolated by protoplast fusion. Among the hybrids tested, 10 strains showed increased activity of some or all components of cellulases, xylanases and amylase up to more than two times. Isozyme pattern of the hybrids were analyzed by polyacrylamide gel electrophoresis and isoelectric focusing followed by activity staining, which showed that some of the hybrids have isozyme patterns unidentical to either of the two parents. By measuring the DNA contents and the sizes ofthe conidia, the karyotypes of the hybrids were estimated to be aneuploid near to haploid, diploid or triploid. It was concluded that the unclear transfer technique is much more efficient in the formation of interspecific hybrids than protoplast fusion and is very useful for the improvement of Aspergillus strains.

  • Transformed, hybrid strains of the yeast Saccharomyces capable of simultaneous secretion of both glucoamylase and ${\alpha}-amylase$ have been produced. These strains can carry out direct, one-step assimilation of starch with conversion efficiency greater than 93% during a 5 day growth period. One of the transformants converts 92.8% of available starch into reducing sugars in only 2 days. Glucoamylase secretion by these strains results from expression of one or more chromosomal STA genes derived from Saccharomyces diastaticus. The strains were transformed by a plasmid(pMS12) containing mouse salivary ${\alpha}-amylase$ cDNA in an expression vector containing yeast alcohol dehydrogenase promoter and a segment of yeast $2{\mu}$ plasmid. The major starch hydrolysis product produced by crude amylases found in culture broths is glucose, indicating that ${\alpha}-amylase$ and glucoamylase act cooperatively.

URP-PCR 분석에 의한 느타리 단핵 계통간 교잡주의 핵 DNA 유전 (Nuclear DNA inheritance of intraspecific somatic hybrids by mono-mono cross in Pleurotus ostreatus based on URP-PCR analysis)

  • 느타리버섯류(Pleurotus spp.) 우량 품종개발에 많이 이용되는 교잡육종법 중에서 단핵-단핵간(mono-mono) 교잡에 관한 특성을 구명하기 위하여 느타리 6계통 및 사철느타리 1계통으로 단핵-단핵간 7조합 85개 교잡주를 얻어 교잡율, 핵 DNA 패턴 양상, 자실체의 갓 색깔과 수량성을 분석한 결과는 다음과 같다. 단핵-단핵간 교잡율은 50~93.75%로 나타났으며, 단핵간 85 교잡주의 핵 DNA 양상을 분석한 결과 양친주의 핵을 공유하고 있어 DNA 패턴은 양친의 중간이지만 유전유사도는 어느 한쪽 친주와 조금 더 가까운 양상을 나타냈다. 계통간교잡주 모두 양친의 핵이 공존하는 DNA 패턴을 나타내었지만 양친 중 한쪽 친과 유연관계가 가까운 것으로 나타났다. 사철느타리와 느타리간 교잡주는 유사도가 사철느타리에 가까웠고, 느타리간의 교잡주도 한쪽 모균주에 가까운 유연관계로 나타났다. 단핵-단핵간 교잡에서 자실체 갓 색은 사철느타리와 느타리간 교잡주는 대부분 양친주의 중간정도의 색을 나타냈으나 양친주 중 어느 한 쪽 친주에 좀 더 가까운 갓 색을 띄는 경향을 나타냈다. 자실체 수량성은 느타리간의 교잡주는 양친과 유사한 것이 82 %, 양친보다 높은 것이 0%, 양친보다 낮은 것이 18%였다. 본 연구는 느타리 계통간 교잡주의 핵 DNA 양상과 자실체 특성을 구명하였다. 단핵-단핵간 교잡법의 장점을 충분히 활용하여 앞으로 육종방법으로서 느타리버섯류의 우량 품종을 개발하는데 유용하게 이용될 수 있을 것으로 기대된다.

Genotyping of Agaricus bisporus Strains by PCR Fingerprints

  • Agaricus bisporus, commonly known as the button mushroom, is the most widely cultivated species of edible fungi. Low frequency of recombination ratio and homokaryotic or monokaryotic spore on meiotic basidia form obstacles for breeding programs. Since the first hybrid varieties for white button mushrooms were released in Europe, new varieties released afterwards were either identical of very similar to these first hybrids on morphologies. Therefore, different DNA markers have been used to define unique varieties of A. bisporus strains. Aim of this study is to assess the genetic diversity of different A. bisporus strains in Korea. Twelve UFP (Universal fungal primer, JK BioTech. Ltd), 12 simple sequence repeat (ISSR) and 30 SSR primers were used to assess genetic diversity of monokaryotic and dikaryotic Agaricus bisporus strains including other 19 Agaricus spp. Of them, four UFP, four SSR primers, $(GA)_8T$, $(AG)_8YC$, $(GA)_8C$ and $(CTC)_6$ and seven SSR markers produced PCR polymorphic bands between the Agaricus species or within A. bisporus strains. PCR polymorphic bands were inputted for UPGMA cluster analysis. Forty five strains of A. bisporus are genetically clustered into 6 groups, showing coefficient similarity from 0.75 to 0.9 among them. In addition, genetic variations of monokaryotic and dikaryotic Agaricus bisporus strains were partially detected by PCR technologies of this study. The varieties, Saea, saedo, Saejeong and Saeyeon that have recently been developed in Korea were involved in the same group with closely genetic relationship of coefficient similarity over 0.96, whereas, other strains were genetically related to A. bisporus strains that were introduced from USA, Eroupe and Chinese.

육계 맹장 유산균의 성장특성에 관한 연구 (Study on Growth Characteristics of Lactobacillus Isolated from Broiler Cecum)

  • This study was investigated to observe Lactobacillus spp. population of poultry digestive organ, duodenum, ilium, cecum and colon by growing steps and to select of valuable lactobacilli as probiotics. Two strains of male broiler commercial chickens, Hybrid and Ross, were used to evaluate population of Lactobacillus spp. in intestinal tracts. Three strains of Lactobacillus were identified, and bile salts environment. The number of lactovacilli was the lowest in duodenum compared to other intestinal tracts which had similar population. Population of Lactobacillus was maintained constantly regardless growing steps after one week of age. Identification of Lactobacillus from cecum resulted in L.reuteri BC5, L. crispatus BC7, L.reuteri BC9. All strains was depressed in pH 1 and 2, although two strains could survive for one hour at pH 2. And they could survive at pH 4 for 4hours. In bile salts tolerance, L.reuteri BC5, L.crispatus BC7 were maintained for 2 hours, but the growth reduced from 2hours. Growth of L.reuteri BC9 was increased continuously. In conclusion, Lactobacillus of intestinal tracts were established at first week, and maintained constant population. They were influenced on severe acidic condition and bile salts. Cecal Lactobacillus has different growth charcteristics by strains.

  • Reciprocal crosses were conducted between three strains of Taiwan Country chickens, developed in the National Chung-Hsing University, and two strains of Single Comb White Leghorns, developed in the Taiwan Livestock Research Institute. Traits studied were growing performances, laying performances, egg quality traits and traits concerning disease resistance, including resistance to Marek's disease virus and immune responses to Newcastle disease virus vaccine and to sheep red blood cell. Results indicated that laying performances of Taiwan country chickens were much inferior to White Leghorns, but they matured earlier, their eggs had better shell strength and larger proportion of yolk, and their general disease resistance was much better than White Leghorns. Heterosis were found in laying performances and egg quality traits. The heterosis in laying traits was so large that the hybrid laid as many eggs and as large eggs as did pure strains of White Leghorns. Strategies on the improvement of native chickens and the utilization of genetic merits of native chickens were also discussed.

Bacillus 속 분리균 2종의 내재형 Plasmids 특성분석 (Characterization of Endogeneous Plasmids from Two Bacillus Isolates)

  • In order to obtain the suitable plasmids for constructing plasmid vectors of Bacillus species, endogeneous plasmid DNAs were screended from thermo-tolerant soil bacteria. Based on agarose gel electrophoresis patterns of the isolated plasmid DNAs, two strains harboring small-size plasmids were selected. The isolated were identified to belong to the genus Bacillus on the basis of their morphological and biochemical properties, and named Bacillus sp. 3-3 and 77-8, respectively. The restriction endonuclease maps were determined for four plasmids including two plasmids from each Bacillus isolates. It is interesting that Bacillus sp. 3-3 and 77-8 have an identical plasmid according to the restriction maps. The three kinds of hybrid plasmids constructed by introducing each plasmid of two isolates into a Escherichia coli plasmid vector. pUCCm18 containing chloramplenicol resistance gene active in Bacillus strains, could be replicated in B. subtilis and B. licheniformis. These plasmids are very stable in B. subtilis, suggesting that the Bacillus plasmids identified in this work would be useful for development of new cloning vectors for Bacillus strains.

표고골목 해균인 주홍꼬리버섯을 방제하기 위한 저항성 표고균주 선발 (Control of Diatrype stigma occurred on the bed-log of Shiitake by resistant Shiitake strains)

  • 표고골목 해균인 주홍꼬리버섯을 방제하기 위해 저항성 표고균주 선발을 시도하였다. 저항성 표고균주 선발 시험에서, 사용된 77개 균주 중 67개 균주가 저항력을 가지는 것으로 확인되었다. 이들 67개 균주 중 KFRI 5를 포함한 13개 균주가 주홍꼬리버섯균의 접근을 막는데 가장 큰 영향을 미쳤고 KFRI 180을 포함한 7개 균주가 주홍꼬리버섯의 영역으로 가장 많이 침범했다. 교잡을 통해 만든 31개 균주 중 KFRI 537을 포함한 8개 균주가 주홍꼬리버섯균의 접근을 막는데 가장 큰 영향을 미쳤고 KFRI 545를 포함한 4개 균주가 주홍꼬리버섯의 영역으로 가장 많이 침범했다. 온도와 접종순서의 변화에 따른 각 균주의 저항력 차이를 확인하기 위해 PDA 평판배지와 상수리나무톱밥을 넣은 시험관배지를 사용했다. PDA 평판배지 시험결과, 대부분의 처리구에서 KFRI 137은 주홍꼬리버섯에 대해 뛰어난 접근 억제력을 보였고 KFRI 180은 뛰어난 침범력을 보였다. 교잡균인 KFRI 545, 546, 547은 모균주들인 KFRI 488과 405에 비해 높은 저항력을 보였다. 시험관배지 시험결과, 표고균과 주홍꼬리버섯균이 대치를 하는 시점부터 $25^{\circ}C$에 두었을 때 모든 균주가 높은 저항력을 나타냈다. KFRI 393은 $25^{\circ}C$에서 지속 배양할 때 다른 균주에 비해 높은 저항력을 나타냈다. 또한 모든 균주들에서, $25^{\circ}C$ 배양 후 두 균이 대치를 할 시기에 $14^{\circ}C$로 옮겨 배양한 처리구는 다른 처리구들에 비해 낮은 저항력을 보였다. 이때의 저항력 정도는 각 균주 최고치의 7-20%에 불과했다. 또한 KFRI 259는 PDA 평판배지에서와 달리, 주홍꼬리버섯의 영역을 침범했다.

  • Ten promising polyvoltine mulberry silkworm strains(SDMGl, SDMG2, SDMG3, SDMG4, SDMWl, SDMW2, RMWl, RMW2, RMW3 and RMW4) that are superior in quantitative and qualitative traits have been synthesized in the polyvoltine breeding laboratory of Andhra Pradesh State Sericulture Research & Development Institute, Hindupur through systematic hybridization and appropriate selection methods. After the genotypes were found homozygous for the desired traits, they have been crossed with 3 bivoltine testers $(APS8, APS4 and {NB_2}{D_4})$ and thirty new hybrid combinations were developed for the assessment of their hybrid performance. Phenotypic expressions of economically important quantitative and qualitative traits of fist filial generation were measured and subjected for statistical analysis. Evaluation Index and Subordinate Function methods were employed for the assessment of hybrid performance since they are widely used in silkworm hybrid evaluation. Total of seven poly${\times}$bivoltine combinations, which ranked high in both the methods, were selected as potential combinations for further field test. These combinations also ranked significantly higher than the control hybrid (APMl${\times}$APS8).

  • 가잠의 교배별 잡종강세율의 비교를행한 결과. 1. 이계통간의 $F_1$($가_{육}$ $\times$$중_{백}$ )과 ($일_{백}$ $\times$$중_{백}$ )은 동계통간의 $F_1$보다 기잠체중의 V.R 및 타계량형질의 V.R에 있어서 높은 잡종강세율(V.R)를 나타냈다. 2. 이계통간의 $F_1$($가_{육}$ $\times$$중_{백}$ )과 ($일_{백}$ $\times$$중_{백}$)의 비교에서는 $가_{육}$ $\times$$중_{백}$ 교배형식이 전반적으로 우수한 잡종강세율(V.R)을 나타냈다. 3. 기잠체중의 잡종강세율(V.R)은 모체의 영향을 받아 의잠부터 용까지의 추이가 Smooth하게 나타나지 않고 계통간에 상당한 차이를 보였다.
