Characterization of Fe (III)-Reducing Bacteria Isolated from the Sediment of Chunho Reservoir (천호지 저질토에서 분리한 철환원세균의 특성)

  • Microbial Fe (III) reduction is important for the biogeochemical cycle in the sediment of freshwater system. Also, the Fe (III) reducing mechanism make a model of oxidizing organic compounds and reducing toxic heavy metals, such as chrome or uranium. Thirty-seven strains which have Fe (III) reducing activity were isolated from sediments in lake Soyang and Chunho reservoir. The initial concentration of Fe (II) was the highest in sediments of lake Soyang. However, the highest Fe (III) reducing activity was shown in Chunho reservoir. All isolates were tested for Fe (III) reducing activity. Strains C2 and C3, which were isolated from sediments of Chunho reservoir, showed the highest activity. These strains were tested to see if they utilize various electron donors such as glucose, yeast extract, acetate, ethanol and toluene. Significantly, glucose and yeast extract were used as electron donors. Also these strains were conformed to use humid acid and nitrate as electron accepters. The 16S rRNA sequences of strains C2 and C3 were closely related to Aeromonas hydrophila with 95% similarity.

Evaluation of Various Oligotrophic Media for Cultivation of Previously Uncultured Soil Bacteria (난배양성 토양세균의 배양법 평가 및 신 분류군의 순수분리)

  • We evaluated cultivation methods to obtain pure cultures of previously uncultivated bacteria from soil. Soil bacteria (suspensions) were inoculated onto various oligotrophic media with one of the following additives: 1) soil extract; 2) anthraquinone disulfonate (humic acid analogue); 3) acyl homoserine lactones (quorum-signaling compounds); 4) catalase (for the protection of bacteria from exogenous peroxides). After the relatively long period (60 days) of incubation with elevated concentrations of $CO_2$ (5%, v/v), the media containing catalase showed the highest colony count. We purified 147 randomly selected colonies from the media and the isolates were subjected to the phylogenetic analyses of their 16S rRNA gene sequences. Phylogenetic analysis revealed that approximately 30% of the isolates might belong to novel species or novel family, suggesting that the media and incubation conditions used could be useful for the cultivation of as-yet-uncultured bacteria. Especially, bacteria belonging to the phylum Acidobacteria, ubiquitous bacterial taxon known as an uncultured bacterial group (at least difficult to culture from environmental samples), were successfully cultured in this study.

Fate of 14C - Carobofuran in Rice Plant and Paddy Soil (수도체(水稻體) 및 담수토양중(湛水土壤中) $^{14}C$-Carbofuran의 행적(行跡)에 관한 연구(硏究))

  • A study was undertaken to investigate the absorption, translocation and metabolism of carbofuran in rice paddies. Ring-3-$^{14}C-carbofuran$ applied onto the paddy soil surface was rapidly absorbed and translocated into rice plants. Within 2 days after treatment, it was observed that carbofuran reached shoot tips and accumulated. More than 15% of total radioactivity was recovered in rice plant from 3 to 20 days after treatment. In organic soluble fraction of rice plant extract, 3-hydroxycarbofuran was the major metabolite recording 43% and 4% of total organic soluble radioactivity in shoot and root at 20 days respectively. 3-Ketocarbofuran and phenolic metabolites including carbofuran phenol, 3-hydroxycarbofuran phenol and 3-ketocarbofuran phenol were also detected in the organic soluble fractions. Some glycosidic conjugates of carbofuran metabolites were found in water soluble fraction of rice plant extract and 3-hydroxycarbofuran was the most abundant aglycone. Radioactivity in paddy soil was rapidly decreased until 3 days after treatment and then maintained almost constant level. A significant portion (42∼56 %) of the total radioactivity remained in soil as nonextractable residue from 5 to 20 days after treatment. The nonextractable radioactivity was mainly located in soil organic matter distributing in humin, fulvic acid and humic acid fractions with the decreasing order. Evolution of $^{14}CO_2$ from ring cleavage of $3-^{14}C-carbofuran$ was negligible recording only 1.8% of total radioactivity during 20 days after treatment.

  • Analyses of ancient DNA (aDNA) from archaeological and historical skeletal material are characterized by low quality. Many soil contaminants such as humic acid, fulvic acid, and bone collagen are often co-extracted with aDNA and inhibit amplification by polymerase chain reaction (PCR). In this study, we compared with two methods of DNA extraction by phenolchloroform extraction and silica-bead extraction. In addition, we applied new protocol, ultra sonication based silica-bead extraction method to extract aDNA from some ancient human skeletal remains. This method was more effective by both mitochondrial DNA (mtDNA) and amelogenin gene amplification.

Study of new adsorption isotherm model and kinetics of dissolved organic carbon in synthetic wastewater by granular activated carbon (입상활성탄에 의한 합성폐수의 용존유기물질의 새로운 흡착등온 모델 및 운동학적 흡착 연구)

  • In this study, we conducted the adsorption equilibrium and batch experiments of dissolved organic carbon (DOC) in the wastewater by granular activated carbon (GAC). The components of organic compound were Beef extract (1.8 mg/L), Peptone (2.7 mg/L), Humic acid (4.2 mg/L), Tannic acid (4.2 mg/L), Sodium lignin sulfonate (2.4 mg/L), Sodium lauryle sulfate (0.94 mg/L), Arabic gum powder (4.7 mg/L), Arabic acid (polysaccharide) (5.0 mg/L), $(NH_4)_2SO_4$ (7.1 mg/L), $K_2HPO_4$ (7.0 mg/L), $NH_4HCO_3$ (19.8 mg/L), $MgSO_4{\cdot}7H_2O$ (0.71 mg/L), The adsorption characteristics of DOC in synthetic wastewater was described using the mathematical model through a series of isotherm and batch experiments. It showed that there was linear adsorption region in the low DOC concentration (0~2.5 mg/L) and favorable adsorption region in high concentration (2.5~6 mg/L). The synthetic wastewater used was prepared using known quantities of organic and/or inorganic compounds. Adsorption modelling isotherms were predicted by the Freundlich, Langmuir, Sips and hybrid isotherm equations. Especially, hybrid isotherm of Linear and Sips equation was a good adsorption equilibrium in the region of the both the low concentration and high concentration. In applying carbon adsorption for treating water and wastewater, hybrid adsorption equation plus linear equation with Sips equation will be a good new adsorption equilibrium model. Linear driving force approximation (LDFA) kinetic equation with Hybrid (linear+Sips) adsorption isotherm model was successfully applied to predict the adsorption kinetics data in various GAC adsorbent amounts.

Amino Acid Composition and characteristic of dissolved organic Compounds in the Yellow Sea (황해의 용존 유기물 특성 및 아미노산 조성)

  • Dissolved free amino acid (DFAA) dissolved hydrolyzable amino acid (DHAA) and D/L amino acid racemic ratio in the dissolved organic compounds were studied to investigate the biogeochemical characteristics of dissolved organic compound in the Yellow Sea. Concentration of total DFAA ranged from 0.06 uM to 0.26 uM in the study area. DFAA composition showed that aspiratae, glutamate, serine, glycine and alanine were predominant. According to characteristics of functional group of amino acid, these belonged to hydroponic group. C-18 short column cartridge (Sep-Pak) activated by methanol was used to extract organic macromolecules in the seawater. In operational scheme, macromolecules were divided into two fractions. Geomacromolecule fraction eluted with 50% methanol was used to extract organic macromolecules in the seawater. In operational scheme, macromolecules were divided into two fractions. Geomacromolecule fraction eluted with 50% methanol was moderately hydrophilic and showed characteristics of humic substance in the seawater. Biomacromolecule fraction eluted with 100% methanol was hydrophobic and most abundant in the surface seawater samples. DHAA was much higher than DFAA in this study area. DHAA ranged from 2.05 uM to 6.19 uM in the B-fraction and from 8.13 uM to 24.46 uM in the G-fraction. DHAA was higher in the surface water than in the bottom water where the vertical stratification developed well. The result of HPLC analysis of D/L amino acid showed that low racemic ratio was found in the B-fraction. It implies that the B-fraction is relatively younger than the G-fraction and freshly derived from biosphere.
