PURPOSE: The coexistence of age-related skeletal muscle mass loss and obesity poses a substantial health risk for individuals because it combines the detrimental effects of muscle mass reduction associated with aging and the health complications from obesity. This study aimed to identify the incidence rate and key influencing elements among Korean men in their thirties. METHODS: A cross-sectional study involving 934 male participants was performed using complex sampling analysis. Various influencing elements were investigated, including age, height, weight, body mass index, waist circumference, skeletal muscle mass index, smoking and drinking behaviors, systolic and diastolic blood pressure, fasting glucose levels, triglyceride, and cholesterol levels. RESULTS: The incidence rate was 2.90%. The key influencing elements were age, height, weight, body mass index, waist circumference, skeletal muscle index, systolic blood pressure, fasting glucose, triglyceride, and total cholesterol (p < .05). CONCLUSION: This study identified the incidence rate and key influencing element for CALSMO among Korean younger community-dwelling men.
The study examined clinical effect of the 'Sexiang Shuhuo Jing' on serum CK and LDH activities and skeletal muscle ${\alpha}-actin$ mRNA expression concentration 140days after skeletal muscle injury in rats. The clinical research consisted of observing and measuring the serum CK, LDH activities and skeletal muscle ${\alpha}-actin$ mRNA expression, at the time of injury and during recovery. All experimental data were analyzed by repeated measurement with ANOVA on of SPSS(11.5v), accepting level for all significances was above ${\alpha}\;=.05.$ The results were as follows: That skeletal muscle injury in rats there existed a substantial increase serum CK, LDH activities and expression of skeletal muscle ${\alpha}-actin$ mRNA And Sexiang Shuhuo Jing treatment group's serum CK, LDH activities lower and faster recovery than control group. The 1 st day after skeletal muscle injury, Sexiang Shuhuo Jing treatment group's skeletal muscle ${\alpha}-actin$ mRNA expression was much more higher than control group, after 2 day's faster recovery normal level than control group. There existed a substantial increase again serum CK, LDH activities and skeletal muscle ${\alpha}-actin$ mRNA expression 3rd days after injury in control group. But in Sexiang Shuhuo Jing treatment group's can't be found that.
A high prevalence of protected horticulture farmer's work-related musculo-skeletal disorders (MSDs) have been reported in precedent studies. One of the tasks required ergonomic intervention to reduce the musculo-skeletal risks is the task of product transporting. The purpose of this study is to evaluate quantitatively the spinal load of operator using manual vehicles to predict and prevent musculo-skeletal risks. Spinal load in operators using 4 kinds of manual vehicle were analyzed. Before evaluating spinal load on operator using the manual vehicles by bio-mechanical approach, it is needed to validate human model. In this study, ADAMS LifeMOD human model shows satisfactory results, comparing with already validated model's results or measured results. While Operators pushed the manual vehicles(wheelbarrow, Trolley, 2 wheel cart, and 4 wheel cart) contained loads that were 0 N and 800 N, their spinal loads(compression force, shear force) were evaluated. The compression force demonstrated under the NIOSH action limits - 3410N - for all 4 manual vehicle's operators(McGill 1997; Marras 2000). However, the lateral shear force demonstrated over the University of Waterloo - 500N - for all 3 manual vehicle's operators except 4Wheel cart (Yingline and McGill, 1999). Therefore, operators have risks in prevalence of the musculo-skeletal disorders due to shear force. The findings of this study suggest that it need to be determine the spinal load, especially lateral shear force in designing the manual vehicles in the future.
PURPOSE: This study was conducted to understand the adaptive responses of different modes of physical exercises utilizing skeletal muscle and the comprehensive relevance of AMPK signaling that can be activated by physical exercise as a potential molecular target in human health problems such as neuromuscular disorders (NMDs). METHODS: Most of the contents in this review article are based on recent publications concerning the main topics of interest. The reference literatures cited were obtained by basic searches of overseas academic databases such as PubMed and ScienceDirect using EndNote X7.8. RESULTS: The phenotypic adaptive responses of skeletal muscle during endurance- and resistance-based exercise training (ET and RT respectively) appear to be distinct. To explain the adaptive responses in each single mode of exercises (ET, RT) along with combined exercise training (CT), AMPK signaling is proposed as an important molecular link among those differential modes of exercise and a promising molecular target of NMDs. CONCLUSION: Based on the available evidence, intracellular AMPK signaling activated by diverse stimuli including physical exercise can be a potential and promising therapeutic target for the prevention, amelioration or cure of various human health problems including NMDs and may also be beneficial for physical rehabilitation and emergency situations that may elicit acute metabolic stresses.

BACKGROUND/OBJECTIVES: Although several recent studies have reported the anti-cancer effects of extracts or components of Citrus unshiu peel, which has been used for various purposes in traditional medicine, the molecular mechanisms for their effects remain unclear. In the present study, the anti-cancer activity of a water-soluble extract of C. unshiu peel (WECU) in MDA-MB-231 human breast carcinoma cells at the level of apoptosis induction was investigated. MATERIALS/METHODS: Cytotoxicity was evaluated using the MTT assay. Apoptosis was detected using DAPI staining and flow cytometry analyses. Mitochondrial membrane potential, reactive oxygen species (ROS) assay, caspase activity and Western blotting were used to confirm the basis of apoptosis. RESULTS: The results indicated that WECU-induced apoptosis was related to the activation of caspase-8, and -9, representative initiator caspases of extrinsic and intrinsic apoptosis pathways, respectively, and caspase-3 accompanied by proteolytic degradation of poly(ADP-ribose) polymerase and down-regulation of the inhibitors of apoptosis protein family members. WECU also increased the pro-apoptotic BAX to anti-apoptotic BCL-2 ratio, loss of mitochondrial membrane potential and cytochrome c release from mitochondria to cytoplasm. Furthermore, WECU provoked the generation of ROS, but the reduction of cell viability and induction of apoptosis by WECU were prevented when ROS production was blocked by antioxidant N-acetyl cysteine. CONCLUSIONS: These results suggest that WECU suppressed proliferation of MDA-MB-231 cells by activating extrinsic and intrinsic apoptosis pathways in a ROS-dependent manner.
본 연구는 고지방식이로 유도한 비만 동물모델에서 아연의 식이를 통한 보충 급여가 혈당 조절과 골격 근육의 ZIP7의 작용에 미치는 영향에 대해 살펴보았다. 고지방식이를 공급한 HF군은 정상 대조군에 비하여 단위 체중당 골격 근육 무게가 유의하게 감소하였으며, 혈당은 유의적으로 증가하였다. 고지방식이와 함께 아연을 보충 공급한 HF+Zn군은 아연을 보충하지 않은 HF군과 비교하여, 공복 혈당과 경구 포도당 부하 후 혈당 증가 면적이 유의하게 감소하였다. 또한, HF+Zn군은 HF군에 비해 골격 근육의 ZIP7 단백질 수준이 유의하게 증가하였으며, AKT 활성과 GLUT4 단백질 수준도 유의하게 증가하는 것으로 나타났다. 이상의 결과를 종합해 볼 때, 아연 보충은 비만으로 인한 고혈당 증세를 완화하는 효과를 나타내며, 이는 골격 근육에서의 ZIP7 아연 수송체에 의한 당 대사 조절과 관련이 있을 것으로 생각된다.
Calumenin was previously identified as a high affinity Ca$\^$2+/ binding protein in mouse cardiac sarcoplasmic reticulum (SR). For the present study, a 48 kDa skeletal homologue of calumenin was identified by sucrose-density gradient of rabbit skeletal SR membranes, concanavalin A treatment, 2D-gel electrophoresis, $\^$45/Ca$\^$2+/ overlay, Stains-all staining, and MALDI-TOF analysis. We attempted to clone the skeletal calumenin by RT-PCR based on mouse cardiac and human calumenin sequences. The deduced amino acid sequence (315 residues) of the skeletal calumenin showed high identity to mouse cardiac calumenin (90%). As seen in the cardiac calumenin, the deduced sequence contains a 19 amino acid N-terminal signal sequence and a HDEF C-terminal sequence, a putative retrieval signal to ER. Also, the skeletal calumenin contains one N-glycosylation site, three PKC phosphorylation sites, eight casein kinase 2 phosphorylation sites, and 6 EF-hand domains. GST-calumenin showed a conformational change and increased mobility in the presence of Ca$\^$2+/ in SDS-PAGE. Three calumenin interacting proteins (ryanodine receptor 1, glycogen phosphorylase, and phosphofructo kinase) were identified by pull-down assay with GST-calumenin and solubilized SR. All the interactions were Ca$\^$2+/dependent. The present results suggest that calumenin plays an important role in Ca$\^$2+/ homeostasis of muscle cells.

Purpose: The aim of this study was to evaluate the transfer of different occlusal forces in various skeletal malocclusions using finite element analysis (FEA). Methods: Three representative human cone-beam computed tomography (CBCT) images of three skeletal malocclusions were obtained from the Department of Orthodontics, Yonsei University Dental Hospital, Seoul, South Korea. The CBCT scans were read into the visualization software after separating bones and muscles by uploading the CBCT images into Mimics (Materialise). Two separate three-dimensional (3D) files were exported to visualize the solid morphology of skeletal outlines without considering the inner structures. Individual dental impressions were taken and stone models were scanned with a 3D scanner. These images were integrated and occlusal motions were simulated. Displacement and Von Mises stress were measured at the nodes of the FEA models. The displacement and stress distribution were analyzed. FEA was performed to obtain the 3D deformation of the mandibles under loads of 100, 150, 200, and 225 kg. Results: The distortion in all three skeletal malocclusions was comparable. Greater forces resulted in observing more distortion in FEA. Conclusions: Further studies are warranted to fully evaluate the impact of skeletal malocclusion on masticatory performance using information on muscle attachment and 3D temporomandibular joint movements.
The purpose of this article was to contribute to the knowledge of physiological and pathological changes of aging skeletal muscles, and of therapic method. By aging there were changes of distribution of muscle fibers, the loss of muscle mass, the loss of the number of muscle fibers, the loss of glycolysis capacity, the decrease of the oxidative enzymes and muscle vascularization in the skeletal muscles. And as a pathological change, the exhaustive maximal exercise increased oxidative stress that led to oxidative damage which were shown to be implicated in promoting aging. The property of intensity and duration exercise is important not only in keeping human health and physical fitness from oxidative stress, but also for the maintenance of well-being and quality of life.
The purpose of this study is to understand the structure and biomechanics of the skeletal muscle. The skeletal muscle takes 40 to 45% of the whole body. Stable posture requires a balance of muscle. However, when the muscle strength is unbalanced, movement initiates. The power generated by the muscle is a primary means to adjust the equilibrium of posture and movement. The structural unit of the skeletal muscle is a long cylindrical type muscle fiber which contains hundreds of nucleus. The thickness of muscle fiber is about $10-100{\mu}m$, and its length is about 1-50cm. Muscle fiber is composed of myofibril that is covered with plasma membrane which is called sarcolemma. In understanding the movement of human body, it is important to comprehend the movement of bone and joint and the tension of muscle. Understanding the structure and biomechanics of muscle also provides basic information on clinical treatment of patients.
