Comparison of Resistance to ${\gamma}$-Irradiation between Cryptosporidium parvum and Cryptosporidium muris Using In Vivo Infection

  • In the genus Cryptosporidium, there are more than 14 species with different sizes and habitats, as well as different hosts. Among these, C. parvum and C. hominis are known to be human pathogens. As C. parvum can survive exposure to harsh environmental conditions, including various disinfectants or high doses of radiation, it is considered to be an important environmental pathogen that may be a threat to human health. However, the resistance of other Cryptosporidium species to various environmental conditions is unknown. In this study, resistance against ${\gamma}$-irradiation was compared between C. parvum and C. muris using in vivo infection in mice. The capability of C. muris to infect mice could be eliminated with 1,000 Gy of ${\gamma}$-irradiation, while C. parvum remained infective in mice after up to 1,000 Gy of ${\gamma}$-irradiation, although the peak number of oocysts per gram of feces decreased to 16% that of non-irradiated oocysts. The difference in radioresistance between these 2 Cryptosporidium species should be investigated by further studies.

  • Following ginsenoside-Rb1-hydrolyzing assay, strictly anaerobic bacteria were isolated from human feces and identified as Prevotella oris. The bacteria hydrolyzed ginsenoside Rb1 and Rd to $20-O-{\beta}-D-glucopyranosyl-20(S)-protopanaxadiol$ (I), ginsenoside Rb2 to $20-O-[{\alpha}-L-arabinofuranosyl (1{\rightarrow}6)-{\beta}-D-glucopyranosyl] - 20(S)-protopanaxadiol$ (ll) and ginsenoside Rc to $20-O-[{\alpha}-L-arabinofuranosyl (1{\rightarrow} 6){\beta}-D-g1ucopyranosyl]-20(S)-protopanaxadiol$ (III) like fecal microflora, but did not attack ginsenoside Re nor Rgl (Protopanaxatriol-type). Pharmacokinetic studies of ginseng saponins was also performed using specific pathogen free rats and demonstrated that the intestinal bacterial metabolites I-111, 20(S)- protopanaxatriol(IV) and 20(S)-protopanaxadiol(V) were absorbed from the intestines to $blood(0.4-5.1\;{\mu}g/ml)$ after oral administration with total saponin(1 g/kg/day).

Bacteriocin을 생산하는 장내 유산균의 분리 및 Bacteriocin 특성조사 (Isolation of Bacteriocin-producing Lactic Acid Bacteria from Human Intestines and the Characteristics of their Bacteriocins)

  • Lactobacillus strains were isolated from volunteer's feces, including from newly-born infants and adults in their 20's, by using differential MRS-BPB plates. Total 56 presumptive Lactobacillus strains were isolated and the bacteriocin productions by the isolates were examined by agar diffusion method. Six bacteriocin-producing strains were confirmed. Among them, two isolates, HU-1 and H22-3, showed the most outstanding antimicrobial activities, which were not affected by pH adjustments or catalase treatments of culture. HU-1 was originated from a two-years old boy and H22-3 was originated from a newly-born infant. HU-1 and H22-3 had the same morphology(short rod) when examined by scanning electron microscope, and the same biochemical traits including growth temperature range, salt tolerance and sugar-fermenting abilities. But the growth-inhibition spectrum and plasmid profiles of HU-1 and H22-3 were different. Both strains inhibited the growth of various Gram (+) microorganisms including Listeria monocytogenes. Micrococcus luteus, and Staphylococcus aureus in addition to many species of lactic acid bacteria, indicating the production of broad-spectrum bacteriocins. Bacteriocins produced by HU-1 and H22-3 were stable up to 90℃, 15 min heat treatments. Their activities were not affected by pepsin or trypsin treatments but destroyed by proteinaseK or pronase treatments.

  • Bacteroides 속 균주들은 척추동물의 분변 등에서 분리된 것으로 알려져 있다. 본 연구에서는 건강한 한국인 분변으로부터 Bacteroides sp. KGMB 02408 균주를 분리하였으며 PacBio Sequel 플랫폼을 이용하여 Bacteroides sp. KGMB 02408 균주의 유전체서열을 분석하였다. 유전체는 G + C 구성 비율이 39.5%이고, 5,005개의 유전자와 rRNA 18개 tRNA 74개로 구성되었으며, 염색체의 크기는 5,771,427 bp였다. 또한, 유전체 분석 결과를 통해 산화-환원 효소와 메나퀴논 생합성 및 그와 관련된 다양한 유전자를 발견하였다.

  • Olsenella 속 균주들은 척추동물의 구강, 반추위 및 분변 등에서 분리된 것으로 알려져 있다. 본 연구에서는 건강한 한국인 분변으로부터 Olsenella sp. KGMB 04489 균주를 분리하였으며 PacBio Sequel 플랫폼을 이용하여 Olsenella sp. KGMB 04489 균주의 유전체서열을 분석하였다. 유전체는 G + C 구성 비율이 65.5%이고, 1,838개의 유전자와 rRNA 13개, tRNA 52개로 구성되었으며, 염색체의 크기는 2,108,034 bp였다. 또한, 유전체 분석 결과를 통해 가수분해효소와 항생제 합성 및 내성과 관련된 다양한 유전자를 발견하였다.

Effect of Lentinus edodes on the Growth of Intestinal Lactic Acid Bacteria

  • As the growth factor of lactic acid bacteria, LD (trehalose) was isolated from Lentinus edode5 by using silica gel column chromatography. LD induced the growth of Bifidobacteria breve and Lactobacillus brevis, which were isolated from human feces. LD selectively induced the growth of lactic acid bacteria among total microflora. When total intestinal microflora were cultured in the medium containing LD, it stimulated the growth of lactic acid bacteria and inhibited harmful enzymes, ${\beta}$-glucosidase, ${\beta}$-glucuronidase, and tryptophanase, of intestinal bacteria. LM, which was a monosaccharide from L. edooles, induced the growth of lactic acid bacteria but it seems to be invaluable in vivo. LH isolated from L. edodes by Sephadex G-100 column chromatography was not effective for the growth of lactic acid bacteria.

Characterization of a potential probiotic strain Lactobacillus plantarum MY4

  • The cells of Lactobacillus plantarum MY4 isolated from the human feces were treated for 24 h in artificial bile after incubation for 2 h in artificial gastric juice and final number of the strain was reached to around $3.1{\times}10^8$ CFU/ml. In test of API ZYM kit, ${\beta}$-glucuronidase or ${\beta}$-glucosidase were not produced by L. plantarum MY4. However, ${\beta}$-galactosidase were weakly produced by it, which they would be alleviated the lactose intolerance. L. plantarum MY4 were resistant to antibiotics such as nisin, tetracycline, streptomycin, rifamycin, doxycycline, roxithromycin, chloramphenicol, nystatin, erythromycin, ciprofloxacin and gentamycin. L. plantarum MY4 was affected by alcohol concentration up to 8%, but more than 16%, their growth was not affected significantly. L. plantarum MY4 was shown to inhibit the growth of Listeria monocytogenes ATCC 19111 completely within 24 h of incubation, which indicates its bactericidal nature. Thus, L plantarum MY4 show promise as a probiotic strain because of its characteristics.

산채류가 장내세균의 In Vitro 생육에 미치는 영향 (Effects of Edible Herbs on the Growth of In Vitro Intestinal Microorganisms)

  • This study was aimed to screen edible herbs which control the composition of intestinal microflora. With in vitro experiments, we screened the water or ethanol extracts of about 60 edible herbs and wild plants in terms of the inhibition activity on the growth of the harmful Clostridium perfringens and growth promoting activity for the beneficial Bifidobacteria. The water extracts of mugwort and small water dropwort inhibited the growth of Cl.perfringens both in agar diffusion method and broth culture. On the other hand, the water extracts of petasites, mugwort, yellow day-lily and bitter cress have shown the promotion effect on the growth of Bifidobacterium longum. In the culture test using human feces as starter, the extracts of the above selected herbs increased the population of Bifidobacteria and Lactobacillus while they reduced the numbers of Cl.perfringens and E.coli.

  • Campylobacter jejuni has been recognized as one of the causes of human gastroenteritis. The feces of a variety of reservoir animals contain c. jejuni as commensals in the intestinal tracts, and are fundamental source of contamination. The intestinal organisms contaminate carcasses, equipment tools hands of the processing line workers and air of the processing facility. Once the contamination happens in the slaughterhouses or the meat processing facilities, it is very difficult to keep the carcasses free from the infection of c. jejuni. Various disinfectants are effective in minimizing the number of Campylobacter infections in the processing facilities by washing contaminated carcasses, tools, and hands. Direct contact with infected animals has been incriminated in transmission of infection caused by C. jejuni. Freezing, cooling with dry air and gamma irradiation are an effective way for preserving the meat and eliminating the transmission, but broad and enforced studies are needed for the practical use.

And the contents in Woehyung pyon are divided into head, face, eyes, ears, nose, mouth and tongue, teeth thoat from upper to lower part of the body.
