Increased effects of Bee Venom on aromatase expression and activity in the human osteoblastic cells (인간 골아세포에서 aromatase 효소의 발현과 활성에 대한 봉독의 증강효과)

  • 본 연구는 osteoblastic cells에서 estogen 의 생합성을 유도하는 aromatase의 activity에 대한 봉독(蜂毒)작용을 측정하여, 봉독치료시 Arthritis의 진행 억제 및 estogen의 의한 bone formation의 효과여부를 검증하기 위해 실행하였다. 사용된 세포주로는 Osteoblastic phenotype으로 분화가 유도되는 Human leukaemic cell line FLG 29.1 및 the primary first-passage osteoblastic cells (hOB cells)이며, 이들을 각각 배양하고 각각의 RNA를 isolation한 뒤 PCR 증폭을 하였다. Aromatase에 대한 활성인자인 TPA와 TGF-${\beta}1$ 및 봉독을 이용하여 aromatase의 expression 및 activity에 대해 미치는 영향을 측정한 바, aromatase expression은 FLG 29.1 cell와 hoB cells에서, 50nM TPA 24시간 처리, 봉독 2 ~ 4시간 처리와 TGF-${\beta}1$ 3시간 처리로 유도한 결과 TPA와 TGF-${\beta}1$의 경우는 서로 유사하였고, 봉독에서 상대적으로 높게 나타났다. Aromatase activity는 FLG 29.1 cell, hoB cells에서 24시간 incubation한 결과, 모든 실험에서 일정하게 선상증가를 보였다. $5{\mu}{\ell}/m{\ell}$ BV에서 TPA와 TGF-${\beta}1$보다 뚜렷하게 증가하였으며, 0.5mM Bt2-cAMP, 50nM dexametasone처리에서는 유의성이 없었다. Estrogen 생합성을 촉매하는 aromatase activity BV가 처리에서 현저하게 증가하였기에, Rheumatis arthritis의 bone destruction에 대해 BV가 효과적인 역할을 할 것으로 보여진다.

Study on the Development of Cosmetic Emulsion Cream for Patients with Atopic Dermatitis using Scutellaria Baicalensis (황금(黃芩)을 이용한 아토피성 피부용 한방화장품 제형화에 관한 연구)

  • Objectives : This study was conducted to determine if Scutellaria baicalensis can be used in cosmetic emulsion cream for patients with atopic dermatitis. Methods : Scutellaria baicalensis extract was obtained with the use of butylene glycol through the pressurized solvent extraction(PSE). The antioxidative activity was assessed through SOD-like activity measurement and skin irritating potential was tested using human patch test. Antimicrobial activity was measured by the clear zone formed against Staphylococcus aureus and Escherichia coli and the rheological effects on the emulsion creams were examined using oscillation test. Results : The SOD-like activity increased dose-dependently and was about 90% at 1,000ppm of Scutellaria baicalesis extract. And Scutellaria baicalensis extract did not show any potential to be irritating to the human skin, but it could not be used as an antimicrobial agent for its poor antimicrobial activity against Staphylococcus aureus. The complex modulus decreased by 1,000 pascals and the loss angle also decreased by 20% with the addition of Scutellaria baicalensis extract into the cosmetic emulsion creams, that is, the extract can confer more elastic property on the vehicle. Conclusion : From those results, Scutellaria baicalensis extract can be effectively used as an antioxidant and reinforces the elastic skincare film formed by the application of cream for patients with atopic dermatitis.

Muscular Activity Analysis in Lower Limbs from Motion and Visual Information of Luge Simulator based Virtual Reality (가상현실 루지 시뮬레이터의 동작과 영상정보별 인체 근육활성도 분석)

  • In this paper, capture motion and visual information from a virtual reality luge simulator to analyze muscular activity in the lower limbs. The Luge Simulator consists of a motion platform with a pneumatic module for weight distribution. We recruited luge athletes and healthy subjects and made real-time surface EMG measurements to estimate the muscular activity in the lower limbs according to the motion protocol of a simulator, and a test was conducted for each subject. The results indicated that the rectus femoris had the highest muscular activity according to the level of the slope and velocity of the luge. The soleus muscle showed a high level of activity during a turn in the luge according to the direction. We found that the development of a virtual reality sports simulator based on physical reaction results could bring positive effects to optimize reality and human cenesthesia.

Inhibitory Effects of Dietary Schisandra Supplements on CYP3A Activity in Human Liver Microsomes

  • Schisandra chinensis and its fruits have been used as a traditional herbal medicine to treat liver dysfunction, fatigue, and chronic coughs. Several in vitro and in vivo studies suggested that dibenzocyclooctadiene lignans present in Schisandra fruits strongly inhibit CYP3A4 activity. However, reports on the inhibitory potential of dietary Schisandra supplements against CYP3A activity are limited despite their increasing consumption as dietary supplements. In this study, we evaluated the CYP3A-inhibitory potential of four dietary Schisandra supplements in human liver microsomes. At a concentration of 0.05 mg/mL, Schisandra supplements from Nature's Way, Swanson, Planetary Herbals, and Only Natural inhibited CYP3A activity by 93.9, 70.8, 33.6, and 24.8%, respectively. Nature's Way, which exhibited the strongest inhibition against CYP3A, had the highest contents of gomisin B and gomisin C, which potently inhibit CYP3A activity. The in vivo pharmacokinetics of this product should be examined to determine whether the clinical relevance of inhibiting CYP3A activity by dietary Schisandra supplementation.

  • WiFi sensing integration enables non-intrusive and is utilized in applications like Human Activity Recognition (HAR) to leverage Multiple Input Multiple Output (MIMO) systems and Channel State Information (CSI) data for accurate signal monitoring in different fields, such as smart environments. The complexity of extracting relevant features from CSI data poses computational bottlenecks, hindering real-time recognition and limiting deployment on resource-constrained devices. The existing methods sacrifice accuracy for computational efficiency or vice versa, compromising the reliability of activity recognition within pervasive environments. The lightweight Compact Convolutional Transformer (CCT) algorithm proposed in this work offers a solution by streamlining the process of leveraging CSI data for activity recognition in such complex data. By leveraging the strengths of both CNNs and transformer models, the CCT algorithm achieves state-of-the-art accuracy on various benchmarks, emphasizing its excellence over traditional algorithms. The model matches convolutional networks' computational efficiency with transformers' modeling capabilities. The evaluation process of the proposed model utilizes self-collected dataset for CSI WiFi signals with few daily activities. The results demonstrate the improvement achieved by using CCT in real-time activity recognition, as well as the ability to operate on devices and networks with limited computational resources.

Taurine Transporter Activity in the Human Colon Carcinoma cell Line(HT-29) is Decreased during Cell Differentiation (인체 소장상피세포주(HT-29)의 분화단계에 따른 타우린수송체 활성의 변화)

  • Previous studies on the effect of age on intestinal taurine transport in animals have invariably shown a decline in the activity of the transport system with increasing age. In the present study changes in taurine transporter activity were observed during cell differentiation in the human colon carcinoma cell line HT-29 This cell line exhibits various enterocytic characteristics when differentiated and therefore has frequently been used to study the characteristcs and regulation of nutrient and drug absorption in the small intestine at the cellular level. Pre-treatment of the cells with $\beta$-alanine(10mM) reduced the taurine transport activity to 33% of the value for the control cells(p<0.05) which implies that taurine and $\beta$-alanine share a common $\beta$-amino acid transport system for their celluar uptake in the HI-29 was continued until 21 days post seeding. Kinetic studies of the taurine transporter were conducted in the HT-29 cell line with varying taurine concentration(5-60$\mu$M) in the uptake medium Both Vmax and the Michaelis-Menten constant(Km) of taurine transporter were decreased as differentiation of the HT-29 cell line was progressed ; Vmax of the taurine transporter in cells incubated for 4, 14 and 21 days post seeding was 2.79$\pm$3.4m 16.89$\pm$1.74, and 0.85$\pm$0.08 and 0.32$\pm$0.01nmol.mg protein-1 .30min-1 respectively(p<0.001) and Km was 42.3$\pm$3.4, 16.89$\pm$1.74, and 11.2$\pm$3.0$\mu$M respectively (p<0.01) These results indicate that the activity of sodium dependent active taurine transport system in the HT-29 cell line is decreased as confluent cells are differentiated. This phenomenon in cell culture system corresponds well with the earlier observation of lower intestinal taurine transport activity in suckling rats compared to that in adult animals although direct relationship of cell differentiation with in vivo aging process needs further verification.

Inhibitory effects of Arisaematis rhizoma(天南星) on cell proliferation in HeLa cell (천남성(天南星)이 HeLa Cell의 증식억제(增殖抑制)와 apoptosis에 미치는 영향(影響))

  • Purpose : This study was undertaken to evaluate the inhibitory effects of Arisaematis rhizoma on the cell proliferation in HeLa cells. Methods : The cultured cell after treatment in the different duration in 24, 48, 72 hours with solution of 1%. 5%, 10% Arisaematis rhizoma was quantified by trypan blue exclusin method. The control group was treated with 2% FBS in the different duration in 24, 48, 72 hours. We examined DNA of activated caspase by FACS analysis, caspase-3 activity, DNA fragmentation by DNA laddering, activity of HeLa Cells by the XTT assay, activity of MAP kinase by RT-PCR analysis. Results : After 72 hours culture, the growth activities of 1%, 5%, 10% Arisaematis rhizoma-treated Hela cell were significantly reduced with control group, respectively. After 24 hours culture, the ratio of cells showing caspase activity by FACS analysis were increased in 1%, 5%, 10% Arisaematis rhizoma-treated Hela cell. It were also increased in 48 hours culture of 10% and 72 hours culture of 5%, 10% Arisaematis rhizoma-treated Hela cell. In 24, 48 and 72 hours culture, DNA fragmentations of 5%, 10% Arisaematis rhizoma-treated Hela cell were obviously observed. These results meaned that Arisaematis rhizoma induces apoptosis of HeLa cells. It was supported by increased caspase-3 activity and decreased MAP kinase activity according to time periods and concentrations of Arisaematis rhizoma solution. Conclusion : The study shows that Arisaematis rhizoma has inhibitory effect on cell proliferation and induction capacity of apoptosis of human cevical carcinoma cell line, HeLa cells, in vitro. These results suggest that Arisaematis rhizoma should be useful for treatment of human cevical carcinoma.

Effect of Continuous Exposure to Reactive Oxygen Species on ${\gamma}$-Glutamyltranspeptidase Expression and Activity in HepG2 Cells (HepG2 세포에서 지속적인 활성 산소 노출이 ${\gamma}$-Glutamyltranspeptidase 발현과 활성에 미치는 영향)

  • The adverse health effects of a number of environment pollutions are related to the formation of free radicals. Induction of antioxidant defensive system in the response to an oxidative attack is an essential element of the cell to survive. CYP2E1 is easily induced by organic solvents and induces continuous formation of reactive oxygen species (ROS). ${\gamma}$-Glutamyltranspeptidase (${\gamma}$GT) plays an important role in glutathione metabolism and xenobiotic detoxification. To evaluate the characteristic of oxidative stress which induces GGT expression and to understand human antioxidant defensive response against oxidative stress induced by CYP2E1, we studied regulation of ${\gamma}$GT enzyme expression in response to various oxidative stresses in human HepG2 cells. The ${\gamma}$GT activity was not modified after exposure of acute oxidative stress inducing agents (ferric nitrilotriacetate, cumene hydroperoxide, ADP-Fe, O-tetradecanoylphorbol-13-acetate, tumor necrosis factor-alpha). To induce continuous exposure of cells to ROS, HepG2 cells were transfected by human CYP2E1 gene transiently. The CYP2E1 activity was verified with chlorzoxazone hydroxylation. Transfection of CYP2E1 showed continuous 60% increase in intracellular ROS and 240 % increase in microsomal ROS. CYP2E1 overexpressing cells showed increased ${\gamma}$GT activity (2.5-fold). The observed enhancement of ${\gamma}$GT activity correlated with a significant increase of ${\gamma}$GT mRNA (2.1-fold). Treatment with antioxidant strongly prevented the increase in ${\gamma}$GT activity. The CYP2E1 overexpression did not modify toxicity index and increased glutathione levels. These results show that continuous exposure of cells to ROS produced by CYP2E1 up-regulates ${\gamma}$GT; This may be one of the adaptive antioxidant responses of cells to oxidative insult. Present study also suggests that the induction of ${\gamma}$GT could be used as a marker of oxidative stress induced by exposure to organic solvents.

Tempo-oriented music recommendation system based on human activity recognition using accelerometer and gyroscope data (가속도계와 자이로스코프 데이터를 사용한 인간 행동 인식 기반의 템포 지향 음악 추천 시스템)

  • In this paper, we propose a system that recommends music through tempo-oriented music classification and sensor-based human activity recognition. The proposed method indexes music files using tempo-oriented music classification and recommends suitable music according to the recognized user's activity. For accurate music classification, a dynamic classification based on a modulation spectrum and a sequence classification based on a Mel-spectrogram are used in combination. In addition, simple accelerometer and gyroscope sensor data of the smartphone are applied to deep spiking neural networks to improve activity recognition performance. Finally, music recommendation is performed through a mapping table considering the relationship between the recognized activity and the indexed music file. The experimental results show that the proposed system is suitable for use in any practical mobile device with a music player.

  • Paraoxonase, an antioxidant enzyme, exclusively located on HDL is well known for both hydrolysis of organophosphate and prevention against LDL oxidation. It have been reported that PON1 decrease its activity under oxidative stress and that PON1 activity is lower in subject with higher vulnerability to organophosphate poisoning. The aim of our study is to examine the effect of oxidative system on paraoxon-hydrolzing activity and to elucidate the plausible mechanisms responsible for the decline of HDL-asscociated PON1 activity in vivo system.
