Hot- Water Soluble and Insoluble Materials of Waxy Black Rice Starch (찰흑미(상해항혈나) 전분의 열수가용성 및 불용성 물질)

  • Some structural characteristics of hot-water soluble and insoluble starches (waxy black rice starch) were investigated. The hot-water soluble material content of waxy black rice starch was higher (16.6%) than that (13.4%) of Shinsunchalbyeo starch heated at 98$^{\circ}C$ for 8 min. The Amax and absorbance at 625 nm for hot-water soluble and insoluble material of waxy black rice starch were lower than those of Shinsunchalbyeo. Elution patterns of hot water soluble and insoluble materials by gel permeation chromatography (Sepharose CL-2B) were similar in both samples.

  • We investigated the optimum conditions for conversion of water-insoluble components of basidiocarps of Ganoderma lucidum to water-soluble components by hydrolyzing with chitinase. We also tried it with Ganoderma luciclum residue remaining after extracting hot water-soluble components of Ganoderma lucidum. After hydrolyzing under optimum conditions (20 ppm chitinase, 2% Ganoderma lucidum or 6% Ganoderma lucidum residue, at pH 3 and at $ 35^{\circ}C$), the contents of total water-soluble components (polysaccharide or protein) were measured, and it was found that the contents of water-soluble components increased to 1.5-2.7 fold. Michaelis constant, $K_m$ and maximum rate, $V_max$ calculated by Lineweaver-Burk plot for hydrolysis of Ganoderma lucidum were 1.75% and 0.02%/min respectively and those for hydrolysis of Ganoderma lucidum residue were 53.15% and 0.53%/min respectively The protein-bound polysaccharide was isolated after hydrolysis and molecular weights were measured by Sepharose CL-4B gel filtration and compared with the molecular weights of polysaccharide before hydrolysis.

Negative Effects of Water Extracts from Pinus densiflora Sawdust on Mycelium Growth of the Shiitake Mushroom Lentinula edodes

  • Extracts from sawdust of Pinus densiflora were showed antifungal activity against Lentinula edodes. It was extracted by hot water and then successively extracted by n-hexane, ethyl acetate, and methanol. The yields of the n-hexane-soluble, ethyl acetatesoluble, methanol-soluble and methanol-insoluble fractions of water extracts were 8.2%, 10.6%, 32.0%, and 49.2%, respectively. The ethyl acetate-soluble fraction showed the greatest antifungal activity against L. edodes: 41.5% inhibition at 1,000 ppm. However, there were not significant differences of antifungal activities between n-hexane-soluble fraction and methanol-soluble fraction at a concentration of 1,000 ppm. The hot water extracts showed 23.5% of antifungal activity against L. edodes at a concentration of 1000 ppm. The four antifungal compounds were separated from ethyl acetate fraction by thin layer chromatography.

Changes in Pectic Substances of Korean Pickled Cucumbers with Different Preparation Methods (담금 방법을 달리한 오이지의 숙성 중 펙틴질의 변화)

  • This investigation was undertaken for the purpose of studying the changes of pectic substances in alcohol insoluble solids(AIS) extracted from cucumbers pickled in salt. The preparation methods were brining the cucumbers in 10% NaCl solution (bolied, method A), 20% NaCl solution(bolied, method B) and 20% NaCl solution(cool to $25^{\circ}C$ after boiling, method C). The composition ratio of hot water soluble pectin(HWSP) in AIS decreased during brining, while that of 0.4% sodium hexametaphosphate soluble pectin(NaSP) increased remarkably. The contents of Ca and Mg in AIS increased regardless of preparation methods used. By DEAE-cellulose chromatography, HWSP from fresh and brined cucumbers were divided into six fracitons. 0.2M peak was the main fraction in fresh cucumber, whereas 0.05M and 0.2M peak were the main fraction in pickled cucumbers. On the other hands, NaSP from fresh and brined cucumbers were divided into five fractions and 0.4M peak was the main fraction. However, the composition ratios of galacturonic acid and neutral sugar of 0.4M fraction in brined cucumbers were higher than those of fresh cucumber.

Antioxidative Effect of Ethanol Fraction for Several Korean Medicinal Plant Hot Water Extracts (몇 가지 생약재의 열수 추출물에 대한 Ethanol 분획물의 항산화 효과)

  • The hot water extracts of Paeonia japonica, Paeonia moutan, Eucommia ulmoides, Bupleurum falcatum, Cornus officinalis were freeze dried and dissolved with 70% ethanol. These extracts were fractionated with ethanol soluble fraction(ESF) and ethanol insoluble fraction(EIF). To compare oxidative stability effect of these extracts with that of BHA, linoleic acid with 3,000ppm extracts was incubated at 35$^{\circ}C$. Induction period of BHA was 9 days but that of Paeonia japonica ESF and Scutellaria baicalensis ESF was 12, 9 days respectively. And the higher antioxidant activity was observed for ESF than EIF of these extracts.

Physicochemical Properties of Rice Flours as Influenced by Soaking Time of Rice (쌀의 수침시간에 따른 쌀가루의 이화학적 특성)

  • This investigation was undertaken for the purpose of studying the physicochemical properties of rice flour made by various soaking time(1,4,6,8,12 hours) of rice. As the soaking time was longer, the lightness of rice flours increased, on the contrary its redness and yellowness decreased. Rice flours had higher swelling power and solubility by increasing soaking time, but water binding capacity decreased upon soaking of rice. Hot water insoluble amylose of rice flours was decreased, whereas hot water soluble amylose was increased as the soaking time increased. The amylograph indices indicated that initial pasting temperature of rice flours decreased, while maximum viscosity increased as the soaking time increased.

  • Two flavons, which are a new flavon (the Authors are going to call it "CLEMATISIN") and a yellow crystalline flavon, are isolated from leaves of Clematis brachura Maxim. Clamatisin : A colorless needle crystal which has a Slightly sweet taste, mp 225 .deg.C (decomp). It is positive (Cherry red) for the Mg-HCl(Hg) reaction and is positive (purple) for the $FeCL_{3}$ reagent. It is soluble easily in methanol, ether, acetone, prydine, ethylacetate, hot water and alkali-solutions. It is soluble slightly in cold water and is insoluble in chloroform, benzene and toluence. According to the results of elementry analysis and molecular weight determination the formula of clematisin agrees with $C_{18}H_{18}O_{7}$ when dried at $80^{\circ}C$, crystalline clematisin (from water) contains one molecule of crystalline water. The following derivatives are prepared ; Clematisin-oxim; a colorless needle crystal, mp 215-216 .deg.C, Clematisin methylate; prepared by diazomethan. mp 191-$192^{\circ}C$ a colorless needle crystal. Acethylmethylate; a colorless powder, It is not sharp in melting point and melts at approximately $215^{\circ}C$, Yellow Crystalline Flavon; mp 285-$286^{\circ}C$ (dexo-mp.), yellow needle crystal. It has a slightly sweet teste and shows positive reaction Acetate; a colorless needle crystal, mp $168^{\circ}C$.

Non-Starch Polysaccharides of Cell Walls in Glutinous Rice, Rice and Black Rice (점미, 백미, 흑미 세포벽의 비전분성 다당류의 성분분석)

  • The non-starch polysaccharides in the cell wall of rice, glutinous rice, and black rice, were analyzed. They were fractionated into fractions; water-soluble, hot writer-soluble, ammonium oxalate-soluble, sodium hydroxide-soluble, potassium hydroxide-soluble, and the alkali-insoluble, according to the solvent solubility. The dietary fiber contents were 5.4% in glutinous rice, 4.2% in rice and 7.5% in black rice. The sodium hydroxide soluble fibers were abundant in each kind of rice, especially 4.01% in black rice. The water soluble fibers were 80% of dietary fiber in glutinous rice, 66% in rice, 86% in black rice. It was supposed that the content of the water soluble fibers in rice was increased by pounding. Pectic substances in water soluble fibers, hot water soluble fibers, and ammonium oxalate soluble fibers fraction, were 2.4% in glutinous rice fraction,1.59% in rice, and 1.12% in black rice. Alkali soluble fibers were considered as hemicellulose. Black rice contained 5.80% of hemicellulose, which was more than twice as much as glutinous rice(2.58%) and rice(2.22%). Alkali insoluble fibers were considered as cellulose, which showed no considerable difference. Among samples content of uronic acid in glutinous rice, rice and black rice were 0.9%, 0.66%, 1.8% respectively. Uronic acid of black rice was twice more than other samples tested. The fraction of black rice that uronic acid was extracted at most was the fraction of sodium hydroxide. Mono saccharides of the fraction was the glucose, the arabinose, the xylose.

Studies on the Processing of Crude Drugs(VIII): The Changes of Physico-chemical Parameter and Biological Activities by Processing of Evodia Fruit (한약수치에 관한 연구(제 8보): 오수유 탕포법)

  • We have studied the physico-chemical change and pharmacological transformation of traditional herbal medicines with processing. Evodia fruit (EF) has processed for the purpose of detoxification and reduction of its bitter taste. There are various methods of processing EF in Oriental medicinal reference. Among them, we processed EF according to the method of Sang-han theory, the most famous medicinal reference. We processed EF with washing in hot water, and then dried. Processed EFs(PEFs) were prepared according to the above procedure through repetition of 1,3,5 and 7 times respectively. The contents of weight loss, water extract, diluted ethanol extract, ether extract, total ash, acid insoluble ash, alkaloids and limonin in non-processed EF (NPEF) and PEFs were examined. The weight loss, and contents of water extract and dilute ethanol extract in PEF showed decrease in proportion to increase of washing time, while the contents of ether extract, total ash and acid insoluble ash showed little change as compared with those of NPEF. And the content of evodiamine and rutaecarpine was not changed distinctly. However, the content of limonin decreased in the final processed material in proportion to increase of washing times with hot water. And the intense of bitter taste in PEF was also reduced. These results were ascribed to the flowing-out of the water-soluble portion. The biological activities of NPEF and PEF were also investigated. In the test of DPPH scavenging effect, xanthine oxidase inhibition effect and TBA-Rs effect, PEF was more effective than NPEF in vitro. Also, both NPEF and PEF showed potent analgesic and anti-inflammatory effects against in mice. Especially, PEF by 3 times washing with hot water was more effective than other PEFs. From these results, it is considered that PEF should be prepared by 3 times washing with hot water.

Studies on the constituents of syringa oblata var. dilatata(Nakai) Rehder (Syringa oblata var. dilata (Nakai) Rehder의 성분 연구)

  • Syringa oblata var dilatata (Nakai) Rehder is a oleaceous, deciduous shrub indigenous in the calcareous zones of Korea. The author researched the constituents of the leaves of the plant, especially the bitter principles which may be used as amara. According to the literature a bitter principle Syringin and Mannitol were isolated from the leaves of S. vulgaris L. And later on B. Power indicated that syringin and mannitol were widely distributed in oleaceous plants. The fresh leaves of the plant wer extracted with hot water, the filtered clear liquid was mixed with solution of lead acetate. The lead precipitate was filtered off and the filtered liquid was freed from lead by H$_{4}$S. The filtrate thus obtained was evaporated, and from the residue colorless needles, M. P. 166.deg., were obtained. It was soluble in water, in hot alcohol, and insoluble in ether and had a sweet taste. The results of the elementar analysis, M. P. and other characteristics agreed with that of d-mannitol. Finally it was proved to be identical with d-mannitol through conversion of it into Hexaacetyl mannitol, M. P. 124.deg., Triformal mannitol, M. P. 227.deg., Tribenzal mannitol, M. P. 224.deg., which exhibited no depression, when mixed with authentic specimens. The experiments to isolate bitter principles of the plant are in progress.
