Remote Impedance-based Loose Bolt Inspection Using a Radio-Frequency Active Sensing Node

  • This paper introduces an active sensing node using radio-frequency (RF) telemetry. This device has brought the traditional impedance-based structural health monitoring (SHM) technique to a new paradigm. The RF active sensing node consists of a miniaturized impedance measuring device (AD5933), a microcontroller (ATmega128L), and a radio frequency (RF) transmitter (XBee). A macro-fiber composite (MFC) patch interrogates a host structure by using a self-sensing technique of the miniaturized impedance measuring device. All the process including structural interrogation, data acquisition, signal processing, and damage diagnostic is being performed at the sensor location by the microcontroller. The RF transmitter is used to communicate the current status of the host structure. The feasibility of the proposed SHM strategy is verified through an experimental study inspecting loose bolts in a bolt-jointed aluminum structure.

The Membrane-Bound Protein, MoAfo1, Is Involved in Sensing Diverse Signals from Different Surfaces in the Rice Blast Fungus

  • To establish an infection, fungal pathogens must recognize diverse signals from host surfaces. The rice blast fungus, Magnaporthe oryzae, is one of the best models studying host-pathogen interactions. This fungus recognizes physical or chemical signals from the host surfaces and initiates the development of an infection structure called appressorium. Here, we found that protein MoAfo1(appressorium formation, MGG_10422) was involved in sensing signal molecules such as cutin monomers and long chain primary alcohols required for appressorium formation. The knockout mutant (ΔMoafo1) formed a few abnormal appressoria on the onion and rice sheath surfaces. However, it produced normal appressoria on the surface of rice leaves. MoAfo1 localized to the membranes of the cytoplasm and vacuole-like organelles in conidia and appressoria. Additionally, the ΔMoafo1 mutant showed defects in appressorium morphology, appressorium penetration, invasive growth, and pathogenicity. These multiple defects might be partially due to failure to respond properly to oxidative stress. These findings broaden our understanding of the fungal mechanisms at play in the recognition of the host surface during rice blast infection.

Debonding monitoring of CFRP strengthened RC beams using active sensing and infrared imaging

  • This study attempts to develop a real-time debonding monitoring system for carbon fiber-reinforced polymer (CFRP) strengthened structures by continuously inspecting the bonding condition between the CFRP layer and the host structure. The uniqueness of this study is in developing a new concept and theoretical framework of nondestructive testing (NDT), in which debonding is detected without relying on previously-obtained baseline data. The proposed reference-free damage diagnosis is achieved based on the concept of time reversal acoustics (TRA). In TRA, an input signal at an excitation point can be reconstructed if the response signal measured at another point is reemitted to the original excitation point after being reversed in the time domain. Examining the deviation of the reconstructed signal from the known initial input signal allows instantaneous identification of damage without requiring a baseline signal representing the undamaged state for comparison. The concept of TRA has been extended to guided wave propagations within the CFRP-strengthened reinforced concrete (RC) beams to improve the detectibility of local debonding. Monotonic and fatigue load tests of large-scale CFRP-strengthened RC beams are conducted to demonstrate the potential of the proposed reference-free debonding monitoring system. Comparisons with an electro-mechanical impedance method and an inferred imaging technique are provided as well.

Digital Watermarking Using Psychoacoustic Model

  • A digital watermarking technique applying psychoacoustic model for audio signal is proposed in this paper. In the watermarking scheme, the pseudo-random bit stream used as a watermark signal is embedded into the audio signal in both speech and music. The strength of the embedded signal is subject to the human auditory system in such a way that the disturbances on host audio signal are beyond the sensing of human ears. The experimental results show that the quality of the watermarked audio signal, in term of signal to noise ratio, can be improved up to 3.2 dB.

  • In this study, we developed the real-time controller for control loading system (CLS) of aircraft simulator. The CLS is given the forces as inputs: the exerted force by a pilot, which is determined according to the position of the control stick, and the calculated force by the host computer. And then CLS makes the pilot feel the back loading force by supplying the motor drive with the actuator signal. The developed real-time controller for CLS is organized into the five parts which are the position sensing part including a encoder, the A/D converter part for the analog load cell signal, the communication interface part to communicate with the host, the D/A converter for the actuator signal, and the CPU DSP2812 to carry out a control algorithm. We constructed the test control loading system and carried out the experiment with the developed real-time controller. The experimental results showed that the real-time controller generates the back loading forces similar to the desired back loading force graph.

Quorum Sensing and Quorum-Quenching Enzymes

  • To gain maximal benefit in a competitive environment, single-celled bacteria have adopted a community genetic regulatory mechanism, known as quorum sensing (QS). Many bacteria use QS signaling systems to synchronize target gene expression and coordinate biological activities among a local population. N-acylhomoserine lactones (AHLs) are one family of the well-characterized QS signals in Gram-negative bacteria, which regulate a range of important biological functions, including virulence and biofilm formation. Several groups of AHL-degradation enzymes have recently been identified in a range of living organisms, including bacteria and eukaryotes. Expression of these enzymes in AHL-dependent pathogens and transgenic plants efficiently quenches the microbial QS signaling and blocks pathogenic infections. Discovery of these novel quorum quenching enzymes has not only provided a promising means to control bacterial infections, but also presents new challenges to investigate their roles in host organisms and their potential impacts on ecosystems.

The cAMP/Protein Kinase A Pathway and Virulence in Cryptococcus neoformans

  • The basidiomycete fungus Cryptococcus neoformans is an important pathogen of immunocompromised people. The ability of the fungus to sense its environment is critical for proliferation and the generation of infectious propagules, as well as for adaptation to the mammalian host during infection. The conserved cAMP/protein kinase A pathway makes an important contribution to sensing, as demonstrated by the phenotypes of mutants with pathway defects. These phenotypes include loss of the ability to mate and to elaborate the key virulence factors capsule and melanin. This review summarizes recent work that reveals new targets of the pathway, new phenotypic consequences of signaling defects, and a more detailed understanding of connections with other aspects of cryptococcal biology including iron regulation, pH sensing, and stress.

센서 응용을 위한 사이클로덱스트린-분자 상호작용의 전기화학적 검출 (Study on Electrochemical Detection of Cyclodextrin-molecule Interactions for Sensor Applications)

  • 사이클로덱스트린(cyclodextrin)은 환형다당류(cyclic oligosaccharide) 분자들의 일종으로서, 독성을 거의 지니지 않으며, 다른 분자를 포획할 수 있는 기능을 갖고 있다. 따라서, 이 분자들은 유해물질이나 유독가스를 제거하는데 활용되어 왔다. 본 연구에서는, 이 분자들이 다른 분자를 포획하여 host-guest 화합물을 형성할 수 있다는 점에 착안하여, 이 화합물의 생성 시에 수반되는 전기화학적 변화를 감지하는 방법론을 탐구하고자 한다. 먼저, host-guest 화합물의 형성은 자외선 분광기를 통해서 고찰한다. 사이클로덱스트린의 농도를 바꿔가면서 화합물의 형성을 모니터링한다. 그리고, 실질적인 검출은 금전극에 표면 처리 과정을 거쳐 사이클로덱스트린 분자를 도입하고, 이 전극에 모델분자인 메틸파라벤(methyl paraben)을 도입하여 전기화학적 변화를 감지하는 방식으로 도모하였다. 그 결과, host-guest 화합물 형성 시에 전하의 전이가 일어나고, 이를 전기화학적 측정 방식으로도 검출할 수 있다는 가능성을 실험적으로 보여주었다. 이는 무독성 분자인 사이클로덱스트린의 활용도를 넓힐 수 있는 의미있는 결과로 기대된다.

Investigation of Quorum Sensing-Dependent Gene Expression in Burkholderia gladioli BSR3 through RNA-seq Analyses

  • The plant pathogen Burkholderia gladioli, which has a broad host range that includes rice and onion, causes bacterial panicle blight and sheath rot. Based on the complete genome sequence of B. gladioli BSR3 isolated from infected rice sheaths, the genome of B. gladioli BSR3 contains the luxI/luxR family of genes. Members of this family encode N-acyl-homoserine lactone (AHL) quorum sensing (QS) signal synthase and the LuxR-family AHL signal receptor, which are similar to B. glumae BGR1. In B. glumae, QS has been shown to play pivotal roles in many bacterial behaviors. In this study, we compared the QS-dependent gene expression between B. gladioli BSR3 and a QS-defective B. gladioli BSR3 mutant in two different culture states (10 and 24 h after incubation, corresponding to an exponential phase and a stationary phase) using RNA sequencing (RNA-seq). RNA-seq analyses including gene ontology and pathway enrichment revealed that the B. gladioli BSR3 QS system regulates genes related to motility, toxin production, and oxalogenesis, which were previously reported in B. glumae. Moreover, the uncharacterized polyketide biosynthesis is activated by QS, which was not detected in B. glumae. Thus, we observed not only common QS-dependent genes between B. glumae BGR1 and B. gladioli BSR3, but also unique QS-dependent genes in B. gladioli BSR3.

USN 센싱 시스템에 기초한 다중 생체신호 분석 시스템 (Multimodal Biological Signal Analysis System Based on USN Sensing System)

  • 본 논문에서는 피실험자의 체온, 맥박, 호흡수, 혈압을 수집할 수 있는 무선 센서를 이용하여 수집된 생체 신호를 분석하는 시스템을 제안하였으며, 생체 신호를 분석하기 위하여 전문가 시스템을 사용하여 역전파 신경망을 설계 하였다. 제안된 시스템은 하드웨어 (UStar-2400 ISP, UStar-2400, Wireless sensors) 부분과 소프트웨어 부분 (Knowledge Base module, Inference Engine module, User Interface module)으로 구성되며 소프트웨어 부분은 HOST PC에 삽입된다. 그리고 시스템의 정확도를 개선하기 위하여 전방향 에러 정정 시스템(LDPC)을 구현하였다. 지식기반 모듈에서 100개의 데이터 집합을 사용하여 역전파 신경망을 훈련하였으며, 지식기반 모듈의 128개의 데이터 집합을 사용하여 실험한 결과 약 95%의 정확도를 가졌으며, 무선 센서를 사용하여 13명의 학생을 대상으로 실험한 결과 약 85%의 정확도를 획득하였다.
