GENETIC SUSCEPTIBILITIES OF CYTOCHROME P450 1A1, 2E1, AND N-ACETYLTRANSFERASE 2 TO THE RISKS FOR KOREAN HEAD AND NECK CANCER PATIENTS (한국인 두경부암종 환자에서 Cytochrome P450 1A1, 2E1 및 N-acetyltransferase 2 효소의 다형성 분석에 따른 유전적 감수성에 대한 연구)

  • Individual genetic susceptibilities to cancers may result from several factors including differences in xenobiotics metabolism to chemical carcinogens, DNA repair, altered oncogenes and suppressor genes, and environmental carcinogen exposures. Among them, genetic polymorphisms of metabolizing enzymes to chemical carcinogens have been recognized as a major important host factors in human cancers. They have two main types of enzymes: the phase I cytochrome P-450 mediating enzymes (CYPs) and phase II conjugating enzymes. The purpose of this study is to determine the frequencies of genotypes of phase I (CYP1A1 and CYP2E1) and phase II (NAT2) metabolizing enzymes in healthy control and head and neck cancer patients of Korean and to identify the relative high risk genotypes of these metabolizing enzymes to head and neck cancer in Korean. The author has analyzed 132 head and neck cancer patients and 113 healthy controls using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The results were as following; 1. The frequencies of genotypes of CYP1A1, CYP2E1 and NAT2 in healthy control were as following; CYP1A1 exon 7 polymorphism; Ile/Ile: Ile/Val: Val/Val = 59.3%: 36.3%: 4.4% CYP2E1 Pst I polymorphism, C1/C1: C1/C2: C2/C2 = 61.1%: 32.1%: 6.2% NAT2 polymorphism; F/F: F/S: S/S = 43.4%: 48.7%: 8.0% 2. In analysis of phase I enzyme, Val/Val genotype in CYP1A1 exon 7 polymorphism and C2/C2 genotype in CYP2E1 Pst I polymorphism were associated with relative high risks to head and neck cancers (Odds' ratio: 2.09 and 1.37, respectively). 3. Among the genotypes of NAT2 enzyme polymorphism, S/S genotype of NAT2 enzyme had 1.03 times of relative risk to head and neck cancers. 4. In combined genotyping of CYP1A1, CYP2E1, and NAT2 enzymes polymorphisms, the patients with Val/Val and C1/C1, C2/C2 and fast acetylator, and Val/Val and fast acetylator had higher relative risks than the patients with each baseline of combined genotypes (Odds' ratio: 2.82, 1.98 and 2.1, respectively). These results suggest the combined genotypes of Val/Val and C1/C1, C2/C2 and fast acetylator, and Val/Val and fast acetylator were more susceptible to head and neck cancers in Korean. And genotyping of metabolizing enzymes could be useful for predicting individual susceptibility to head and neck cancer.

  • With the increasing speed of internationalization of corporations under WTO system foreign direct investment is considered as the important means of international cooperation as well as avoiding trade barrier. From this point of view the study on foreign direct investment of Korean corporations is of great importance. Many studies till now, however, are concerned with the macroscopic analysis of foreign direct investment of Korean corporations, not with the establishment of strategy appropriate to the environment of host country and the investment performance. In order to meet the problems mentioned above this thesis aims at accomplishing the following three tasks. (1) synthetic assessment of existing theories of foreign direct investment. (2) systematic adjustment of factors of environment in the foreign direct investment activities. (3) analysis of above factors in the foreign direct investment of Korean textile industry and proper proposal of strategic model for the individual corporation. The results of analysis according to the aim of the study are as follows: (1) To lead foreign direct investment successfully, variable and effective information-presentation should be offered with priority. (2) The investment strategy through regional diversification is required. (3) The localization of decision making has to be attempted assertively. (4) The global network system is needed for more recommendable foreign direct investment. (5) More aggressive and outgoing attitude like developing into the local market and obtaining the high-technology is desperately needed.

Immune reconstitution after allogeneic hematopoietic stem cell transplantation in children: a single institution study of 59 patients

  • Purpose: Lymphocyte subset recovery is an important factor that determines the success of hematopoietic stem cell transplantation (HSCT). Temporal differences in the recovery of lymphocyte subsets and the factors influencing this recovery are important variables that affect a patient's posttransplant immune reconstitution, and therefore require investigation. Methods: The time taken to achieve lymphocyte subset recovery and the factors influencing this recovery were investigated in 59 children who had undergone HSCT at the Department of Pediatrics, The Catholic University of Korea Seoul St. Mary's Hospital, and who had an uneventful follow-up period of at least 1 year. Analyses were carried out at 3 and 12 months post-transplant. An additional study was performed 1 month post-transplant to evaluate natural killer (NK) cell recovery. The impact of pre- and post-transplant variables, including diagnosis of Epstein-Barr virus (EBV) DNAemia posttransplant, on lymphocyte recovery was evaluated. Results: The lymphocyte subsets recovered in the following order: NK cells, cytotoxic T cells, B cells, and helper T cells. At 1 month post-transplant, acute graft-versus-host disease was found to contribute significantly to the delay of $CD16^+/56^+$ cell recovery. Younger patients showed delayed recovery of both $CD3^+/CD8^+$ and $CD19^+$ cells. EBV DNAemia had a deleterious impact on the recovery of both $CD3^+$ and $CD3^+/CD4^+$ lymphocytes at 1 year post-transplant. Conclusion: In our pediatric allogeneic HSCT cohort, helper T cells were the last subset to recover. Younger age and EBV DNAemia had a negative impact on the post-transplant recovery of T cells and B cells.

Efficacy of Prophylactic Entecavir for Hepatitis B Virus-Related Hepatocellular Carcinoma Receiving Transcatheter Arterial Chemoembolization

  • Background and Aims: Hepatitis B virus (HBV) reactivation was reported to be induced by transcatheter arterial chemoembolization (TACE) in HBV-related hepatocellular carcinonma (HCC) patients with a high incidence. The effective strategy to reduce hepatitis flares due to HBV reactivation in this specific group of patients was limited to lamivudine. This retrospective study was aimed to investigate the efficacy of prophylactic entecavir in HCC patients receiving TACE. Methods: A consecutive series of 191 HBV-related HCC patients receiving TACE were analyzed including 44 patients received prophylactic entecavir. Virologic events, defined as an increase in serum HBV DNA level to more than 1 log10 copies/ml higher than nadir the level, and hepatitis flares due to HBV reactivation were the main endpoints. Results: Patients with or without prophylactic were similar in host factors and the majorities of characteristics regarding to tumor factors, HBV status, liver function and LMR. Notably, cycles of TACE were parallel between the groups. Ten (22.7%) patients receiving prophylactic entecavir reached virologic response. The patients receiving prophylactic entecavir presented significantly reduced virologic events (6.8% vs 54.4%, p=0.000) and hepatitis flares due to HBV reactivation (0.0% vs 11.6%, p=0.039) compared with patients without prophylaxis. Kaplan-Meier analysis illustrated that the patients in the entecavir group presented significantly improved virologic events free survival (p=0.000) and hepatitis flare free survival (p=0.017). Female and Eastern Cooperative Oncology Group (ECOG) performance status 2 was the only significant predictors for virological events in patients without prophylactic antiviral. Rescue antiviral therapy did not reduce the incidence of hepatitis flares due to HBV reactivation. Conclusion: Prophylactic entecavir presented promising efficacy in HBV-related cancer patients receiving TACE. Lower performance status and female gender might be the predictors for HBV reactivation in these patients.

Occurrence of Fruit Flies (Diptera: Tephritidae) in Fruit Orchards from Myanmar (미얀마 과수원에서 과실파리 발생에 관한 연구)

  • Population of fruit flies was monitored by using methyl eugenol trap during 2010-2011 in Yezin, Myanmar. Population numbers were analyzed with meteorological factors including rainfall, temperature, relative humidity and duration of sunshine. Samples of mango, guava, and jujube fruits were collected from orchards. The fruits were kept in containers so that the species of flies infesting the fruit could be identified when the adult insects emerged and to assess damage caused by fruit flies. Regression analyses indicated that populations of fruit flies were observed to be positively correlative with rainfall, minimum temperature and relative humidity, and negatively correlative with the duration of sunshine. Eleven species of fruit flies, Bactrocera arecae, B. carambolae, B. correcta, B. dorsalis, B. kandiensis, B. latilineola, B. malaysiensis, B. neocognata, B. raiensis, B. verbascifoliae, and Carpomya vesuvina, were identified. B. correcta and B. dorsalis were the most abundant and accounted for 29.3% and 28.6% of total emerged adults in the different fruit samples. The highest percentage of fruit damage was observed on guava ($59{\pm}15.4$), followed by mango ($35.5{\pm}12.1$) while the lowest was recorded on jujube ($18.5{\pm}7.9$).

An Analysis of the Locational Motives for the Korean Auto Industry′s Investment in the U.S. - Case Study of Hyundai Motor Manufacturing Alabama - (우리나라 자동차산업의 미국투자 입지동기 분석 - 현대자동차 미 앨라배마 투자 사례를 중심으로 -)

  • Foreign direct investment (FDI) by firms has various motives in terms of the strategy for firms' long-term growth. This research uses the case study of Hyundai Motor Manufacturing Alabama to analyze the motives of the Korean auto industry's FDI in the U.S. and the locational factors that determined the selection of the site. This paper starts from the question of why Hyundai made the decision to invest in the U.S., which is not favorable in terms of production cost, especially considering that its exports to the U.S. have been on the increase. The results indicate that the strongest motive for the decision to invest in the U.S. was to ameliorate the trade friction between Korea and the U.S. Given that Hyundai depends on the U.S. market to a large extent for its exports, the foremost motive was to use local production in order to reduce the serious trade imbalance in the automobile sector between Korea and the U.S. in order that trade friction with the U.S. may be avoided and causes for trade disputes may be reduced. Other motives such as improving the access to local consumers were of secondary importance. After the selection of the country in which to invest, however, various factors were considered in the decision regarding the location of the plant, and incentives by local governments ultimately played a key role in this decision. The results imply that the Korean manufacturing industry's investment in the U.S. cannot be explained by traditional FDI theories and instead was greatly driven by a strategic defensive motive. In addition, the results confirm that the design of an appropriate incentive structure by host governments is important for attracting FDI.

Analysis of Siting Criteria of Overseas Geological Repository (II): Hydrogeology (국외 심지층 처분장 부지선정기준 분석 (II) : 수리지질)

  • Geology, hydrogeology, and geochemistry are the main technical siting factors of a geological repository for spent nuclear fuels. This paper evaluated the siting criteria of overseas geological repository with related to hydrogeologic properties, such as hydraulic conductivity, partitioning coefficient, dispersion coefficient, boundary condition, and water age. Each country establishes the siting criteria based on its important geological backgrounds and information, and social environment. For example, Sweden and Finland that have decided a crystalline rock as a host rock of a geological repository show different siting criteria for hydraulic conductivity. In Sweden, it is preferable to avoid area where the hydraulic conductivity on a deposition hole scale (~30m) exceeds $10^{-8}m/s$, whereas Finland does not decide any criterion for the hydraulic conductivity because of limited data for it. In addition, partitioning coefficients should be less than 10-1 of average value in Swedish crystalline bedrock. However, the area where shows 100 times less than average partitioning coefficients of radionuclides in crystalline rock should be avoided in Sweden. In German, the partitioning coefficients for the majority of the long-term-relevant radionuclides should be greater than or equal to $0.001m^3/kg$. Therefore, it is strongly required to collect much and exact information for the hydrogeologic properties in order to set up the siting criteria.

Bacterial Growth-inhibiting Activity of Amniotic Fluid Against E. coli (양수의 대장균에 대한 세균증식 억제효과)

  • The amniotic fluid provides a medium in which the fetus can readily move, cushions him against possible injury and helps him maintain an even temperature. Besides above mentioned functions, investigators reported that human amniotic fluid contains host-resistance factors which prevent bacteria from producing infectious disease and this activity shows difference among human racial groups or bacterial genera, species and strains. 40 amniotic fluid specimens from Korean women in their second and third trimesters of pregnancy were examined for inhibiting the growth of Escherichia coli. And various factors which might affect bacterial growth inhibiting activity such as pH, initial inoculum size, concentration of amniotic fluid, and heat resistance, were also tested using a strongly inhibitory amniotic fluid specimen. Finally plate diffusion tests were carried out using other strongly inhibitory amniotic fluid. The following results were obtained: 1. Of the 40 fluid samples examined, 18 specimens(45%) had inhibitory activity and samples from women in their second trimester of pregnanancy showed non-inhibitory activity(2 specimens). 2. The pH of the fluids varied between 7.43 and 8.33. There was no correlation between pH and inhibitory activity. 3. No. 19 amniotic fluid showed bacteriostatic activity after 24 hours incubation when an inoculum of $10^2$ organisms per milliliter was used, but non-inhibitory with an inoculum of $10^3$ and $10^4$ bacteria per milliliter. 4. The content of amniotic fluid in culture media influenced E. coli growth. At 90 percent, E. coli was inhibited growth but at 10 percent and 50 percent. 5. Inhibitory activity of No. 19 amniotic fluid was retained after heating to $50^{\circ}C$ for 30 minutes or 100^{\circ}C$ for 30 minutes. 6. Plate diffusion tests with No. 27 amniotic fluid showed that 0.7ml amniotic fluid gave clear zone of growth inhibition around the central well but 0.2ml and 0.1ml amniotic fluids were not.

Changes in sedimentary structure and elemental composition in the Nakdong Estuary, Korea (낙동강 하구역 퇴적구조 및 원소조성 변화에 관한 연구)

  • To understand the sedimentary environment of Scirpus planiculmis habitat (Myeongji and Eulsuk tidal flats) in the Nakdong Estuary, this study analyzed the statistical parameters (sorting, skewness, and kurtosis) of grain size data and the major (Al, Fe, Mn, Mg, Ca, Na, K, Ti, and P), minor (Li, Sc, V, Cr, Co, Ni, Cu, Zn, Sr, Zr, Cs, Pb, Th, and U), and rare earth elements (REEs) in sediment cores. For Myeongji, the sediment structure of the upper part of the cores was poorly sorted, more finely skewed, and more leptokurtic due to construction of the West gate. By contrast, the Eulsuk cores all differed due to the contrasting floodgate operation patterns of the West and East gates. The linear discriminate function (LDF) results corresponded to the statistical parameters for grain size. At the Eulsuk tidal flat (sites ES05 and ES11), elemental distributions were representative of Al-, Fe- and Ca-associated profiles, in which the elements are largely controlled by the accumulation of their host minerals (such as Na- and K-aluminosilicate and ferromagnesium silicate) and heavy detrital minerals at the sites. Detrital minerals including the aluminosilicates are major factors in the elemental compositions at ES05, diluting the REE contents. However, clay minerals and Fe-oxyhydroxides, as well as REE-enriched heavy minerals, appeared to be controlling factors of the elemental composition at ES11. Therefore, the mineral fractionation process is important in determining the elemental composition during sedimentation, which reflects the depositional condition of riverine-saline water mixing at both sites.

Graft Considerations for Successful Anterior Cruciate Ligament Reconstruction (성공적인 전방십자인대 재건술을 위한 적절한 이식건의 선택)

  • Several factors need to be considered for a successful anterior cruciate ligament (ACL) reconstruction, such as preoperative planning, operation technique, and postoperative rehabilitation. Graft choice, fixation, preparation method, maturation, incorporation to host bone, and graft tension should also be considered to achieve a good outcome after an ACL reconstruction. Factors to consider when selecting a graft are the graft strength, graft fixation, fixation site healing, and donor site morbidity, as well as the effects of initial strength, size, surface area, and origin of the graft on its potential for weakening during healing. There are two types of graft for an ACL reconstruction, autograft or allograft. Several autografts have been introduced, including the bone-patellar tendon-bone, hamstring tendon, and quadriceps tendon-bone. On the other hand, each has its advantages and disadvantages. The recent increased use of allografts for an ACL reconstruction is the lack of donor site morbidity, decreased surgical time, diminished postoperative pain, and good availability of source. Despite this, there are no reports suggesting that an allograft may have a better long-term outcome than an autograft. Allografts have inherent disadvantages, including a longer and less complete course of incorporation, remodeling, biomechanically inferiority to autograft, the potential risk of an immunogenic reaction and disease transmission. Higher long-term failure rates and poorer graft maturation scores were reported for allografts compared to autografts. An autograft in an ACL reconstruction should remain the gold standard, although the allograft is a reasonable alternative. If adequate length and diameter of autograft can be obtained for an ACL reconstruction, an autograft with adequate graft fixation and postoperative rehabilitation should be chosen instead of an allograft to achieve better results.
