This study was undertaken to understand development stage of ovary and changes of hormone concentrations in the Israeli carp, Cyprinus carpio from Ferbruary to May. The results obtained in these experiments are as follows: 1. Serum LH levels began to increase sharply in March, coinciding with the onset of rapid ovarian development. 2. LH levels were well correlated with changes in gonadosomatic index. 3. Dramatic increase in gonadosomatic index occured during the months of March. 4. Ripe stage(Stage VII) rapidly increase in March. 5. Early perinucleolus oocyte rapidly develop into late perinucleolus oocytes in March. 6. The vitellogenic phase begins as these late perinucleolus oocytes become transformed into early maturing oocytes through the accumulation of yolk. 7. The cytoplasm completely fills with yolk as oocytes reach the late maturing stage. 8. Changes in the microscopic appearances of the ovaries were well correlated with changes in both gonadosomatic index and macroscopic appearance. 9. It is concluded from these observations that LH plays a major role in sexual maturating of the Israeli carp.
Intensive farming methods that do not guarantee animal welfare can induce stress in pigs. Stress, in turn, can reduce their disease resistance and influence their hormones and metabolites in such a manner that productivity is negatively affected. This study was conducted to compare the stress related factors and blood characteristics of pigs raised on conventional farms and those raised on animal welfare farms. We measured the levels of cortisol, epinephrine and norepinephrine, biochemical parameters in blood and glycogen, L-lactate and heat shock protein 70 (HSP70) in muscle, as physiological markers of indicating the stress in conventional farm pigs (Control, n=10) and animal welfare farm pigs (Welfare, n=10). We found that there was a significant difference in the albumin-globulin ratio (A/G ratio), as well as the albumin (ALB), blood urea nitrogen (BUN) and aspartate aminotransferase (AST) levels between the two farms. Epinephrine was significantly higher in conventional farm, while level of norepinephrine was higher in animal welfare farm. There was no significant difference in cortisol, which is known as a stress hormone, across the two groups of farms. Muscular glycogen content was significantly high in animal welfare farm pigs. While L-lactate tended to be low in the animal welfare farm pigs, the difference between them and the conventional farm cohorts was not significant. HSP70 showed high levels of expression in conventional farm. Thus, we suggested that blood parameter results showed a stress response in the livers of conventional farm, and that catecholamine hormones, glycogen, L-lactate and HSP70 can be used as physiological factors of assessing animal welfare.

Objective: Polycystic ovary (PCO), a diagnostic component of polycystic ovary syndrome (PCOS), requires either an ovarian volume (OV) criterion or a follicle number per ovary (FNPO) criterion. This study investigated the association of OV and FNPO criteria with various manifestations of PCOS. Methods: This cross-sectional study was conducted at a university hospital among 100 patients newly diagnosed with PCOS (according to the revised Rotterdam criteria). Fasting blood samples were collected to measure glucose, total testosterone (TT), luteinizing hormone (LH), follicle-stimulating hormone (FSH), lipid, insulin, and hemoglobin A1c levels. An oral glucose tolerance test was performed. Transabdominal or transvaginal ultrasound of the ovaries was done, depending on patients' marital status. All investigations were conducted in the follicular phase of the menstrual cycle. OV >10 mL and/or FNPO ≥12 indicated PCO. A homeostasis model assessment of insulin resistance (IR) value ≥2.6 indicated IR, and metabolic syndrome (MS) was defined according to the international harmonization criteria. Results: Seventy-six participants fulfilled the OV criterion, 70 fulfilled the FNPO criterion, and 89 overall had PCO. Both maximum OV and mean OV had a significant correlation with TT levels (r=0.239, p=0.017 and r=0.280, p=0.005, respectively) and the LH/FSH ratio (r=0.212, p=0.034 and r=0.200, p=0.047, respectively). Mean OV also had a significant correlation with fasting insulin levels (r=0.210, p=0.036). Multivariate binary logistic regression analysis showed that IR (odds ratio [OR], 9.429; 95% confidence interval [CI], 1.701 to 52.271; p=0.010) and MS (OR, 7.952; 95% CI, 1.821 to 34.731; p=0.006) had significant predictive associations with OV alone, even after adjustment for age and body mass index. Conclusion: OV may be more closely related to the androgenic and metabolic characteristics of PCOS than FNPO.

This study aims to explore the impact of Rehmannioside D (RD) on ovarian functions of rats with diminished ovarian reserve (DOR) and its underlying mechanisms of action. A single injection of cyclophosphamide was performed to establish a DOR rat model, and fourteen days after the injection, the rats were intragastrically administrated with RD for two weeks. Rat estrus cycles were tested using vaginal smears. Ovarian tissues were histologically evaluated, the number of primordial, mature, and atretic follicles was calculated, and the apoptotic rate of granulosa cells. Follicle-stimulating hormone (FSH), luteinizing hormone (LH), and estradiol (E2) levels were determined by ELISA assays. Protein levels of Forkhead Box O1 (FOXO1), KLOTHO, Bcl-2, and Bax were investigated in ovarian tissues of DOR rats. The binding between FOXO1 and KLOTHO was verified by ChIP assay. High-dose administration of RD into DOR rats improved their estrus cycles, increased ovarian index, enhanced the number of primordial and mature follicles, reduced the number of atretic follicle number, and ovarian granulosa cell apoptosis in addition to inhibiting FSH and LH levels and upregulating E2 expression. FOXO1 and KLOTHO were significantly suppressed in DOR rats. FOXO1 knockdown partially suppressed the protective effects of RD on DOR rats, and KLOTHO overexpression could restore RD-induced blockade of DOR development despite knocking down FOXO1. FOXO1 antibody enriched KLOTHO promoter, and the binding between them was reduced in DOR group compared to that in sham group. RD improved ovarian functions in DOR rats and diminished granulosa cell apoptosis via the FOXO1/KLOTHO axis.

Objective: The number of bovine mammary epithelial cells (BMECs) is closely associated with the quantity of milk production in dairy cows; however, the optimal levels and the combined effects of hormones and essential amino acids (EAAs) on cell proliferation are not completely understood. Thus, the purpose of this study was to determine the optimal combination of individual hormones and EAAs for cell proliferation and related signaling pathways in BMECs. Methods: Immortalized BMECs (MAC-T) were treated with six hormones (insulin, cortisol, progesterone, estrone, 17β-estradiol, and epidermal growth factor) and ten EAAs (arginine, histidine, leucine, isoleucine, threonine, tryptophan, lysine, methionine, phenylalanine, and valine) for 24 h. Results: Cells were cultured in a medium containing 10% fetal bovine serum (FBS) as FBS supplemented at a concentration of 10% to 50% showed a comparable increase in cell proliferation rate. The optimized combination of four hormones (insulin, cortisol, progesterone, and 17β-estradiol) and 20% of a mixture of ten EAAs led to the highest cell proliferation rate, which led to a significant increase in cell cycle progression at the S and G2/M phases, in the protein levels of proliferating cell nuclear antigen and cyclin B1, cell nucleus staining, and in cell numbers. Conclusion: The optimal combination of hormones and EAAs increased BMEC proliferation by enhancing cell cycle progression in the S and G/2M phases. Our findings indicate that optimizing hormone and amino acid levels has the potential to enhance milk production, both in cell culture settings by promoting increased cell numbers, and in dairy cows by regulating feed intake.

Blood samples collected from green (chelonia mydas), loggerhead (caretta caretta), and Kemp's ridley (Lepidochelys kempii) sea turtles were analyzed by radioimmunoassay to detect seasonal variations in thyroid hormones and compare levels in wild and captive individuals. Sexual dimorphism in the annual cycle of thyroxine was observed in adult Kemp's ridley. No differences were observed between immature male and female green and loggerhead sea turtles. The level of triiodothyronine was consistently low relative to thyroxine in all species investigated, and captive sea turtles exhibit higher plasms thyroid levels than do wild ones.

The effects of orchildectomy with/without testosterone replacement in wale rats, and ovarectomy with estrogen replacement in female rats on lipid peroxidation were studied in male and female rats fed with diets fortified with 20% w/w, soybean oil or palm oil for 4 months. Serum, liver and heart homogenates were assayed for malonaldehyde and conjugated diene levels. Orchidectomy was found to reduce levels of lipid peroxidation products in the serum, liver and heart. Testosterone replacement did not increase the lipid peroxidation products to levels in the non-orchildectomised rats, while estrogen did not influence lipid peroxidation significantly. Palm oil decreased, but soybean oil increased lipid peroxidation in the liver and heart of both the castrated and sex hormone-replaced male and female rats.
Influence of the blockade of the three major pressor systems-sympathetic nervous system (SNS), renin-angiotensin system (RAS) and vasopressin system-on the pressor responsiveness to norepinephrine (NE), angiotensin II (AII), and vasopressin (VP) as well as on basal blood pressure (BP) levels was investigated in urethane-anesthetized rabbits. To block the SNS and RAS, chlorisondamine (CS) and pirenzepine (PZ), sympathetic ganglionic blockers, and enalapril (ENAL), an inhibitor of angiotensin converting enzyme, respectively were used. And for suppressing the VP system bremazocine (BREM), a kappa opiate receptor agonist shown to suppress plasma levels of VP, was employed. Each of CS (0.4 mg/kg), ENAL (2 mg/kg), and BREM (0.25 mg/kg) produced almost same levels of steady hypotensive state. The hypotensive effect of BREM was significantly attenuated by desmopressin, a synthetic VP-like analogue, suggesting the hypotension being at least in part due to suppression of plasma levels of VP. CS, ENAL and BREM elicited further fall of the BP which had been lowered by ENAL or BREM, CS or BREM, and CS or ENAL, respectively. The hypotension produced by both CS and PZ together with either of ENAL or BREM was more marked than that produced by the three drugs other than CS. CS potentiated the pressor response not only to NE but to AII and VP. The pressor effect of AII was increased by ENAL and BREM, too. The pressor response to VP was also enhanced by BREM. Blockade of ${\alpha}-adrenergic$ receptors with phentolamine or phenoxybenzamine potentiated the pressor response to AII and that to VP. The results on basal BP levels indicate that the three major pressor systems are all participating in control of BP, but SNS has the greatest potential for supporting BP. The finding that blockade of one of the pressor systems induced enhanced pressor responsiveness to the pressor hormone of that particular system as well as to the pressor hormone(s) of the other systems(s) provides evidence for important interactions among the three major pressor systems.
Two hundred sixteen crossbred ($Landrace{\times}Yorkshire$) castrates with an average weight of $7.4{\pm}0.3kg$ were used in a $3{\times}3$ factorial treatment array. The treatments were three levels of Herb mixture (HM; 0, 0.40 and 0.80 g/kg BW/day) and three levels of dietary nutrient (17.30% CP, Level-1; 17.90% CP, Level-2; and 18.50% CP, Level-3). The influence of HM intake and nutrient level on growth performance and ADG in 0.40- and 0.80-HM pigs increased significantly (p<0.01) as nutritional level was elevated. Although very little enhancement of ADG was observed at Level-1, peak ADG occurred in 0.8-HM treated pigs at Level-3. Feeding of 0.80 g HM/kg/d to pigs consuming Level-1 diet resulted in a 8.7% increase in ADG compared with control pigs, whereas the increase in ADG as a result of 0.80-HM with Level-3 treatment was 39%. ADFI in Level-2 pigs improved linearly (p<0.01) as HM level was increased. Treatment with HM resulted in a 12.0% increase ranging 4.7 to 20% in the ADFI compared with respective controls. ADFI at all nutritional level was significantly higher in 0.80-HM pigs (p<0.02). F/G in Level-2 pigs improved significantly as HM was fed (p<0.01), and in HM-0.80 pigs was also significantly improved as nutritional level was increased (p<0.05). Pigs fed HM had higher bone mineral density (BMD) at Level-1, longer dorsal spine length (DSL) at level-2 (p<0.05) than pigs fed basal diets. Pigs fed HM tended to higher BMD and DSL than those fed basal diets. The level of GH secretion declined with age. There was no difference between treatments (p>0.05) in the serum growth hormone at the same age. The GH was higher in pigs fed HM than those fed basal diets and increased in all pigs after 2wks feeding. A positive effect of added Herb-Mix on growth performance in weaned pigs was demonstrated by measuring the serum growth hormone, bone mineral density and length of dorsal spine.
