Tributyltin chloride (TBTCl) toxicity on the oxygen consumption rate and histological changes of gill in the equilateral venus, Gomphina veneriformis (Bivalvia: Veneridae) (대복, Gomphina veneriformis 아가미의 조직학적 변화와 산소소비율에 미치는 TBTCl의 독성)

  • This study was conducted to find out biological response of bivalves exposed to tributyltin chloride(TBTCl). The results of the study confirmed that TBTCl induce the reduction of oxygen consumption rateand histopathological feature in the gill structure of equilateral venus, Gomphina veneriformis. The experi-mental groups consisted of a control and 3 TBTCl exposure groups (0.4, 0.6 and 0.8 yg TBTCl L') and theexperimental period was 36 weeks. For histological analysis, gill tissues were fixed in Bouin's fluid andthen stained H-E stain, AB-PAS (PH 2.5) reaction and Masson's trichrome stain after having serially sec-tioned the tissue by paraffin method at thickness of 4-6 (an. The oxygen consumption rate was not signifi-cantly different between the control and exposure groups at 4 weeks, but in all exposure groups at 28 weeks,it was significantly different to the control. Gill of G. veneriformis had demibranch that attached two sheetsof lamellae and a lamella was composed of numerous filaments, numbering 25 on average. The frontal fila-ment zone had three types of cilia; frontal, latero-frontal and lateral depending on locations while the lateralcilia were the longest and largest in number. The mucous cells observed in filaments were more abundant in(542c) in AB-PAS (PH 2.5) reaction. Gill exposed to TBTCl was extended hemolymph sinus and increased hemocytes at 4 weeks, and then it showed increases of mucous cells and partially disappearance of frontalcilia. In the group of 0.8 yg TBTCl L' at 12 weeks, hypertrophy of frontal and latero-frontal epithelia wasobserved. Also it observed m decrease of mucous cell containing weekly acid mucosubstance and appearedpartially destruction muscle fiber bundle, In the groups of 0.4 and 0.6 ug TBTCl L' at 36 weeks, it appearedpartially modification of epithelia and in 0.8 us TBTCl L' group, observed filaments that come out chiti-nous rod from disappearance of frontal and latero-frontal epithelia.

Inhibitory effects of herbal medicine on coccidiosis (한약재의 첨가 급여에 따른 콕시듐 억제 효과)

  • This study examined the efficacy of natural ingredient formulated herbal medicine as feed additives against coccidiosis in broiler chicken. Medicinal herbs used in this study were Artemisia annua, Pulsatilla chinensis, Clematis chinensis and Scutellaria baicalensis. Two week old chicks were administered with 1% ingredient formulated herbal medicine. Observations of the efficacy were confirmed by examining the lesion score, histopathological appearance of cecum, shedding of fecal oocyst and production index in farm. S. baicalensis of the most effective substance were analyzed by UPLC. The results are as follows; among various types of Chinese medicine, the most efficacious ingredient was found to be pure bicalein. The major index substance is bicalein and $13{\mu}g/ml$ per gram was found. Feeding of chickens with S. baicalensis induced the coccidiostat effect and reduced the fecal shedding of E. tenella oocyst by 14%. Experimental results on a farm, feed conversion ratio and weight gain and production rate in the group with natural ingredient formulated additives than in control group increased. The rate of weight gain (%) in each experimental group showed 108% and 126% increase while the mortality rate was 1.4% lower than the control group. The investigation showed that the most efficacious ingredient was found to be S. baicalensis among herbal medicine products for coccidiostats. However, as it was found in the farm experiment that pure S. baicalensis alone is economically unviable. The additives with cocktailed of efficacious ingredient are suggested to be implemented for effective protection against coccidiosis.

A pathological study of spontaneous tumors in SPF Ktc : ICR mice (SPF Ktc : ICR 마우스의 자연발생 종양에 대한 병리학적 연구)

  • The study was carried out to obtain the basic data for types, incidence and histopathological features of the spontaneous tumors of the specific pathogen free (SPF) Ktc : ICR mice bred in a barrier system in Technology Research Center, Korea Research Institute of Chemical Technology. One hundred of the mice consisted of 50 males and 50 females were examined for 18 months. 1. The overall incidence rate of spontaneous tumors was 51(51%) of 100 heads tested. The male mice showed slightly higher incidence(28%) than the female(23%), and the incidence rate and the number of affected organs were increased with the increasing age of mice. 2. The incidence rate of primary tumor was 59(59%) of 100 heads tested, consisted of 30 cases (50.8%) of benign tumors and 29 cases(49.2%) of malignant tumors. Among the malignant tumors twenty cases were metastasized to various organs. 3. In tumor incidence rates by systems and organs, the male mice showed the high incidence rate in the liver (18%), hematopoietic system (16% ) and lung (14%), while the female mice, in the hematopoietic system(18%), lung(12%), liver(8%) and uterus(8%). 4. The tumors showing the particularly low incidence rates (<1.0%) were rhabdomyosarcoma in the skeletal muscle, malignant schwannoma in the peripheral nerve, cortical adenoma in the adrenal gland, transitional cell carcinoma in the urinary bladder, tubular cell adenoma in the kidney and adenoma in the pituitary gland and harderian gland.

Ultrastructural Change of Osmoregulatory Cells during Seawater Adaptation in Rainbow Trout (Oncorhynchus mykiss) (무지개송어의 해수순치과정에 일어나는 삼투조절세포의 미세구조)

  • There were observed the histomorphological alterations such as chloride cell hyperplasia, branchial lamellar epithelial separation, the increased cellular turnover of chloride cells, glomerular shrinkage and blood congestion in rainbow trout (Oncorhynchus mykiss) during the seawater adaptation. The ultrastructure by scanning electron microscope (SEM) indicated that the gill secondary lamella of rainbow trout exposed to seawater, were characterized by rough convoluted surfaces during the adaptation. There were observed a large number of mitochondria with the elongate and well-developed cristae in chloride cells exposed to seawater by transmission electron microscope (TEM). The presence of two mitochondria- rich cell types is discussed with regard to their possible role in the hypoosmoregulatory changes which occur during seawater-adaptation. Glomerulus shrinkage and blood congestion were occurred higher in nephrons of seawater-adapted fish than those living in freshwater. Our findings demonstrated that rainbow trout tolerated moderately saline environment and the increased body weight living in seawater was relatively higher than that living in freshwater in spite of histopathological changes.

Carnoy's Solution Application for Patient Preliminarily Diagnosed with Keratocystic Odotogenic Tumor: Case Report (각화낭성치성종양으로 의심되는 환자의 Carnoy's Solution 적용: 증례보고)

  • The odontogenic keratocyst (OKC) was originally classified as a developmental cyst, and OKC is histologically divided into orthokeratic and parakeratic types. According to revised histopathological classification of odontogenic tumors by the World Health Organization (2005), the term, keratocystic odontogenic tumor (KCOT) has been adopted to describe parakeratic OKC. The KCOT is noted for its capacity to recur after treatment. The aggressive, or potentially aggressive, nature of KCOT has led to alternative treatments. Among the treatments is noted the use of Carnoy's solution as adjunctive intralesional therapy having a low rate of recurrence. This study suggests a possible benefit of Carnoy's solution against recurrence of KCOT.

Pathological Interrelations of Soil-Borne Diseases in Cucurbits Caused by Fusarium Species and Meloidogyne incognita

  • Pathological interrelations of two soil-borne diseases in cucurbits (watermelon, oriental melon, shintosa and cucumber) caused by Fusarium isolates (FI) and the root-knot nematode (RKN), Meloidogyne incognita were characterized by the fusarium disease severity index (DI), RKN gall index (GI) and eggmass index (EI) in inoculation tests using FI and RKN. Virulence of FI as determined by DI at 4 weeks after inoculation was mostly in the higher order of Fusarium proliferatum F6, F5 and Fusarium oxysporum f. sp. melonis or Fusarium oxysporum f. sp. niveum with no significant differential interactions among the cucurbits and RKN co-infection. Significant increases of DI due to RKN coinfection were noticed in watermelon and oriental melon infected with F. proliferatum isolates, suggesting the DI increase due to RKN coinfection may depend upon the virulence of FI relative to aggressiveness of RKN on the cucurbits. For the coinfection of FI and RKN, GI and EI were mostly reduced logarithmically with the increase of DI, largely more in EI than GI, in all cucurbits except for shintosa. Microscopic examination of the root tissues showed histopathological features characteristic to infection types; formation of fungal hyphae and/or spores and plant defense structures (tyloses and mucilage) in variable degrees and formation of giant cells at variable developmental stages and with variable cytoplasmic depletion or degeneration which were visualized in relations with the values of DI, GI and EI. These findings will be helpful to develop control strategies of the soil-borne disease complex based on their pathological characteristics.

The Therapeutic Effect of Hovenia dulcis Thunberg Extracts and 6 Types of Herbal Extracts on $CCl_4-Induced$ Acute Liver and Kidney Damages in Rats (헛개나무추출액 및 생약성분의 혼용이 사염화탄소 투여로 유발된 흰쥐의 급성 간 및 신장손상의 회복에 미치는 영향)

  • This study was performed to investigate the therapeutic effects of Hovenia dulcis Thunberg (HDT) and HDTmix extracts on the biochemical analysis, histopathology and histomorphometry of liver and kidney in carbon tetrachloride $(CCl_4)$ administrated rats. Extract was prepared by autoclave ($121^{\circ}C$, 15 psi, 3 hours) and filtered with nylon cloth and filter paper then freezing dried. In blood chemistry analysis, HDTmix group, aspartate aminotransferase and alanine aminotransferase were significantly (p<0.01) decreased compared to the $CCl_4$ group, on 3rd day, respectively. In histologic and histomorphometry analysis, the $CCl_4-related$ hepatopathies and nephropathies were dramatically decreased (3rd, 5th day), and well corresponded to the histopathological changes significantly (p<0.01) decreases of degenerative regions, degenerative cells and glomeruli were detected in liver and kidney with significantly decreases of $CCl_4$ group. HDTmix group, quite similar effects on the liver and kidney were observed compared to that of HDT extracts group but more favorable efficacies were detected especially HDTmix also inhibit the hepatopathies (1 day), in which HDT extract does not showed any inhibit effects.

Growing pigs developed different types of diabetes induced by streptozotocin depending on their transcription factor 7-like 2 gene polymorphisms

  • The different polymorphisms of the transcription factor 7-like 2 (TCF7L2) gene promote variances in diabetes susceptibility in humans. We investigated whether these genotypes also promote differences in diabetic susceptibility in commercial pigs. Growing pigs (Landrace, both sex, 50-60 kg) with the C/C (n=4) and T/T (n=5) TCF7L2 genotypes were identified and intravenously injected with streptozotocin (STZ, 40 mg/kg) twice in weekly intervals, then a high-energy diet was offered. Oral glucose tolerance tests, blood analyses and the homeostasis model assessment-insulin resistance (HOMA-IR) index calculations were performed. The animals were sacrificed at the end of 12 weeks of treatment to reveal the pancreas histomorphometry. The results showed that all of the treated pigs grew normally despite exhibiting hyperglycemia at two weeks after the induction. The glycemic level of the fasting or postprandial pigs gradually returned to normal. The fasting insulin concentration was significantly decreased for the T/T carriers but not for the C/C carriers, and the resulting HOMA-IR index was significantly increased for the C/C genotype, indicating that the models of insulin dependence and resistance were respectively developed by T/T and C/C carriers. The histopathological results illustrated a significant reduction in the pancreas mass and insulin active sites, which suggested increased damage. The results obtained here could not be compared with previous studies because the TCF7L2 background has not been reported. Growing pigs may be an excellent model for diabetic in children if the animals are genetically pre-selected.

Practical Understanding of Gross Examination Techniques (육안검사기술의 실무적 이해)

  • Gross examination techniques (GETs) of specimens collected from cancer surgery or endoscopy comprise the act of recording visual information about cancer for accurate histopathological diagnosis and collecting sections of the lesion to create microscopic specimens. GETs must include concise and accurate expressions, appropriate structuring, sufficient resections, error-free standardization of important information, and photo-diagramming of complex specimens. To increase the satisfaction of pathological interpretation, it is a task that must be performed accurately and carefully to gain confidence on a theoretical and practical basis with a sufficient understanding of gross examination. Based on the experience of clinical pathologists in the field of GETs, additional specimen types should be identified as viable candidates. Also, their needs and concerns regarding treatment should be carefully considered. In addition, departments at each institution should review the national focus on clinical partnerships, continuous professional training, diagnostic errors, and value-based healthcare provision.

Clinical study of keratocystic odontogenic tumors

  • The odontogenic keratocyst (OKC) was originally classified as a developmental cyst, and OKCs were histologically divided into orthokeratotic (O-OKCs) and parakeratotic (P-OKCs) types. Clinical features differ between O-OKCs and P-OKCs with P-OKCs having a tendency to recur after surgical treatment. According to the revised histopathological classification of odontogenic tumors by the World Health Organization (2005), the term keratocystic odontogenic tumor (KCOT) has been adopted to describe P-OKCs. In this retrospective study, we examined 186 KCOTs treated at the Maxillofacial Surgery Department of the Tokyo Medical and Dental University Hospital from 1981 through 2005. The patients ranged in age from 7 to 85 years (mean, 32.7) and consisted of 93 males and 93 females. The most frequently treated areas were the mandibular molar region and ramus. The majority of KCOTs in the maxillary region were treated by enucleation and primary closure. The majority of KCOTs in the mandibular region were enucleated, and the wound was left open. Marginal resection was performed in the 4 patients with large lesions arising in the mandible. In patients who were followed for more than a year, recurrences were observed in 19 of 120 lesions (15.8%). The recurrences were found at the margins of the primary lesion in contact with the roots of the teeth or at the upper margins of the mandibular ramus. Clinicians should consider aggressive treatment for KCOTs because the recurrence rate of P-OKCs is higher than that of other cyst types such as O-OKCs, dentigerous cysts, primordial cysts that were non-keratinized, and slightly keratinized stratified squamous epithelium. Although more aggressive treatment is needed for KCOTs as compared to other cystic lesions, it is difficult to make a precise diagnosis preoperatively on the basis of clinical features and X-ray imaging. Therefore, preoperative biopsy is necessary for selecting the appropriate treatment for patients with cystic lesions.
