The Effects of ChondroT on Thrombus in FeCl3-induced Rats (ChondroT 투여가 FeCl3로 유발된 혈전에 미치는 영향)

  • Objectives This study was designed to investigate the effects of ChondroT on thrombus in FeCl3-induced rats. Methods We exposed FeCl3 to rat's carotid artery to induce thrombus. Specimens were divided in 5 groups; Intact, Control, ASA10 (aspirin 10 mg/kg), CT100 (ChondroT 100 mg/kg), and CT200 (ChondroT 200 mg/kg), each n=6. We investigated thromboxane, platelet activating factor (PAF), histological change, lipid metabolism, transaminase, leukocyte, erythrocyte and thrombocyte level. Results In ASA10, CT200 groups, there was significants decrease in both thromboxane level and total cholesterol level, compared to control group and there were significant histological changes of blood vessel, compared to control group. In CT200 group, there was significant decrease in PAF level, compared to control group (p<0.05). In ASA10, CT200 groups, triglycerides level tended to decrease, compared to control group. Conclusions Based on these results, it could be suggested that ChondroT was effective on thrombus in FeCl3-induced rats, and further study is needed to conduct a rigorous clinical research.

  • Histological changes occurred by freezing storage and subsequent thawing, and changes of muscle fiber after heating and drying of the eel (Anguilla japonica) were observed under microscope . The results are as follows : (1) After one month of freezing storage the muscular tissue produced considerable number of extracellular and intracellular ice crystals. The sample stored at the temperature of $-40^{\circ}C$ produced ice crystals inside the muscle cells while sample stored at $-20^{\circ}C$, outside. (2) No changes were observed in the hypodermic fat after thawing regardless of storage temperature, while insufficient recovering of muscle cells were detected in the muscular tissue. Muscular tissues which have been stored $-20^{\circ}C$ showed severe change in shape due to dehydration. (3) Microscopic observation on muscle homogenate showed loss of transparency due to free-zing, disfiguration and contraction by drying and water seperation, and elasticity by heating.

Effects of Starvation on the Morphometric Characteristics and Histological Changes in Chum Salmon (Oncorhynchus keta) Fry

  • A 26 day experiment was conducted to determine the effects of feeding and starvation on the survival, morphology, and histology in chum salmon (Oncorhynchus keta) fry. We included three experimental groups: starved, fed, and initial. The survival and growth rates were lower in the starved group than in the fed group (P < 0.05). In the starved group, survival began to decline after 16 days, and all fish had died after 26 days. We determined the effects of starvation on the morphometric parameters using the truss and classical dimensions. The dimensions in the head region were larger in the starved group than in the initial and fed groups. In contrast, the truss dimensions of the fed group were larger than those of the initial and starved groups. Starvation reduced the heights of the hepatocyte nuclei and of the intestinal epithelium (P < 0.05). The starved group also showed atrophy of the digestive structures and shrinkage of the foregut and midgut. Starvation led to the degeneration and atrophy of the exocrine pancreas, in which the lumen was markedly diminished and the folds of mucosa were less apparent. The hepatocyte morphology in the starved group was abnormal compared with that of the initial and fed groups, with highly compact, irregularly shrunken nuclei. Melanomacrophages were randomly distributed in the kidneys of the starved group, and their abundance increased rapidly during the experiment. In contrast, neither the initial nor fed group had any melanomacrophages. These results suggest that the nutritional parameters used in this study are useful indices of nutritional status in chum salmon.

Effects of Different Light Spectra on the Oocyte Maturation in Grass Puffer Takifugu niphobles

  • In order to examine the effects of four different light spectra (white, red, green, and blue) on the oocyte maturation, the change of reproductive parameters, via brain-pituitary-gonad (BPG) axis in grass puffer, were investigated. After exposure four different light spectra for 7 weeks, the abundance of gonadotropin-releasing hormone (GnRH) mRNA which is a type of seabream (sbGnRH) and two different subunit of gonadotropin hormones mRNAs, follicle-stimulating hormone ($fsh{\beta}$) mRNA and luteinizing hormone ($lh{\beta}$) mRNA, were analyzed in the brain and pituitary. Histological analysis showed that the mature oocyte ratio in the green spectrum was higher than other light spectra-exposed groups. Gonadosomatic index (GSI) and oocyte developmental stage were also investigated in the gonad based on histological observations. GSI value with the presence of yolk stage oocytes was significantly increased in the green spectrum-exposed group when compared to that of the other light-exposed groups (white, red, and blue) (p<0.05). The abundances of sbGnRH mRNA and $fsh{\beta}$ mRNA in the green spectrum-exposed group were also significant higher than those of the other light spectra-exposed groups (p<0.05). These results indicate that the maturation of oocyte in grass puffer can be accelerated by exposure to the spectrum of green. To better understand the molecular mechanism for the maturation of oocyte in grass puffer, further study examining the relationship between oocyte development and its related genes is required.

Studies on the Cyst Occurred in the Mesosalpinx, Mesovarium and Fimbria in the Gilts and Sows (자돈(雌豚)의 난관간막(卵管間膜), 난소간막(卵巢間膜) 및 난관채(卵管綵)에 형성(形成)된 낭종(囊腫)에 관한 연구(硏究))

  • Gross and histological findings were obtained on cysts formed in the reproductive organs, particularly in the mesosalpinx, mesovarium, and fimbria, of 144 gilts and 37 sows. Of the 181 gilts and sows, 63 (34.8%) were found to have cysts which totaled 89. The number of cysts totaled 38 for the gilts and 51 for the sows, with relative frequencies of 18.1% and 48.6%, respectively. The total number of cysts and the incidence were greater in the sows than in the gilts. With regard to the location of cysts, the highest incidence was found in the mesosalpinx (59.6%). The incidence of cysts in the mesovarium and fimbria was relatively low, with 16.9% and 23.6%, respectively. Most cysts were spherical or oval in shape, and below 5.9mm in diameter. A pedunculated cyst was also seen. The bilateral, incidence of the cysts was lower than unilateral, although the incidence was approxmately the same between the gilts and the sows. With regard to the sexual cycle, the high incidence of cysts was found in the follicular phase than in the luteal phase. The cysts putting pressure upon the oviduct were classified into 3 types according to the shape of epithelium and lamina propria. In type I the epithelium had ciliated cells and the lamina propria formed folds. I type 2 ciliated cells existed, but no folds were formed. In type 3, neither ciliated cells nor folds were seen, the histological change, including partial atrophy and lack of folds in the tubal mucosa were seen.

Clinical significance of serum alanine aminotransferase and lifestyle intervention in children with nonalcoholic fatty liver disease

  • Purpose: This study aimed to investigate the clinical significance of serum alanine aminotransferase (ALT) levels in children with nonalcoholic fatty liver disease (NAFLD) and the effect of lifestyle intervention on NAFLD. Methods: The clinical data of 86 children diagnosed with NAFLD were reviewed retrospectively. Forty-six patients belonged to the elevated ALT group and 40 to the normal ALT group. The clinical parameters of patients with NAFLD were also compared based on the status of ALT levels after lifestyle intervention. Results: Patients with elevated ALT had significantly higher body mass index (BMI) scores than those with normal ALT (P<0.05). Of all the patients with elevated ALT, 89% exhibited moderate or severe degree of fatty change in the liver on ultrasonographic examination, whereas most patients with normal ALT exhibited mild or moderate degree changes. Liver biopsy was performed in 15 children with elevated ALT and all showed mild histological changes. Of all patients with elevated ALT, 49% achieved normal ALT levels after lifestyle intervention. Those with more severe histological changes tended to have continuously increasing ALT levels. There was no correlation between the normalization of posttreatment ALT level and BMI, as well as ultrasonographic findings at diagnosis. Conclusion: ALT elevation in NAFLD is highly associated with higher BMI scores and more severe degree of fatty changes on ultrasonographic examination. Lifestyle intervention can significantly improve ALT in children with NAFLD. The degree of histologic changes appears to be a predictor of the treatment response to NAFLD.

Changes in Plasma Steroid Hormone Levels and Gonad Development by the Control of Photoperiods and Water Temperatures on Timing of Sexual Maturity of Rockfish (Sebastes schlegeli)

  • Plasma steroid hormone levels in the viviparous rockfish (Sebastes schlegeli) were examined in relation to gonadal histology under controlled photoperiods and water temperatures. To investigate those effects in S. schlegeli the photoperiod was maintained at 15L:9D in June and then it was gradually decreased to 9L: 15D in October. It was then gradually increased to 12L:12D in January, followed by 14L:I0D in February. The water temperature was $19-20^{\circ}C$ in July. From August to October, it was from $18^{\circ}C\;to\;12^{\circ}C$. Then, it was dropped to a low of $19-11^{\circ}C$ in November to December and then gradually increased to $14-15^{\circ}C$ in February. In females, both plasma $estradiol-l7\beta\;$ (E2) and testosterone (T) levels from August to February showed a similar pattern in both the treatment and the control groups. In the treatment group, the peaks of plasma E2 and T were observed in November, and the peaks were closely correlated to histological observations. Oocytes contained many yolk globules (final vitellogenic oocytes), and oocytes at the migratory nucleus stage increased in size. Plasma levels of progesterone did not change much throughout the experimental period. However, in the control group, the peaks of E2, T, and progesterone were observed in February. These results indicate that the controlled photoperiod and water temperature accelerated sexual maturity, corresponding to the advancement of plasma E2 and T peaks by approximately 3 months. In males, plasma T levels showed a similar pattern from August to October in the treatment and control groups, though levels in the treatment group were higher than those in the control group. From histological observations, the treatment group copulated one month earlier.

Survival, Hematological Characteristics, Hemocyte Mortality and Histological Changes of Korean Rockfish Sebastes schlegeli Held in Live Fish Containers (활어 수송용 컨테이너에 수용된 조피볼락(Sebastes schlegeli)의 생존율, 혈액성상, 혈구 사멸률 및 조직학적 변화)

  • This study investigated the survival rates and physiological responses of Korean rockfish, Sebastes schlegeli, in live fish containers ($8^{\circ}C$, 33 psu) for 18 days. The survival rate was 99% and 97% in the control and experimental groups, respectively. Hematocrit and hemoglobin did not differ significantly between the control and experimental groups. Glucose and cortisol rose immediately on the first day of containment, but both gradually normalized. Aspartate aminotransferase and alanine aminotransferase did not differ significantly between the two groups after recovery. $NH_3$ continued to rise after the first day, but decreased to a level not significantly different from that of the control group during the recovery period. Plasma ions and osmolality did not change abnormally. The hemocyte population was not significantly different from that of the control. The ratios of apoptotic and necrotic cells indicated no specific variation in hemocyte viability. The histological changes in the skin and gills were not significantly different from those seen in the control. The experimental data obtained in this study suggest that live fish containers may be used to transport Korean rockfish without significantly affecting their physiology or survival.

A Study for Normal Development of the Posterior Cranial Fossa in the Chick Embryos (gestation 14-20 days) with MR Images and Histopathology (정상 닭배자(배양14-20일)의 후두와 발생과정에 대한 연구 : 자기공명영상 및 해부병리학적 소견)

  • The objective was to use MR imaging to provide a template of posterior fossa development during the late stages in the chick embryos. The MR findings were then correlated with histological data. Fourteen normal formalin-fixed embryonic specimens with a gestational age of 14 to 20 days were examined with 1.5 Tesla unit MRl using a conventional clinical magnet and pulse sequences. The MR findings were correlated with the whole-mount histological specimens. Resolution of the morphological features of posterior fossa development in embryos greater than 14 days gestational age was possible. Development of cerebellum, brain stem, 4th ventricle and bony posterior fossa was documented. In the 14-day-old embryos, a premordial cerebellum was visualized in the enlarged bony posterior fossa, and it covered the the roof of the primitive fourth ventricle. The bony posterior fossa grows at the same rate along the supratentorial skull. The supratentorial skull and the rostral part of the brain grows at the same rate. The cerebellum begins to grow later than the rostral part of the brain. In the 19- to 20-day-old embryos, MRl revealed the rapid development of the cerebellar hemispheres, along with an increase in volume manifested by the more typical mushroom-shaped configuration observed in the newly hatched. At this stage, the cerebellum almost completely filled the posterior fossa and covered the entire fourth ventricle. The brain stem grew steadily, but the volume change was too subtle to evaluate. Features of cerebellar histogeneis were beyond the resolution of MRl. However, there were lots of artifacts in the features of the bony posterior fossa. An MR template of normal posterior fossa development would be useful to avoid confusion of normal development with abnormal development and to identify the expected developmental features when provided the estimated gestational age of a embryo.

Effects of Gamma Irradiation on the Mid-gut of Hyphantria Cunea (흰불나방의 중장(中腸)에 미치는 $\gamma$선(線) 조사(照射)의 영향(影響))

  • In this paper, the author studied the histological changes of the midgut cells of fall webworms(Hyphantria cunea Drury) through 1.75-7 krad of the whole body gamma irradiation according to their metamorphosis by comparing the contol group with the irradiated one through an optical microscope. Here the results were as follows: 1. The epithelium of midgut was composed of columnar, goblet and regenerative cells. 2. The effects of gamma irradiation were varied with the dosages and the stages during the metamorphosis. 3. The degree of histological change mode by irradiation was increased with the dosages. 4. Radiosensitivity was the highest in both last-stage larva and 8-day-old pupae.
