A simplified model for predicting microsegregation during columnar-dendritic solidification of binary alloys is developed, in which back diffusion, dendrite arm coarsening and dendrite tip undercooling are simultaneously incorporated. The inclusion of tip undercooling is accomplished by modifying the initial conditions of the existing solute diffusion model, in such forms that tip undercooling depresses the beginning of solidification below the liquidus temperature, and that the secondary arm spacing evolves in accordance with the minimum undercooling theory. Sample calculations for the well-known benchmark system show that the present predictions not only consist with the extablished limiting cases, but also agree favorably with the available experimental data within a reasonable tolerance. In particular, a typical decreasing trend in the eutectic fraction at high cooling rates is successfully resolved. Comparison of the individual and combined effects of characteristic parameters in reference with the limiting cases reveals the interactions among parameters. Every parameter plays the role of reducing the eutectic fraction, and the degree of influence depends primarily on the cooling rate. Coarsening enhances the effect of tip undercooling, while suppressing that of back diffusion. A vigorous back diffusion seems to restrain the apperance of the undercooling effect. Overall, each contribution of the three parameters to microsegregation is estimated to be of the same order, which suffices to justify the present study.
Two strains of yeast (RA-74-2 and RA-912) showing superior fermenting ability at a high temperature were isolated from soils and wastewaters by an enrichment culture method. Based on the morphological and physiological charateristics, the two strains were identified as Saccharomyces cerevisiae and Kluyveromyces marxianus, respectively. RA-74-2 was able to grow upto $43^{\circ}C$ and sustain similar fermenting ability in the temperatures range from 30 to $40^{\circ}C$. In addition, the sugar- and ethanol-tolerance of RA-74-2 were 30% (w/v) glucose and 10% (v/v) ethanol, which appeared to be higher than those of nine other industrial yeast strains currently being used in the alcohol factories. The thermotolerant ethanol fermenting yeast RA-912 showed identical growth in the temperatures range from 35 to $45^{\circ}C$ and was resistant to various heavy metals. The quality and quantity of byproducts of the isolated yeast strains in fermentation broth after fermentation at $40^{\circ}C$ and $45^{\circ}C$ were similiar with those obtained at $30^{\circ}C$. These results show that RA-74-2 can be adopted for the ethanol fermentation process where the expenses for cooling system is significant, and suggest that RA-912 may be applied in either SSF(simultaneous saccharification and fermentation) or Flash-fermentation process and RA-912 may be used as a gene donor for the development of thermotolerant ethanol-fermenting yeasts.
Glutaraldehyde was used as a cross-linking agent for immobilization of purified ${\alpha}$-amylase from Exiguobacterium sp. DAU5. Befitting concentration of glutaradehyde and cross-linking time is the key to preparation of cross-linking chitosan beads. Based on optimized immobilization condition for ${\alpha}$-amylase, an overall yield of 56% with specific activity of 2,240 U/g on chitosan beads and 58% with specific activity of 2,320 U/g on chitosan-carbon beads was obtained. The optimal temperature and pH of each immobilized enzyme activity were $50^{\circ}C$ and 50 mM glycine-NaOH buffer pH 8.5, respectively. Those retained more than 75 and 90% of its maximal enzyme activity at pH 7.0-9.5 and after incubation at $50^{\circ}C$ for 1 h, respectively. In addition, the immobilization product showed higher organic-solvent tolerance than free enzymes. The mode of hydrolyzing soluble starch revealed that the ${\alpha}$-amylase possessed high hydrolyzing activity. These results indicate that chitosan is good support and has broad application prospects of enzyme immobilization.
With the objective of identifying the commercial potential of new direct-fed microbials, several temperature-tolerant strains were isolated from cane molasses at $39^{\circ}C$ and tested for their tolerance to pH, bile salts, and a mixture of volatile fatty acids (acetic acid:propionic acid:butyric acid=6.5:2.0:1.5). It was found that the isolated strain DY 252 grew very well up to pH 2.0 and was resistant to relatively high concentrations of bile salts. Among the strains tested, DY 252 was least inhibited by the addition of volatile fatty acids to the growth medium at $39^{\circ}C$. Accordingly, it would appear that strain DY 252, identified as yeast Issatchenkia orientalis, may be a potential candidate for use as a microbial feed additive.
The objective of this study is to predict the roll wear in hot rod rolling process. In this study hot rod rolling process for round and oval passes has been investigated. In order to predict the roll wear, the wear model is reformulated as an incremental form and then wear depth of roll is calculated at each deformation step on contact area using the results of finite element analysis, such as relative sliding velocity and normal pressure at contact area. Archard's wear model was applied to predict the roll wear. To know the thermal softening of DCI (Ductile Cast Iron) roll according to operating conditions, high temperature micro hardness test is executed and a new wear model has been proposed by considering the thermal softening of DCI roll expressed in terms of the main tempering parameter curve. 3D wear program developed in this study might be used for adjusting the gap of rolls to set up a suitable rolling schedule for keeping dimensional tolerance of the product.
The band-width of disk drive servo system is rapidly increasing for the robustness to external disturbance as the track density is increasing. The increase of the band-width may cause mechanical resonance of an actuator. In disk drive servo system, a notch filter is usually used to suppress the mechanical resonance of the actuator. However, the resonance frequency differs from drive to drive because of manufacturing tolerance and varies with temperature even within a single drive. The variation of resonance frequency degrades the suppression performance of the notch filter. In this paper, we present an adaptive digital notch filter that identifies the resonance frequency of the disk drive servo actutaor precisely and adjusts automatically its center frequency. For this, we design an adaptive FIR digital filter for the estimation of the resonance frequency. The estimation filter identifies the resonance frequency adaptively using the output signal generated from the servo system, which is excited with an excitation signal including all the expected resonance frequency components. We prove mathematically the convergence of the resonance frequency estimation filter. Furthermore, in order to demonstrate the practical use of our work, we present some experimental results using a commercially available disk drive.
Micro engine that includes Micro scale combustor is fabricated. Design target was focused on the observation of combustion driven actuation in MEMS scale. Combustor design parameters are somewhat less than the size recommended by feasibility test. The engine structure is fabricated by isotropic etching of the photosensitive glass wafers. Electrode formed by electroplating of the Nickel. Photosensitive glass can be etched isotropically with almost vertical angle. Bonding and assembly of structured photosensitive glass wafer form the engine. Combustor size was determined to be 1 mm scale. Movable piston is engraved inside the wafer. Ignition was done by nickel spark plug which was electroplated with thickness of 40 ${\mu}{\textrm}{m}$. The wafers were bonded by epoxy that resists high temperature. In firing test due to the bonding method and design tolerance pressure buildup by reaction was not confirmed. But ignition, flame propagation and actuation of micro structure from the reaction was observed. From the result basement of design and fabrication technology was obtained.
To study the functioning of HSP70 in Escherichia coli, we selected NtHSP70-2 (AY372070) from among three genomic clones isolated in Nicotiana tabacum. Recombinant NtHSP70-2, containing a hexahistidine tag at the amino-terminus, was constructed, expressed in E. coli, and purified by $Ni^{2+}$ affinity chromatography and Q Sepharose Fast Flow anion exchange chromatography. The expressed fusion protein, $H_6NtHSP70$-2 (hexahistidine-tagged Nicotiana tabacum heat shock protein 70-2), maintained the stability of E. coli proteins up to 90$^{\circ}C$. Measuring the light scattering of luciferase (luc) revealed that NtHSP70-2 prevents the aggregation of luc without ATP during high-temperature stress. In a functional bioassay (1 h at 50$^{\circ}C$) for recombinant $H_6NtHSP70$-2, E. coli cells overexpressing $H_6NtHSP70$-2 survived about seven times longer than those lacking $H_6NtHSP70$-2. After 2 h at 50$^{\circ}C$, only the E. coli overexpressing $H_6NtHSP70$-2 survived under such conditions. Our NtHSP70-2 bioassays, as well as in vitro studies, strongly suggest that HSP70 confers thermo-tolerance to E. coli.
The electrochemical performance and Cr poisoning behavior of $La_{1-x}Ba_xCo_{0.9}Fe_{0.1}O_{3-{\delta}}$ (LBCF, x = 0.3, 0.4, 0.5) and $La_{0.6}Sr_{0.4}Co_{0.2}Fe_{0.8}O_{3-{\delta}}$ (LSCF) cathodes were investigated for solid oxide fuel cells (SOFCs). The polarization resistance of the LBCF/GDC/LBCF symmetrical cell was found to decrease with increasing Ba content (x value). This phenomenon might be associated with the high oxygen vacancy concentration in the LBCF sample, with x = 0.5. In addition, there was no chromium poisoning in the LBCF cathode. On the other hand, the polarization resistance of the LSCF cathode was found to significantly increase after exposure to gaseous chromium species; it appears that this result stemmed from the formation of $SrCrO_4$ phase. Therefore, it can be expected that LBCF can be a durable potential cathode material for intermediate-temperature solid oxide fuel cells (IT-SOFC).
In this paper, we focused on the self-healing concrete using new nano-capsules. Three types of nano-capsules with respect to availability, high strength and temperature tolerance are used; type 1 is URF and polyethylene (PE) as shell and nano titanium oxide (TiO2) as core, type 2 is URF and PE as shell and nano silica oxide (SiO2) as core, type 3 is PE as shell and nano silica oxide (SiO2) as core. The concrete samples mixed by nano-capsules with three percents of 0.5, 1 and 1.5. Based on experimental tests and the compressive strength of samples, the URF-PE-SiO2 is selected for additional tests of compressive strength before and after recovery, ultrasonic test, ion chlorine and water penetration depths. After careful investigation, it is concluded that the optimum value of URF-PE-SiO2 nano-capsules is 0.5% since leads to higher compressive strength, ultrasonic test, ion chlorine and water penetration depths.
