Polyamine 농도에 미치는 효소에 관한 연구 대두의 유묘에서 IAA가 Lysine Decarboxylase와 Diamine Oxidase의 활성과 Polyamine함량에 미치는 영향 (Studied on Enzymes Responsible for Polyamine Level Effects of IAA on the activities of lysine decarboxylase and diamine oxidase and polyamine level in seedlings of soybean)

  • The effect of IAA on lysine decarboxylase(LDC)activity, diamine oxidase (DAO) activity and the polyamine level in seedlings of soybean were investigated. The seedlings of soybean were treated with IAA (10-8M, 10-6M, 10-4M) and LDC activity, DAO activity and polyamine levels of cotyledon, elongating hypocotyl, elongated hypocotyl and the root were measured after 4, 6, 8, 10 days. Increased concentration of IAA stimulated the LDC activity and increased the level of polyamine in the seedlings, which reducing the DAO activity and polyamine level was increased in the seedlings. The light also stimulated the IAA effect on polyamine level and DAO activity, and polyamine level was higher in the seedlings grown in the light than those in the dark. Cadaverine was the most abundant polyamine in the seedlings of soybean. The levels of diamines, cadaverine, putrescine were higher in the elongated hypocotyl and root than in the elongating hypocotyl, but the levels of spermidine and spermine were relatively higher in the cotyledon than in the other parts. Accumulation of diamines by high concentration of IAA is postulated to be a phenomenon of inhibition in seedlings growth.

Predicting standardized ileal digestibility of lysine in full-fat soybeans using chemical composition and physical characteristics

  • Objective: The present work was conducted to evaluate suitable variables and develop prediction equations using chemical composition and physical characteristics for estimating standardized ileal digestibility (SID) of lysine (Lys) in full-fat soybeans (FFSB). Methods: The chemical composition and physical characteristics were determined including trypsin inhibitor activity (TIA), urease activity (UA), protein solubility in 0.2% potassium hydroxide (KOH), protein dispersibility index (PDI), lysine to crude protein ratio (Lys:CP), reactive Lys:CP ratio, neutral detergent fiber, neutral detergent insoluble nitrogen (NDIN), acid detergent insoluble nitrogen (ADIN), acid detergent fiber, L* (lightness), and a* (redness). Pearson's correlation (r) was computed, and the relationship between variables was determined by linear or quadratic regression. Stepwise multiple regression was performed to develop prediction equations for SID of Lys. Results: Negative correlations (p<0.01) between SID of Lys and protein quality indicators were observed for TIA (r = -0.80), PDI (r = -0.80), and UA (r = -0.76). The SID of Lys also showed a quadratic response (p<0.01) to UA, NDIN, TIA, L*, KOH, a* and Lys:CP. The best-fit model for predicting SID of Lys in FFSB included TIA, UA, NDIN, and ADIN, resulting in the highest coefficient of determination (R2 = 0.94). Conclusion: Quadratic regression with one variable indicated the high accuracy for UA, NDIN, TIA, and PDI. The multiple linear regression including TIA, UA, NDIN, and ADIN is an alternative model used to predict SID of Lys in FFSB to improve the accuracy. Therefore, multiple indicators are warranted to assess either insufficient or excessive heat treatment accurately, which can be employed by the feed industry as measures for quality control purposes to predict SID of Lys in FFSB.

고라이신(QPM) 주요 계통과 교잡계의 생육특성 및 아미노산 조성 변이 (Growth Characteristics and Variation in Amino Acids Composition of Quality Protein Maize Lines)

  • 본 연구는 non-QPM인 KS140, QPM인 CML 계통 및 교잡계의 생육 특성과 단백질, 지방산 등 일반성분, 아미노산 조성을 분석하여 국내 적응 QPM 신품종 육성을 위한 기초연구 자료로 활용하고자 수행하였다. 1. 출사일수는 CML 계통이 78~90일, CML 교잡계가 81~87일 이었고 평균 출사일 수는 KS140, CML 계통 및 교잡계 모두 85일 이었다. CML계통 중 CML191이 간장과 착수고 모두 155 cm, 72 cm로 가장 낮았고, CML529의 간장이 242 cm, CML164의 착수고가 139 cm로 가장 높았다. 과피색은 KS140이 노란색, CML 계통은 흰색과 노란색이었고, 교잡계는 CML 계통이 흰색인 경우에는 노란과 흰색의 중간색을 나타내었다. 2. CML 계통의 단백질 함량은 9.1~12.1% 범위로 CML153이 가장 적었고 CML191이 가장 많았다. CML 교잡계의 단백질 함량은 9.1~11.1% 범위로 KS140/CML170이 가장 적었고, KS140/CML188이 가장 많았다. 3. 지방산의 조성비는 KS140, CML 계통과 교잡계 모두 C18:2 (linoleic acid)가 가장 높았고 C18:1 (oleic acid), C16:0 (palmitic acid), C18:0 (stearic acid), C18:3 (linolenic acid) 순이었다. CML 계통의 평균 포화지방산 비율은 21.4%였으며 불포화지방산 조성비는 78.6%로 KS140 보다 포화지방산 조성비가 다소 높았다. 4. 아미노산 조성비 중 KS140, CML 계통 및 교잡계에서 Glutamic acid 함량이 가장 높았으며 lysine의 조성비는 non-QPM인 KS140이 2.51%, QPM인 CML 계통 평균이 4.83%였으며 CML155가 6.92%로 가장 높았다. CML 교잡계 평균은 3.46%였고 KS140/CML164와 KS140/CML170의 lysine 조성비는 각각 4.18%과 4.99%로 교배하기 전의 QPM계통인 CML164, CML170보다 높았다. 5. 아미노산 조성에 따라 시험 계통들은 크게 두 개 그룹으로 나눌 수 있었으며 CML 계통도 두 개의 그룹으로 나누어졌다. CML155, 180, 181, 191을 포함하는 Group은 lysine을 비롯하여 histidine, glycine, threonine, serine의 조성비가 높았고, CML153, 157, 164, 170, 177을 포함하는 Group은 아미노산 조성비는 상대적으로 낮았다.

Transesterification Using the Cross-Linked Enzyme Aggregate of Photobacterium lipolyticum Lipase M37

  • Biodiesel is methyl and ethyl esters of long-chain fatty acids produced from vegetable oils or animal fats. Lipase enzymes have occasionally been used for the production of this biofuel. Recently, biodiesel production using immobilized lipase has received increased attention. Through enhanced stability and reusability, immobilized lipase can contribute to the reduction of the costs inherent to biodiesel production. In this study, methanol-tolerant lipase M37 from Photobacterium lipolyticum was immobilized using the cross-linked enzyme aggregate (CLEA) method. Lipase M37 has a high lysine content (9.7%) in its protein sequence. Most lysine residues are located evenly over the surface of the protein, except for the lid structure region, which makes the CLEA preparation yield quite high (~93%). CLEA M37 evidences an optimal temperature of $30^{\circ}C$, and an optimal pH of 9-10. It was stable up to $50^{\circ}C$ and in a pH range of 4.0-11.0. Both soluble M37 and CLEA M37 were stable in the presence of high concentrations of methanol, ethanol, 1-propanol, and n-butanol. That is, their activities were maintained at solvent concentrations above 10% (v/v). CLEA M37 could produce biodiesel from olive oil and alcohols such as methanol and ethanol. Additionally, CLEA M37 generated biodiesel via both 2-step methanol feeding procedures. Considering its physical stability and reusability, CLEA M37 may potentially be used as a catalyst in organic synthesis, including the biodiesel production reaction.

  • The seed, or grain, of modern cultivars of Lupinus angustifolius, commonly known as Australian sweet lupins (ASL), is an established feed resource for the intensive animal industries of Australia, Japan, Korea and several other countries in Asia and Europe. Since the introduction of ASL to the world marketplace about 25 years ago, researchers in many countries have found them to be a valuable component of the diet of beef and dairy cattle, sheep, pigs, poultry, finfish and crustaceans. The seed of ASL contains ~32% crude protein (CP) (~35% DM basis) and 5% oil. The main storage carbohydrates in the seed are the ${\beta}$-galactans that comprise most of the cell-wall material of the kernel and the cellulose and hemicellulose of the thick seed coats. ASL seeds contain about 40% non-starch polysaccharides (NSP) and a negligible amount of starch. This makes them an excellent ingredient for ruminant diets, as the risk of acidosis is very low. The seed of modern cultivars of domesticated Lupinus species contain negligible amounts of lectins and trypsin inhibitors so they do not require preheating before being used as an ingredient in feeds for monogastric species. They have a high digestibility coefficient for protein, >90% for most species, but a low energy digestibility, ~60%, which is mostly due to the high content of NSP. The low content of methionine (0.22%) and of lysine (1.46%) is typical of the legumes. The lysine availability for pigs is >70%. Lupin kernels contain ~39% CP (~42% DM basis), 6% oil and 30% NSP. They have a higher digestible energy for pigs and finfish and a higher metabolisable energy for poultry than whole seed. Commercial operations rarely achieve complete separation of kernel from hull and it is more likely that the kernel fraction, called splits or meats, will contain ~36% CP. The replacement of soybean meal or peas with ASL in cereal-based diets for most intensively reared animals, birds and fish is possible provided lysine, methionine and digestible energy levels are kept constant. This makes ASL economically competitive in many, but not all, circumstances.

Preparation of Lysine-Coated Magnetic Fe2O3 Nanoparticles and Influence on Viability of A549 Lung Cancer Cells

  • Objective: To explore the effect of lysine-coated oxide magnetic nanoparticles (Lys@MNPs) on viability and apoptosis of A549 lung cancer cells. Methods: Transmission electron microscopy (TEM), vibrating sample magnetometer (VSM) and Zeta potentiometric analyzer were employed to characterize Lys@MNPs. Then Lys@MNPs and lung cancer A549 cells were co-cultured to study the effect of Lys@MNPs on cell viability and apoptosis. The pathway of Lys@MNPs entering A549 cells was detected by TEM and cell imaging by 1.5 T MRI. Results: Lys@MNPs were 10.2 nm in grain diameter, characterized by small size, positive charge, and superparamagnetism. Under low-dose concentration of Lys@MNPs (< $40{\mu}g/mL$), the survival rate of A549 cells was decreased but remained higher than 95% while under high-dose concentration ($100{\mu}g/mL$), the survival ratewas still higher than 80%, which suggested Lys@MNPs had limited influence on the viability of A549 cells, with good biocompatibility and and no induction of apoptosis. Moreover, high affinity for cytomembranes, was demonstrated presenting good imaging effects. Conclusion: Lys@MNPs can be regarded as a good MRI negative contrast agents, with promising prospects in biomedicine.

Enzymatic synthesis of sugar esters of L-lysine and L-aspartic acid

  • The enzymatic synthesis of conjugates of lysine and aspartic acid with D-glucose was studied. Optimase M-440 showed the very poor regioselectivity in the transesterification of $N{\alpha}$,$N'{\varepsilon}-di-t-Boc-L-Lys-OTFE$ and N-t-Boc-L-Asp-diOTFE with D-glucose. The acylation of glycosidic -OH and primary -OH of D-glucose occurred. However, Optimase M-440 catalyzed only the acylation of primary -OH group in the transesterification of $N{\alpha}$,$N'{\varepsilon}-di-t-Boc-L-Lys-OTFE$ and N-t-Boc-L-Asp-diOTFE with ${\alpha}-$ and ${\beta}-methylglucopyranoside$ in high yields without any other transesterification. Optimase M-440 also discriminated carboxyl groups of N-t-Boc-L-Asp-diOTFE.

바지락젓의 유리(遊離)아미노산(酸) (Changes in the Free Amino Acids During the Fermentation of Clam, Tapes japonica)

  • Clam, Tapes japonica, has been widely used and occupies an important position in the markets. More than thirty kinds of fermented fish and shellfish are presently available in the markets in korea. This study was attempted to establish the basic data for evaluating the taste compounds of the clam. The free amino acids as taste compounds during the fermentation of the clam, Tapes japonica, were analyzed by an amino acid autoanalyzer. Alanine (17.0%), glycine (15.5%), taurine (13.9%) were high in amount in the fresh clam and then glutamic acid, arginine, tyrosine, asparagine, lysine, threonine, serine and leucine were the next in order. Such amino acids as methionine, phenylalanine, histidine and cystine were low in amounts. The total free amino acid nitrogen in the fresh clam was 45.5% of its extract nitrogen. During the fermentation of the clam, the kinds of the free amino acids were the same with the fresh samples although the amounts were changed. It is believed that glutamic acid, aspartic acid, glycine, alanine, lysine, and leucine may play an important role as the taste compounds in the fermented clam.

Immobilization of L-Lysine on Zeolite 4A as an Organic-Inorganic Composite Basic Catalyst for Synthesis of α,β-Unsaturated Carbonyl Compounds under Mild Conditions

  • Lysine (Lys) immobilized on zeolite 4A was prepared by a simple adsorption method. The physical and chemical properties of Lys/zeolite 4A were investigated by X-ray diffraction (XRD), FT-IR, Brunauer-Emmett-Teller (BET), scanning electron microscopy (SEM), transmission electron microscopy (TEM) and UV-vis. The obtained organic-inorganic composite was effectively employed as a heterogeneous basic catalyst for synthesis of ${\alpha},{\beta}$-unsaturated carbonyl compounds. No by-product formation, high yields, short reaction times, mild reaction conditions, operational simplicity with reusability of the catalyst are the salient features of the present catalyst.

  • Pnicillinase를 분필하는 Streptomyces bobili YS-40균주의 plasmid DNA를 phemol방법으로 유출하여 Bocillus subtilis IAM 12118균주에 형질 전환시켜서 penicillin 내성 plasmid DNA가 표현되게 하였다. 형질전환에 미치는 최적 pH와 온도는 각각 7.0, 3$0^{\circ}C$으로 나타났다. DNA와 Competent cell을 20분 이상 접촉시킴으로서 형질전환이 잘 일어났으며 DNA의 농도가 증가할수록 transformant의 수도 증가했다. DNA inhibitor 로 사용되는 lysine 의 첨가로 형질전환의 빈도가 약 6배정도 상승되었으며 chloramphenicol은 형질전환의 빈도에 별 영향을 미치지 않았다.
