Optimization of Betacyanin Production by Red Beet (Beta vulgaris L.) Hairy Root Cultures. (Red Beet의 모상근 배양을 이용한 천연색소인 Betacyanin 생산의 최적화)

  • Optimal conditions for the production of natural color, betacyanin were investigated by varying light intensity, C/N ratio, concentrations of phosphate and kinds of elicitors. Batch cultivation was employed to characterize cell growth and betacyanin production of 32 days. The maximum specific growth rate, ${\mu}$$\sub$max/, was 0.3 (1/day) for batch cultivation. The maximum specific production rate, q$\^$max/$\sub$p/, was enhanced 0.11 (mg/g-cell/day) at 3 klux. A light intensity of 3 klux was shown to the best for both cell growth and betacyanin production. The maximum specific production rate was 0.125 (mg/g-cell/day) at 0.242 (1/day), the maximum specific growth rate. The dependence of specific growth rate on the light lintensity is fit to the photoinhibition model. The correlation between ${\mu}$ and q$\sub$p/ showed that the product formation parameters, ${\alpha}$ and ${\beta}$$\sub$p/ were 0.3756 (mg/cell) and 0.001 (mg/g-cell/day), respectively. The betacyanin production was partially cell growth related process, which is different from the production of a typical product in plant cell cultures. In C/N ratio experiment, high carbon concentration, 42.1 (w/w) improved cell growth rate while lower concentration, 31.6 (w/w) increased the betacyanin production rate. The ${\mu}$$\sub$max/ and q$\^$max/$\sub$p/ were 0.26 (1/day) and 0.075 (mg/g-cell/day), respectively. Beta vulgaris L. cells under 1.25 mM phosphate concentration produced 10.15 mg/L betacyanin with 13.46 (g-dry wt./L) of maximum cell density. The production of betacyanin was elongated by adding 0.1 ${\mu}$M of kinetin. This also increased the cell growth. Optimum culture conditions of light intensity, C/N, phosphate concentration were obtained as 5.5 klux, 27 (w/w), 1.25 mM, respectively by the response surface methodology. The maximum cell density, X$\sub$max/, and maximum production, P$\sub$max/, in optimized conditions were 16 (g-dry wt./L), 12.5 (mg/L) which were higher than 8 (g-dry wt./L), 4.48 (mg/L) in normal conditions. The ${\mu}$$\sub$max/ and q$\^$max/$\sub$p/ were 0.376 (1/day) and 0.134 (mg/g-cell/day) at the optimal condition. The overall results may be useful in scaling up hairy root cell culture system for commercial production of betacyanin.

Yield characteristics and medium composition of winter mushroom in cultivation farms in Korea (국내 팽이버섯 재배농가의 배지조성 및 수량 특성)

  • This study was conducted to provide basic data for setting a standard medium for winter mushroom cultivation. Investigation of medium composition in winter mushroom farms in Korea revealed that the types of medium used for each farm were slightly different and that the mixing ratio of the medium also varied. All farmers used corncob, rice bran, and beet pulp as the main media, and calcium carbonate and ground oyster shells were used at 1.0~3.8% to adjust the pH of the medium. Analysis of the physicochemical properties of the mixed media showed nitrogen content of 1.28~1.52%, carbon content of 45.7~48.5%, and C/N ratio of 30.7~37.9. The content of inorganic components was significantly different in each farm, depending on the type and amount of minerals in the mixed medium. Mycelial growth was the fastest at Farm 2, reaching 6.5 cm within 34 days of culture, followed by Farm 4 at 5.7 cm, whereas Farm 1, 3, and 5 showed similar growth. Mycelial density was high without any significant difference between farms. Survey of yield according to medium composition in each farm showed the highest yield in Farm 2 at 173.4 g/bottle, followed by Farm 1 at 168.4 g/bottle, whereas Farm 3 and 4 showed similar yield. However, Farm 4 had the lowest yield at 145.4 g/bottle. Therefore, mushroom yield was different depending on the medium composition in the farm, and thus selection of a medium that can be used universally in multiple mushroom farms was considered possible.

Effect of Milk Vetch Utilization Rice Cultivation to Reduce Application Amount of Nitrogen at Plowing Time in Paddy Field (자운영 후작(後作) 벼 재배시(栽培時) 경운시기별(耕耘時期別) 질소시비량(窒素施肥量) 절감효과(節減效果))

  • This study was carried out to find out the effect of Milk vetch(Astragalus sinicus L.) on growth, and yield of rice, physicochemical properties of soil, reduction rate of nitrogen fertilization, and soil improvement under the different plowing time with Milk vetch cultivated in paddy field, plowing at maximum blooming, last blooming, fruiting stages. The fresh weight of Milk vetch at each plowing time of maximum blooming, last blooming and fruiting stage was 22,500, 20,000, $12,500kg\;ha^{-1}$ respectively. Content of total nitrogen at three plowing times was 2.95, 2.66, and 2.47% and the C/N ratio were 15.7, 18.0, and 19.2, respectively. Physico-chemical properties of soil were improved in cultivated milk vetch, the content of T-N. OM and porosity ratio were increased while the content of $P_2O_5$ and bulk density, solidphase ratio were decreased compared to noncultivated milk vetch. Content of $NH_4-N$ in soil was highest plowing at maximum blooming stage and appeared an increasing tendency according to increased nitrogen level. Amount of nitrogen fertilizer by rice was highest plowing at maximum blooming stage and appeared an increasing tendency according to increased nitrogen level. Nitrogen-use efficiency was high in $33kg\;ha^{-1}$ nitrogen level at three plowing times. The number of spikelets per $m^2$ was high in plowing at maximum blooming stage, last blooming stage and the percentage of ripeness was high in fruiting stage of milk vetch. So the rice yield was increased 9%, 8% in $55kg\;ha^{-1}$ nitrogen level plowing at maximum blooming stage, last blooming stage and 1% in $77kg\;ha^{-1}$ nitrogen level plowing at fruiting stage compared to conventional cultivation.

  • The potential of using GIS in analyzing pest surveillance data was explored. The Spatial Analysis System (SPANS) was used to construct a spatial data base to study pest distributions using pest surveillance data collected from 152 stations in South Korea. The annual spatIal distributions of the striped rice borer(SRB), Chdo suppressalis, showed that high densities started to expand in the early 1980s, reaching a peak in 1988. The pattern change appears to be related to cultivation of japonica and indica-japonica hybrid varieties in South Korea. Japo7l!ca varieties have longer duration resulting in the SRB haVlng more time to mature and hibernate in wmter. The locus of SRB spread appears to be located in the mid-west region near lri, Chun~ Buk Province. High brown planthopper (BPH) populations in South Korea are often related to the early immigration and temperature. The simulated distribution of PPH densities in September using these two factors was compared with the actual distribution obtained using 1990 data. The two density maps corresponded closely excepL for differences in the south eastern valley. By overlaying the simulated map layer with the elevation and rice area maps, more specific BPH risk zones could be identified.

Effects of Planting Dates on Growth and Yield of Soybean Cultivated in Drained-Paddy Field

  • This study was carried out to determine adequate planting date, to compare the growth characteristics between early and late maturing cultivars, and to provide the data for the cultivation techniques of soybean [Glycine max (L.) Merr.] in double cropping system with winter crops on paddy field in Korea. Cultivars were planted on 26 May, 16 June, and 7 July with a planting density of $70cm(row\;widtb)\;{\times}\;10cm$ (planting spacing). Seed yield of soybean planted on June 16 and July 7 was approximately $37\%\;and\;53\%$, respectively, less than that of conventional planting date of May 26 in Pungsan-namulkong, and planted on June 16 and July 7 was about $30\%\;and\;37\%$, respectively, less then that of conventional planting date of May 26 in Hanamkong. The number of pods and seeds per plant decreased as planting date delayed. Seed weight increased in Pungsan-namulkong but decreased in Hannamkong as planting date delayed. The flowering date was late in delayed planting plots, but it was shorted for days from emergence to flowering and from emergence to maturity. The plant height of Hannamkong was greater than Pungsan-namulkong from the emergence to flowering stages, but in contrast, it was greater in Pungsan-namulkong than Hannamkong after flowering stage (50d after emergence) when it planted on May 26. There were no significant differences between two soybean cultivars at planting dates of June 16 and July 7. Leaf number, leaf area, and dry matter were also reduced by late planting, and Both of them were shown in high reduction at the later planting. There was a high significant difference at the flowering $(r\;=\;0.87^{**})$ and pod formation $(r\;=\;0.91^{**})$ stages between leaf dry matter and seed yield. Crop growth rate (CGR) was greater at $R2\~R3$ growth stages compared to $R3\~R4\;or\;R4\~R5$ growth stages in two soybean cultivars and the greatest CGR was obtained at planting date of May 26 in two soybean cultivars except for R4-R5 growth stage in Pungsan-namulkong. There was a highly significant positive difference between the seed yield and the leaf area index (LAI) across R3 to R4 and R2 to R3 stages. The photosynthetic rate $(P_N)$ of the uppermost leaf position had no significant difference among planting dates and between two soybean cultivars. However, $P_N$ of the $7^{th}$ leaf position increased as the planting date delayed.

Effects of Decreasing Methods of Salt Content in Root Zone on Soil Properties and Crop Grwoth at the Newly Reclaimed Tidal Soil (신간척지(新干拓地)에서 근권(根圈)의 염농도(鹽濃度) 저하(低下) 방법(方法)이 토양특성(土壤特性)과 작물생육(作物生育)에 미치는 영향(影響))

  • This experiment was conducted to find out the useful data for upland crop cultivation in the newly reclaimed tidal land. Poseung silty clay loam soil was selected, and cotton(Mogpo VII) and tall fescue were cultivated under different drainage systems and soil ameliorator applications. Soil hardness and bulk density were decreased by subsurface drainage and plastic film installed at 40cm depth of the soil. Red earth application was also effective to loosen the soil, but zeolite and gypsum made the subsoil compact. Water content of the soil was high in surface drain than that of subsurface drain or plastic film curtain plot during dry season. The water content was in order of plastic film curtain, surface drain and subsurface drain. Electrical conductivity(EC) was decreased to lower than 0.4 Simens $meter^{-1}$ ($SM^{-1}$) in the subsurface drain during rainy season, and the EC of subsurface drain was maintained a quater to an half of surface drain. The yield of cotton and tall fescue were high in order of subsurface drain, plastic film curtain and surface drain plot. The yields of cotton were increased to 36-73 % by ameliorator application, and the red earth application was more effective for tall fescue growth compare to gypsum and zeolite.

Evaluating the Characteristics of Growth and Seedling Quality of Tetradium daniellii (Benn.) T. G. Hartley using Five Different Container Types (용기 종류에 따른 쉬나무 용기묘의 생장 및 묘목품질 특성)

  • There is an increasing demand for Tetradium daniellii seedlings due to their uses as alternative energy, for ecological restoration, and as a honey plant. This study was conducted to determine the optimum container for superior seedling production of T. daniellii. Experiments were performed using five plastic container types (500, 350, 320, 300, and 250 ml) for forestry facility cultivation. The height and root collar diameter growth of T. daniellii seedlings were significantly high in the 350-ml container. High growth appeared primarily in the container with a larger cavity volume and lower growing density. Root development was most active in full sunlight. The maximum dry matter production was observed in the 350-ml container, which was similar to the results of height and root collar diameter growth. QI, an index showing the quality of a seedling, was maximum at 0.97 in the 350-ml container. In conclusion, the 350-ml container is optimum for superior seedling production of T. daniellii.

  • In the present study, the vegetation was classified using the phytosociological method and canonical-correlation analysis (CCA) was implemented to analyze correlation between community structure and environmental factors in the natural habitats of forest byproducts, especially medicinal plants, such as Cudrania tricuspidata, Sorbus commixta, and Hovenia dulcis, in 2021-2022 to provide primary ecological data to establish environmental conditions for wild vegetable cultivation. A total of 11 plots in five regions, 8 plots in three regions, and 17 plots in 5 regions were selected for the natural habitats of C. tricuspidata in southern Korea, S. commixta in high mountains, and H. dulcis in valleys of central Korea, respectively. The importance value in each community was respectively analyzed as follows, in C. tricuspidata community, the importance value of C. tricuspidata (61.10) was the highest, followed by Celtis sinensis, Pinus thunbergii, Neolitsea aciculata, Styrax japonica, Carpinus coreana, Quercus serrata, and Q. acutissima. In Sorbus commixta community, Q. mongolica (57.21) was the highest, followed by, S. commixta (42.58), Betula ermani, Tilia amurensis, A. pseudosieboldianum, A. tschonoskii var. rubripes, Cornus controversa, Magnolia sieboldii, and Taxus cuspidata. In H. dulcis community, H. dulcis (64.58) was the highest, followed by Zelkova serrata, Cornus controversa, A. mono, Q. serrata, C. cordata, and Juglans mandshurica. As the result of the analysis on DBH of the major species having the high importance value, in C. tricuspidata community, C. tricuspidata, C. sinensis, Neolitsea aciculata, and C. coreana show the density of normal distribution, so the dominant status of these species is likely to continue. In S. commixta community, S. commixta show the density of reverse J-shaped curve, so the dominant status of these species is likely to be stable, and Q. mongolica, B. ermani and T. amurensis, show the density of normal distribution, so the dominant status of these species is likely to continue. In H. dulcis community, C. cordata, and J. mandshurica show the density of reverse J-shaped curve, so the dominant status of these species is likely to be stable, and H. dulcis, Z. serrata, C. controversa and A. mono had a formality distribution, suggesting a continuous domination of these species over the other species for the time being. The results of CCA ordination analysis using 11 environmental factors and 30 communities of three taxa classified by TWINSPAN analysis revealed that the altitude showed the strongest correlation with the vegetation. C. tricuspidata community was distributed on the moderate and gentle northeastern slope at low altitude with the highest pH, C.E.C, Ca2+, and Mg2 and various P2O5, whereas S. commixta community was distributed on the steep slope at high altitude with the highest O.M and T-N and lower P2O5, Ca2+, Mg2+, C.E.C and pH, which is the opposite tendency of the environment of C. tricuspidata community. H. dulcis community was distributed on the gentle northern slope at lower altitude with an average pH, O.M, T-N, Ca2+, Mg2+, and C.E.C, except higher P2O5.

Planting Evaluations for the Landscaping Tree and Application Plan by Assessment Grade in the City Park - A Case Study of Haedoji Park, Songdo, Incheon Metropolitan City - (도시공원 조경수목 식재 평가 및 평가등급 적용 방안 - 인천광역시 송도 해돋이공원을 대상으로 -)

  • This study aims to present an evaluation scheme to improve the problems in tree shapes and tree growth which were identified through shape and growth assessments of landscape trees currently planted in Haedoji Park, Songdo, Incheon Metropolitan City after plant structure status and propriety review for funtion of space and concept of planting. Suitability the planting concept was evaluated according to function of space. The result indicated that the shade planting areas accounted for 29.5% of the the shade spaces area. 58.7%, respectively planting areas of visual landscape. And 11.8%, respectively planting areas of buffer. Because the planting areas was lacked according to the park established spatial configuration of central facilities and the result of plant young trees. Plant structure status required consideration with plant structure, density, size, growth status for improve planting function. The tree assessment was performed on a total of 28 species and 600 trees of which 22 species and 209 trees were planted in the buffer zone, 8 species and 71 trees in the shade zone, 16 species and 266 trees in the visual landscape zone, and 4 species and 54 trees in the ecological landscape zone. The trees were divided into grades based on their assessment score and were statistically grouped by the functional zone in where they are planted and by tree species to verify their significance. The tree shape assessment was an average of 56.6 points and the tree growth assessment was an average of 76.0 points. Using the results of the tree assessments, the tree standards for each functional space were identified and a concept of optimum planting and cultivating was applied. When applying the shape assessment results by zone to the concept of planting, since trees for buffering require high functionality they received E's, the lowest grade; as trees for ecological landscaping require diverseness and naturalness, they received D's; since trees for shading require utilization, they received C's as trees with branching at the main stems were considered; and since trees for visual landscaping required aesthetical value, they received A's and B's. When applying the growth assessment results by zone to the concept of planting, based on planting foundations of favorable and poor, for buffering, visual landscaping, and ecological landscaping, trees from grades A to E could be planted, and for shading, trees from grades A to C could be planted. For a cultivation plan that could improve the growth of the trees, we proposed that the topography of the land be selected considering the tree's characteristics and that a method of pot seeding be used. Also, to improve the shape of the trees, we proposed that poles be used to improve the growth of vertically-straight stems, an appropriate planting density be applied for reasonable branch growth, manage tree shape to maintain good crowning, and better manage fertilization to maintain a reasonable crown density.

Comparative Evaluation of Muddy Water Occurrence Possibility in Dam Reservoir Using GIS (GIS를 이용한 댐 저수지의 흙탕물 발생 가능성 비교 평가)

  • The muddy water occurrence possibility of reservoir were analyzed by considering GIS based soil erosion model, sediment delivery ratio and effective reservoir capacity. For the purpose, the weakness factors for the establishment of countermeasures of basin were analyzed by evaluating input factors of RUSLE model based on spatial data such as DEM, soil map, landcover map and so on. The potential of soil erosion was estimated considering highland upland. The sediment yields of Chungju-Dam and Soyanggang-Dam showed the highest result in sediment yield using sediment delivery ratio with considering basin area. The sediment concentration of Imha-Dam and Chungju-Dam showed the highest value as 0.791 $kg/m^3/yr$ and 0.526 $kg/m^3/yr$ respectively in sediment concentration with considering effective reservoir capacity. Especially, sediment yield of Imha-Dam was about 2.36 times lower than Soyanggang-Dam, but the sediment concentration was 1.90 times higher preferably, because the effective reservoir capacity of Imha-Dam was about 4.48 times lower. This study calculated sediment concentration using the 10 years mean rainfall event and could consider the aspects of soil, terrain, landcover, cultivation condition and effective reservoir capacity of each basin effectively through the results. Therefore, these quantitative sediment concentration data could be used to estimate the potential of high density turbid water for reservoir and applied with effective tools for the management of reservoir.
