Effect of Warming Temperature on the Viability of Bovine IVP Blastocyst Cryopreserved by Glass Micropipette (GMP) Vitrification

  • The purpose of this study was to investigate the warming temperature and exposed time on the post-thaw survival rate and viability of bovine blastocyst cryopreserved by GMP vitrification. Groups of three bovine IVP blastocysts were sequentially placed into vitrification solution before being loaded into the GMP straws and immersed into LN$_2$within 20 to 25 sec. The warming rate was increased 2 times of warming temperature for improvement of post-thaw survival rates. The frozen embryos were warmed either at 35 or 70$^{\circ}C$ for 1 or 2 sec and then diluted in sucrose solution. Post-thaw blastocysts were serially washed in 0.25 and 0.15 M sucrose in holding medium (HM: TCM199 supplemented with 10% FCS) and TCM-199 for each 5 min, respectively, and then cultured in TCM199 for 24 h. The rate of re-expanded blastocyst was significantly different fer 35 and 70$^{\circ}C$ warming temporature (76.4 vs. 89.3%; P<0.05). The rate of re-expanded blastocyst at 70$^{\circ}C$ for 1 sec was significantly higher than that for 2 sec (91.1 vs. 70.9%; P<0.05). The number of nuclei counted were significantly different among control, 35 and 70$^{\circ}C$ (121${\pm}$8.5 vs. 104${\pm}$11.7 vs. 114${\pm}$10.3; P<0.05). These results indicated that the increasing of warming rate can provide high survival rates of bovine IVP blastocysts. Especially, the best viability of post-thaw blastocyst could be thaw at 70$^{\circ}C$ for 1 sec. The warming temperature and exposed time far warming was considered to be limiting factors to the viability of bovine IVP embryos. he purpose of this study was to investigate the warming temperature and expose.

  • Background: This study has mainly focused on finding pharmacological effects of ginsenosides that can reduce the unwanted side effects of the cytotoxic anticancer drugs and are highly effective on prostate cancer, colorectal cancer, liver cancer, hormone-dependent breast cancer, triple-negative breast cancer, and brain cancer (neuroblastoma). Methods: Minor and rare ginsenosides (GS) of Rh2 which have a high absorption ability and excellent pharmacological actions were treated with the 6 different types of cancer cell lines and their anticancer activities were investigated by analyzing gene expressions associated with various cancers through qPCR and other relevant methods. Results: In cancer cells exposed to Rh2, cell viability and cell migration were reduced, and apoptosis was induced. Each cancer cell was divided into three groups according to the cell proliferation response by Rh2; 1) A group in which the cell viability decreases inversely to an increase in Rh2 treatment concentration; 2) A group in which the cell viability rapidly decreases in Rh2 treatment above a certain level of concentration; 3) A group in which the cell viability was not suppressed below 20-30% even with 100 μL of Rh2, the highest concentration used in this study. Conclusions: It was shown that Rh2 has a significant effect on inhibiting the proliferation of prostate cancer cells and hormone-dependent breast cancer cells.

Differential Wnt11 Expression Related to Wnt5a in High- and Low-grade Serous Ovarian Cancer: Implications for Migration, Adhesion and Survival

  • Wnt is a powerful signaling pathway that plays a crucial role in cell fate determination, survival, proliferation and motility during development, in adult tissues and cancer. The aims of the present study were three fold: i) to assess Wnt11 immunoexpression and its possible relationship with Wnt5a in high- and low-grade human serous ovarian cancer (HGSC and LGSC) specimens; ii) to assess Wnt11 expression levels in Wnt5a overexpressing SKOV-3 cells; iii) to reveal the role of Wnt11 in viability, adhesion, migration and invasion of SKOV-3 cells using recombinant human Wnt11 (rhWnt11). Immunohistochemistry revealed a significant difference in Wnt11 expression between HGSC and LGSC groups (p=0.001). Moreover, a positive correlation was observed between Wnt5a and Wnt11 expression in the HGSC (r=0.713, p=0.001), but not the LGSC group. The expression of Wnt11 was decreased by 35% in Wnt5a overexpressing cells (SKOV-3/Wnt5a) compared to mock controls. Similarly Wnt11 expression levels were decreased by 47% in the presence of exogenous Wnt5a compared to untreated cells. In the presence of rhWnt11, 31% increased cell viability (p<0.001) and 21% increased cell adhesion to matrigel (p<0.01) were observed compared to control. Cell migration was increased by 1.6-fold with rhWnt11 as revealed by transwell migration assay (p<0.001). However, 45% decreased cell invasion was observed in the presence of rhWnt11 compared to control (p<0.01). Our results may suggest that differential Wnt11 immunoexpression in HGSC compared to LGSC could play important roles in serous ovarian cancer progression and may be modulated by Wnt5a expression levels.

Increased Stability of Bacillus polyfermenticus SCD in Low pH, High Temperature and High Glucose Concentration via Three Layer Coating (Bacillus polyfermenticus SCD의 Three Layer Coating에 의한 pH, 열, 높은 glucose 농도에 대한 안정성효과)

  • Bacillus polyfermenticus SCD derived from Bacillus sp., which is commonly called as Bisroot$^{ⓡ}$. The goal of this study, is to Increase stability of Bacillus polyfermenticus SCD in low pH, high temperature and high glucose concentration via three layer coating. The viability of coated Bacillus polyfermenticus SCD increased to 30%, 20%, 14% in the condition of pH 2 4 6 than that of uncoated Bacillus polyfermenticus SCD at 37$^{\circ}C$ for 4 h. Final viability of the coated Bacillus polyfermenticus SCD in 80$^{\circ}C$ increased to 40% than that of uncoated Bacillus polyfermenticus SCD. In high glucose concentration coated Bacillus polyfermenticus SCD is more stable than uncoated about 50%. In conclusion, the three layer coated Bacillus polyfermenticus SCD is very stable for low pH, high temperature and high glucose concentration.

Protective Effects of Chungkookjang Extract on High Glucose Induced Oxidative Stress in LLC-PK1 Cells

  • This study was designed to investigate the protective effect of a methanol extract of Chungkookjang (CKJ) on high glucose induced oxidative stress in LLC-$PK_1$ cells (renal tubular epithelial cells), which are susceptible to oxidative stress. Freeze dried CKJ powder was extracted with methanol, and the extract solution was concentrated, and then used in this study. To determine the protective effect of CKJ extract, oxidative stress was induced by exposing of LLC-$PK_1$ cells to high glucose (30 mM) or normal glucose (5 mM) for 24 hr. Exposure of LLC-$PK_1$ cells to high glucose for 24 hr resulted in a significant (p<0.05) decrease in cell viability, catalase, SOD and GSH-px activity and a significant (p<0.05) increase in intracellular ROS level and thiobarbituric acid reactive substances (TBARS) formation in comparison to the cells treated with 5 mM glucose. CKJ extract treatment decreased intracellular ROS level and TBARS formation, and increased cell viability and activities of antioxidant enzymes including catalase, SOD and GSH-px in high glucose pretreated LLC-$PK_1$ cells. These results suggest that CKJ extract may be able to protect LLC-$PK_1$ cells from high glucose-induced oxidative stress, partially through the antioxidative defense systems.

Inhibitory Effects of (-) Epigallocatechin Gallate and Quercetin on High Glucose-induced Endothelial Cytotoxicity

  • Functional damage to microvascular endothelial cells by hyperglycemia is thought to be one of the critical risk factor.; in the impaired wound healing seen with diabetes mellitus. It is also thought that oxidative stress plays a significant role in this endothelial cell dysfunction. The present study examined the differential effects of flavonoids on endothelial cell dysfunction under high glucose conditions. Human endothelial cells exposed to 30 mmol/L glucose for 7 d were pre-treated with various flavonoids and pulse-treated with 0.2 mmol/L $H_2O_2$ for 30 min. High glucose markedly decreased cell viability with elevated oxidant generation and nuclear condensation. $H_2O_2$ insult exacerbated endothelial cytotoxicity due to chronic exposure to high glucose. (-)Epigallocatechin gallate and quercetin improved glucose-induced cell damage with the disappearnnce of apoptotic bodies, whereas apigenin intensified the glucose cytotoxicity. In addition, cell viability data revealed that these flavonoids of (-)epigallocatechin gallate and quercetin substantially attenuated both high glucose- and $H_2O_2$- induced dual endothelial damage. These results suggest that (-)epigallocatechin gallate and quercetin may be beneficial agents for improving endothelial cell dysfunction induced by high glucose and may prevent or reduce the development of diabetic vascular complications.

A Novel Behavior, Bang-Sensitive Paralysis, Associated With The shibire Locus of Drosophila melanogaster

  • The Drosophila temperature-sensitive mutant shibire (shi) is paralyzed at restrictive temperature by a reversible block in synaptic transmission. To explore the functional relationship among shi gene products, viability and temperature-sensitive paralytic behavior were quantitaively analyzed for four shi alleles, shi$^{ts1}$, shi$^{ts2}$, shi$^{ts4}$, and shi$^{ST139}$, and their heteroallelic combinations. The hemizygous combination of shi alleles over deficiency was not completely lethal. shi$^{ts2}$ exhibited distinctively higher viability than other alleles. A novel behavior, bang sensitivity, was also found in shi/Df(1). This bang-sensitive paralytic behavior was compared with that of the typical bang-sensitive mutant flies. Heterozygotes, shi/+, are more severe in temperature sensitivity than deficiency hemizygotes, Df(1)/+. Heteroallelic combinations of shi were less sensitive to high temperature than homozygotes. Among all allelic combinations, shi$^{ts2}$/shi$^{ts4}$ showed an unexpected extreme reduction in temperature sensitivity. The results of allelic interactions among 4 shi alleles suggest that the shi mutations examined behave as antimorphic alleles and that the gene product of shi are likely to function in multimeric forms.

Anti-oxidative Activity and Trace Component of a Sprout Serum (새싹 추출물의 항산화 작용과 미량 성분)

  • In this study, the anti-oxidative effects and components of sprout serum were investigated. In the buds, high levels of zinc, iron and manganese were analyzed in addition to copper, magnesium, and potassium. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity of sprout serum was evaluated with $IC_{50}$. $IC_{50}$ of sprout serum was $58.0\;mgmL^{-1}$, and that of vitamin C was $3.7\;mgmL^{-1}$. In the test of superoxide scavenging activity of sprout serum, the activity was dependent on the concentration of serum. In this case, the $IC_{50}$ was 2.0 wt%. Cell viability was detected by using the MTT method. Cultured human fibroblast was treated with 15 mM $H_2O_2$ and cell viability was 70% in case of control. However, the effect of treating 0.5% of sprout serum was similar to that of 0.0001% of vitamin C.

Viability of Probiotic Bacteria in Yogurt Supplemented with Enzyme-Bioconverted Ginseng, Ascorbic Acid, and Yeast Extract (효소처리인삼, 아스코르브산, 효모추출물이 첨가된 요구르트에서 프로바이오틱 세균의 활성)

  • The effects of yogurt supplementation with enzyme-bioconverted ginseng (EBG), ascorbic acid, and yeast extract on the bacterial counts of Streptococcus thermophilus, Lactobacillus acidophilus LA-5, and Bifidobacterium BB-12 were investigated to develop healthy yogurts with high probiotic counts during storage. In addition, the colors and viscosities of the yogurts were determined. EBG, ascorbic acid, and yeast extract did not affect S. thermophilus counts. EBG and ascorbic acid enhanced the viabilities of L. acidophilus LA-5 and Bifidobacterium BB-12 during storage. Yeast extract improved growth of L. acidophilus LA-5 and Bifidobacterium BB-12 during fermentation. EBG turned the yogurt into brown color. We conclude that supplementation of yogurt with EBG, ascorbic acid, and yeast extract may enhance its health-promoting functions by increasing the viability of probiotics, which can thus promote consumption of the yogurt.

The Effects of Increased Temperature on Yield Properties, Antioxidant Contents, and Pollen Viability of Adzuki Bean (Vigna angularis L.) Responses in Temperature Gradient Greenhouse and Growth Periods (온도구배온실에서 온도상승이 생육시기별 팥의 수량, 항산화 성분, 화분 임성에 미치는 영향)

  • The quality and yield of crops produced using field cultivation are expected to decrease due to the recent global climate change caused by extreme weather. The plant reproductive stage associated with crop yields is a highly vulnerable period to global warming caused by high temperatures. This study analyzed the adzuki bean's yield properties, antioxidant contents, and pollen viability of adzuki bean (Vigna angularis L.) under high-temperature stress and growth periods in a temperature gradient greenhouse that forms 0 to 4℃ above the outside temperatures. As the main variety of red beans cultivated in Korean farms, the "Arari" red bean was grown in the rain shield greenhouse and the temperature gradient greenhouse from 2021 to 2022 in Milyang, Korea. Compared to 2022, it showed a 0 - 1.0℃ lower temperature during the whole growth period in 2021. However, its average temperatures were 0 - 3.7℃ higher in the vegetative stage and 0.4 - 2.4℃ higher in the anthesis stage in 2021. The lowest yield (6.8 ± 0.7 g) was at the highest temperature (T4: low, 23.6℃; average, 28.5℃; high, 35.8℃) during the anthesis stage in 2021. The temperatures of the mature stage were 1.7 - 3.9℃, which were higher in 2022 than in 2021, although the low temperatures of 2022 were lower than in 2021. The yields of the mature stage in 2022 increased more than in 2021 because of the high temperature of the mature stage. The growth and yield were good at 40.5℃ in the vegetative stage. However, growth was poor when the average temperature was 27.0℃ or higher, and yields decreased during the flowering period. Total polyphenol and flavonoid contents were increased, and the pollen viability was 40.75% in the whole growth period at high temperature (T4: low, 22.9℃; average, 28.8℃; high, 36.9℃). These results showed that the antioxidant levels increased when the antioxidant component was affected at higher temperatures than at normal. In contrast, the pollen viability-related yield decreased as the temperature increased. Our results are the basic data for field growers and the breeding of thermos-tolerance in adzuki beans to prepare for the changeable future climate.
