Development of Marine Virus-like Particles Live/Dead Determination Method for the Performance Evaluation of Ballast Water Treatment System (선박평형수처리장치 성능 평가를 위한 해양 바이러스 생사판별 방법 개발)

  • To prepare more stringent regulations for USCG Phase II ballast water management, this study investigated the staining efficiency of SYBR Green I(SGI) and SYBR Gold(SG) on the virus-like particle (VLP). A dye with high staining efficiency was applied to the treated water that was passed through the ballast water treatment system (BWTS). VLP staining was observed most clearly under the 100-fold and 200-fold dilution of the stock solution when the volume of filtered samples was 0.5 mL to 2 mL. The staining efficiency of SGI and SG did not show a significant difference. On the other hand, the green fluorescence of viruses in the sample stained with SGI was more pronounced than in the samples stained with SG (expressed yellow fluorescence), making it easier to observe. The abundance of VLP in the test water and control water treatments that did not pass through the two types of BWTS (electrolysis type, UV + electrolysis type) was approximately 109 - 1010 VLP 100 mL-1. In contrast, no stained VLP was observed in the treated water treatments. Moreover, SGI was confirmed to be effectively stained under various salinity conditions, including seawater, brackish water, and freshwater. Further verification tests and development of staining methods under various BWTS are required, but the SGI staining method is believed to be a good alternative to the VLP live/dead determination of the USCG Phase II type approval test.

Seasonal changes in phytoplankton community related with environmental factors in the Busan coastal region in 2014 (2014년 부산 연안 해역에서 계절적 환경특성에 따른 식물플랑크톤 군집의 변화양상)

  • To assess the influence of environmental factors on the phytoplankton community structure and total phytoplankton biomass during four seasons in 2014, we investigated the abiotic and biotic factors at 25 stations in the Busan coastal region. The phytoplankton community and total phytoplankton biomass were strongly dependent on the discharge from the Nakdong River, and the high density of phytoplankton was related with the introduction of the Tsushima Warm Current (TWC), particularly in the thermohaline fronts of the fall season. The relationship between the salinity and nutrient (Dissolved inorganic nitrogen=DIN: R2=0.72, p<0.001 and Dissolved inorganic silicon=DSi: R2=0.78, p<0.001) highly correlated with the river discharge, implying that those nutrients have played a crucial role in the growth of diatom and cryptophyta. The total phytoplankton biomass was highest in the summer followed by autumn, spring, and winter. Diatom and cryptophyta species were dominant species during the four seasons. Additionally, there were strong positive correlations between Chlorophyll a and total phytoplankton biomass (R2=0.84, p<0.001), cryptophyta (R2=0.76, p<0.001) and diatom (R2=0.50, p<0.001), respectively. In particular, we found that there were significant differences in the nutrients, phytoplankton community compositions, and total phytoplankton biomass between the inner and the outer coastal region of Busan, depending on the amount of river discharge from the Nakdong River, particularly during rainy seasons. Therefore, the seasonal change of TWC and river discharge from the Nakdong River serve an important role in determining phytoplankton population dynamics in the Busan coastal region.

Assessment of Eutrophication Using Trophic State Index and Water Quality Characteristics of Saemangeum Lake (새만금호의 수질 특성 및 영양상태지수를 이용한 부영양화 평가)

  • We evaluated the eutrophication of Saemangeum Lake, which causes abnormal growth of algae, using the Carlson index. Eutrophication characteristics of Saemangeum Lake were analyzed. For the study, water quality surveys were conducted at 7 stations in Saemangeum Lake every month in 2021. The concentration of Chl.a was slightly higher in the Mankyeong water system in winter, and slightly higher in the Dongjin water system in spring and summer, but overall, except for some periods, the concentration was similar to or lower than the lake water quality environmental standard of class 3. COD showed water quality similar to or above the lake quality environmental standard of grade 4 in both the Mankyeong and Dongjin water systems in the summer and Autumn. TOC concentrations were within lake water quality standard 3 at all sites. Total phosphorus concentrations exceeded the lake water quality standard of Class 4 and were higher in January and August after rainfall. In the correlation analysis between water quality factors, the correlation of organic matter, total phosphorus, and total nitrogen to salinity was relatively high. This reflected the water quality characteristics of freshwater, brackish water, and seawater areas due to seawater inflow through the drainage gate and freshwater inflow through upstream rivers. According to the characteristics of eutrophication fluctuations in Saemangeum Lake by trophic state index, the indices of Chl.a, SD, and TN showed water quality in the early stage of eutrophication, while the TP index showed a severe eutrophication state. The magnitude of the eutrophication index among water quality components was TSI(TP) > TSI(TN) > TSI(SD) > TSI(CHL) in all water systems. Quadrant analysis of the deviation of TSI(CHL) from TSI(TP) and TSI(SD) on a two-dimensional plane showed that there was no limiting effect of total phosphorus on algal growth in all water systems. In addition, the factors af ecting light attenuation appeared to be dominated by small particulate matter from outside sources.

Variation of Water Qualities Due to Freshwater Introduction to Tidal Flat: A Mesocosm Study (메조코즘을 이용한 갯벌의 담수화과정 중 수질 변화)

  • A mesocosm, an artificial tidal flat ecosystem, was constructed outdoors to simulate in situ physical and biochemical environmental conditions of natural tidal flat as much as possible. During the experiment from February to August 2004, the study was focused on the biogeochemical variations of superficial water and porewater after introduction of freshwater into the mesocosm. The mesocosm has three experimental conditions; SW-M-T: maintaining the saline water of approximately 20 psu; FW-M-T: complete exchange of freshwater ul the mesocosm with continuous mixing of water column: FW-NM-T: complete exchange of saline water to freshwater in the mesocosm without mixing of water column. Mass extinction of benthic macrofauna appeared due to drastic decrease of porewater salinity from 20 psu to less than 10 psu between the 63th and 91st day of freshwater introduction in FW-M-T and FW-NM-T. Throughout the periods, 7/8 of bivalves and 2/3 of polychaete populations have been extinguished in the sediment. In FW-NM-T, as temperature rises, both evident decrease of DO in water column and active release of DIP from sediment were observed. ${NO_3}^-$ was removed from water column into sediment throughout the periods. Therefore extremely low ${NO_3}^-$ was found during late spring and summer. Whereas ${NH_4}^+$ exhibited only $1/2{\sim}1/8$ of ${NO_3}^-$ concentration. Unexpectedly even after mass extinction of benthic macrofauna, we were not able to find high ${NH_4}^+$. This mesocosm study suggests that when fresh water introduce to natural tidal flat, its sediment activity functions as a potential source of DIP, but a sink of ${NO_3}^-$.

Study on Forage Cropping System Adapted to Soil Characteristics in Reclaimed Tidal Land (간척지 토양특성에 알맞은 사료작물 작부체계 연구)

  • This study was conducted to find out the optimum cropping system for the stable production of forage crops in the newly reclaimed land located at Gwanghwal and Gyehwa region of Saemangum reclaimed tide land from October, 2009 to October, 2011. Whole crop barley (WCB), Rye, Italyan-ryegrass (IRG) as winter crops and Corn, Sorghum${\times}$sudangrass hybrid (SSH) as summer crops were cultivated. Soil chemical properties, nutrient uptake, feed value, growth and yield were examinated. The testing soil was showed saline alkali soil where the contents of organic matter, available phosphate and exchangeable calcium were very low, while exchangeable sodium and magnesium were higher. Changes of soil salinity during the growing season of forage crops were less than 0.2%, and the growth of forage crops was not affected by salt injury. Standing rates of winter crops were higher in the order of Rye, WCB, and IRG, while the dry matter yield of winter crops was higher in the order of IRG, Rye and WCB. The highest crude protein (CP) content was recorded in IRG, and total digestive nutrients (TDN) contents were increased in the order of WCB, IRG, and Rye. The TDN content was higher in corn, whereas other feed value was higher in SSH. The content of mineral nutrients on stem, leaf and grain in IRG, Corn were high. After experiment pH was lowed, contents of exchangeable magnesium, sodium and organic matter were decreased while contents of total nitrogen, available phosphate and exchangeable potassium, calcium were increased. Winer crops and summer crops after continually cultivating in cropping system, fresh matter yield increased, compared to WCB-Corn (74,740 kg $ha^{-1}$), IRG-SSH 10%, IRG-Corn 7%, Rye-SSH 6%, Rye-Corn and WCB-SSH 3%. Dry matter yield increased, compared to WCB-Corn (20,280 kg $ha^{-1}$), IRG-SSH 7%, Rye-SSH 6%, IRG-Corn/Rye-Corn/WCB-SSH 3%. The TDN yield increased, compared to WCB-Corn (13,830 kg $ha^{-1}$), IRG-SSH 2%, WCB-SSH and IRG-Corn 1%. Therefore, we suggest that the crop combination of IRG-SSH and WCB-SSH would be preferred for silage stable production.

Light and Electron Microscopy of Gill and Kidney on Adaptation of Tilapia(Oreochromis niloticus) in the Various Salinities (틸라피아의 해수순치시(海水馴致時) 아가미와 신장(腎臟)의 광학(光學) 및 전자현미경적(電子顯微鏡的) 관찰(觀察))

  • This study was taken to examine the light microscopic and ultrastructural changes of gill and kidney of female tilapia{Oreochromis niloticus) adapted in 0%o, 10%o, 20%o, and 30%o salt concentrations, respectively, by light, scanning and transmission electron microscope. The results obtained in these experiments were summarized as follows: Gill chloride cell hyperplasia, gill lamellar epithelial separation, kidney glomerular shrinkage, blood congestion in kidneys and deposition of hyalin droplets in kidney glomeruli, tubules were the histological alterations in Oreochromis niloticus. Incidence and severity of gill chloride cell hyperplasia rapidly increased together with increase of salinity, and the number of chloride cells in gill lamellae rapidly increased in response to high external NaCl concentrations. The ultrastructure by scanning electron microscope(SEM) indicated that the gill secondary lamella of tilapia(Oreochromis niloticus) exposed to seawater, were characterized by rough convoluted surfaces during the adaptation. Transmission electron microscopy(TEM) indicated that mitochondria in chloride cells exposed to seawater, were both large and elongate and contained well-developed cristae. TEM also showed the increased chloride cells exposed to seawater. The presence of two mitochondria-rich cell types is discussed with regard to their possible role in the hypoosmoregulatory changes which occur during seawater-adaptation. Most Oreochromis niloticus adapted in seawater had an occasional glomerulus completely filling Bowman's capsule in kidney, and glomerular shrinkage was occurred higher in kidney tissues of individuals living in 10%o, 20%o, 30%o of seawater than in those living in 0%o of freshwater, and blood congestion was occurred severer in kidney tissues of individuals living 20%o, 30%o of seawater than in those living in 10%o of seawater. There were decreases in the glomerular area and the nuclear area in the main segments of the nephron, and that the nuclear areas of the nephron cells in seawater-adapted tilapia were of smaller size than those from freshwater-adapted fish. Our findings demonstrated that Oreochromis niloticus tolerated moderately saline environment and the increased body weight living in 30%o was relatively higher than that living in 10%o in spite of histopathological changes.
