  • It is now generally accepted that individuals at increased risk for cardiovascular disease may be identified by certain traits or habbits. The factors such as high blood pressure, elevated blood cholestrol, age, sex and obesity are associated with increseaed frequency of disease. The blood cholesterol level lowering will decrease cardiovascular disease risk. The regression of atherosclerosis can be achieved by lowering the level of circulating cholesterol. Those things are connected with the quantity and quality of protein, fats, carbohydrates, especially soluble and non-soluble fiber, magnesium and calcium. The lipoprotein and lipid metabolism are connected with the lipid transport. The factors on lipid absorption and blood serum lipid pattern of human are exist. The factors have a variety of materials with different chemical and physical properties. The soluble fiber diet make a low blood and liver lipids. Many kind of soluble fiber results in a lowering of blood cholesterol and triglyceride levels. The cholesterol lowering effects of dietery fiber may be a results of alterations of in intestinal handling of fats, hepatic metabolism of fatty acid or triglyceride acid metabolism of lipoprotein. It is investigated that the high density lipoprotein (HDL) is inversely related to coronary artery disease. It has been postulated that HDL may be an important factor in cholesterol efflux from the tissues, therby reducing the amount of cholesterol deposited there. Alternatively, the HDL may pick up cholestyl ester and phospholipid during normal VLDL lipolysis in the plasma. The HDL levels are relatively insensitive to diet. At present time, the cause-and -diet effect of HDL's inverse relation to CHD remains unclear.

고 불포화 지방산 식이가 인체 혈청 Cholesterol의 Esterification과 Transfer에 미치는 영향 (Effects of High Polyunsaturated Fat Diet on Human Plasma Cholesterol Esterification and Transfer)

  • Lecithin Cholesterol Acyltransferase(LCAT)와 cholesteryl ester transfer protein(CETP)은 간과 간외조직간의 혈청 cholcsterol 항상성을 유지하는데 중요한 기전인 reverse cholestrol transport system(RCT)에 매우 중요한 인자들이다. 본 연구의 목적은 RCT 기전의 식이 지질 효과를 추정하여 항 고지혈증 치료식이 연구의 중요한 자료를 제공하는데 있다. 4주의 인체대사 실험에서, 새당사 12명의 여성들은 실험 1주 전에 고 포화지질로써 적응시기를 거친 후, 각각 6명씩 high PUFA(corn oil)군과 high SFA(butter)군에 무작위로 배정되었다. Butter군은 total-34(%), esterified-(96%), $HDL_3$-(23%), LDL-(20%) 및 VLDL+LDL-(35%) cholestetol을 감소시켰다. Corn oil군은 esterified(2.5%) 및 LDL-(15%) cholesterol과 triglycerde(27%)를 감소시켰다. Corn oil과 butter fat군간의 식이 효과 차이는 total-(p=0.0001), esterified-(p=0.0001), total HDL-(p=0.005), $HDL_2$-(p=0.01) 및 LDL-(p=0.0001) cholesterol에서 유의적이다. LCAT activity는 두 군에서 변화가 없으나, 4주 후 CETP activity는 butter군에서 2.5배 증가하였다. 이는 VLDL+LDL cholesterol 농도가 butter군에서 증가한 결과와 일치한다. LCAT activity는 corn oil군에서 증가된 HDL의 apo A-I 농도와 free cholesterol과 정의 상관관계가 높은 반면, CETP activity는 total cholesterol과 LDL 및 VLDL+LDL cholesterol과 정의 상관관계가 높았다.

Dimethylhydrazine으로 처리한 쥐에서 식이의 Calcium 함량과 지방종류에 따라 혈장 Cholesterol 수준과 Cholesterol 대사에 미치는 영향 (Effect of Dietary Calcium and Fat on Plasma Cholesterol Level and Cholesterol Metabolism in 1, 2-dimethylhydrazine-treated Rats)

  • The study was designed to observe the effect of dietary calcium and fats on plasma cholesterol level, hepatic microsomal fluidity and HMG-CoA reductase activity as well as the excretion of fecal bile acids and neutral sterols in 1, 2-dimethylhydrazine(DMH)-treated rats. Male Sprague Dawley rats, at 7 weeks of age, were divided into 2 groups, 0.3% and 1.0% Ca levels and each group again subdivided into 2 groups of corn oil and perilla oil. Each rat was intramuscularly infused with DMH for 6 weeks to give total dose of 180mg/kg body weight and also fed experimental diet containing 15%(w/w) different fit and Ca(0.3% or 1.0%) for 20 weeks. High dietary calcium(1.0%) did not significantly influence on plasma cholesterol as well as hepatic microsomal fluidity and HMG CoA reductase activity, but significantly reduced the excretion of total bile acid per gram of faces and increased the excretion of total neutral sterol. However, high dietary Ca reduced the excretion of secondary bile acid(deoxycholic and lithocholic acids) which was known as promoter for colon cancer. Perilla oil rich in n-3 ${\alpha}$-linolenic acid significantly decreased plasma cholesterol by increasing hepatic microsomal fluidity compared with corn oil, but did not influence on HMG CoA reductase activity. Perilla oil did not influence on fecal excretion of total and primary bile acids, but reduced the excretion of secondary bile acids. Therefore, it could be recommended to consume more fish product and food rich in calcium and use more perilla oil in meal preparation to prevent from coronary hear disease and colon cancer especially when high fit diet has been practiced. Korean Curcuma longa L. induces lipolysis and regulates leptin in adipocyte cells and rats

  • BACKGROUND/OBJECTIVES: Turmeric (Curcuma longa L.) has been reported to have many biological functions including anti-obesity. Leptin, peptide hormone produced by adipocytes and its concentration is increased in proportion to the amount of the adipocytes. In the present study, we examined the effects of Korean turmeric on the regulation of adiposity and leptin levels in 3T3-L1 adipocytes and rats fed a high-fat and high-cholesterol diet. MATERIALS/METHODS: Leptin secretion, free fatty acid and glycerol contents in 3T3-L1 adipocytes were measured after incubation of cells with turmeric for 24 hours. Rats were divided into four experimental groups: a normal diet group (N), a high-fat and high-cholesterol diet group (HF), a high-fat and high-cholesterol diet group supplemented with 2.5% turmeric extracts (TPA group) and a high-fat and high-cholesterol diet group supplemented with 5% turmeric extracts (TPB group). Serum samples were used for the measurement of leptin concentration. RESULTS: Contents of free fatty acid and glycerol showed concentration dependent increase in response to turmeric extracts. Effects of turmeric extracts on reduction of lipid accumulation in 3T3-L1 cells were examined by Oil Red O staining. Treatment with turmeric extracts resulted in increased expression levels of adipose triglyceride lipase and hormone-sensitive lipase mRNA. The concentration of leptin from 3T3-L1 adipocytes was significantly decreased by turmeric. Proportional abdominal and epididymal fats weights of the turmeric 5% supplemented group, TPB has significantly decreased compared to the HF group. The serum levels of leptin in the TPA and TPB groups were significantly lower than those of the HF group. CONCLUSIONS: Based on these results, we suggested that Korean turmeric may contribute to the decreasing of body fat and regulating leptin secretion.

Effects of Diets on Serum and Liver Lipid Levels and Fatty Acid Composition of Liver Phospholipids in Rats

  • Serum and liver lipid levels and fatty acid composition of liver phospolipids (PL) were investigated in 36 rats which consumed either one of five different dietary fats or a high carbohydrate diet for 4 weeks. As the sources of five dietary fats, concentrated cicosapentaenoic acid(EPA), fish oil (FO), perilla oil(PO), corn oil(CO) and beef tallow (BT) were provided to the rats. As a control group, cron starch (CS) replaced dietary fat. The FO group showed lower serum total cholesterol (TC), high density lipiprotein cholesterol(HDL-C) and serum PL levels than those of the CO group(p<0.05). There were no significant differences in serum TC and serum HDL-C levels between the polyunsatured fatty acid(PUFA) groups and the EPA, FO and PO groups. The CS group showed the highest level serum TC. Compared with the CS group, both the EPA and CO groups showed significantly lower atherogenic indices(AI). However, there were no significant differences in AI among different dietary fat groups. No significant differences in liver triglyceride (TG) , TC and PL levels were detected among the six experimental groups. Phosphatidylcholine(PC) and phosphatidylethanolamine(PE) composed 30-40% and 15-20% of total liver PL, respectively. The fatty acid composition of liver PC and PE reflected dietary fatty acid composition . Compared to the different dietary fat based diets used in our study, the high carbohydrate diet had the most adverse effects on serum lipid profiles. However, we can not conclude from this result that long chain n-3 PUFA diets such as the EPA and FO based diets have more beneficial effects on serum lipid profiles than n-6 PUFA diet such as the CO based diet or shorter chain n-3 PUFA diets like the PO based diet.

고지방식이를 섭취한 흰쥐에서 김치유산균분말의 비만 억제 및 지질 저하 효과 (Weight Reduction and Lipid Lowering Effects of Kimchi Lactic Acid Powder in Rats Fed High Fat Diets)

  • 김치유산균추출물의 비만억제 및 지질 저하 효과를 20%(w/w) 고지방식이를 섭취시킨 횐 쥐에서 살펴보았다. 제한식이를 하였음에도 불구하고 HFD의 체중은 ND에 비해 약 50%증가 하였고, HFK10 및 HFK20에서는 HFD에 비해 각각 13%그리고 15%(p<0.01) 감소하였다. HFD의 복부지방의 함량은 ND에비해 98% 증가하였고(p<0.01), HFK10 및 HFK20에 서 각각 42%, 48% 유의적으로 감소하여(p<0.01) 체중 저하 효과 보다 복부지방 축적을 억제하는 효과가 더 높은 것으로 나타났다. 혈장 중성지방의 농도는 HFD가 ND에 비해 약 55% 증가하였고(p<0.01), HFK10 및 HFK20은 HFD에 비해 26%, 및 35% 농도 의존적으로 감소하였다(p<0.01). 혈장 총 콜레스테롤 농도 역시 HFK10 및 HFK20에서 농도의존적으로 유의적인 감소를 보였고(p<0.01) LDL 농도는 HFD가 ND보다 61% 정도 증가하였고(p<0.01), HFK10 및 HFK20에서 약 8%, 33%감소하여(p<0.01)총 콜레스테롤 보다 LDL의 감소가 더 현저하였다. HDL은 증가하여 김치유산균추출물의 첨가에 의한 지질 개선 효과가 관찰되었다. 김치 유산균 추출물의 간 기능 개선 효과는 고지방 식이에 의해 유발된 지방간 현상으로 정상대조군에 비해 상승되었던 혈장 AST 및 ALT 활성이 감소됨으로서 관찰되었다. 분변의 지방농도를 HFD와 HFK10 및 HFK20간에 비교하였을 때 중성지방 농도는 각 62%와 111% 정도 현저하게 증가하였고, 콜레스테롤 농도는 HFK20에서 약 31% 정도 증가하였다(p<0.05). 이상의 결과에서 사균의 형태인 김치유산균 추출물은 비만을 억제하고 혈장 지질을 저하시키는 효과가 있음이 확인되었으며, 간 기능 보호효과도 확인되었다. 이러한 김치유산균 추출물의 효과는 장내에서 지방의 흡수를 억제하기 때문으로 생각된다.

The effects of high-fat diets composed of different animal and vegetable fat sources on the health status and tissue lipid profiles of male Japanese quail (Coturnix coturnix japonica)

  • Objective: The current study aimed to investigate the impact of high-fat diets composed of different animal and vegetable fat sources on serum metabolic health markers in Japanese quail, as well as the overall lipid content and fatty acid profiles of the edible bird tissues following significantly increased dietary lipid supplementation. Methods: Fifty seven male quail were divided into six groups and fed either a standard diet or a diet enriched with one of five different fats (22% coconut oil, lard, palm oil, soybean oil, or sunflower oil) for 12 weeks. The birds were subjected to an oral glucose tolerance test following the feeding period, after which they were euthanized and blood, liver, breast, and thigh muscle samples collected. Total fat content and fatty acid profiles of the tissue samples, as well as serum uric acid, triglyceride, cholesterol, total protein, albumin, aspartate transaminase, and total bilirubin concentrations were assessed. Results: High-fat diet feeding had no significant effects on the glucose tolerance of the birds. Dietary fatty acid profiles of the added fats were reflected in the lipid profiles of both the liver and breast and thigh muscle tissues, indicating successful transfer of dietary fatty acids to the edible bird tissues. The significantly increased level of lipid inclusion in the diets of the quail used in the present study was unsuccessful in increasing the overall lipid content of the edible bird tissues. Serum metabolic health markers in birds on the high-fat diets were not significantly different from those observed in birds on the standard diet. Conclusion: Thus, despite the various high-fat diets modifying the fatty acid profile of the birds' tissues, unlike in most mammals, the birds maintained a normal health status following consumption of the various high-fat diets.

동충하초 (Cordyceps Militaris)가 고지방식이를 섭취한 흰쥐의 지질대사, 단백질농도 및 효소활성에 미치는 영향 (The Effects of Cordyceps Militaris on Lipid Metabolism, Protein Levels and Enzyme Activities in Rats Fed a High Fat Diet)

  • 동충하초 (Cordyceps militaris)의 자실체와 균사체가 고지방 식이를 섭취한 성숙된 흰쥐의 영양 생리활성에 미치는 영향을 평가하고자, 생후 35주령의 흰쥐에 표준식이를 급여한 정상군, 표준식이에 15% 돈지를 첨가한 식이를 급여한 고지방군, 고지방 식이에 자실체 및 균사체를 각각 3%씩 첨가한 식이를 급여한 군 등 4군으로 나누어 5주간 사육하였다. 실험동물의 체중증가, 간의 무게 및 식이효율은 고지방군에 비하여 자실체군 및 균사체군이 유의하게 감소되어 정상군과 비슷한 수준으로 체중과 간의 무게를 정상으로 유지시켜 주는 효과가 나타났다. 식이섭취량은 고지방군과 동충하초군들이 비슷한 수준을 유지하였다. 신장, 췌장 및 심장의 무게는 정상군과 각 실험군이 비슷하였다. 간의 총 지질과 콜레스테롤 농도 및 간과 혈청의 중성지질 농도는 고지방군에 비하여 자실체군 및 균사체군이 유의하게 감소되었다. 혈청의 총 콜레스테롤 및 인지질 농도는 정상군에 비하여 각 실험군이 유의하게 증가되었다. 혈청의 HDL-콜레스테롤 농도는 정상군에 비하여 자실체군 및 균사체군이 유의하게 증가되었다. 고지방군에 비하여 자실체군의 총 콜레스테롤에 대한 HDL-콜레스테롤 비율은 유의하게 증가되었고, 동맥경화지수는 감소되었다. 혈청의 총 단백질 농도는 정상군에 비하여 고지방군 및 균사체군이 유의하게 증가되었다. 혈청의 알부민, 혈당 및 혈색소 농도는 정상군과 각 실험군이 비슷하여 정상 수준을 유지하였다. 혈청의 glutamic pyruvic transaminase, glutamic oxaloacetic transaminase 및 ${\gamma}$-glutamyltr-anspeptidase의 활성은 정상군과 각 실험군이 유사한 경향을 보였다. Lactic dehydrogenase 활성은 고지방군에 비하여 자실체군 및 균사체군은 유의하게 증가되어 정상군과 비슷하였다. Alkaline phosphatase 활성은 정상군에 비하여 각 실험군이 유의하게 감소되었다. 이상의 결과로 보아 고지방 식이에 3%동충하초의 자실체와 균사체 섭취에 의한 체중과 간의 무게가 정상수준을 유지하였고, 간의 총 지질과 콜레스테롤 농도 및 간과 혈청의 중성지질 농도를 낮추는 효과가 있는 것으로 나타났다.

황기홍국소스를 처리한 계육이 고지방 및 고콜레스테롤 식이 흰쥐의 체중과 혈청 및 간 조직의 지질 함량에 미치는 영향 (Effects of Chicken Treated with Hwangki-Beni Koji Sauces on Body Weight, Serum and Hepatic Lipid Profiles of Rats Fed High Fat and High Cholesterol Diets)

  • 황기열수추출물(WEH)과 홍국 70% 에탄올추출물(EEB) 및 Monascus pilosus 액침배양액(LBK)을 동결건조한 등량 혼합물을 일반 베이스소스에 0.225% 혼합한 황기홍국소스(HBS)에 침지한 계육의 식이가 고지방-고콜레스테롤 병행식이 흰쥐의 체중, 장기중량, 혈청지질 함량에 미치는 영향을 조사하였다. 실험군은 정상군(NC), 고지방-고콜레스테롤 대조군(HFC), 고지방-고콜레스테롤 식이에 베이스소스를 처리한 계육을 15% 첨가한 식이군(HFC-BS) 및 고지방-고콜레스테롤 식이에 황기홍국소스를 처리한 계육을 15% 첨가한 식이군(HFC-HBS)으로 구분하여(5 마리/군) 5주간 사육하였다. 식이섭취량은 HFC-HBS군이 HFC 및 HFCBS군에 비하여 높았으나 식이효율과 사육후의 체중은 낮아 체중감소효과가 확인되었다. ALT 및 AST의 활성도 정상군에는 미치지 못하나 HFC군 및 HFC-BS군에 비하여 현저한 감소를 보였다. HFC-HBS군은 HFC-BS군에 비하여 혈청 triglyceride 함량은 34.06%, total cholesterol 함량은 6.28% 및 LDL-cholesterol 함량은 31.00%가 감소하였으며 HDL-cholesterol 함량은 1.72배 증가되었고 동맥경화지수는 2.52배 감소하였다. 간 조직의 총 지질, 중성지질 및 총콜레스테롤 함량도 HFC-HBS군이 HFC-BS군에 비하여 각각 9.53%, 5.25% 및 22.20%가 감소되었다. NC군은 정상적인 간 조직구조를 나타낸 반면 HFC군은 거의 모든 간세포가 multiple micro vesicles 형태의 지방을 함유하는 것으로 나타났다. 또한 HFC-BS군에서는 HFC군과 비교해 볼 때 소적형 지방축적은 감소되었으나 염증세포와 쿠퍼세포가 활성화된 모습을 보인 반면 HFC-HBS군은 HFC-BS군과 비교해 볼 때 간 실질세포 내 지방들은 아주 작은 포말형으로 나타나며 정상적인 간 실질세포의 형태가 잘 나타나 고지방-고콜레스테롤 식이로 인한 간세포 손상을 크게 완화시킨 것으로 사료된다.

고지방식이를 투여한 생쥐에서 마황과 향부자의 항비만 효능 비교 연구 (The comparative studies on anti-obesity effects of Ephedrae Herba and Cyperi rhizoma in high fat diet fed mice)

  • Objectives : This comparative study was to investigate on anti-obesity effects of Ephedrae Herba and Cyperi rhizoma in high fat diet(HFD) fed mice. Methods : Male C57BL/6J mice were fed a normal diet(normal group, N), high fat 45 cal% diet[HFD, control group, C), HFD with Ephedrae Herba(EH group) and Cyperi rhizoma(CR group) extracts fed for 5 weeks. We were observed as follows : changes of body weight, amount of diet intake, weight of total visceral fats, levels of obesity-related hormones and blood lipids. Results : The change of body weight after EH and CR oral administration significantly more decreased in EH group than that of control group. The FFR(Food Efficiency Ratio) was decreased in EH group, but more increased in CR group than that of control group. The weight of periepididymal and perirenal fats were significantly decreased in EH and CR groups compared to the control group. The levels of serum leptin and insulin were significantly decreased in EH group, and the level of serum adiponectin was increased in EH group compared to control group. The levels of serum triglyceride and total cholesterol were significantly decreased in EH and CR groups, and HDL-cholesterol levels was significantly increased in EH group compared to control group. Conversely in CR group, its values showed the opposite effect. The staining density of lipid droplets within the hepatocytes was widely distributed in CR and control groups, but in EH group, its density was weakly stained. Conclusions : These experimental results suggest that Ephedrae Herba shows conspicuous anti-obesity effect, and Cyperi rhizoma shows weak anti-obesity effect.
