식이성 유발 고지혈증이 체내지질대사 및 신기능에 미치는 영향 (Effects of High fat and High Cholesterol Diet on Kidney Function)

  • The effects of diet induced hyperlipidemia on kidney function were studied in Sprague-Dawley rats fed high fat diet containing 20% beef tallow and high cholesterol diet containing 5% cholesterol for 8, 12, 16 weeks, respectively. The concentrations of serum total lipid, total cholesterol and LDL-cholesterol were significantly high cholesterol diet groups during all experimental periods (p<0.05). HDL-cholesterol concentration was the lowest value in high cholesterol diet group of 16 weeks(p<0.05). Triglyceride concentration was not affected by experimental diets. Serum total protein, albumin and creatinine concentration tended to higher in high cholesterol diet groups than those in high fat diet groups. And serum urea-N concentration was higher in high fat diet group of 16 weeks than that in other diet groups. Urinary total protein and urea-N were higher in high cholesterol diet groups than those in other diet groups regardless of experimental period period. There was no significant difference in urinary creatinine concentratin among diet groups(p<0.05). GFR was lower in high cholesterol diet groups than that in high fat diet groups at 8, 16 weeks, respectively. Wet weight per body weight, total lipid, triglyceride, total cholesterol concentations of liver tissue were apparently high in high cholesterol diet groups(p<0.05). Kidney wet weight per body weight wer not affected by experimently diets, total lipid concentration of kidney tissue was significantly high in high fat diet groups of 12 weeks(p<0.05), kidney tissue triglyceride concentrations of high cholesterol diet groups of 12, 16 weeks apparently low, and total cholesterol concentration of kidney tissue was higher in experimental diet groups than that of control groups at 12, 16 weeks(p<0.05). Fecal excretion, total lipid, triglyceride and cholesterol concentrations of feces were markedly high in all high cholesterol diet groups except high fat diet group of 16 weeks. The results of light microscopic examination indicated that glomerulosclerosis was not obsrved in rats fed experimental diets.

  • This study was conducted to investigate the hypocholesterolemic effects of porphyran and insoluble dietary fiber isolated from laver in rats fed high fat diet containing 1% cholesterol, 0.25% sodium cholate and 12% lard. Rats were fed, ad libitum, diets containing 5% diet fiber as cellulose(normal control or high fat control), porphyran or insoluble dietary fiber for 4 weeks. Among the groups fed high fat diet, liver weight was significantly lower in high fat porphyran group than high fat control. Plasma GOT, GPT, total cholesterol, cholesteryl ester, LDL-cholesterol and liver total cholesterol concentration were significantly lower in high fat porphyran group than high fat control. The feeding of porphyran significantly increased fecal cholesterol and bile acid excretion. The feeding of insoluble dietary fiber had no significant effect on either plasma or liver cholesterol levels, although fecal cholesterol level in the insoluble dietary fiber group was significantly higher than that in the high fat control. The results indicate that porphyran isolated from the laver may exert their hypocholesterolemic effect by increasing excretion of fecal bile acid and cholesterol.

Effect of Red Pepper Seeds Powder on Lipid Composition in Rats Fed High-Fat.High-Cholesterol Diets

  • The purpose of the present study was to evaluate the effects of red pepper seeds powder on lipid metabolism in rats fed high fat high cholesterol diet. Rats were divided into five experimental groups: normal diet group, high fat high cholesterol diet group, high fat high cholesterol diet with 5% red pepper seeds powder supplemented group (SA group), high fat high cholesterol diet with 10% red pepper seeds powder supplemented group (SB group) and high fat high cholesterol diet with 15% red peeper seeds powder supplemented group (SC group). The serum triglyceride (TG) and cholesterol contents, and LDL-cholesterol and atherogenic index (AI) of the red pepper seed powder supplemented groups were significantly decreased compared to the HF group. The serum HDL-cholesterol contents of the red pepper seed powder supplemented groups were increased compared to the HF group. However, there was no significant difference in the serum HDL-cholesterol among all experimental groups. The hepatic TG and cholesterol contents of the red pepper seed powder supplemented groups were significantly decreased compared to the HF group. The fecal total cholesterol and triglyceride contents of the red pepper seeds powder supplemented groups were significantly increased compared to the HF group. These results suggest that supplementation of red pepper seed powder may have a pronounced impact on markers of lipid metabolism in serum and liver of rats fed high fat high cholesterol diets.

서목태(Rhynchosia nulubilis) 추출물이 고지방 및 고콜레스테롤 식이 급여 흰쥐의 혈청 지질 농도에 미치는 영향 (Effects of Methanol Extracts from Rhynchosia nulubilis on serum Lipid Concentrations in Rats fed High Fat and High Cholesterol Diet)

  • The effects of the Rhynchosia nulubilis extracts on serum lipid concentrations were evaluated in rats. Forty-two male Sprague-Dawley rats weighing $100{\pm}10g$ were divided into six groups and fed high fat and high cholesterol diets for four weeks. Experimental groups were administered with following diets; basal fat diet(control) and basal and high fat diets with 1% cholesterol and Rhynchosia nulubilis extract(40mg/kg, 400mg/kg and 4000mg/kg). The concentrations of serum triglyceride in rats fed the Rhynchosis mulubilis extract, high fat, 1% cholesterol, and RN-HFC(-40, -400, -4000mg/kg) groups were lower than those in other groups. The concentrations of total cholesterol in the RN-HFC (-40, -400, -4000mg/kg) groups were lower than those in fat diet groups. The concentrations of HDL-cholesterol in serum of the RN-HFC(-40, -400, 4000mg/kg) groups were significantly higer than those of other groups. The levels of LDL-cholesterol in serum of the RN-HFC(-40, -400, -4000mg/kg) groups were tended to be lower than those of other groups. These results suggest that Rhynchosia nulubilis extract may reduce elevated levels of serum lipid concentrations in rats fed high fat diets.

  • This study investigated the effect of Dolichos lablab extract on the lipid metabolism of rats fed a high fat and cholesterol diet. S-D male rats (n=48) were given 40, 400, or 4000 mg/kg of Dolichos lablab extract and fed a high fat and high cholesterol diet for four weeks (DL-HFC group). As a result, serum levels of total lipids, total cholesterol, and triglycerides were higher in the high fat and high cholesterol group (HFC-group) than the control, and significant differences were observed between the control and the DL-HFC (40, 400, 4000 mg/kg) group. The fat absorption rate increased with a high fat diet feeding, but was higher in the DL-HFC (40, 400, 4000 mg/kg) group.

Effects of Cordyceps militans Cultivated on Rice on Lipid Metabolism in Rats Fed High Fat-cholesterol Diets

  • Dongchunghacho rice, produced by cultivating Dongchunghacho fungus on rice, could be an effective functional food because it offers added value to rice and thus increases rice consumption. However, the physiological effect of Dongchunghacho rice has not been reported yet although there is increasing consumers demand. Therefore, we investigated the effect of Dongchunghacho rice (unpolished rice cultivated with Cordyceps militaris) on lipid metabolism in hyperlipidemic rats. Forty of 8 wk-old male Sprague-Dawley rats were divided into four groups after a 1 week of adaptation period and fed either a normal diet (66% polished rice diet, NC), high fat (12 g/100 g)-high cholesterol (1 g/100 g) diet with 53% polished rice (HC), or high fat-high cholesterol diets supplemented with 30% of the total rice as either unpolished rice (DR) or Dongchunghacho rice (DR). After 4 weeks, rats fed the Dongchunghacho rice diet with high fat and cholesterol had dramatically lower plasma LDL cholesterol concentrations and atherogenic indexes and higher plasma HDL cholesterol levels compared with the rats consuming polished rice or unpolished rice with high fat and cholesterol diet. Dongchunghacho rice led to less total lipid and total cholesterol accumulation in liver. However, these significant reductions in plasma or hepatic lipid profiles were not closely correlated with fecal total lipid or total cholesterol excretion. The plasma concentration of total cholesterol and triglycerides were not affected by Dongchunghacho rice. This hypolipidemic effect of Dongchunghacho rice seemed to be unrelated to unpolished rice itself, because the plasma and hepatic lipid profiles of DR group were not different from that of the HC group. These results suggest that unpolished rice containing cultivated Cordyceps militaris can improve plasma and hepatic lipid profiles in rats fed with high fat-high cholesterol diet.

  • The present studies were designed to observe the effects of both dietary fat levels and P/S ratio on lipid components in plasma and tissues of young rats when cholesterol was supplemented at 1%(w/w) to four dietary groups providing total fat as 10%( LF ) or 45% ( HF ) of calories and P/S ratio as 0.2 or 4.0, Low Fat-0.2 : Low Fat-4.0 : High Fat-0.2 : High Fat-4.0. Plasma total cholesterol was increased almost to the same level in all dietary groups after the cholesterol supplement but the value of HF -0.2 was slightly higher than that of LF -0.2. Plasma TG was also increased in all dietary groups when cholesterol was supplemented but was more significantly increased in LF group than in HF group. HDL-cholesterol was slightly increased by cholesterol supplement but there was no effect by the total dietary fat level of previous diet. However, plasma HDL-cholesterol, total cholesterol and TG levels were slightly reduced in high fat diet of P/S 4.0. Total cholesterol per g -liver was higher in HF group than in LF group before and after cholesterol supplement even though it was increased more in LF group with cholesterol addition. Liver cholesterol was also higher in rats fed diets of P/S 4.0 at high fat level before and after cholesterol supplement. In contrast, total cholesterol per g-muscle was reduced by cholesterol supplement in all groups and it was significantly higher in LF than in HF. There was no significant effect in liver TG by total fat level and P/S ratio when cholesterol was supplemented. Muscle TG was lowered in all groups by cholesterol supplement but no effect by total fat level and P/S ratio.

해조류 갈래곰보가 고지방 식이를 급여한 흰쥐의 혈액 및 간장 지질 농도에 미치는 영향 (Effect of Meristotheca papulosa on Lipid Concentration of Serum and Liver in Rats Fed High Fat Diet)

  • This study was carried out to investigate the effect of Meristotheca papulosa on lipid concentration of serum and liver in rats fed high fat diet for 4 weeks. Experimental groups ere divided into normal group (5% corn oil), high fat diet group (5% corn oil+20% lard), high fat diet and 5% or 10% Meristotheca papulosa treated group. The weights of kidney, heart and tests were not significantly different among the group, but weight of liver was significantly decreased by supplement of Meristotheca papulosa (10%). The contents of serum total lipid, trigly-ceride, total cholesterol, free cholesterol, cholesterol ester and atherogenic index were decreased, while HDL-cholesterol was increased by Meristotheca papulosa supplementation compared to high fat diet group. Meristotheca papulosa supplement has obvious effects on lowering hepatic total lipid, triglyceride and total cholesterol. Our findings suggest that Meristotheca papulosa supplement can improve lipid metabolism in high fat fed rats.

Anti-obesity Effect of Monascus pilosus Mycelial Extract in High Fat Diet-induced Obese Rats

  • This study was carried out to investigate the dietary effects of Monascus pilosus mycelial extract on obesity in high-fat with cholesterol-induced obese rat models. It was observed that M. pilosus mycelial extract contains $25.85{\pm}1.98mg%$ of total monacolin K without citrinin by highperformance liquid chromatography (HPLC). The rats were randomly divided into 2 groups; normal control and a high-fat with cholesterol diet group. The high-fat with cholesterol diet group was fed a 5L79 diet with an added 15% lard and 1% cholesterol supplemented diet for 3 weeks for induction of obesity. After induction, obesity was confirmed by checking obesity indexes, the animals were divided into 4 groups (n=5); first, the normal control (NC), and then taken from the obese model of rats, a high-fat with cholesterol diet obesity control group (HF), 0.5% M. pilosus mycelial extract supplemented high-fat with cholesterol diet group (MPMs), 2% conjugated linoleic acid supplemented high-fat with cholesterol diet group (CLA) for 7 weeks. Body weight gains, obesity indexes, and body fat contents in the experimental groups (MPMs and CLA) were decreased compared with HF group. Feed Efficiency Ratio (FER) in MPMs was significantly lower than that of HF without change of feed intake. These results suggested that the anti-obesity effects of the M. pilosus mycelial extracts (MPMs) could prevent obesity induced by high-fat with cholesterol diet possibly via inhibition of lipid absorption.

  • The effects of indigestible dextrin on lipids in serum, tissue, and feces were investigated in two experiments carried out with 64 male Sprague-Dawley rats for eight weeks. Experiment I diet(normal fat diet) containing 5% corn oil and experiment II diet(high fat diet) containing 18% fat(13% beef tallow and 5% corn oil) and 1% cholesterol were divided into four groups with 0.5% cellulose(0.5CL : control), 10% cellulose(10CL), 10% Indigestible dextrin I(ID-I) and 10% indigestible dextrin II(ID-II), respectively. ID-I and ID-II diets produced a decrease in serum total lipid and cholesterol concentrations in both normal and high fat diets but more significantly in the groups fed high fat diet. ID-I, ID-II, and 10CL of high fat diet decreased serum glucose concentration. ID-I and ID-II diets also decreased liver cholesterol and triglyceride concentrations in rats fed normal and high fit diets. The fecal excretion of cholesterol in ID-II/high fat diet was significantly higher than that of 0.5CL diet. Bile acid excretion was increased in the rats fed 10CL and ID-II diets of normal and high fat diets. These observations indicat the hypolipidemic and hypocholesterolemic effects of indigestible dextrin on the rats. These effects are mediated by the increase of facal cholesterol and bile acid excretion. (Korean J Nutrition31(6) : 981-990, 1998)
