곤약 글루코만난을 첨가한 식빵 급여가 고지방식이로 유도한 비만 흰쥐의 체중 및 혈청지방에 미치는 영향 (Effects of White Bread with Konjac Glucomannan on Body Weight and Serum Lipids on Rats with Diet-Induced Obesity)

  • 곤약 글루코만난(liquid konjac)을 동결건조한 식빵에 첨가한 사료를 고지방식이로 유도한 비만 흰쥐에 6주간 급여하였을 때의 체중, 혈청지질, 비만지수에 미치는 영향을 조사하였다. 실험군은 정상군(NC), 고지방식이 비만 대조군(HF), 고지방식이 비만+곤약 글루코만난을 10% 되게 혼합하여 동결건조 한 식빵을 사료에 10% 혼합한 식이군(KGM1) 및 비만+곤약 글루코만난을 20%되게 혼합하여 동결건조한 식빵을 사료에 20% 혼합한 식이군(KGM2)으로 구분(군당 7마리)하여 6주간 사육하였다. KGM1군과 KGM2군은 HF군에 비하여 혈청 중성지질, 총지질, 총콜레스테롤, LDL-콜레스테롤 함량 및 atherogenic index(AI)는 감소하였으나 HDL-콜레스테롤 함량은 증가하였다. KGM1군과 KGM2군은 HF군의 체중에 비하여 감소하였으며 곤약 글루코만난 첨가 식빵의 비율이 높아질수록 체중감소율이 높았다. 체지방 함량은 NC군 및 HF군에서는 실험식이 6주후에 크게 증가하였으나 KGM1군은 유의적인 변화가 없었고, KGM2군은 감소하였다. 따라서 곤약 글루코만난 첨가 식빵은 고지방식이로 유도된 비만 흰쥐에서 체지방 함량을 감소시키는 효과가 있었다.

Effects of Purslane Extract on Obesity and Diabetes in High-Fat Diet-Induced Obese Mice

  • The frequency of obesity has risen dramatically in recent years but only few safe and effective drugs are currently available. In addition, obesity can induce type 2 diabetes (T2DM), hyperlipidemia and fatty liver disease. Recently, protective effect of purslane extract (PE) on obesity has been reported, but little is known about the role and mechanism of PE in obesity. This study aimed to evaluate the effect of PE on obesity and diabetes in obese mice. In addition, the effect of PE was compared with anti-obesity and diabetes drugs. High-fat diet (HFD)-induced obese mice were treated for 8 weeks with drugs as follows: PE, orlistat, metformin, voglibose or pioglitazone. While PE mixed with normal diet did not have any effects on BW in non-obese mice, PE mixed with HFD significantly reduced BW gain, insulin resistance, and glucose intolerance, without affecting food intake and appetite in obese mice. The effect was comparable to the effects of anti-obesity and diabetes drugs. Furthermore, PE significantly increased the activity of hepatocellular anti-oxidant enzymes, leading to protection of liver from oxidative stress in obese mice. These results suggest that PE treatment may be a useful tool for preventing obesity and complication of obesity.

비만1호방(肥滿1號方)이 고지방식이(高脂肪食餌)로 비만(肥滿) 유도(誘導)된 백서(白鼠)에 미치는 영향 (The Inhibitory Effects of Bimanbang-1(肥滿1號方) on the Obese-mouse Fed High-fat Diet)

  • Objective: This study investigated the effects of Bimanbang-1 (肥滿1號方: here in after referred to BMB-1) on the obese gene and obese inhibitory. Methods: C57BL/6 mice were induced by high fat diet. Mice were divided into three groups (normal, high fat diet with control, high fat diet with BMB-1 extract) and fed for 15 weeks. Items of this experimental study are as follows: body weight change, final body weight, the weight change of the adipocytes in body, the level change of LFT, NEFA and creatinine, the expression of ${\beta}3AR$ and leptin gene in primary adipocytes, the production change of leptin in primary adipocytes, the expression of ${\beta}3AR$, leptin and $TNF-{\alpha}$ in adipocytes tissue. Result: 1. All experimental groups showedthat the weight change decreased considerably and the high density group showedthat the final weight decreased considerably. 2. The high density group showed that the amount of the adipocyte in weight decreased considerably. 3. All experimental groups showedthat the amount of ALT decreased considerably, and AST decreased in the high density group. However, the amount of creatinine and glucose did not increase considerably. 4. All experimental groups showed that the amount of total cholesterol, LDL-cholesterol, triglyceride, and NEFA decreased, and HDL-cholesterol increased considerably. 5. The high density groups showedthat the amount of leptin decreased considerably. 6. All experimental groups showed that the revelation of ${\beta}3AR$ in primary adipose cell and 3T3-L1 cell increased considerably, and that the revelation of leptin and $TNF-{\alpha}$ in primary adipose cellsand 3T3-L1 cells decreased considerably. 7. All experimental groups showed that the size of adipocyte in adipocytes tissue decreased. 8. All experimental groups showed that the adipose vacuoles in liver tissue decreased considerably. Conclusion: The findings suggest that Bimanbang-1 causes weight loss and histological change, thus it may be effective to treat obesity.

라스베라트롤 투여가 고지방식이 비만쥐의 지방조직에서의 inflammasome과 대식세포 마커에 미치는 영향 (Resveratrol Ameliorates High-fat-induced Metabolic Complications by Changing the Expression of Inflammasome Markers and Macrophage M1 and M2 Markers in Obese Mice)

  • 본 연구 목적은 고지방식이 유도 비만 쥐의 피하지방조직에서 라스베라트롤 투여가 대식세포 침윤관련 염증인자에 미치는 영향을 규명하고자 하였다. 본 연구를 위해 정상식이군, 고지방식이군, 고지방식이+라스베라트롤 투여군으로 분류한 후, 라스베라트롤 투여군은 15주간 25 mg/kg 농도로 Dimethyl Sulfoxide에 용해하여 투여하였으며, 비교군은 Dimethyl Sulfoxide 용액만을 투여하였다. 연구결과 고지방식이군은 정상식이군에 비하여 체중이 유의하게 증가하였고, 라스베라트롤 투여군에서 고지방식이 군보다 NLRP3. ASC, Casepase1 mRNA 발현이 감소하였다. 또한 염증마커로 알려진 IL-18 mRNA 발현이 라스베라트롤 투여군에서 정상식이군과 고지방식이군보다 낮게 나타났다. 대식세포 침윤 마커인 F480, CD86 mRNA 발현에서도 라스베라트롤 투여군에서 고지방식이 군보다 유의한 감소를 보였다. 따라서 라스베라트롤 투여는 고지방식이 유도 비만 상황에서 대식세포 침윤 염증과 inflammasome에 긍정적인 영향을 미치는 것으로 보여진다.

Anti-Obesity and Hypolipidemic Effects of Dietary Levan in High Fat Diet-Induced Obese Rats

  • We found previously that dietary high fat caused obesity, and levan supplementation to the regular diet reduced adiposity and serum lipids. In the present study, we examined the effects of levan [high-molecular-mass $\beta$-(2,6)-linked fructose polymer] supplement on the development of obesity and lipid metabolism in rats fed with high-fat diet. Thus, to determine whether the dietary levan may have the anti-obesity and hypolipidemic effects, 4-wk-old Sprague Dawley male rats were fed with high-fat diet for 6 wk to induce obesity, and subsequently fed with 0, 1, 5, or 10% levan supplemented high-fat diets (w/w) for another 4 wk. For the comparison, a normal control group was fed with AIN-76A diet. Supplementation with levan resulted in a significant reduction of high-fat-induced body weight gain, white fat (i.e., epididymal, visceral, and peritoneal fat) development, adipocyte hypertrophy, and the development of hyperinsulinemia and hyperlipidemia in a dose-dependent manner. Serum triglyceride and free fatty acid levels were greatly reduced by levan supplementation. Serum total cholesterol level was reduced, whereas the HDL cholesterol level was increased by dietary levan. The expression of uncoupling protein (UCP) was increased by dietary high fat, and was further induced by levan supplementation. The mRNA level of UCP1, 2, and 3 in brown adipose tissue (BAT) and UCP3 in skeletal muscle was upregulated in rats fed with dietary levan. In conclusion, upregulated UCP mRNA expression may contribute to suppression of development of obesity through increased energy expenditure. The present results suggest that levan supplementation to the diet is beneficial in suppressing diet-induced obesity and hyperlipidemia.

보기감비탕(補氣減肥湯)이 고지방사료 식이로 비만이 유발된 생쥐에게 미치는 영향 (Inhibitory Effects of Bogigambi-tang on the Obese-mouse Induced by High Fat Diet)

The following results have been obtained All experimental group have shown that the weight and the final increase of weight have decreased considerably. All experimental group have shown that the amount of the adipocyte in weight has decreased considerably. All experimental group have shown that the amount of leptin has decreased considerably. All experimental group have shown that the revelation of ${\beta}$3AR in primary adipose cell and 3T3-L1 cell has increased considerably, and that the revelation of leptin in primary adipose cell and 3T3-L1 cell has decreased considerably, All experimental group have shown that the size of adipocyte in adipocytes tissue has decreased. The high density group have shown that the adipose vacuoles in liver tissue has decreased considerably, and that the cell nucleuses has similar with normal group.

산사의 장내 미생물 조절을 통한 항비만 효과 (Anti-obesity Effect of Crataegus pinnatifida through Gut Microbiota Modulation in High-fat-diet Induced Obese Mice)

  • Objectives This study was performed to evaluate anti-obesity effects of Crataegus pinnatifida (CP) on high-fat-diet induced obese mice. Methods The experimental animals were divided into four groups: normal diet (NOR) group, high fat diet (HFD) group, HFD+Xenical (XEN) group, and HFD+CP (CP) group. NOR group was fed a normal diet and the other three groups were fed high fat diet during the experiment. After the first two weeks of diet, XEN group and CP group were administered with XEN or CP for seven weeks, respectively. After that, we measured body weight, liver weight, fat weight, food intake, and serum concentrations of lipids and liver enzymes. Also the liver, intestine, fat tissue was removed to estimate the obesity-related mRNA expressions and the stool sample was collected to analyze the gut microbiota. Results We found that body weight, fat weight, and triglyceride level were decreased significantly in CP group compared to HFD group. Also CP significantly suppressed gene expressions associated with lipogenesis and inflammation, and increased gene expressions of browning of white adipose tissue and mitochondrial biogenesis. Moreover, it shifted the microbial diversity closer to that of NOR group and increased Firmicutes/Bacteriodetes ratio. Conclusions These results suggest that CP decrease body weight, fat weight and serum triglyceride. Also it inhibit inflammation and adipogenesis, altering gut microbial diversity and abundance. In conclusion, CP could be used as a therapeutic drug for obesity via gut microbiota modulation.

Effect of combined mulberry leaf and fruit extract on liver and skin cholesterol transporters in high fat diet-induced obese mice

  • Obesity is an epidemic disease characterized by an increased inflammatory state and chronic oxidative stress with high levels of pro-inflammatory cytokines and lipid peroxidation. Moreover, obesity alters cholesterol metabolism with increases in low-density lipoprotein (LDL) cholesterols and triglycerides and decreases in high-density lipoprotein (HDL) cholesterols. It has been shown that mulberry leaf and fruit ameliorated hyperglycemic and hyperlipidemic conditions in obese and diabetic subjects. We hypothesized that supplementation with mulberry leaf combined with mulberry fruit (MLFE) ameliorate cholesterol transfer proteins accompanied by reduction of oxidative stress in the high fat diet induced obesity. Mice were fed control diet (CON) or high fat diet (HF) for 9 weeks. After obesity was induced, the mice were administered either the HF or the HF with combination of equal amount of mulberry leaf and fruit extract (MLFE) at 500mg/kg/day by gavage for 12 weeks. MLFE treatment ameliorated HF induced oxidative stress demonstrated by 4-hydroxynonenal (4-HNE) and modulated the expression of 2 key proteins involved in cholesterol transfer such as scavenger receptor class B type 1 (SR-B1) and ATP-binding cassette transporter A1 (ABCA1) in the HF treated animals. This effect was mainly noted in liver tissue rather than in cutaneous tissue. Collectively, this study demonstrated that MLFE treatment has beneficial effects on the modulation of high fat diet-induced oxidative stress and on the regulation of cholesterol transporters. These results suggest that MLFE might be a beneficial substance for conventional therapies to treat obesity and its complications.

Dietary Supplementation of Mushroom Water Suppresses Fat Accumulation in High Fat Diet Induced-Obese Female Mice and Enhances Immune Cell Development in Non-Obese Mice

  • Extracted mushroom water showed an ability to suppress the accumulation of body fat in female mice after feeding 5 weeks with high fat diet. Particularly, in parametrial and mesenteric adipose, it significantly reduced 44 and 47% of weight, respectively. In non-obese mice, maturated NK cell ($CD11b^{hi}CD27^{lo}$) population were increased ($70.9{\pm}3.8%$) in mushroom water fed mice compared to control ($61.4{\pm}4.3%$) and NK cell population were augmented in mushroom fed mice compared to control.

삼황탕(三黃湯)이 고지방식이, MSG 및 난소적출로 유발된 흰쥐의 비만에 미치는 영향 (Effects of Samhwang-tang(Sanhuang-tang) on Obese Rats Induced by High Fat Diet, MSG and Ovariectomy)

  • Objectives : Obesity is a metabolic disease resulted from various causes including nutritional disproportion, hormonal imbalance, and hypothalamic nuclei disorder. Samhwang-tang(Sanhuang-tang, SHT) is composed of 5 herbs (Ephedrae Herba, Scutellariae Radix, Typhae pollen, Acori Rhizoma and Ansu Semen) and is frequently used for obese patients. This study investigated anti-obesity effects of SHT on the rats. Methods : Obesity was induced by high fat diet (HFD), monosodium L-glutamate injection (MSG) and ovariectomy (OVX) in the Sprague-Dawley rats. Water extract of SHT was treated for 2 weeks, then changes of body weight gain and epididymal fat cell size were measured. In OVX animals, c-Fos and neuropeptide Y (NPY) expressions in hypothalamic nuclei were observed with immunohistochemistry. Results : SHT treatment demonstrated significant decrease of body weight gain in obesity induced by HFD and OVX, but not in obesity induced by MSG. SHT treatment demonstrated significant decrease of fat cell size in obesity induced by HFD, but not in obesity induced by MSG. On c-Fos and NPY expressions in hypothalamic nuclei, SHT treatment resulted significant decrease of immuno-reactivity of c-Fos and NPY in obesity induced by OVX. Conclusions : These results suggest that SHT has a weight loss effect against obesity induced by nutritional disproportion of diet intake and hormonal imbalance, especially without central nervous system disorder.
