Differentiation in Nitrogen-Converting Activity and Microbial Community Structure between Granular Size Fractions in a Continuous Autotrophic Nitrogen Removal Reactor

  • The differentiations in nitrogen-converting activity and microbial community structure between granular size fractions in a continuous completely autotrophic nitrogen removal over nitrite (CANON) reactor, having a superior specific nitrogen removal rate of $0.24g/(g\;VSS{\cdot}h)$, were investigated by batch tests and high-throughput pyrosequencing analysis, respectively. Results revealed that a high dissolved oxygen concentration (>1.8 mg/l) could result in efficient nitrite accumulation with small granules (0.2-0.6 mm in diameter), because aerobic ammonium-oxidizing bacteria (genus Nitrosomonas) predominated therein. Meanwhile, intermediate size granules (1.4-2.0 mm in diameter) showed the highest nitrogen removal activity of $40.4mg/(g\;VSS{\cdot}h)$ under sufficient oxygen supply, corresponding to the relative abundance ratio of aerobic to anaerobic ammonium-oxidizing bacteria (genus Candidatus Kuenenia) of 5.7. Additionally, a dual substrate competition for oxygen and nitrite would be considered as the main mechanism for repression of nitrite-oxidizing bacteria, and the few Nitrospira spp. did not remarkably affect the overall performance of the reactor. Because all the granular size fractions could accomplish the CANON process independently under oxygen limiting conditions, maintaining a diversity of granular size would facilitate the stability of the suspended growth CANON system.

  • The characteristics of Lake Cheonhoji water and sediment were investigated in oder to utilize these as fundamental materials for the management of lake water quality. The hydrographic properties of Lake Cheonhoji which are relatively low chance of nutrients loading from the watershed and a long retention time of lake water, lead to the probability of high lake productivity. It was also observed that lake water showed stratification during summer and complete mixing during fall, even though water depth was relatively shallow. The trophic state was eutrophic to hypertrophic from summer to late fall. The overall properties of the sediment were oligohumic, high ignition loss and high composition of NAIP and Resid.-P, which might serve as potential pollution sources of lake water quality. In laboratory scale experiments, it was observed that leaching potential of nutrients in the sediment was greatly dependant upon water temperature and dissolved oxygen. Finally, water pollution in Lake Cheonhoji was considered to be largely due to the adverse cycle of uncontrollable eutrophication, which resulted in the subsequent occurrence of dead algae and animal plankton, organic sedimentation, reduction of dissolved oxygen and nutrients leaching, which again reinforced the cycle of eutrophication in the lake.

Candida tropicalis DS-72에 의한 Xylose로부터 Xylitol의 생산

  • A high xylitol producing yeast was isolated from the sludge of xylose manufacturing factory and then identified as Candida tropicalis DS-72 according to physiological properties. The strain was able to produce xylitol in a high concentration up to 72g/l from 100g/l xylose in 32 hours. Medium optimization for xylitol production by C. tropicalis DS-72 was performed. Effect of various nitrogen sources on xylitol production was investigated. Of nitrogenous compounds, yeast extract was the most suitable organic nitrogen nutrient for the enhancement of xylitol production. However, inorganic nitrogen resulted in a low cell concentration and did not produce xylitol. Effect of inorganic salts such as KH$_{2}$PO$_{4}$, and MgSO$_{4}$, 7H$_{2}$O on xylitol production was also studied. Optimal medium was selected as xylose 100g/l, yeast extract 10g/l, KH$_{2}$PO$_{4}$, 5 g/l and MgSO$_{4}$, 7H$_{2}$O 0.2 g/l. Xylitol of 88 g/l was produced from 100 g/l xylose in 30 hours using the optimal medium in a flask. In a fermentor, a fed-batch culture with 300g/l xylose was carried out. A final xylitol concentration of 240 g/l in the culture could be obtained in 43 hours of culture time by maintaining the high level of dissolved oxygen during growth phase and limiting the dissolved oxygen in the same culture during production phase. This result corresponded to a xylitol yield of 80% and a xylitol productivity of 5.58 g/1-h.

Variation of Phosphorus Concentration and Redox Potential in a Paddy Field Plot During Growing Season (영농기 필지논에서의 인 (P) 농도와 산화환원전위 (Eh)의 변화 특성)

  • The purpose of this study is to investigate characteristics of total phosphorus (TP) and phosphate phosphorous ($PO_4$-P) concentrations in ponded water and redox potential (Eh) in paddy soil during the growing season. The TP and $PO_4$-P concentrations showed twice peak values after basal dressing and tillering fertilization. The ratio of $PO_4$-P to TP showed low values (0.07~0.18), indicating that most of phosphorus is particlulate. The $PO_4$-P concentrations significantly decreased with dissolved oxygen (DO) concentrations. The Eh showed high values (179~636 mV) under non-ponded aerobic condition, but low values (74~112 mV) under ponded anaerobic condition The TP and $PO_4$-P concentrations in ponded water increased shortly after tillering fertilization even if phosphorus was not applied. This may be due to the release of dissolved phosphorus from the bottom sediment and its associated algal and water flea blooms under anaerobic condition. Therefore, proper water management should be needed shortly after tillering fertilization.

Inhibition of Pitting Corrosion Failure of Copper Tubes in Wet Sprinkler Systems (스프링클러 구리배관의 공식 파손 억제)

  • The inhibition of pitting corrosion failure of copper sprinkler tubes in wet sprinkler systems was studied. First, an apparatus and technology for removing air in the sprinkler tubes by vacuum pumping and then filling the tubes with water were developed. Using this apparatus and technology, three methods for inhibiting the pitting corrosion of the copper sprinkler tubes installed in several apartment complexes were tested. The first one was filling the sprinkler tubes with water bubbled by high-pressure nitrogen gas to reduce the dissolved oxygen concentration to lower than 2 ppm. In the second method, the dissolved oxygen concentration of water was further reduced to lower than 0.01 ppm by sodium sulfite. In the third method, the sprinkler tubes were filled with benzotriazole (BTAH) dissolved in water. The third method was the most effective, reducing the failure frequency of the sprinkler tubes due to pitting corrosion by more than 80%. X-ray photoelectron spectroscopy analyses confirmed that a Cu-BTA layer was well coated on the inside surface of the corrosion pit, protecting it from corrosion. A potentiodynamic polarization test showed that BTAH should be very effective in reducing the corrosion rate of copper in the acidic environment of the corrosion pit.

Distribution of Dissolved Organic Carbon (DOC) in the Southwestern East Sea in Summer

  • In the summer of 2008 (August 4-14), vertical and horizontal distributions of inorganic nutrients and dissolved organic carbon (DOC) were measured in the southwestern East Sea. Concentrations of DOC were determined for the first time in the southwestern East Sea using the high-temperature combustion oxidation (HTCO) method, and results were compared with those measured by another laboratory. Concentrations of DOC ranged from 58 to 104 ${\mu}M$ in the upper 200 m, showing a typical decreasing pattern with depth. Generally, concentrations of DOC were relatively lower, with higher nutrient concentrations, in the upper layer of the coastal upwelling zone. Concentrations of DOC ranged from 54 to 64 ${\mu}M$ in the deep Ulleung Basin (200-1500 m), and were higher than those in the Pacific and Atlantic oceans. In association with rapid vertical ventilation of the euphotic, this difference indicates a larger accumulation of semi-labile DOC in the deep East Sea than in the major oceans. A correlation between apparent oxygen utilization (AOU) and DOC in the deep ocean of the East Sea revealed that only a small portion (<10%) of the sinking DOC, relative to the sinking particulate organic carbon (POC), contributes to microbial degradation. Our results present an important data set of DOC in the East Sea, which plays a critical role in carbon cycle modeling and sequestration.

  • An advanced mobile technology is described for saturating water with oxygen. The MTR(Mass Transfer Reactor) is 2~10 times more effective in dissolving oxygen in water compared to most other systems used in aquaculture, because it can generate extremely small(0.005~0.05mm) bubbles. New fish farming facilities could make use of this technology to build deeper ponds for raising multiple species with optimized conditions for DO(dissolved oxygen), food, light, etc. The proposed technology offers higher DO levels with minimal operating costs. It is easy to use and maintain, with a high reproducibility. Accordingly, the MTR can be industrially applied in the treatment of fish waste and reduction of water consumption during fish farming.

The Effect of Dissolved Oxygen on the Estival Mass Mortality of sea squirt, Halocynthia roretzi(Drasche) (여름철 우렁쉥이 대량폐사에 대한 용존산소의 영향)

  • Mass mortality of Halocynthia roretzi has been occurred in culturing grounds of southern coast of Korea during the long period of summer drought and the unusal high water temperature in 1988. Especially Halocynthia roretzi was damaged severely in 2 years group. Therefore we verified these phenomena by observing the environmental factors of culturing grounds and by evaluating oxygen consumption rate in indoor experiment. In culturing grounds, the mortality rate was high in Namhae Mizo area where water temperature was the highest and the lowest in dissolved oxygen concentration as low as 3.76m/l in field observation. In indoor experiment, the higher the water temperature, the more mortality was occurred and the salinity above 30 had no effect on its mortality. In oxygen consumption rate experiment, the smaller needs more oxygen than larger one, i. e., oxygen demand per unit weight was higher in 2 years group than that of 3 years. The theoretical and the actual minimum oxygen demands(MOB) for preventing asphixation was 3.7ml/kg/h at $20^{\circ}C$ and 6.3ml/kg/h gat $25^{\circ}C$, But actual MOD was 4.3ml/kg/h at $25^{\circ}C$ in indoor experiment. It seemed that mass mortality of Halocynthia roretzi in summer was caused by metabolism disorder due to insufficient oxygen level influenced by the high water temperature and the high density of Halocynthia roretzi, which results in the paucity of oxygen especially in 2 years groups.

  • To study photocatalytic mechanism of metal doped $Ti0_2$, we investigated photodecomposition effect, photocurrent effect and antibacterial effect. When aluminium content was 2 wt %, photodecomposition effect was better than the others. Silver doped thin films had high photocurrent efficiency and antibacterial effect. This reactions were caused by dissolved oxygen in solution and oxygen adsorbed on surface of thin films.

Phosphorus removal by lime-natural mineral dissolved solutions

  • In previous studies, solely ferric (Fe3+) and calcium (Ca2+) ions were commonly used for removal of PO4-P (considered as T-P in this study) in wastewater via chemical precipitation. Herein, the removal of total phosphorus (T-P) in wastewater was performed using various mineral and lime dissolved solutions. The dissolution kinetics of different minerals (feldspar, olivine, elvan, illite, sericite, and zeolite) and lime was compared and used their solutions for T-P removal of real wastewater. The highest T-P removal (almost 90%) was obtained by the lime dissolved solution and followed by zeolite, illite, feldspar, and others. We observed a significant co-relationship (R of 0.96) between the amount of initial Ca2+ and T-P removal. This was induced by formation of hydroxyapatite-like mineral via Ca-P precipitation reaction at high pH solution. Furthermore, additional removal of suspended solid (SS) and chemical oxygen demand (COD) was achieved by only lime dissolved solution. Finally, the lime-feldspar dissolved solutions were prepared at different ratios (10-50%), which showed a successive T-P removal up to two times by samples of 40 and 50%.
