Fed Batch Culture of Hybridoma for Reduction of Lactate and Higher Monoclonal Antibody Productivity (젖산 생성 감소 및 단일군항체 생산성 향상을 위한 하이브리도마의 유가배양)

  • Accumulation of toxic waste byproducts in hybridoma culture can limit cell growth and monoclonal antibody (MAb) productivity, and one of the major toxic metabolites is lactate produced via glycolytic pathway of glucose metabolism. The factors affecting the glucose to lactate conversion rate were investigated. The conditions of high initial glucose concentration and high growth rate stimulated glucose to lactate conversion rate. The glucose-controlled fed-batch culture was investigated, and 19% reduction in lactate formation and 41% enhancement of MAb titer could be achieved by fed-batch culture.

Uncertainty Avoidance, Individualism and the Readiness of Business-to-Consumer E-commerce

  • This study aims to test whether and to what extend national culture affects the readiness of business-to-consumer (B2C) e-commerce across the world. We regress two sets of data at national level: (1) the ratings of two main dimensions of Hofstede's national culture framework, uncertainty avoidance and individualism, and (2) UTCTAD B2C e-commerce index value. A sample of 83 countries was tested in this study. Several control variables at national level are included in our regression model. We find that these two cultural dimensions have an effect on the readiness of B2C e-commerce in various ways. We find that countries with high individualism score have high index value of B2C e-commerce development. We discuss the implications of these findings for B2C e-commerce developing strategies. We then call for designing relative policies with full consideration on national culture to promote the development of B2C e-commerce. In addition, we identify the limitations of the study and propose recommendations for future research. This study is the first one to use UNCTAD data on B2C e-commerce to explore the relationships between two dimensions of Hofstede's national culture and the readiness of B2C e-commerce and adds to the knowledge of literature in this research field.

An Ethnographic Study on CosPlay Group in Korea II - Analysis on CosPlay Culture in Korea and Japan - (한국 코스프레 집단의 문화기술지적 연구 II - 한국과 일본의 코스프레 문화에 대한 비교 분석 -)

  • This study examines the social meaning of the CosPlay, the growth potential of CosPlay culture and its effect on the related industry through the perspectives and language of the youths who enjoy CosPlay, based on the ethnographic research. Also, this study presents a comparative description of Korea and Japan CosPlay culture by the ethnographic methodology whose purpose is to define relationship of cause and effect with phenomenon. For further step, this study plans to emphasize the need to link culture, clothes and related industry in order to create a cultural environment where diversity co-exists. CosPlay is the mania culture of Japan that emulated the Halloween party of the West and that developed the party into a unique form. In Korea, this practice was accepted for the first time among a handful of youths, starting from the 1990s, after which, it was introduced to the masses while holding CosPlay related events. While CosPlay is succeeded as an industry in Japan, CosPlay in Korea is considered childish play due to the Korean culture of considering cartoon as a childish and low class genre which is enjoyed by youths. CosPlay in Korea faces the following changes: aging of the members who comprised the CosPlay culture at the initial stage; population increase, centered on middle and high school students; interest of the government and the businesses that wish to produce economic wealth by organizing CosPlay events into events for youths; and changes in the environment that comprised the surrounding of the CosPlay culture. CosPlay is an honest play that demonstrates one's effort on the stage through performance. Moreover, most of the middle and high school students who comprise the CosPlay culture demonstrate similar characteristics as mania type of people when it came to the reason that they enjoy CosPlay. However, they did not consider CosPlay culture as an important aspect of their lives. Instead, most of them said that they participate to relieve stress. Thus, they have the potential to move onto another form of youth culture that may appear more attractive to them. To them, it is not the CosPlay culture that is important, but the fact that CosPlay provides a forum where they can freely engage in play.

The Relationship Between 7S Factors of the Nursing Organizational Culture and Organizational Effectiveness (간호 조직문화 7S 요인과 조직 유효성의 관계)

  • Purpose: The Purpose of this study was to identify the relationship between 7S factors of the nursing organizational culture and organizational effectiveness. Method: The data were gathered from the self-reported questionnaires of 717 nurses who work for eight different general hospitals located around Seoul and Kyounggi province. The period of data collection was from November 12 to December 7, 2002. For data analysis, descriptive statistics, clustering analysis, and t-test with SPSS Program were used. Result: The nurses who highly perceived 7S factors of nursing organizational culture showed higher job satisfaction and organizational commitment in comparison with the nurses who lowly perceived 7S factors of nursing organizational culture. And the nurses who highly perceived 7S factors of nursing organizational culture showed higher organizational citizenship behavior in comparison with the nurses who lowly perceived 7S factors of nursing organizational culture. Among subdimension of organizational citizenship behavior, altruism and civic virtue were significant. Conclusion: From the above results, the high group with 7S factors of nursing organizational culture has strong culture, therefore nursing organization with strong culture is very implicative to enhance the organizational effectiveness.

Controlling Mammalian Cell Metabolism in Bioreactors

  • Animal cells in culture typically convert most of the glucose they consume into lactate. The accumulation of lactate, however, is commonly cited as one of the factors that inhibit cell growth and limit the maximum cell concentration that can be achieved in culture. The specific production of lactate and the amount of glucose converted to lactate can be reduced when cells are grown in a fed-batch culture in which the residual glucose concentration is maintained at low levels. Such a fed-batch culture was used to grow and adapt hybridoma cells into a low-lactate-producing state before changing into continuous culture. The cells reached and maintained a high viable cell concentration at steady state. In a similar manner, cells that were initially grown in batch culture and a glucose-rich environment reached a steady state with a cell concentration that is much lower. The feed composition and dilution rates for both cultures were similar, suggesting steady state multiplicity. From a processing perspective the desired steady state among those is the one with the least metabolite production. At such seady state nutrient concentration in the feed can be further increased to increase cell and product concentrations without causing the metabolite inhibitory effect typically seen in a cell culture. Controlling cell metabolism in a continuous culture to reduce or eliminate waste metabolite production may significantly improve the productivity of mammalian cell culture processes.

Increased SOX2 expression in three-dimensional sphere culture of dental pulp stem cells

  • Mesenchymal stem cells in the dental pulp exhibit a tendency for differentiation into various dental lineages and hold great potential as a major conduit for regenerative treatment in dentistry. Although they can be readily isolated from teeth, the exact characteristics of these stem cells have not been fully understood so far. When compared to two-dimensional (2D) cultures, three-dimensional (3D) cultures have the advantage of enriching the stem cell population. Hence, 3D-organoid culture and 3D-sphere culture were applied to dental pulp cells in the current study. Although the establishment of the organoid culture proved unsuccessful, the 3D-sphere culture readily initiated the stable generation of cell aggregates, which continued to grow and could be passaged to the second round. Interestingly, a significant increase in SOX2 expression was detected in the 3D-spheroid culture compared to the 2D culture. These results indicate the enrichment of the stemness-high population in the 3D-sphere culture. Thus, 3D-sphere culture may act as a link between the conventional and 3D-organoid cultures and aid in understanding the characteristics of dental pulp stem cells.

Analysis of School Foodservice Managers' Job Satisfaction by Organizational Culture and Commitment in Jeju (제주지역 학교급식 관리자의 조직문화 및 조직몰입에 따른 직무만족도 분석)

  • The purpose of this study was to examine the job satisfaction of foodservice managers at schools in Jeju, Korea, according to the organizational culture and commitment of the school foodservice and administrative departments. The subjects included 144 (98.6%) dieticians from Jeju schools. The data were analyzed by descriptive analysis, reliability analysis, factor analysis, ttests, ANOVA, and Pearson's correlation coefficients, using the SPSS Win 12.0 program. In terms of organizational culture, the dieticians perceived the 'human relations model' and 'internal process model' as the strongest types for the school foodservice departments and administrative departments, respectively. However, for both types, a gap existed between the organizational culture that was perceived and that which was expected. In particular, the subjects expected that the 'human relations model' and 'open systems model' were the most important organizational culture types. In terms of the subjects' organizational commitment scores, loyalty and pride scored highest whereas unity scored lowest. For the level of job satisfaction, the performed work itself scored highest and was deemed most important. Additionally, organizational culture was positively correlated to the factors affecting organizational commitment and job satisfaction. Finally, the surveyed managers had high demands for a human relations-oriented organizational culture to enhance job satisfaction.

Production and characterization of ultra-high-molecular weight poly(3-hydroxybutyrate) by recombinant Escherichia coli

  • An efficient fermentation strategy for the high level production of ultra-high-molecular weight poly(3-hdyroxybutyrate) (PHB) was developed. Although the cell and PHA concentrations obtained by flask cultures at different initial pH (6.0 or 6.9) were almost same level, the molecular mass of PHB produced were quite different along with the initial pH. When a recombinant Escherichia coli XL1-Blue harboring pJC2 containing the Alcaligenes latus PHB biosynthesis genes was cultivated in flask culture (pH 6.0), the PHB having a very high molecular weight of 22 MDa could be produced while only below 1 MDa at initial pH 6.9. The ultra-high-molecular weight PHB could be synthesized to high concentration of 89.8 g/L resulting in the PHB productivity of 2.07 g/L-h by simple fed-batch culture. In this study, we report that PHB having various molecular mass can be produced by employing metabolically engineered E. coli strains harboring the plasmids of different copy numbers containing the A. latus phbCAB genes.

Self-concept of High School Girls in Relation to Their Clothing Selection Behavior

  • This study aims to figure out the levels of self-concept, to reveal the aspects of clothing selection behavior, and to clarify the relation of self-concept to clothing selection behavior in high school girls. Questionnaire was used to collect data. The subjects were made up of 298 second-grade high school girls from four parts in Taegu. The findings of this study are as follows : 1. The level of physical self, personal self, family self, and social self of high school girls are shown as above average. 2. In general trend of their clothing selection behavior, the scores related to practicality, economy, exhibitionism except fashionability are shown as high. 3. It seems that high school girls have a tendency to firstly weigh exhibitionism, and then economy, practicality, fashionability are followed one after another. 4. Physical self is shown as significantly different in fashionability, exhibitionism among clothing selection behavior.

Multiplexer as selector to select different speed (Normal speed, High speed and Super high speed) to display CAR at different speed to color TV system

  • The article presents a concept of designing a Multiplexer circuit which acts as a "Selector" and that becomes capable to select different speed created at different TTL Gate configurations; Standard TTL(Normal Speed), High Speed TTL(High Speed), Schottky TTL(Super High Speed) and further connect the selected Gate speed to the CAR shape created using C-Programming at Computer Graphics and finally CAR shape display at different speed to the color TV. The Multiplexer supporting efficient and more reliable selection criteria using "Logical based selection criteria" and further the output from multiplexer is provided to CAR shape created using c-programming and finally CAR shape is display to color TV system. Basic purposes and assumptions regarding the design and development of this system as well as a description of its operation have been presented.
