  • A study was carried out to investigate the effects of safflower seed powder on the improvement of lipid metabolism in high fat and high cholesterol fed rats. Male Sprague Dawley rats of 10 weeks old, weighing 325$\pm$5g, were divided into two groups; the control group(C group, high fat(10% lard) and high cholesterol(1% cholesterol)) and safflower seed group(S group, 10% safflower seed powder), they were fed experimental diets for 6 weeks. Food intake, body weight gains and organ weight had little differences between the groups. Concentration of lipoprotein in serum was remarkably lower in S group than in C group. Serum cholesterol levels were significantly lower in S group(72.94$\pm$4.08 mg/dl) than in C group(89.41$\pm$4.19mg/dl). The level of serum HDL cholesterol was higher in S group than in C group. The level of serum LDL C was significantly lower in S group than in C group. The ratio of HDL cholesterol to total cholesterol were higher in the S group than in the C group, too. The value of atherogenic index(AI) was determined to be low in S group. The content of liver triglyceride and cholesterol in the S group was lower than that of C group. ACAT activities which involves in cholesterol esterification in liver, was not significantly different between two groups.

Effects of Jebikong (Dolichos lablab) Extract on Serum Lipid Metabolism in Rats Fed a High Fat Diet (제비콩 추출물이 고지방 식이 급여에 의한 흰쥐의 혈청 지질 함량에 미치는 영향)

  • This study investigated the effect of Dolichos lablab extract on the lipid metabolism of rats fed a high fat and cholesterol diet. S-D male rats (n=48) were given 40, 400, or 4000 mg/kg of Dolichos lablab extract and fed a high fat and high cholesterol diet for four weeks (DL-HFC group). As a result, serum levels of total lipids, total cholesterol, and triglycerides were higher in the high fat and high cholesterol group (HFC-group) than the control, and significant differences were observed between the control and the DL-HFC (40, 400, 4000 mg/kg) group. The fat absorption rate increased with a high fat diet feeding, but was higher in the DL-HFC (40, 400, 4000 mg/kg) group.

Effect of Lotus Root(Nelumbo nucifera G.) on Lipid Metabolism in Rats with Diet - Induced Hypercholesterolemia (연근이 고콜레스테롤혈증 흰쥐의 지질대사에 미치는 영향)

  • This study was performed to investigate the effects of lotus root ethanol extinct (LRE) on 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase activity in vitro, and lipid metabolism in the serum and liver of rate fed normal or high cholesterol diet in vivo. LRE (200 mg/kg/day and 400 mg/kg/day) was administered only to rats with fed high cholesterol diet for 6 week. We divided into 6 groups: normal diet group (NC), high cholesterol diet group (1% cholesterol and 0.25% sodium cholate)(HC), LRE 200 mg/kg treated group (NC-LREL), LRE 400 mg/kg teated group (NC-LREH), high cholesterol diet and LRE 200 mg/kg treated group (HC-LREL), and high cholesterol diet and LRE 400 mg/kg teated group (HC-LREH). LRE significantly inhibited the HMG-CoA reductase activity in a concentration-dependent manner in vitro. The body weight gain and liver weight of the high cholesterol diet group were higher than the normal diet group whereas the groups administered LRE were gradually decreased. The high cholesterol diet group increased serum triglyceride, total cholesterol, free cholesterol and LDL-cholesterol levels, and decreased atherogenic index, HDL-cholesterol and phospholipid levels as compared with the normal diet group. LRE administrated groups were increased in serum HDL-C/T-C, HDL-cholesterol and phospholipid levels, and decreased serum triglyceride, total cholesterol, free cholesterol, and LDL-cholesterol levels as compared with the high cholesterol diet group. These effect of LRE within the high cholesterol diet groups were concentration-dependent manners. There were no differences in the levels of serum triglyceride, total cholesterol, phopholipid, HDL-cholesterol and free cholesterol between normal diet groups. The hepatic LRE administrated groups than in the high cholesterol diet group. Teken together, it is suggested that LRE exerts hypocholesterolemic effect by reducing serum cholesterol concentration in rats with high cholesterol diet-induced hypercholesterolemia.

Preparation of Mulberry Leaf Extract by Adding Mugwort and Pine Needle and Effects on Lipid Composition in Rats Fed High Cholesterol Diets

  • This study investigated the effects of feeding mulberry leaf extracts on lipid composition in rats fed high cholesterol diets. An initial 30-person sensory evaluation of preparations containing various concentrations of mulberry leaf extract showed that a preparation containing 9% mulberry leaf extracts was the most highly preferred. In addition, subsidiary materials of pine needle extracts and mugwort extracts were added to weaken the unpleasant smell of mulberry leaf extract A preparation containing 9% mulberry leaf extract with 3% mugwort extract and 7% pine needle extract was given highest preference scores by the 30-person panel. When comparing the functional ingredients contents of the various preparations of mulberry leaf extracts, such as GABA, DNJ and flavonoids, no significant differences were found as a result of adding subsidiary materials (pine needle and mugwort extracts). Sprague-Dawley male rats weighing l00$\pm$10g were randomly assigned to one normal diet group, and to four high cholesterol diet groups containing 1% cholesterol, to elucidate the functionality of the mulberry leaf extract The four high cholesterol diet groups were classified into: a mulberry leaf extract diet group free of subsidiary materials (EB group); a mulberry extract diet group with pine needle extracts (EP group); a mulberry leaf extract diet group with mugwort extracts (EM group); and a control group (HC group). The mulberry leaf extracts were provided as drinking water; the diet and water were fed ad libitum. Hepatic cholesterol and triglyceride levels were higher, by 279% to 475%, in the high cholesterol groups compared to the normal diet groups, but were significantly lower in the three groups supplied with mulberry leaf extracts, compared with the high cholesterol control. There were no changes in functionality of the mulberry leaf extract preparations due to the addition of subsidiary materials. In conclusion, preparations of mulberry leaf extracts were shown to improve lipid metabolism in rats fed a high cholesterol diet, by reducing hepatic and plasma triglyceride and cholesterol levels. Also human palatability of the mulberry leaf preparation was improved by adding subsidiary materials such as pine needle and mugwort extracts.

Effect of Salvia plebeia Water Extract on Antioxidant Activity and Lipid Composition of Rats Fed a High Fat-High Cholesterol Diet (배암차즈기 열수 추출물 첨가가 고지방-고콜레스테롤 식이를 급여한 흰쥐의 항산화 활성과 지질조성에 미치는 영향)

  • This study examined the antioxidant activity and lipid composition of rats fed a high fat-high cholesterol diet containing Salvia plebeia water extract. Three levels (50 mg/kg, 200 mg/kg, 500 mg/kg) of Salvia plebeia water extract were administered to seven weeks old male Sprague Dawley rats for four weeks. The activity of glutathione peroxidase in serum was high in all groups that received the Salvia plebeia water extract (p<0.05). The glutathione reductase activity was high (p<0.05) in groups that received 50 mg/kg and 500 mg/kg of Salvia plebeia water extract. When rats received Salvia plebeia water extract, the content of MDA (malondialdehyde) in the serum and liver was low in all groups. In addition, the serum total lipid, triglyceride, total cholesterol and LDL-cholesterol contents decreased significantly (p<0.05). Total cholesterol and triglyceride levels in the liver were significantly lower in the group that received the Salvia plebeia water extract than in the control group (p<0.05). Salvia plebeia water extract improves lipid metabolism and it almost meets with the increase of antioxidant activity and peroxide formation reduction. Based on these results, supplemented of a high fat-hypercholesterol diet with Salvia plebeia can increase antioxidant activity and depress peroxide formation in serum, as well as liver cholesterol and triglycerides.

Effects of Iksugangjiryung on Hyperlipidemia in Rats (익수강지령(益壽降脂靈)이 흰쥐의 고지혈증 병태에 미치는 영향)

  • This study was designed to investigate the hyperlipidemia-healing effect of water extract from Iksukangjiryung, it was performed on the hyperlipidemia of rats induced by Triton WR-1339 and high cholesterol diet. We prepared two types of hyperlipidemia model in rats induced by Triton WR-1339 and high cholesterol diet. The amount of total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, lipid peroxide, glutathione and the activity of superoxide dismutase(SOD) were measured. The water extract from Iksukangjiryung showed significant decreasing effects on total cholesterol, LDL- cholesterol, triglyceride, lipid peroxide in the both model. And it showed significant increasing effects on the activity of SOD in both model and amount of glutathione in Triton WR-1339 model. These results suggest that water extract from Iksukangjiryung has healing efficacy on hyperlipidemia induced by Triton WR-1339 and high cholesterol diet.

  • This study was performed to examine the effects of Liriopis Tuber extracts to high cholesterol diet on lipid metabolism in rats. Male Sprague-Dawley rats of $90{\pm}10\;g$ were divided into six groups; basal diet (normal group), 1% high cholesterol diet (control group), high cholesterol diet supplemented with 3%, 5% water extracts (C3W, C5W group) and high cholesterol diet supplemented with 3%, 5% MeOH extracts (C3M, C5M group). These experimental diets were fed for 4 weeks. The lipid contents on serum, liver, feces were lower Lriopis Tuber extracts groups than control group. Especially, the effects of water extract was better than MeOH extract.

Effect of Meristotheca papulosa on Lipid Concentration of Serum and Liver in Rats Fed High Fat Diet (해조류 갈래곰보가 고지방 식이를 급여한 흰쥐의 혈액 및 간장 지질 농도에 미치는 영향)

  • This study was carried out to investigate the effect of Meristotheca papulosa on lipid concentration of serum and liver in rats fed high fat diet for 4 weeks. Experimental groups ere divided into normal group (5% corn oil), high fat diet group (5% corn oil+20% lard), high fat diet and 5% or 10% Meristotheca papulosa treated group. The weights of kidney, heart and tests were not significantly different among the group, but weight of liver was significantly decreased by supplement of Meristotheca papulosa (10%). The contents of serum total lipid, trigly-ceride, total cholesterol, free cholesterol, cholesterol ester and atherogenic index were decreased, while HDL-cholesterol was increased by Meristotheca papulosa supplementation compared to high fat diet group. Meristotheca papulosa supplement has obvious effects on lowering hepatic total lipid, triglyceride and total cholesterol. Our findings suggest that Meristotheca papulosa supplement can improve lipid metabolism in high fat fed rats.

The effects of Brassica juncea L. leaf extract on obesity and lipid profiles of rats fed a high-fat/high-cholesterol diet

  • BACKGROUND/OBJECTIVES: Obesity is a global health problem of significant importance which increases mortality. In place of anti-obesity drugs, natural products are being developed as alternative therapeutic materials. In this study, we investigated the effect of Brassica juncea L. leaf extract (BLE) on fat deposition and lipid profiles in high-fat, high-cholesterol diet (HFC)-induced obese rats. MATERIALS/METHODS: Male Sprague-Dawley rats were divided into four groups (n = 8 per group) according to diet: normal diet group (ND), high-fat/high-cholesterol diet group (HFC), HFC with 3% BLE diet group (HFC-A1), and HFC with 5% BLE diet group (HFC-A2). Each group was fed for 6 weeks. Rat body and adipose tissue weights, serum biochemical parameters, and tissue lipid contents were determined. The expression levels of mRNA and proteins involved in lipid and cholesterol metabolism were determined by reverse transcription polymerase chain reaction and western blot analysis, respectively. RESULTS: The HFC-A2 group showed significantly lower body weight gain and food efficiency ratio than the HFC group. BLE supplementation caused mesenteric, epididymal, and total adipose tissue weights to decrease. The serum levels of triglyceride, total cholesterol, and low-density lipoprotein cholesterol were significantly reduced, and high-density lipoprotein cholesterol was significantly increased in rats fed BLE. These results were related to lower glucose-6-phosphate dehydrogenase, acetyl-coA carboxylase, and fatty acid synthase mRNA expression, and to higher expression of the cholesterol $7{\alpha}$-hydroxylase and low density lipoprotein-receptor, as well as increased protein levels of peroxisome proliferator-activated receptor ${\alpha}$. Histological analysis of the liver revealed decreased lipid droplets in HFC rats treated with BLE. CONCLUSIONS: Supplementation of HFC with 3% or 5% BLE inhibited body fat accumulation, improved lipid profiles, and modulated lipogenesis- and cholesterol metabolism-related gene and protein expression.

Influence of Panax notoginseng on the Atherosclerosis Induced by High-cholesterol Feed in Rats (고콜레스테롤 식이로 유발된 동맥경화병태흰쥐의 혈관조직내 지질과산화 및 산화스트레스에 대한 삼칠근의 영향)

  • Panax notoginseng exhibit several beneficial effects including anti-oxidant effects. P. notoginseng is used as a therapeutic agent to stop haemorrhages and a tonic to promoted health in Korean and Chinese medicine. The pharmacokinetic profiles of the main P. notoginseng are still not accurately investigated. The exact mechanism of the anti-oxidant activitys of water extracts of P. notoginseng, however, has not been determined. in present study, I examined the effects of water extracts of P. notoginseng on high cholesterol diet atherosclerosis-induced rats in serum and abdominal aorta. A total of 3-week old 9 male rats of Sprague-Dawley were divided into 3 groups and fed with the basal diet (normal group), high cholesterol diet (atherosclerosis induced group) for 8 weeks, high cholesterol diet supplemented with water extracts of P. notoginseng (P. notoginseng group) for 4 weeks. And rats were sacrificed, serum lipid level, abodominal aortic anti-oxidant activities and lipid peroxide were measured. These results indicated that serum total cholesterl, LDL-cholesterol, triglycerides concentration significently lowered in P. notoginseng group than high cholesterol diet group. But HDL-cholesterol concentraion significently higher in P. notoginseng group than high cholesterol feed group. And abdominal aortic xanthine oxidase activity was significantly reduced by dietary water extracts of P. notoginseng supplementation (p<0.05) Also abdominal aortic superoxide dismutase, catalase, glutathione peroxidase activities were significantly increased by dietary water extracts of P. notoginseng supplementation (p<0.05) Especially, abdominal aortic level of lipid peroxide tended to increase in high cholesterol feed group, but water extract of P. notoginseng intake reduced the value (p<0.05).
