n-Hexane에 의한 삼백초 추출물의 항균 및 항바이러스 활성 (Antimicrobial and antiviral activity of Saururus chinensis extract by n-Hexane)

  • This study was conducted to investigate the antimicrobial, antivirus properties of Saururus chinensis extracts. The n-hexane extracts from Saururus chinensis showed the active antimicrobial activity against gram-positive bacteria. Minimum inhibitory concentration (MIC) of Saururus chiensis n-hexane extracts was 1.25 mg/ml against B. subtilis and 2.5 mg/ml against S. aureus. The cytotoxicity effects on MDBK (Madin-Darby bovine kidney) cell were observed at the various n-hexane extract concentrations. In $TCID_{50}$ assay, 0.6 mg/ml of n-hexane extracts decreased BVD (bovine viral diarrhea) virus by 1.4 log, whereas other extracts did not show antiviral activity. In this study, The results suggested that n-hexane extracts and fractions of Saururus chinensis can be a candidate materal of feed additive to chemical antibiotics and antivirus substances.

Effect of Saliva miltiorrhiza Bunge on Antimicrobial Activity and Resistant Gene Regulation against Methicillin-Resistant Staphylococcus aureus (MRSA)

  • This study was conducted in an effort to evaluate the antimicrobial activity and antibiotic-resistant gene regulation from Saliva miltiorrhiza Bunge on methicillin-resistant Staphylococcus aureus (MRSA). A variety of solvent fractions and methanol extracts of S. miltiorrhiza Bunge were tested in order to determine its antimicrobial activities against S. aureus and MRSA. As a result, the hexane fraction of S. miltiorrhiza Bunge evidenced the highest levels of antimicrobial activity against S. aureus and MRSA. The MICs of the hexane fraction against various MRSA specimens were $64. The hexane fraction evidenced inhibitory effects superior to those of the chloroform fraction. The results showed inhibition zones of hexane (16 mm) and chloroform (14 mm) fractions against MRSA KCCM 40511 at $1,000{\mu}g/disc$. The hexane and chloroform fractions inhibited the expression of the resistant genes, mecA, mecR1, and femA in mRNA. Moreover, the results of Western blotting assays indicated that the hexane and chloroform fractions inhibited the expression of the resistant protein, PBP2a. These results reveal that the hexane and chloroform fractions of S. miltiorrhiza Bunge may prove to be a valuable choice for studies targeted toward the development of new antimicrobial agents.

감잎 핵산획분의 항돌연변이 효과와 항돌연변이 물질의 GC-MS를 이용한 동정 (Antimutagenic Effects and Compounds Identified from Hexane Fraction of Persimmon Leaves)

  • 감잎의 메탄올 추출물이 Salmonella typhimurium TA100에서 aflatoxin B$_1$(AFB$_1$), DMAB, MNNG, 그리고 4-NQO의 돌연변이 유발성을 억제시키는 효과가 있었다. 메탄올 추출물을 다시 hexane, chloroform, ethyl acetate, butanol, 그리고 수용성층으로 분획하여 각 획분의 수득율과 항돌연변이 효과를 조사하였다. Hexane, butanol, 그리고 수용성 획분의 수득율이 높았으며, 이중 hexane 획분이 AFB$_1$, DMAB, MNNG, 그리고 4-NQO에 대한 Salmonella typhimurium TA100에서 항돌연변이효과가 가장 크게 나타났다. Hexane 획분을 silica gel column과 thin layer chromatography (TLC)법으로 연속분리하여 TLC상에서 8개의 bands로 분리하였다. 그중 항돌연변이 효과가 가장 컸던 band를 hexane/ethylacetate (1 : 1, v/v)로 추출한 다음 그중에 존재하는 화합물을 GC-MS를 이용하여 분리동정하였다. 활성획분에서는 1'-oxocannabinol, 3$\beta$-acetoxy-17-methyl-5$\alpha$-18 (13-17) abeoard-rost-13-ene, 4-methoxy-2'6'-dinitro-3,5-di-t-butylbiphenyl, 8, 9-dihydro-5,6-dimethoxy-dibenz[c,h]isoquino [2, 1, 8-1ma]carbazole-11, 16-dione 등이 분리동정되었다.

천화분 메탄올 추출물의 멜라닌 형성 억제 효과 (Inhibitory Effect of Methanolic Extract from Radix Trichosanthis on Melanin Synthesis)

  • Objectives: In order to investigate the relationship of Radix Trichosanthis components and the melanin synthesis, the author has analyzed the cell viability and tyrosinase activity, melanin content and morphologic changes in n-hexane, EtOAc, n-BuOH, and H2O fraction. Methods: At first, in order to determine the concentration of the Radix Trichosanthis component, the author investigated the viability of B16 melanoma cell. To measure the effects of Trichosanthes kirilowii extracts (n-BuOH, n-Hexane, EtOAc, H2O fractions) on the viability of A549 cells, A549 cells were treated with various concentrations (from 0.5 to $25{\;}\mu\textrm{g}/ml$) of components of Trichosanthes kirilowii. After 24hrs, the cell viability was measured by MTT assay. The EtOAc components of Trichosanthes kirilowii decreased the viability of A549 cells in a dose-dependent manner. H2O and n-BuOH components had no cell toxicity till $25{\;}\mu\textrm{g}/ml$, the n-hexane component showed minor cell toxicity at $25{\;}\mu\textrm{g}/ml$ and the EtOAc component cell toxicity was revealed at $5{\;}\mu\textrm{g}/ml$ concentration. Results: 1. The results of tyrosinase activity and the Radix Trichosanthis component; n-hexane and EtOAc components controlled it effectively; the n-BuOH components were less effective. 2. The results of melanin content analysis showed that the n-hexane and EtOAc components effectively inhibited, the n-BuOH fraction inhibited less, and H2O component didn't inhibit the terminal melanin formation. 3. In the n-BuOH and H2O component there were no changes, but in the n-hexane component the melanin content was effectively inhibited. 4. In the EtOAc fraction, although the melanin content was inhibited, the cell count was evidently suppressed, Of all of the Radix Trichosanthis components, the n-Hexane and EtOAc fractions inhibited the melanin synthesis best, but owing to its toxicity, the EtOAc components inhibited the cell count. Conclusion: The above results demonstrated that Radix Trichosanthis n-hexane fraction efficiently inhibited the tyrosinase activity and melanin synthesis.

고들빼기 잎추출물이 흰쥐의 사염화탄소에 의한 간손상에 미치는 영향 (Effects of Godulbaegi Leaf Extracts on $CCl_4$-Induced Hepatotoxicity in Rats)

  • 본 연구는 고들빼기를 분획별로 나누어 수층, butanol 층, hexane층, ethyl acetate층 분획 물들을 1주일간 흰 쥐의 복강내 주사 후 마지막날 사염화탄소를 복강내 1회 투여하여 간손상을 유발시킨 쥐의 간중량, 과산화지질, 항산화효소활성 및 간조직을 관찰하였다 체중에 대한 간중량 백분율은 간손상군중에서 n-hexane 분획물 투여군이 가장 낮게 나타났으며, 혈청 중 GPT활성 역시 n-hexane 분획물 투여군이 거의 정상군과 비슷한 수준으로 떨어 졌다. 간 중 MDA함량은 고들빼기 분획물 투여군들이 모두 낮은 경향이었으며 그 중에서도 n-butanol 분획물 투여군과 n-hexane 분획물 투여군이 대조군에 비해 유의적으로 낮았다 간 중 GST활성은 정상군에 비해 간손상군이 떨어졌으며 간손상군에서는 대조군에 비해 ethyl acetate 분획물 투여군이 유의적으로 높게 나타났다. 간조직 관찰에서는 정상군에 비 해 간손상군의 조직에 염증 및 괴사가 뚜렷이 나타났고 고들빼기 n-hexane 분획물 투여 후 간독성 유발 쥐의 간조직에서는 염증 및 괴사가 확실히 감소한 경향을 볼 수 있었다.

편백나무 건조 중 발생하는 휘발성 유기화합물의 항진균 및 항산화 활성 (Antifungal and Antioxidant Activities of Volatile Organic Compounds Generated During the Drying Process of Chamaecyparis obtuse)

  • 본 연구는 편백나무 건조 중 발생하는 휘발성 유기화합물로 구성된 응축수에 대해 항진균 및 항산화 활성을 평가하였다. 응축수는 hexane과 ethyl acetate로 각각 추출하였으며 추출액의 농도 100-500 mg/L에서 피부사상균에 대한 항진균 활성을 측정하였다. 추출액 농도가 증가할수록 항진균 효과도 증가하였으며 500 mg/L에서는 모든 피부사상균에 대해 높은 항진균 활성을 나타냈다. 추출용매에 따라서는 hexane으로 추출하였을 경우 높은 활성을 나타냈으며, 특히 Trichophyton rubrum에 대한 항진균 활성이 높았다. 항산화 활성을 측정한 결과 25 mg/L 이상의 농도에서는 90% 이상의 항산화 활성을 나타냈으며 추출용매에 따른 차이는 거의 없었다. 하지만 25 mg/L 이하의 농도에서는 hexane 추출물에서 높은 항산화 활성을 나타냈다. 각각의 추출물 구성성분을 분석한 결과 hexane 추출물의 주요 구성성분은 torreyol, alpha-cadinol, tau-cadinol이며, ethyl acetate 추출물의 주요 구성성분은 alpha-amorphene, alpha-cadinol, gamma-cadinene으로 나타났다.

사상자의 항염증작용 (Anti-inflammatory Activities of Torilis japonica Fruit)

  • The methanol extract of Torilis Fructus showed potent anti-edematous effect in carrageenan-induced paw edema in rats. Systematic fractionation of the extract with hexane, chloroform, ethyl acetate and butanol resulted in potent anti-edematous action only in hexane fraction at an oral dose of 237 mg/kg which is corresponding to 4.8 g of raw Fructus. This hexane fraction showed inhibitory effects at 77 mg/kg p.o on vascular permeability in mice, at 8mg/pouch on leucocyte emigration in rats, and at 120 mg/kg p.o. on adjuvant arthritis model in rats. These results demonstrate that hexane fraction of the extract possesses potent anti-inflammatory effect which may support its traditional uses.

Separation of Lipid-Soluble Component to Decrease Thermal Oxidation of Lard from Spinach (Spinacia oleracea)

  • Component having antioxidant activity on lard during heating was separated from hexane extract of spinach, and its characteristic chemical structure was speculated through nuclear magnetic resonance spectroscopy, liquid chromatography-mass spectrometry, and Fourier transform infrared spectrophotometry. Lard was heated with hexane-, ethyl ether-, ethyl acetate-, or ethanol extract of spinach at $180^{\circ}C$ for 20 hr. Hexane extract of spinach, having highest antioxidant activity on lard during heating, was fractionated by silicic acid column chromatography (SACC), and SACC fractions having higher antioxidant activity on lard during heating were further separated by thin layer chromatography (TLC). Isolated compound from SACC fractions of hexane extract of spinach by TLC had sugar moieties and benzene ring along with hydroxy, carbonyl, and alkyl groups in the structure.

공기중에 오염되어 있는 BHC의 정량에 관한 연구 (Study on the Determination of Benzene Hexachloride in Contaminated Air)

  • 공기중에 오염되어 있는 미량의(benzene hexachloride : BHC)를 정량하는 간단하고 예민한 방법을 연구하였다. 시료를 n-hexane에 통과시켜 BHC를 녹인 다음 n-hexane 용액의 흡광도를 245nm에서 측정하고 고체 BHC 표준시료를 기화시킨 후 n-hexane에 통과시키고 같은 방법으로 흡광도를 측정하여 얻은 검량선으로부터 농도를 계산하였다.

  • Effects of the ultrasonic treatment on plant represents antifeeding activity and chitin synthesis inhibition activity against diamondback moth (Plutella xylostella L.) by a leaf-dipping method at a concentration of 5,000ppm. Methanol extracts of leaves of the ultrasonic treatment on plant was sequentially partitioned with n-hexane, chloroform, ethlyacetate, butanol and water fractions, and bioassayed. Antifeeding activities were observed in n-hexane, chloroform and ethlyacetate fractions. N-hexane fraction has 94.6% of strongest antifeeding activity against P. xylostella. From the n-hexane sequentially partitioned showed H6 and H8 fractions of antifeeding activities against P. xylostella. And chitin synthesis inhibition activity against P. xylostella was obtained the methaol, n-hexane and water fractions.
