Screening of Antimicrobial Activity from Differential Extracts of Allii sativi Bulbus (대산의 분획별 추출물에서 항균활성 검색)

  • Allii sativi Bulbus(garlic) have been shown to possess medicinal value, in particular, antimicrobial activity. In this study, we compared the efficacy on some pathogenic bacteria and fungus among several different extracts(water, hexane, ethyl acetate, methanol, chloroform) of Allii sativi Bulbus. Animal pathogenic bacteria and fungus(S. gallinarium: KCTC 2441, S. flexneri: KCTC 2361, E. cloacae: KCTC 2006, K. pneumonia: KCTC 2208, C. albicans: KCTC 1940) were used to test by measurement of minimum inhibitory concentrations(MIC) and disc diffusion. Allii sativi Bulbus were cut and mixed with water at 37℃ about 24 h and filtered, and several different solvents(hexane, chloroform, ethyl acetate, methanol) were respectively added to separate the fraction of each solvent. The antimicrobial(bacteriocidal) and antifungal effect were apparently shown from water extract, hexane and ethyl acetate extract against using strains(Staphylococcus gallinarium, Shigella flexneri, Enterobacter doacae, Klebsiella pneumonia, Candida albicans). Especially, the water extract showed the superior efficacy. And the clear zone size of water extract (11~27 mm) was greater than that of gentamycin, hexane extract and ethyl acetate extract against S. gallinarium. S. flexneri, K. pneumonia and C. albicans. Minimum inhibitory concentrations(MIC) of water extract appeared to around 2.0~7.5 ㎎/㎖ against S. gallinarium, S. flexneri, E. cloacae and K. pneumonia. The greater activity was shown by water extract because the MIC of water extract for C. albicans observed in very low concentration(<1.0 ㎎/㎖) compared to hexane(5.0 ㎎/㎖) and ethyl acetate(10.0 ㎎/㎖). Therefore, these results exhibited that water extract of Allii sativi Bulbus have stronger antimicrobial activity than hexane and ethyl acetate extract, and may be useful as topical medicine of superficial infections causing C. albicans as well as antifungal agents.

The Effect of Flow Rate on the Process of Immiscible Displacement in Porous Media (다공성 매체 내 비혼성 대체 과정에서 주입 유량이 거동 양상에 미치는 영향)

  • A series of experiments using transparent micromodels with an artificial pore network etched on glass plates was performed to investigate the effects of flow rate on the migration and distribution of resident wetting porewater (deionized water) and injecting non-wetting fluid (n-hexane). Multicolored images transformed from real RGB images were used to distinguish n-hexane from porewater and pore structure. Hexane flooding followed by immiscible displacement with porewater, migration through capillary fingering, preferential flow and bypassing were observed during injection experiments. The areal displacement efficiency increases as the injection of n-hexane continues until the equilibrium reaches. Experimental results showed that the areal displacement efficiency at equilibrium increases as the flow rate increases. Close observation reveals that preferential flowpaths through larger pore bodies and throats and clusters of entrapped porewater were frequently created at lower flow rate. At higher flow rate, randomly oriented forward and lateral flowpaths of n-hexane displaces more porewater at an efficiency close to stable displacement. It may resulted from that the pore pressure of n-hexane, at higher flow rate, increases fast enough to overcome capillary pressure acting on smaller pore throats as well larger ones. Experimental results in this study may provide fundamental information on migration and distribution of immiscible fluids in subsurface porous media.

Antimicrobial Activity of the Extract from Pyrola japonica against Bacillus subtilis (노루발풀(Pyrola japonica) 추출물의 Bacillus subtilis에 대한 항균활성)

  • The antimicrobial substance from Pyrola japonica were extracted and isolated. Eighty percent ethanol extract of dried Pyrola japonica was fractionated to hexane, diethyl ether, ethyl acetate and aqueous layer. The hexane-soluble fraction showed the highest inhibitory activity against Bacillus subtilis. Moreover the hexane layer was fractionated into 5 groups by silica gel column chromatography. From the results, group No. 2 ($18{\sim}40$ fractions) showed the highest antimicrobial activity. The group was re-separated to 10 fractions by preparative thin layer chromatography and the peak I as active fraction was isolated by HPLC.

Anti-inflammatory and Anti-cancer Effects of Sterol-rich Fraction from Nannochloropsis oculata by using Saponification (해양미세조류(Nannochloropsis oculata)로부터 saponification을 통한 Sterols 분획물의 항염증 및 항암 효능 평가)

  • The green microalga of the genus Nannochloropsis (class Eustigmatophyceae) is a leading candidate for biofuel production due to its ability to accumulate high oil content (28.7% of cellular ash-free dry weight). We investigated the anti-inflammatory and anticancer activities of sterol-rich fraction from nannochloropsis oculata n-hexane (NOH) extract after saponification of the microalga. Among the fractions with n-hexane, chloroform and ethyl acetate, the n-hexane fraction showed the highest anti-inflammatory activity in LPS-stimulated RAW 264.7 macrophage as well as anticancer activity against human leukemia HL-60 cells without the cytotoxity. And the sterol-rich fraction was obtained from the n-hexane fraction by open silica column under the gradient solvent condition with 100% hexane (1L), hexane : ethyl acetate (20 : 1, 10 : 1, 5 : 1, 1 : 1, v/v). Among the four fractions (NOH-1~4), especially NOH-1 contained the highest content of sterols. NOH1 showed the highest HL-60 (about 85%) and NO inhibitory activities at the concentration of $100{\mu}g/mL$. These results demonstrated that the sterol-rich fraction from N. oculata might be a useful candidate as anti-inflammatory and anticancer agents for anti-inflammatory and anticancer activity.

  • We have measured ${\pi}^*$, $\alpha$ and $\beta$ polarity parameters of 2-propanol / hexane and ethyl acetate / hexane mixed solvents over the entire range of composition. For the ethyl acetate / hexane system, only ${\pi}^*$ and $\beta$ were measured since $\alpha$ of these mixed solvents are defined zero. We have corrected the measured polarity parameters to obtain consistent data with the existing literature data assuming a good linear correlation between the measured and the true values. The variation patterns are consistent with the expected trends based on the chemistry of the solvent components. The general trends of the two solvent systems are summarized as follows. ${\pi}^*$ merely increases as the content of the more polar solvent increases while $\alpha$ and $\beta$ increase with the increase of the content of the more polar solvent, then decrease upon continuing addition of the more polar solvent.

  • This study was performed to evaluate antimicrobial activity of methanol extract from Rosmarinus officinalis L. and their fractions on methicillin-resistant Staphylococcus aureus (MRSA) and resistant gene regulation. The methanol extract of Rosmarinus officinalis L. and their hexane fractions showed the strongest antimicrobial activity against S. aureus and MRSA. To observe the morphological change of MRSA according to the hexane fraction $80{\mu}g$/mL treatment, scanning electron microscope (SEM) of MRSA were measured. The results from SEM showed decreased number of bacteria, lysis and damaged cell wall. Expressions of MecA and penicillinase were substantially decreased in a dose-dependent manner on MRSA that had been treated with methanol extract of Rosmarinus officinalis L. and their hexane fractions.

A Method for Analysis of Styrene Dimer and Trimer in Foods and Containers (식품 및 용기 중 스티렌 다이머 및 트리머의 분석법)

  • A method for quantitative determination of styrene dimer and trimer, which are suspected as endocrine disruptors, in foods and containers was studied. For residual contents of styrene dimer and trimer in two-kinds of containers, which contained instant noodle and yogurt, sample pieces were completely dissolved in tetrahydrofuran. The polymer was precipitated with n-hexane, a portion of supernatant was concentrated for analysis. A sensitive method was also optimized for the quantification of styrene dimer and trimer in foods such as instant cup noodle and yogurt by using GC/MS. Limits of Detection were about 3.2-87.0 ppb for styrene dimers and trimers. The highest recovery was obtained by extraction with acetone/hexane(1:1), followed by florisil clean-up. The levels of styrene dimer and trimer migrated by food simulants were much higher in heptane.

Washing effect of fingermark contaminated with flammable liquids (인화성액체에 오염된 지문의 세척 효과)

  • The effect of washing fingerprints deposited on glass that were contaminated with a flammable liquid (gasoline, kerosene, diesel, and thinner) was studied by washing with hexane or heptane. The fingerprints were visualized using fuming cyanoacrylate, followed by basic yellow 40 staining. After comparing the washing effect, by dividing one fingerprint into four sections, it was confirmed that the ridge detail was damaged by dissolving the fingerprints in flammable liquid. As a result of washing fingerprints contaminated with flammable liquids using hexane or heptane, fingerprints contaminated with gasoline, kerosene, and thinner did not show a washing effect because the ridge detail was damaged at the time of contamination, and only fingerprints contaminated with diesel exhibited improved ridge detail quality. Because hexane and heptane washing damage the ridge detail, it was found that fingerprints contaminated with gasoline, kerosene, and thinner were better enhanced directly without the washing process. In addition, it was found that the amount of the washing solvent and contact time should be minimized when washing fingerprints contaminated with diesel.

Effect of Ginger Fractions for Inhibition of Soybean Oil and Rat Liver Microsomal Lipid Peroxidation (생강 추출획분의 대두유 및 흰쥐 간 마이크로좀 지질 과산화 억제 효과)

  • The content ratios of gingerol in 4 fractions (hexane, ether, ethylacetate and hexane-ether (1:1, v/v) fraction) extracted from ginger (Zingiber officinale Roscoe) were analyzed using high performance liquid chromatography (HPLC). The content ratios of 6-gingerol in 4 fractions were hexane fraction; 49.5%, ether fraction; 20.74%, ethylacetate fraction; 21.43% and hexane-ether (1 : 1, v/v) fraction; 93.70%. Antioxidant activities of soybean oil added 0.2% of each ginger fraction and 0.02% of BHT were determined by peroxide value during storage at 45$^{\circ}C$. And relative antioxidant effectiveness (RAE) was calculated as the ratio of the induction period of a given substrate oil to that of a control oil. RAE of each fraction was hexane fraction; 2.60, ether fraction; 2.33, ethylacetate fraction; 2.07 and hexane-ether (1 : 1, v/v) fraction; 2.75, BHT; 1.74. Inhibition effect of each fraction on the rat liver microsomal lipid peroxidation showed hexane fraction; 93%, ether fraction; 92%, ethylacetate fraction; 86% in 350 g/$m\ell$ and hexane-ether (1 : 1, v/v) fraction; 89% in 20 g/$m\ell$.

  • These experiments were conducted to analyze the characteristics of organic compounds and molecular weight distribution according to the treatment steps of purifying system for pig wastewater. The results obtained are summarized as follow. 1. Raw pig wastewater contained 31 kinds of organic compounds such as acetic acid, 2-butanone, hexane, chloroform, propionic acid, butanoic acid etc. 2. After ozone treatment, 13 kinds of organic compounds such as ethene, pentane, 2-methylopropanal, hexane, furan and so on were detected in the wastewater. 3. After $TiO_2$ treatment, 12 kinds of organic compounds such as ethene, hexane, chloroform, 1-decene, silane and so on were detected from the effluent. 4. After both ozone and $TiO_2$ treatment, portion of molecular weight smaller than 500 Daltons and molecular weight between 500 to 1,000 Daltons in the wastewater increased.
