Distribution of Indicator Organisms and Influence of Storage Temperature and Period in Commercial Plant Food (시판 식물성 식품의 오염지표세균 분포 및 저장온도, 기간별 오염지표세균의 변화)

  • There were few data for the distribution of the indicator organisms in the commercial plant foods, and for the normal flora and for the foodborne agents within the country. First of all it must be investigated the distribution of the indicator organisms. And also it is very important to prepare the sanitation criteria for the plant foods through the microbiological examination and the investigation of tendency to change of the indicator organisms according to the storage temperature and period. The average number of total viable counts for grains was 2.9$\times$105/g, psychrophilic bacteria 2.9$\times$105/g, heterotrophic bacteria 3.1$\times$105/g, heat-resistant bacteria 2.1$\times$103/g, Pseudomonas aeruginosa 23/g. That for beans was 6.3$\times$102/g, psychrophile 34/g, heterotroph 1.7$\times$102/g. That for sesames was 1.4$\times$105/g, coliform 350/g, psychrophile 7.4$\times$104/g, heterotroph 5.8$\times$104/g, Pseud. aeruginosa 2.3$\times$103/g. heat-resistant bacteria 150/g. That for potatoes was 2.0$\times$107/g, coliform 5.0$\times$104/g, psychrophile 1.8$\times$107, heterotroph 1.4$\times$107/g, heat-resistant bacteria 3.3$\times$104/, Staphylococcus 2.7$\times$105/g, fecal streptococcus 4.5$\times$103/g, Pseud. aeruginosa 7.0$\times$103/g. That for mushrooms was 1.2$\times$108/g, psychrophile 9.4$\times$107/g, heterotroph 1.0$\times$109/g, heat-resistant bacteria 1.6$\times$105/g, Pseud. aeruginosa 1.3$\times$103/g. That for vegetables was 5.9$\times$1011/g, coliform 1.8$\times$106g/, Staphylococcus 1.1$\times$1012/g, heterotroph 8.4$\times$1011/g, heat-resistant bacteria 7.6$\times$106/g, Staphylococcus 1.1$\times$107/g, fecal streptococcus 1.1$\times$104/g, Pseud. aerugniosa 5.2$\times$104/g. That for nuts 3.9$\times$104/g, coliform 3.9$\times$103/g, psychrophile 4.0$\times$104/g, heterotroph 3.2$\times$104/g, heat-resistant bacteria 400/g. In commercial grains and beans, SPC, psychrophile, heterotroph and heat-resistant bacteria stored at 1$0^{\circ}C$, 2$0^{\circ}C$, 3$0^{\circ}C$ were constant. Staphylococcus, coliform, Pseud. aeruginosa were decreased a little n grains, but were not detected in beans. In mushrooms, all indicator organisms were increased as time goes on and were increased rapidly at 2$0^{\circ}C$. In sesames, coliform was not detected at all temperature. psychrophile was increased for 7 days, the others were constant. In potatoes, SPC, psychrophile, heat-resistant bacteria, heterotroph had a tendency to increase and the others were constant. In vegetables, indicator organisms were had a tendency to increase, psychrophile, heterotroph were rapidly increased after 7 days. In nuts, SPC, coliform, psychrophile heterotroph, heat-resistant bacteria, Pseud. aeruginosa were constant, staphylococcus and fecal streptococcus were not detected.

Parameterising a Microplankton Model

  • This paper describes and assesses the parameterisation of MP, the microplankton compartment of the carbon­nitrogen microplankton­detritus model. The compartment is 'the microbial loop in a box' and includes pelagic bacteria and protozoa as well as phytoplankton. The paper presents equations and parameter values for the autotroph and microheterotroph components of the microplankton. Equations and parameter values for the microplankton as a whole are derived on the assumption of a constant 'heterotroph fraction'. The autotroph equations of MP allow variation in the ratios of nutrient elements to carbon, and are largely those of the 'cell­quota, threshold­limitation' algal growth model, which can deal with potential control of growth by several nutrients and light. The heterotroph equations, in contrast, assume a constant elemental composition. Nitrogen is used as the limiting nutrient in most of the model description, and is special in that MP links chlorophyll concentration to the autotroph nitrogen quota.

  • The average number of total viable counts for the commercial pork tested was 19/g, coliform 1.8/g, psychrophilic bacteria 15/g, heterotrophic bacteria 12/g, fecal streptococcus 6.2/100 g, Pseudomonas aeruginosa 13/100 g and none of heat-resistant bacteria and Staphylococcus was detected. That for the commercial beef tested was 130/g, coliform 5.2/g, psychrophile 140/g, heterotroph 28/g, Staphylococcus 1.2/g, fecal streptococcus 9.5/100 g, Pseud. aeruginosa 1.9/100 g and heat-resistant bacteria was not detected. That for the commercial chicken tested was 8800/g, coliform 53/g, psychrophile 4600/g, heterotroph 4700/g, fecal streptococcus 9.9/100 g, Pseudo aeruginosa 2.5/100 g. That for milk was 4700/ml, psychrophile 120/ml, heterotroph 420/ml and the others were not detected. That for the commercial cheese was 3.2/g, psychrophile 2.3/g, heterotroph 1.6/g, Staphylococcus l/g, fecal streptococcus 9.1/g. That for fermented milk was $10^{7}/ml$, heatresistant bacteria $10^{6}/ml$, fecal streptococcus 2400/100 ml, lactobacillus $3.2{\times}10^{15}/ml$, in accordance with lactic acid bacteria and the others were not detected. There was not detected any indicator organisms from ham, sausage, butter, eggs and quails in the commercial fooods tested. SPC, coliform, psychrophile and heterotroph in commercial meats stored at $10^{\circ}C$ were increased rapidly as time goes on but heat-resistant bacteria, staphylococcus, fecal streptococcus and Pseudo aeruginosa were constant. At $20^{\circ}C$, SPC, coliform, psychrophile, heterotroph and fecal streptococcus were the highest at 7 days and heat-resistant bacteria, staphylococcus and Pseudo aeruginosa were increased a little. At $30^{\circ}C$, all indicators were increased rapidly for 3 and 7 days and then decreased rapidly. All indicator organisms were increased at the level of 10/g for 14 days in meat products stored at $10^{\circ}C$, but SPC, psychrophile and heterotroph in meat products stored at $20^{\circ}C$ were increased at the level of $lO^5/g$. It showed that the indicators in meat products stored at $30^{\circ}C$ had a tendency to increase at the level of $10^{2}/g$ relative to those stored at $20^{\circ}C$. SPC, psychrophile and heterotroph in milk stored at $10^{\circ}C$ increased up to the level of $10^4/ml$, but coliform, staphylococcus, fecal streptococcus and Pseudo aeruginosa were not detected. As stored at $20^{\circ}C$ and $30^{\circ}C$, they were increased rapidly for 1 or 3 days and then constant for a long time.

  • Biofilm airlift reactor was continuously operated to investigate the competitions between the autotrophs and heterotrophs, ammonia oxidizers and nitrite oxidizers, and Nitrobacter and Nitrospira with real wastewater at a C/N ratio of 0.86. As the reactor achieved complete nitrification microbial distribution was analyzed by FISH/CLSM technique. The results showed that heterotroph was more abundant than nitrifying bacteria. Ammonia oxidizers (17%) and Nitrobacter (7%) prevailed nitrite oxidizers (9%) and Nitrospira (2%), respectively.

  • It is a time consuming work to identify the inhibition of unknown chemicals or industrial wastewater. Thus it is needed to establish a fast assay tool for finding a toxicant source. Biomass activity and ammonia inhibition were measured by DHAINT method. Ammonia inhibition tests were comprised of total ammonia inhibition and free ammonia inhibition. Those inhibitions were carried out by nitrifier and heterotroph each other with nitrifier inhibitor. The ammonia inhibition was proportional to an amount of total ammonia and pH increase. It meaned that a free ammonia played a key role for ammonia inhibition. however both total ammonia and free ammonia should be considered for an accurate assay of the ammonia inhibition. Nitrifier was more sensitive than heterotroph when the ammonia concentration above 3.000mg/L.

BNR 슬러지 유입비율에 따른 SBR의 초기 start-up과정의 영양염류제거 특성

  • 이번 연구에서는 pH 조작이 어려운 소규모 하수 처리장에서의 BNR 슬러지의 접종으로 인한 SBR start-up시 영양 염류 제거 특성 및 변화를 관찰 하였다. 1) 글루코즈를 탄소원으로 사용한 유기물 제거에 있어서 모두 85%이상의 높은 제거 효율을 보였으나 BNR 슬러지 접종 비율이나 유무에 따른 SBR의 유기물 제거에 미치는 영향은 미비한 것으로 나타났다. 2) 유입비율에 따라 암모니아 제거 효율은 BNR 슬러지 첨가 유무에 따라 바뀌었으며, 비율에 상관없이 첨가된 반응조는 7일 이내 $82{\sim}98%$에 달하는 제거율을 보였다. 3) 인 제거효율에 있어서는 유입비율에 따라 인 제거 효율이 증가하였으며, 첨가와 미첨가의 차이는 25%이상의 인 방출량 차이를 나타내었다. 4) 첨가한 BNR 슬러지의 비율이 전체의 40%이상일 때는 50%일 때와 큰 차이를 보이지 못했다. 5) FISH를 통하여 미생물 군집을 확인하였고 각 미생물들 간의 상대적인 비율과 heterotroph의 급격한 증식을 확인할 수 있었다. 6) pH, DO, ORP graph를 통한 Reactor의 실시간 조정 가능성을 볼 수 있었다. 7) BNR 슬러지를 접종하지 않을 경우, PAOs와 질산화 미생물의 성장이 더뎌 영양염류 제거가 어려웠다.

  • Wir haben das Wachstum und Chloropbyllzunahme und -abnahme bei der Autotroph-, Mixotroph-, und Heterotrophanzucht analysiert, und ob Glukose-Chlorose von Stickstoffi-nangel verursacht wird, oder Glukose die Chlorophyllbildung unterdriickt, und wie Lipid und Stdrke gebildet worden ist. Wir wollten bei Glukose-Chlorose Zucker, Stickstoff und Chlorophyll drei Verhiiltnisse untersuchen. Chlorella variegate fehlte Nitrat-Reduktase. Sie wachste nicht mit $KNO_3$ als stickstaffquelle und bildete kein chlorophyll bei Heterotroph und unterdrilckte chlorophyllbildung bei Mixotroph. Akkumulation von Lipid und Stdrke war nicht so viel wie bei Mineralsalz mangelchlorose. Chlorella variegate brauchte nicht $CO_2$ die Oxydationsphosphorylierung aber $O_2$ ist notig. Durch DNP und Fluoracetat blockierte Chlorella variegate die oxydationsph-osphorylieriing. Zell-Vermehrung und Koblenhydratbildung sind reduziert worden, aber nicht normal.

Inhibition Effect of Germ-resistant Sponge on Microbial Growth in Kitchen Hygiene (주방위생에서 항균수세미의 효과)

  • It was intended to investigate the effect of the microbiological kitchen hygiene such as dishclothes and scrubbers. The 8indicator organisms (standard plate counts, coliform, heterotroph, enterococcus, staphylococcus, heat-stable bacteria, psychrotroph, Pseudomonas aeruginosa) were detected highly in dishwaters, dishcloth and scrubber. Coliform and Staphylococcus aureus were appeared on dishcloth dominantly than the scrubber, and the scrubbers were intruded by hetrotrophs and psychrotrophs numerously than dishclothes. The germ-resistant sponge inhibited the growth of the most of test strain, and appeared the about 100% reduction rate after 24 hr, but did not affect Pseudomonas aeruginosa and P. fragi so typically after 24 hr. The anti-microorganism durability of germ-resistant sponge, treated with food soil, was maintained by 10 days, the early stage strain density was founded in 20 days, and the strains grew after 30 days.

Comparison of a Microbiological Model Simulation with Microcosm Data

  • Using nitrogen as the limiting nutrient, the default version of a microplankton-detritus model linked chlorophyll concentration to the autotroph nitrogen. However, phosphorus dynamics were added to simulate the results of a microcosm experiment. Using standard parameter values with a single value of microheterotroph fraction in the microplankton taken from the observed range, the best simulation successfully captured the main features of the time-courses of chlorophyll and particulate organic carbon, nitrogen and phosphorus, with root-mean-square error equivalent to 29% of particulate concentration. A standard version of microbiological model assumes complete internal cycling of nutrient elements; adding a term for ammonium and phosphate excretion by microheterotrophs did not significantly improve predictions. Relaxing the requirement for constant microheterotroph fraction resulted in an autotroph-heterotroph model AH, with dynamics resembling those of a Lotka-Volterra predator-prey system. AH fitted the microcosm data worse than did MP, justifying the suppression of Lotka-Volterra dynamics in MP. The paper concludes with a discussion of possible reasons for the success of the simple bulk dynamics of MP in simulating microplankton behaviour.

Comparison of Labyrinthulid Strains L4 and L75 by Fatty Acid Composition and Characteristics

  • Labyrinthulids are unicellular heterotrophic marine microalgae. Two labyrinthulid strains, L4 and L75, which produce generous amounts of polyunsaturated fatty acids(PUFAs), were isolated from leaves floating in the coastal areas of Quang Ninh province, Vietnam, and Fukuoka Prefecture, Japan, respectively. Both strains had spindle-shaped cells surrounded by ectoplasmic networks. Numerous oil bodies were observed in each cell, mostly in the vicinity of cell membranes. When soybean oil or soybean lecithin was used as a carbon source with peptone as a nitrogen source, the proportion of PUFAs reached 25-30% and 50-56% for L4 and L75, respectively. After 14 days of growth at $25^{\circ}C$, L4 produced 0.3 mg PUFAs/g-agar in PYA-SBO medium and 0.6 mg PUFAs/g-agar in PYA-SBL medium. In comparison, L75 produced 0.2 mg PUFAs/g-agar in both types of media. The differences between the two strains included changes in cellular morphology and the capacity for attaching tightly to fibers when cultured in liquid PYA medium containing 2% SBL. In addition, when the strains were grown under the same conditions, L4 had a higher growth rate and produced more PUFAs than L75.
