Glycyrrhizae radix (GR) is an herbal medicine commonly used in East Asia for treating a variety of diseases, including stomach disorders. In this study, the antidepressant-like activity of GR was investigated using the forced swimming test (FST) in rats. After the FST, the expression of c-Fos and corticotrophin releasing factor (CRF) was assessed by immunochemistry of brain samples from the paraventricular nucleus of the hypothalamus (PVN). The results of the FST showed that a high dose (400 mg/kg) of extract was very effective in reducing immobility(P<0.01), and increased climbing. In addition, treatment with GR (400 mg/kg) significantly decreased the expression of c-Fos and CRF in the PVN, compared to controls. In conclusion, the findings of this study demonstrated that GR effectively reduced behavioral and physiological depression responses in an animal model of depression, suggesting that GR might be useful in the treatment of clinical depression.
Objective : Fatigue is the most common symptom in post-COVID-19 syndrome. We report the case of a patient with post-COVID-19 syndrome with fatigue treated using herbal medicine (Saengkangsasim-tang). Methods : A 64-year-old man had severe fatigue for 4 months after a severe acute respiratory syndrome-coronavirus-2 (SARS-CoV-2) infection. Saengkangsasim-tang was administered to this patient based on the disease pattern identification diagnostic system by Shanghanlun provisions (DPIDS). We used the Numeric Rating Scale (NRS) and Beck Depression Inventory (BDI) to evaluate the effectiveness of Saengkangsasim-tang for fatigue in the patient with post-COVID-19 syndrome. Results : According to DPIDS, the patient was diagnosed with the provision 157. After administration of Saengkangsasim-tang for 30days, the fatigue based on NRS and depression based on BDI were improved. Conclusions : The administration of Saengkangsasim-tang to patients with post-COVID-19 syndrome with fatigue, diagnosed according to the Shanghanlun provision 157, may be effective.
Objective : This study was conducted to investigate the effects of Hwangryunhaedok-Tang and Geongangbuja-Tang on the change of interleukin-6 (IL-6) and tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) level induced by LPS I.C.V. injection in mice. Method : We devided group into 6 mice and 6 mice were assingned to each group. In the normal group only saline was administered intragastrically, and in the control group LPS was injected intracerebroventricularly 1 hr after intragastric administration of saline. In the experiment groups Hwangryunhaedok-Tang(0.5g/kg, 1.0g/kg, 3.0g/kg) was administered intragastrically to mice 1 hr prior to LPS(100mg/mouse) I.C.V. injection.. Also Geongangbuja-Tang (0.5g/kg, 1.0g/kg, 3.0g/kg) was administered intragastrically to mice 1 hr prior to LPS(100mg/mouse) I.C.V. injection. To measure the plasma IL-6 and $TNF-{\alpha}$ level of mice, their blood samples were collected from retro-orbital plexus, immediately centrifuged at $4^{\circ}C$, and plasma was removed and stored frozen at $-83^{\circ}C$ for later determination of IL-6 and $TNF-{\alpha}$. The level of IL-6 and $TNF-{\alpha}$ production was measured by enzyme-linked immunosorbent assay in the plasma. Result : Regarding IL-6 level, The 0.5g/kg and the 1g/kg groups of Geongangbuja-Tang decreased IL-6 level. Especially the 3g/kg control group decreased IL-6 level significantly than the normal group(p<0.01). Regarding $TNF-{\alpha}$ level, the 3g/kg group of Geongangbuja-Tang decreased it significantly(p<0.05). Conclusion : These data revealed that Hwangryunhaedok-Tang might not have the anti imflammatory effect and Geongangbuja-Tang(3g/kg)might have the anti imflammatory effect by reducing the plasma IL-6 and $TNF-{\alpha}$ level in mice LPS Injection.EIM (Eighteen Incompatible Medicaments) is an important component in Oriental pharmacology and is directly related to clinical prescriptions. Medical practitioners argued that the definite cause and meaning of EIM was ambiguous and therefore debated the issue of clinical application of the EIM. This study conducted an in-depth literary research on the origin, meaning and contents of EIM with the purpose to contribute in its efforts to be used clinically. Even after thousands of years have past since establishment of Oriental medicine, EIM is still tabooed and was an obstacle that hindered ideologies. Modern herbal medicine texts claim that the use of EIM can reduce treatment effects and promote poisoning and side effects. However, since long ago, there has been medical practitioners who reject this as false. Recently, poisoning caused by EIM has been claimed to be from the toxicity of the drug itself, rather than the result of interaction between the drugs, and therefore they suggest that EIM is not a forbidden domain. In addition, EIM showed a difference in number depending on the era. However, this can be understood not as a definite number, but instead as a warning to be careful during combination of drugs for use as clinical medicine. Historically, there were very few cases in which EIM was used for clinical tests and thus, the clinical value is not, while others applied EIM directly to their bodies, which showed signs for the usefulness and potential of EIM for us. A more concrete and in-depth study must be made on EIM.
This report describes the studies as follows through the research into the data of Fructus Forsythiae main blended prescriptions from Dongeuybogam. Through investigation on prescriptions from Dongeuybogam with Fructus Forsythiae as an ingredient, the following conclusions were reached. 1. All kinds of infection in a boil and the malignant tumor recorded the largest number of clinical frequency of the prescriptions in therapeutic use when Fructus Forsythiae was taken as a principle medicine. 2. The prescriptions are compounded with Fructus Forsythiae as a principle medicine can apply to eye disease, nose and ear disease, throat disease, trauma, the malignant tumor, skin disease, dental disease and heat pathogenic disease. 3. Various pathogenic factors such as wind-heat, heat, fire, wind, qi, poison, cold are put to practical use. 4. The dosage of Fructus Forsythiae is 2bun(about 0.75 gram) to 3jeon(about 11.25 gram), however 1joen(about 3.75 gram) has been taken the most for clinical application. 5. With examination the formulae of presciptions containing Fructus Forsythiae, I found the basic prescriptions as well as the medicines are in conjunction with Fructus Forsythiae and the theoretic grounds of formula in combining Fructus Forsythiae as a group of medicine for prescription through this research.
Objective : This study aimed to report the improvement of a patient with anorexia by treatment with Jinmu-tang (Hyunmu-tang) based on the disease pattern identification diagnostic system (DPIDS) of the Shanghanlun provisions. Methods : We evaluated the progress of symptoms, patient compliance, and presence of side effects after the patient was administered Jinmu-tang. The clinical response was estimated according to the number of meals a day, the size of meals, the number of complaints of abdominal pain in a week, and a Likert scale. Results : According to the DPIDS, the patient was diagnosed according to provision 316 with soyinbing. After administration of Jinmu-tang for 45 days, the number of meals a day and the size of meals increased, the number of complaints of abdominal pain in a week decreased, and the Likert scale score decreased from 3 to 0. Conclusions : This case report suggests that the word "腹痛" (abdominal pain) in the 316th Shanghanlun provision indicates anxiety about abdominal pain, which affected anorexia in this case.
Objectives : Hyperthermia is a widely used therapeutic tool for cancer therapy and a well-known inducer of apoptosis. Although the Cinnamomi cortex (CC) is a potent anticancer agent for several human carcinomas, it is less potent in the human U937 cell line. To explore any enhancing effects of CC with hyperthermia induced apoptosis, this study investigated the combined effects and apoptotic mechanisms of hyperthermia and CC in U937 cells. Methods : U937 cells were heat treated at $43^{\circ}C$ for 30 min with or without pre-treatment for 1h with CC and then incubated at $37^{\circ}C$ with 5% $CO_2$. Cell viability was analyzed by MTT assay and Trypan blue assay. Morphological changes reflecting apoptosis were visualized under microscope. Synergy effect of CC combined with hyperthermia were calculated by Compusyn software. The expression of proteins related to apoptosis and signaling pathways was determined by western blotting. Results : Hyperthermia with CC reduced cell viability and induced apoptosis. Combined hyperthermia and CC treatment markedly augmented apoptosis by upregulating proapoptotic proteins and suppressing antiapoptotic proteins, culminating in caspase-3 activation. Furthermore, the combined treatment, decreased the expression of in Bcl-2 family, cyclin D1, VEGF, MMP2 and MMP9 expression. Conclusion : This study provides compelling evidence that hyperthermia, in combination with CC, is a promising therapeutic strategy for enhancement of apoptosis and suggests a promising therapeutic approach for cancer.
Objective: This study was conducted to investigate the effects of Coptidis Rhizoma on the plasma IL-6 and $TNF-{\alpha}$ level in mice by intracerebroventricular(I.C.V.) injection of Lipopolysaccharide (LPS). Method: 6 mice were assigned to each of the Normal group, the Control group, and the individual Experimental groups. In the Normal group only saline was administered intragastrically, and in the Control group LPS was injected intracerebroventricularly 1 hr after intragastric administration of saline. In the Experimental groups Coptidis Rhizoma(0.5g/kg, 1.0g/kg, 3.0g/kg) was administered intragastrically to mice 1 hr prior to LPS (100ng/mouse) I.C.V. Injection. To measure the plasma IL-6 and $TNF-{\alpha}$ level of mice, their blood samples were collected from retro-orbital venous plexus, immediately centrifuged at $4^{\circ}C$, and plasma was removed and stored frozen at $-83^{\circ}C$ for later determination of plasma IL-6 and $TNF-{\alpha}$. Result: 1. LPS I.C.V. Injection increased plasma IL-6 level significantly in a dose-dependent manner compared with Normal group. (P<0.01) The plasma IL-6 concentration reached a significant maximal level about 1 hr after LPS(100ng/mouse) I.C.V. Injection.(P<0.001) 2. Both the 0.5g/kg(Sample A) and 1.0g/kg(Sample B) groups to which Coptidis Rhizoma was administered intragastrically 1 hr prior to LPS(100ng/mouse) I.C.V. Injection showed insignificant lower plama IL-6 level in 1 hr than Control group(P>0.05), and 3.0g/kg group(Sample C) conversely showed higher plama IL-6 level than Control group. 3. LPS I.C.V. Injection increased plasma $TNF-{\alpha}$ level significantly in a dose-dependent manner compared with Normal group.(P<0.05) The plasma $TNF-{\alpha}$ concentration reached a significant maximal level about 1 hr after LPS(100ng/mouse) I.C.V. Injection.(P<0.001) 4. All Sample groups(0.5g/kg, 1.0g/kg, and 3.0g/kg) to which Coptidis Rhizoma was administered intragastrically with each constituent-dose 1 hr prior to LPS(100ng/mouse) I.C.V. Injection showed significant lower $TNF-{\alpha}$ plama level in 1 hr than Control group.(P<0.001) These data revealed that Coptidis Rhizoma might have anti inflammatory effect by reducing the plasma $TNF-{\alpha}$ level in a dose dependent manner in mice LPS I.C.V. Injection.
